Pigs are the only livestock species encoding a functional protein for both the second isoform of gonadotrophin-releasing hormone (GnRH-II) and its cognate receptor (GnRHR-II). Unlike the classical GnRH system (GnRH-I and GnRHR-I), GnRH-II and GnRHR-II are abundantly produced in porcine testes. Moreover, GnRH-II binding its receptor on Leydig cells stimulates luteinizing hormone-independent testosterone secretion. Interestingly, GnRHR-II is also localised to the connecting piece of mature, ejaculated spermatozoa, whereas GnRH-II is detected in seminal plasma, an interaction possibly influencing the function of sperm. To examine the role of GnRH-II and its receptor in the testis, we produced a swine line with reduced endogenous GnRHR-II levels (GnRHR-II KD). The objectives of this study were to (1) compare sperm characteristics between mature GnRHR-II KD and littermate control boars on the day of collection and following semen extension and (2) determine whether a GnRHR-I and GnRHR-II antagonist alters sperm characteristics after storage of extended semen. In Experiment 1, GnRHR-II KD (n=3) and littermate control (n=3) ejaculates were collected (Day 1) and computer-assisted sperm analysis (CASA) was performed (IVOS II Animal; Hamilton Thorne) to determine measures of sperm motion (motility, progressive motility, slow, and static), morphology (normal morphology, bent tail, coiled tail, distal droplet, proximal droplet (PD), distal midpiece reflex, elongation, and area), and kinematics (length of average path (DAP), length of straight line path (DSL), length of curvilinear path (DCL), average path velocity (VAP), straight line velocity (VSL), curvilinear velocity (VCL), straightness (STR), linearity (LIN), amplitude of lateral head displacement (ALH), beat-cross frequency, and wobble (WOB)). Next, 3 billion sperm were extended with Androstar Plus (80-mL doses; Minitube) and stored at 17°C until Day 7 CASA. Data were analysed with the MIXED procedure of SAS (SAS Institute Inc.). On Day 1, semen doses from GnRHR-II KD boars had reduced DSL, VSL, STR, LIN, and WOB (P<0.05), whereas sperm from control boars possessed more PD (P<0.01). Day 7 CASA revealed that transgenic sperm had reduced DAP, DCL, VAP, and VCL, although sperm from control boars were slower (P<0.05). In Experiment 2, control ejaculates (n=3) were extended as above, treated with increasing concentrations (0, 0.0001, 0.001, 0.01, 0.1, 1, and 10μM) of a GnRH antagonist inhibiting both GnRHR-I and GnRHR-II (SB-75, cetrorelix), and stored at 17°C until Day 7 and 9 CASA. On Day 7, only sperm characteristics in doses treated with 10μM SB-75 were significantly lower (normal morphology, DAP, DCL, VAP, VCL, and ALH) or higher (PD, WOB, and area) than controls. Similar differences (except ALH; P<0.10) for the 10μM SB-75 treatment were detected on Day 9; however, motility, slow, static, STR, and LIN were also reduced (P<0.05). Thus, these data suggest that GnRH-II and its receptor are important to sperm function, representing a potential avenue to improve semen preservation. The study aimed at determining the effect of storage and season on fresh semen of Beni Arouss goats. Ejaculates were collected at monthly intervals from seven mature bucks and were extended at a final concentration of 800 × 106 spermatozoa. ml-1 and stored at 16 °C for 24 h. Semen motility, viability and normal morphology were assessed at 0, 4, 8 and 24 h after collection. Motility and normal morphology parameters were recorded using computer-assisted sperm analysis (CASA) and viability was analyzed using eosin–nigrosin staining. As expected, motility, viability and normal morphology parameters showed a significant reduction within 24 h of storage and during all seasons (p < 0.05). However, semen collected in summer maintained a better quality after 24 h of storage at 16 °C than semen collected during the other periods (p < 0.05). In conclusion, the storage ability of Beni Arouss bucks’ semen stored at 16 °C was significantly higher during the summer.

Abstract Semen quality parameters of infertile and fertile dogs were compared. Sperm concentration and semen motility parameters were measured by the Hamilton-Thorne Semen Analyser IVOS 12.3. The spermatozoal morphology and the percentage of live spermatozoa were examined microscopically. Forty-six dogs of various breeds were examined. Twenty dogs had a conception failure within last year. These dogs had a history of prior normal fertility. Twenty six fertile dogs served as control. All animals underwent a clinical examination as well as ultrasonography. Sperm concentration was significantly lower in infertile dogs than in fertile dogs. For most determined motility parameters (MOT, PMOT, VAP, VSL, VCL, BCF, RAPID, STATIC) significant differences between infertile and fertile dogs were found. The percentage of spermatozoa with normal morphology also significantly differed between both groups. Ultrasonography of the prostate gland and testes revealed no pathological conditions. The testicular degeneration was assumed to be a possible cause of infertility in these dogs. The present study showed that the most sperm CASA motility parameters were significantly lower in infertile dogs in comparison to the fertile ones, and confirmed the usefulness of the Hamilton-Thorne Semen Analyser for a quick and objective analysis of sperm concentration and motility in dogs.

Computer-assisted semen analysis (CASA) systems have been one the most used tools to evaluate sperm kinetics. The objective of this research was to estimate the correlation between sperm motility characteristics evaluated by CASA during 72 hours of cooling with the farrowing rate (FR) and total number of piglets born (TNB) after artificial insemination. Multiparous sows (n=464) were inseminated with semen from seven boars (19.6 ± 1.3 ejaculates/male). Sperm motility parameters were determined immediately after dilution and after 24, 48 and 72 hours of cooling at 15°C: total motility (TM-%), progressive motility (PM-%), curvilinear velocity (VCL-μm/s), straight line velocity (VSL-μm/s), average path velocity (VAP-µm/s), amplitude of lateral head displacement (ALH-µm), flagellar beat cross frequency (BCF-Hz), straightness (STR-%) and linearity (LIN-%). Pearson’s correlation coefficient was applied to analyze the data and the comparison of the means of the sperm characteristics between the boars was done by Tukey’s test at 5% probability. TM and PM at time zero (T0) were significant and had a moderate to high correlation with FR and TNB. After 72 hours of refrigeration, the semen quality was reduced and showed a significant and low correlation of the TM and PM with these same parameters. The boar presenting the lowest value of TM and PM after dilution obtained lower FR and TNB. In conclusion, computer-assisted semen analysis soon after dilution can be used to predict fertility of boars.

 El estudio evaluó la motilidad espermática y su efecto postdescongelación en semen caprino, en dos medios comerciales (Andromed® y TwoStep®) y diferentes protocolos de congelación (medio completo, con adicción del 10% de yema de huevo, semen centrifugado y sobrenadante seminal), se utilizaron machos de la raza alpina de la Universidad Francisco de Paula Santander Ocaña, el semen fue colectado con electroeyaculador, una vez los medios terminados y parte de los contenidos seminales enteros o centrifugados mezclados, se estabilizó por 2 horas, se envasó en pajillas de 0,5 cc y se congela en vapores de nitrógeno por 10 minutos, las pajillas se llevaron al laboratorio de Andrología de la Universidad del Zulia y por medio del sistema C.A.S.A.(Computer Assisted Sperm Análisis) se evaluaron los parámetros de motilidad como velocidad curvilínea (VCL), velocidad rectilínea (VSL), velocidad lineal (VAP), índice de linealidad (LIN), índice de rectitud (STR), índice de oscilación (ALH), Amplitud media del desplazamiento lateral de la cabeza del espermatozoide (BCF), los datos fueron analizados por medio del procedimiento GLM de SAS versión 9.0; los mejores índices de motilidad (VCL, ALH, BCF) fueron expresados enel tratamiento de contenido seminal centrifugado en medio Andromed®. Semen quality assessment is a prime prerequisite for any breeding programme. Computer-Assisted Sperm Analyzer (CASA) can measure the semen quality very accurately and sharply. The research was conducted at American Dairy Limited (ADL) at Vangnahati, Sreepur, Gazipur. A total of 120 ejaculates were collected from three genetic groups of bucks namely Black Bengal (BB), Boer and Jamunapari (JP) with a view to characterize and compare the semen motility and velocity parameters using Computer- Assisted Sperm Analyser. Significantly (p<0.05) higher ejaculate volume was found in Boer buck (1.73±0.16 ml) with the lower in BB buck (0.98±0.14), respectively. But significantly (p<0.05) higher total, forward progressive and fast motility was found in BB (55.47±3.17, 47.23±2.74 and 37.62±2.40%) and lower in JP (40.67±3.37, 30.97±2.91 and 21.81±2.55%), respectively. Significantly (p<0.01) fastest straight line and average path distance was observed in Boer bucks (12.29±1.63 and 15.89±1.95 μm) than Black Bengal and Jamunapari bucks (4.38±1.19 and 7.26±1.57; and 3.14±1.27 and 8.42±1.95 μm), respectively. The distance curved line (DCL) and distance straight line (DSL) was significantly (p<0.05) higher in Boer (29.13±4.87, 12.29±1.63 and 15.89±1.95 _m/sec) and lower in BB (22.07±4.36 and 7.26±1.57 _m/sec), respectively. In case of curvilinear velocity, straight line velocity and average path velocity value was significantly (p<0.01) higher in Boer (127.08±13.88, 50.01±7.68 and 68.13±7.25 _m/sec) with lower in JP (62.01±12.77, 15.75±7.35 and 22.39±9.42) where as linearity and straightness was higher in Boer and lower in BB (p>0.05), respectively. The scrotal circumference had positive correlations with semen volume and curvilinear velocity for all the genotypes. It was concluded that semen of Boer bucks found consistently better than other two genetic groups of bucks. Further, competitive studies with large sample may be performed to evaluate the breed specific differences in available goat breeds. I think this study also disclosed the some rare parameters of semen quality by using Computer-Assisted Sperm Analyzer which was impossible in manually. The effects of a simple saline solution prepared using two different pH (4.4 and 8.5) on sperm motility in burbot were investigated. Results were recorded during a 96-hour chilled storage (4°C) in 24-hour intervals. Measurements were focused on the detailed characteristics of motility using 12 parameters obtained from the Computer-assisted Sperm Analysis (CASA). Significantly higher progressive motility (pMOT), distance average path (DAP), distance curved line, distance straight line (DSL), average path velocity (VAP), curvilinear velocity, straight line velocity, and beat cross frequency (BCF) were observed with the activating solution buffered at pH 8.5 in comparison with pH 4.4. Already after 24 h a significant reduction was measured in pMOT (0 h: 49 ± 24%, 24 h: 12 ± 7%). Similar decreasing tendency was recorded only after 72 h in DAP (0 h: 26 ± 4 µm/s, 72 h: 19 ± 9 µm/s), DSL (0 h: 21 ± 5 µm/s, 72 h: 17 ± 8 µm/s), VAP (0 h: 59 ± 9 µm/s, 72 h: 43 ± 21 µm/s), and BCF (0 h: 28 ± 2 Hz, 72 h: 18 ± 10 Hz). The response of different investigated CASA parameters to different treatments varied in our experiments. According to our studies, numerous burbot sperm motility parameters are sensitive to chilled storage and to low pH of the activating solution. Our results could support the effective sperm quality assessment and successful artificial propagation process in burbot.

In the present study, we tested the effect of treatment with a slow-release implant containing the gonadotrophin-releasing hormone agonist DeslorelinTM (Peptech Animal Health Australia, North Ryde, NSW, Australia) on pituitary and testicular function in mature male dogs. Four dogs were treated with Deslorelin (6-mg implant) and four were used as controls (blank implant). In control dogs, there were no significant changes over the 12 months of the study in plasma concentrations of luteinising hormone (LH) or testosterone, or in testicular volume, semen output or semen quality. In Deslorelin-treated dogs, plasma concentrations of LH and testosterone were undetectable after 21 and 27 days, testicular volume fell to 35% of pretreatment values after 14 weeks and no ejaculates could be obtained after 6 weeks. Concentrations returned to the detectable range for testosterone after 44 weeks and for LH after 51 weeks and both were within the normal range after 52 weeks. Semen characteristics had recovered completely by 60 weeks after implantation. At this time, the testes and prostate glands were similar histologically to those of control dogs. We conclude that a single slow-release implant containing 6 mg Deslorelin has potential as a long-term, reversible antifertility agent for male dogs.

Abstract. Horses are seasonal breeders with a natural breeding season beginning in spring and extending through midsummer. In this study, quantitative and qualitative parameters of chilled stallion semen were compared between fertile and subfertile stallions and between the breeding and the non-breeding season. Semen quality parameters compared included ejaculate volume, sperm concentration, total sperm number, sperm morphology, and computer-assisted semen analysis (CASA)-derived sperm movement characteristics obtained from two groups of warmblood stallions (n=8; four fertile stallions and four subfertile stallions), which differ in the seasonal pregnancy rate 80 %–90 % (fertile) vs. 40 %–60 % (subfertile). A total of 64 ejaculates were collected from the stallions (n=8; four in the breeding season and four in the non-breeding season of each stallion). No significant differences in the semen quality parameters between the fertile and the subfertile stallions in the non-breeding season were observed. However, in the breeding season the proportion of morphologically normal sperm, total motility, progressive motility, average path velocity (VAP), and curvilinear velocity (VCL) were significantly higher in the fertile group (P<0.05) when compared with the subfertile group. In addition, a significant seasonal variation in the proportion of morphological normal sperm was found in the fertile group between the breeding and the non-breeding season (P<0.05). Moreover, significant seasonal variations (P<0.05) in CASA parameters of mean VAP, straight line velocity (VSL), and beat-cross frequency (BCF) were observed in the fertile and the subfertile stallions, which tended to be lower in the non-breeding season. In conclusion, differences between the fertile and the subfertile stallions were observed only in the breeding season, and a few of CASA-derived parameters seemed to be significantly lower during the non-breeding season in both the fertile and the subfertile stallions.

The Baudet du Poitou is a vanishing donkey breed recognized for engendering robust working mules. In Chile, only two pure breed Poitou males exist, which belong to the Chilean army and are used for mule production. We performed an extensive sperm and seminal analysis of these two jackasses aged 3 and 6 years and investigated the use of a simple hypometabolic extender for sperm cryopreservation. Computer-assisted sperm analysis showed high motility, velocity, and linearity in sperm movement. The seminal plasma analysis revealed that sodium and chloride were the main electrolytes, and globulins were the main metabolites. Active and variable enzymatic activity was observed. New information is reported about gamma-glutamyltransferase, aspartate aminotransferase, zinc, and magnesium concentrations in seminal plasma of Poitou donkeys. Ejaculates among jackasses showed some variability due to individual variability and different stages in sexual maturation according to age. The freezability index analysis based in viability, total motility and progressive motility with Botucrio extender (57.1 ± 11.0%; 56.6 ± 20.0%; and 22.6 ± 10.3%, respectively) were significantly higher (p &lt; 0.05, p &lt; 0.0001, and p &lt; 0.0001, respectively) than with HM-0 extender (42,6 ± 11.4%; 14.9 ± 5.1%; and 1.0 ± 2.5%, respectively). We report new information on Poitou donkey semen and cryopreservation in the Southern Hemisphere that could be useful in donkey breeding and conservation programs to develop strategies that improve the effectiveness of population management of this breed.
