23 Sperm quality of Pure Spanish stallions is affected by inbreeding coefficient and age

Inbreeding depression, a genetic condition produced by the mating of close-related individuals, has been associated with a reduction of fertility in several species. However, a loss in sperm quality was also associated with age. In horses, the few existing reports have described a tendency of both parameters to produce a negative effect on sperm quality. However, those reports were performed using a subjective evaluation of sperm motility. In the present study, a total of 692 ejaculates from 86 Pure Spanish stallions (PRE), aged between 3 and 22 years, were evaluated using a computer-assisted methodology to determine the effect of inbreeding in four semen parameters: free-gel volume (V), sperm concentration (C, by haemocytometer), and total (TM) and progressive (PM) sperm motility (by Spermvision sperm class analyser; Minitube). The inbreeding coefficient (F) was estimated using 300 000 PRE pedigree records approximately (minimum pedigree depth, eight equivalent complete generations; range, between 1 and 30.1%). Stallion, age, ejaculate, and season of semen collection were the variables included in the statistical model (general linear model), with ejaculate and season being the variables with a major effect (by variance components analysis). Our results showed that sperm concentration (r=−0.18; P<0.0001) and volume (to a lesser extent) were reduced with advancing age, both showing a major decline after 15 years of age. To the contrary, sperm motility was not affected by age of the stallion. We also found a negative correlation between the inbreeding coefficient and ejaculate volume (r=−0.14; P<0.001), with a marked decrease seen when F was between 7 and 20%. Also, a negative correlation was observed in PM (r=−0.08; P<0.05), although to a lower extent. Conversely, C and TM were not affected by inbreeding depression (P>0.05). In conclusion, our results demonstrated that high levels of inbreeding can compromise severely the sperm quality of the PRE stallion, which, subsequently, may have a negative influence on fertility. Ongoing studies using genomic data will help to detect genetic variants associated with stallion semen quality and how it is influenced by inbreeding in specific genomic regions.

<p class="MDPI17abstract"><strong>Objective: </strong>Reproduction and nutritional status is closely related. Nutritional deficiency or insufficiency directly affects reproductive efficiency. Deficiency of nutrition could affect the sperm quality and the ability to fertilize. The objective of this study was to evaluate the effect of feed additive (Minoxvit) on semen quality of buck.</p><p class="MDPI17abstract"><strong>Methods: </strong>This study used 6 sexually mature bucks, aged 1.5 years old. The bucks were fed daily with 3 kg of freshly chopped king grass, 1 kg of Legume (<em>Calliandra</em> sp.), and 500 g of concentrate. Three bucks were given Minoxvit by 1.25 g/day in the concentrate, while three other bucks were considered as control. Semen was evaluated once a week for 49 days. Semen was evaluated macroscopically and microscopically. The macroscopic evaluation consisted of volume, color, and consistency of semen, while microscopic evaluation consisted of mass motility, sperm motility, viability, and sperm concentration. Data were analyzed using Completely Randomized Design with Tukey test.<strong></strong></p><p class="MDPI17abstract"><strong>Results: </strong>The result showed significantly different (<em>P</em>&lt;0,05) in which bucks semen in Minoxvit addition group had higher semen volume (0.75 ml vs 0.54 ml), mass motility (3.32 vs 2.67), sperm motility (70% vs 58 %), sperm viability (86.67% vs 79.19%), and sperm consentration (2,6x10<sup>9</sup> mL vs 1,7x10<sup>9</sup> mL).<strong></strong></p><p><strong>Conclusions: </strong>This study concludes that the addition of Minoxvit has a positive influence on the quality of buck sperms providing volume, mass motility, individual motility, viability, and concentration of the sperm.</p>

Reproductive performance is one of the key factors in livestock production. It is well known that reproductive traits are influenced by several genetic factors, such as the increase of individual inbreeding levels, which are associated with changes in sperm motility and shape in several species. In horses, the increase in inbreeding is a common problem because of the reduction in effective population size and the increase in selection intensity observed in several breeds. However, studies assessing the effect of high levels of inbreeding on the sperm quality of stallions are scarce. In the present study, we aimed to determine the effect of increased inbreeding levels and age on the sperm motility patterns of Purebred Spanish horses (PRE). We performed kinetic characterisation of 557 sperm samples of 82 PRE stallions aged between 3 and16 years, using computer-assisted sperm analysis (Androvision™, Minitube). We evaluated 5 parameters in 6 different fields per sample: curved line velocity (VCL, µm/s), velocity average path (VAP, µm/s), velocity straight line (VSL, µm/s), amplitude of lateral head displacement (ALH, µm), and beat-cross frequency (BCF, Hz). We determined the pedigree-based inbreeding coefficient (Fped) based on ∼300,000 PRE pedigree records to evaluate the inbreeding effect. Individuals were separated into 2 groups: highly inbred (n=339) and lowly inbred (n=218) according to an F value of 12.5%. Differences between groups were analysed using a generalized linear model. The analysis did not show significant differences (P&gt;0.05) in the variables analysed with respect to the age of stallions. However, VAP, VCL, and AHL were lower in highly inbred than in lowly inbred animals (P&lt;0.05), suggesting less velocity and amplitude of head displacement. In the case of BCF, no significant differences (P&gt;0.05) were observed between the two study groups. In conclusion, age did not affect sperm quality parameters in the age group of stallions analysed. In addition, we demonstrated that high inbreeding coefficient reduced the mean velocity and trajectory pattern of spermatozoa in PRE.

The results of AI depend on many factors, with the quality of semen being one of the most important. Not all male hybrids can meet the requirements for semen quality, because they often have reduced fertility following cryopreservation. Thus, it is necessary to improve semen processing before use in AI. The aim of the study was to evaluate the effectiveness of using the “swim-up” flotation method to improve sperm quality of hybrid males of the Ovis genus. Semen from interspecific hybrid rams (1/4 Argali×3/4 Romanov, n=15; 1/8 Argali×7/8 Romanov, n=15) was freshly obtained, frozen–thawed, and processed by the swim-up method. Evaluation of sperm motility was determined using computer-assisted semen analysis. Statistical analysis was performed using SPSS vs.15.0 (ANOVA and t-test; SPSS Inc.). Semen was collected during the breeding season (October–December) via artificial vagina. Assessment of acrosome integrity was determined using differential staining with a Diachem diff-quick kit (NPF ABRIS+). The degree of sperm DNA fragmentation was determined using the acridine-orange test. The sperm freezing/thawing cycle was accompanied by sperm damage and an increase in the proportion of immobile sperm from 10 to 58%, with non-progressive movement increasing from 9 to 19.3%. The number of spermatozoa with abnormal morphology doubled, and the DNA fragmentation index increased from 16 to 26%. Use of the swim-up procedure allowed us to sort progressively motile spermatozoa. The content of progressively motile spermatozoa in the samples obtained from the supernatant was 86%, which was 2.3 times higher than in frozen–thawed sperm (P≤0,01). The obtained results show the effective use of the swim-up procedure to determine the quality of semen in hybrid rams. Abstract Purpose: The length of sexual abstinence seems to influence sperm quality. However, few data on the relevance of abstinence time in pathological sperm samples are available. With our study, we look for associations of abstinence length and semen quality. Methods: We studied semen samples from 4423 men undergoing fertility evaluation. Sperm concentration, percentage of progressively motile spermatozoa, total motile sperm count, percentage of spermatozoa with normal morphology, were compared after each day and 0-2, 3-7 and >7 days of abstinence. Results: We found that a longer abstinence time was related to higher sperm concentration in normal semen samples (P<0.001) and in semen samples with any sperm pathology (P<0.001, P=0.004) with the exception of oligozoospermia (P=0.125). Longer abstinence time was also associated with significantly reduced progressive motility in normal samples (P<0.001) and in cases of teratozoospermia (P<0.001). In normal samples a higher percentage of sperm were morphologically normal after a shorter abstinence period (P=0.03); in oligoasthenoteratozoospermia (OAT) samples, this was the case after a longer abstinence period (P=0.013). Conclusion: A longer abstinence time is associated with higher sperm concentration, whereas sperm motility is optimal after shorter abstinence times; results on morphology are controversial. The recommendation on abstinence time needs to be adjusted in relation to the parameter that needs to be improved.

SensiTemp, a new in vitro maturation (IMV) bull straw concept, presents the advantage of colour changing while the straw is thawed. The colour of frozen straws is blue and straws start to become white when the temperature reaches 33°C, with a complete change of colour at 37°C. The objective of this study is to assess sperm quality after thawing of semen frozen in SensiTemp from 2 bulls, by analysing, in experiment 1, sperm motility and membrane integrity using computer-assisted semen analysis (CASA) and flow cytometry (FC), and, in experiment 2, the in vitro embryo production (IVP) using IVP technologies [IVM, IVF, and in vitro culture (IVC)]. The ejaculates of 2 bulls, selected during preliminary experiments on high in vitro fertility, were harvested at CIA L’Aigle, France, and split ejaculates were frozen in experimental (SensiTemp) and conventional (control) straws. In experiment 1 after thawing semen from the 2 types of straws (5 pooled straws each; 2 replicates), motility was assessed using the IVOS CASA system (Hamilton Thorne Inc., Beverly, MA, USA) and membrane integrity was evaluated through FC with Cytosoft software (Millipore-Guava Technologies Inc., Hayward, CA, USA). In experiment 2, IVF was used to evaluate the non-toxicity of SensiTemp and control straws. Cumulus-oocyte complexes (COC; n = 1178; 4 replicates) collected from slaughterhouse ovaries were matured in IVM medium (TCM-199 with bicarbonate, Sigma-Aldrich, Saint Quentin Fallavier, France; 10 µg mL–1 FSH-LH, Reprobiol, Liège, Belgium; and 10% FCS, Thermo Fisher, Illkirch, France) for 22 h. After fertilization, presumptive zygotes of each group (SensiTemp and control for each bull) were cultured in synthetic oviduct fluid medium (SOF, Minitube, Tiefenbach, Germany) with 1% estrous cow serum (ECS) and 0.6% BSA (Sigma-Aldrich, France) up to 8 days. All cultures were conducted at 38.5C in 5% CO2, and 5% O2. The cleavage and blastocysts rates were evaluated on Days 3 and 7, respectively, for each group. Embryo quality was recorded on Day 7 according to the IETS evaluation. Data from each bull were analysed separately using the chi-squared test (P < 0.05). In experiment 1, neither sperm motility from bull 1 (61.2 and 60.5%) and bull 2 (66.2 and 66.5%) nor membrane integrity from bull 1 (58.6 and 52.2%) and bull 2 (61.0 and 61.9%) were different between SensiTemp and control, respectively. Results from experiment 2 showed no difference (P > 0.05) in cleavage rate between SensiTemp and control for the 2 bulls: 92.1 and 91.7% for bull 1 and 94.2 and 94.6% for bull 2 respectively. The blastocysts rate on Day 7 did not differ (P > 0.05) among groups (47.5, 47.1 and 51.3, 50.4% for SensiTemp and control bull 1 and bull 2, respectively) nor the quality of embryos retrieved in the different groups: 25.4, 23.3, and 30.8, 29.6% in grade 1 embryo for SensiTemp and control bull 1 and bull 2, respectively. Those results demonstrate, in vitro, that the new SensiTemp straws were non-toxic and did not affect the semen quality after thawing nor did the SensiTemp straws affect the ability of sperm cells to fertilize oocytes and produce 8-day-old embryos.

A study was conducted to assess the semen quality using Resazurin Reduction Test (RRT) in Jersey and Jersey cross bulls. A total 36 fresh ejaculates were collected for evaluation RRT, sperm motility, acrosomal integrity and sperm concentration. The mean value of RRT in bulls was 15.77±0.85 minutes.Statistical analysis revealed that RRT was highly significantly (p<0.01) and positively correlated with sperm concentration (r=0.769), sperm motility(r=0.858) and acrosomal integrity (r=0.515), hence it may be helpful in evaluating the quality of semen in bulls.

Background: The infertility rate in India has also increased gradually along with population growth. Based on the geographic, the prevalence of male infertility varies. It is noticed that around the world 60–80 million couples with infertility problem every year. The aim of the study was to assess the correlation between the serum hormones and semen quality in infertile males.Methods: This research study was conducted in 60 infertile males aged from 20–50 years who reported at the Department of Andrology, Kanmani Fertility Centre (Pvt. Ltd.), Chennai, Tamil Nadu, India. The semen and hormone analysis was done in all study participants and its correlation was studied using SPSS (statistical product and service solutions) software.Results: The results demonstrate that among the study participants, 15.0% of them were normozoospermia, asthenozoospermia (26.6%), asthenoteratozoospermia (21.6%), oligoasthenozoospermia (16.6%) and oligoasthenoteratozoospermia (20.0) respectively. The results of Pearson correlation revealed that TSH, FSH, LH and PRL show a negative correlation but TST level shows a positive correlation with semen parameters.Conclusions: This study concludes that the serum hormones concentration was observed abnormal in infertile males which show negative correlation with semen parameters of the study participants.

Abstract Background: This was a cross-sectional study in China, we analyzed the levels of macrophages(Mφ) in semen. The study evaluated the influence of the levels of Mφ in semen on sperm quality.Methods: The subjects were 78 males between 25 and 35 years old. The samples were divided into a low group(Mφ<6×105/ml) and a high group (Mφ>6×105/ml). Evaluation included consideration of the influencing factors of male semen quality, macrophage concentration, sperm motility, morphology, membrane integrity DNA fragmentation index (DFI), anti-sperm antibodies (AsAb), IL-10, and IL-12 in semen.Results: There was no difference in the physical or chemical indices of the semen, sperm concentration, AsAb, IL-10, or IL-12 between the two groups (P>0.05). The percentage of sperm forward motility (PR%), the rate of normal sperm shape, and the integrity of cell membranes in the low group were higher than those in the high group (P<0.05), while the percentage of sperm inactivity (IM%), the rate of sperm head deformity, the rate of deformity in the neck and middle segment, the sperm malformation index (SDI), the abnormal sperm index (TZI), and the sperm DFI in the low group were lower than those in the high group (P<0.05). The concentration of Mφ in the semen was linearly correlated with sperm concentration, sperm PR%, IM%, sperm normal shape rate, head deformity rate, neck and middle deformity rate, SDI, TZI, sperm DFI, and sperm cell membrane integrity (P<0.05), but there was no linear correlation with IL-10 or IL-12 (P>0.05).Conclusions: The concentration of Mφ in semen had no significant correlation with semen volume or sperm concentration, but it did have a significant negative correlation with sperm motility, sperm morphology, cell membrane integrity, and DNA breakage rate. There was no significant correlation with the concentration of IL-10 or IL-12.

Background and Aim: Seasonal variations among geographical regions could influence pigeon semen quality differently. This study aimed to determine the seasonal effect on semen availability and quality of racing pigeons in Thailand to understand and improve breeding management in the country. Materials and Methods: Semen was collected from six fertile captive pigeons once a week during summer (March-June), monsoon (July-October), and winter (November-February) during 2019-2020. The success rate of semen collection and semen quality was determined in each season – by which changes in average temperature, humidity, and photoperiod were obtained. Results: Comparable success rates of semen collection were acquired among different seasons, while varied semen qualities were revealed. The percentages of total motility and progressive motility score of sperm were significantly lowest in summer (66.35±3.40 and 3.88±0.15, respectively) compared to monsoon (85.45±2.91 and 4.67±0.10, respectively) and winter (79.29±1.96 and 4.37±0.10, respectively), while its concentration (×109 sperm/mL) and outputs (×106 sperm) were significantly highest in winter (7.62±0.54 and 91.44±10.83, respectively) compared to summer (4.23±0.41 and 48.45±6.35, respectively) and monsoon (3.57±0.30 and 51.45±7.21, respectively). Besides, semen samples collected from birds housing at an average temperature of <29.5°C demonstrated better sperm motility sperm concentration and total sperm counts than those from at a higher temperature. Conclusion: Winter was regarded as the best season contributing the best semen quality, while summer was the worst. Due to the fluctuation of temperature during summer and winter, the seasonal temperature was implied as the major factor contributing to changes in sperm quality of racing pigeons in Thailand.

Ongole Grade cattle is one of local cattle in Indonesia.This study was aimed to analyze the effect of bull age and season on semen quality of Ongole Grade cattles macroscopically (sperm motility, and sperm concentration) and microscopically (semen ejaculate volume) to support the policy and management on improvement of semen production. Data were collected from 533 ejaculates of 10 bulls aged 3, 4, and 5 yr at Singosari National Artificial Insemination Center(SNAIC). The 4-year-old bulls hadhighestsperm motiliy and semen ejaculate volume. The lowest ejaculate volume was reported in Ongole Gradecattlesaged 3 yr. Sperm motility and concentrationweresignificantly higher (P<0.01) in wet season than dry season. In conclusion, the bull age and season should be considered by Singosari National Artificial Insemination Centerwhen collecting the semen, especially on bulls aged 3 yr old and in dry season.
