Chemical characterization of a PD-1/PD-L1 inhibitory activity fraction of the ethanol extract from Gymnadenia conopsea

Abstract Cissus erosa (Vitaceae), popularly known in Brazil as Cipó-fogo, is a medicinal plant used in the treatment of warts and external ulcers. The present study aimed to evaluate the activity of stems and leaves ethanol extracts of C. erosa against the Dengue and Zika virus by the MTT colorimetric method and to carry on the phytochemical characterization of active extracts by high performance liquid chromatography coupled to mass spectrometry (UPLC-MS). Only the leaves ethanol extract showed anti-Dengue virus activity EC50 18.2 µg/ml (SI > 27.5) and low cytotoxicity for LLCMK2 cells (CC50 > 500 mg/ml). Both extracts (stems and leaves) showed anti-Zika virus activity with EC50 of 45.8 mg/ml and 82.8 mg/ml, respectively. These extracts presented CC50 of 309.2 µg/ml (leaves) and 387.6 µg/ml (stems) to Vero cells. Analysis by CCD and HPLC-DAD detected the presence of triterpenes, steroids, flavonoids and tannins. UPLC-MS analyses of these extracts, allowed the identification of the majority of flavonoids present known as vitexin, isovitexin, orientin, isoorientin and two flavones derivatives, methoxyluteolin-6(8)-C-hexosyl and luteolin-7,4’-di-O-glycosylflavone. The results of the phytochemical studies here described suggest that flavonoids and terpenoids are the substances that contribute to the antiviral activity of the ethanol extracts within this species.

Background: Traditionally, The Algerian medicinal plant Elaeosilenum thapsioides have been used for many diseases. Objective: The present research work is aimed to explore the chemical and biological characterization of essential oil of, Elaeoselinum thapsioides (Desf.) Maire. Methods : The essential oils were obtained by hydrodistillation of different Elaeosilenum thapsioides (Apiaceae) aerial parts samples collected from two different regions (Mahouane and Megres) Setif, Eastern Algeria. The chemical characterization of the obtained essential oils was investigated in the present work for the first time by GC and GC-MS. Besides, they were evaluated for their in-vitro acetylcholinesterase (AChE) inhibitory activity whose enzyme hyperactivity is involved in Alzheimer’s disease. Using Ellman’s spectrophotometric method, additionally, their in-vitro antimicrobial activity was assessed by the disc diffusion method. Both activities were performed at various oil concentrations. Results: The GC/MS analysis of the aerial parts (leaves, stems, flowers, and seeds) essential oils of E. thapsioides revealed the presence of dominant compounds and others in small quantities, identifying 47 chemical molecules. Monoterpene hydrocarbons were the main components, ranging from 72.78 % to 99.13 %. Oxygenated monoterpenes and oxygenated sesquiterpenes ranged between (1.37 % -17.25 %) and (0.12 % -3.53 %) in leaves and stem essential oils. Sesquiterpene hydrocarbons were present in small to large quantities in the essential oils of both populations, with contents ranging from 0.69 % to 13.44 %. For the Isothiocyanates, their presence was recorded in leaves and stem essential oils from Mahouane and Merges with 9.73 % and 3.72 %, consecutively. Indeed, the essential oil of the Mahouane stem showed the highest AChE inhibitory activity among all the tested essential oils. Whereas the highest antibacterial activity was shown by the essential oil obtained from Megres leaves against Bacillus cereus ATCC 11778. Conclusion: The oils exhibited a moderate inhibitory activity in both activities.

SummaryFour members of an Italian family (two with histories of venous thromboembolism) had a qualitative defect of antithrombin III reflected by normal antigen concentrations and halfnormal antithrombin activity with or without heparin. Anti-factor Xa activities were consistently borderline low (about 70% of normal). For the propositus’ plasma and serum the patterns of antithrombin III in crossed-immunoelectrophoresis with or without heparin were indistinguishable from those of normal plasma or serum. A normal affinity of antithrombin III for heparin was documented by heparin-sepharose chromatography. Affinity adsorption of the propositus’ plasma to human α-thrombin immobilized on sepharose beads revealed defective binding of the anti thrombin III to thrombin-sepharose. Hence the molecular defect of this variant appears to be at the active site responsible for binding and neutralizing thrombin, thus accounting for the low thrombin inhibitory activity.

ABSTRACT A protein isolated from ovine pituitary glands has been purified, and its homogeneity assessed by NH2- and COOH-terminal amino acid determination, ultracentrifugation studies, and polyacrylamide gel electrophoresis after carboxymethylation. Its chemical and immunochemical properties are closely similar to those of beef and pork neurophysins, less similar to those of human neurophysins. It contains no tryptophan (like other neurophysins) or histidine (like all except bovine neurophysin-I and human neurophysins). It has alanine at the NH2-terminus and valine at the COOH-terminus. Its amino acid composition is similar to, but not identical with those of porcine and bovine neurophysins.
