AbstractRecent experimental data on the transcription dynamics of eve gene stripe two formation of Drosophila melanogaster embryos occurs in bursts of multiple sizes and durations. That has motivated the proposition of a transcription model having multiple ON states for the promoter of the eve gene each of them characterized by different synthesis rate. To understand the role of multiple ON states on gene transcription we approach the exact solutions for a two state stochastic model for gene transcription in D. melanogaster embryos and derive its bursting limit. Simulations based on the Gillespie algorithm at the bursting limit show the occurrence of bursts of multiple sizes and durations. Based on our theoretical approach, we interpret the aforementioned experimental data as a demonstration of the intrinsic stochasticity of the transcriptional processes in fruit fly embryos. Then, we conceive the experimental arrangement to determine when gene transcription has multiple ON promoter state in a noisy environment.

AbstractCircadian clocks are based on a molecular mechanism regulated at the transcriptional, translational and post-translational levels. Recent experimental data unravel a complex role of the phosphorylations in these clocks. In mammals, several kinases play differential roles in the regulation of circadian rhythmicity. A dysfunction in the phosphorylation of one clock protein could lead to sleep disorders such as the Familial Advanced Sleep Phase Disorder, FASPS. Moreover, several drugs are targeting kinases of the circadian clocks and can be used in cancer chronotherapy or to treat mood disorders. In Drosophila, recent experimental observations also revealed a complex role of the phosphorylations. Because of its high degree of homology with mammals, the Drosophila system is of particular interest. In the circadian clock of cyanobacteria, an atypical regulatory mechanism is based only on three clock proteins (KaiA, KaiB, KaiC) and ATP and is sufficient to produce robust temperature-compensated circadian oscillations of KaiC phosphorylation. This review will show how computational modeling has become a powerful and useful tool in investigating the regulatory mechanism of circadian clocks, but also how models can give rise to testable predictions or reveal unexpected results.

Chlamydial abortion constitutes one of the major causes of infectious abortion in sheep and goats. The disease leads to considerable losses in most sheep and goat-rearing countries, as well as, in Greece. In this paper, a review of the causative agent, pathogenesis, host immune response and epizootiology is presented. After recent changes, that have occured in the taxonomy of chlamydiae, it is necessary to refer to new families, new genera and new species within the order Chlamydiales, with emphasis on new species of Chlamydophila abortus, the causal agent of chlamydial abortion. This study reviews the recent experimental data which indicate the sites of entry of the organism, the way of its dissemination and establishment to the placenta. It also reviews current research on the humoral and cell-mediated immune responses, the role of the IFN-γ in latent infection and this of Tcells in the protective immune response. Finally, particular reference is given on the role of the male in the disease transmission and the role of inter-species contamination in chlamydial abortion.

In this study the calculations of the total fusion reaction cross section have been performed for fusion reaction systems 17F + 208Pb and 15C + 232Th which involving halo nuclei by using a semiclassical approach. The semiclassical treatment is comprising the WKB approximation to describe the relative motion between target and projectile nuclei, and Continuum Discretized Coupled Channel (CDCC) method to describe the intrinsic motion for both target and projectile nuclei. For the same of comparsion a full quantum mechanical clacualtions have been preforemd using the (CCFULL) code. Our theorticalrestuls are compared with the full quantum mechaincialcalcuations and with the recent experimental data for the total fusion reaction  checking the stability of the distancesThe coupled channel calculations of the total fusion cross section σfus, and the fusion barrier distribution Dfus. The comparsion with experiment proves that the semiclassiacl approach adopted in the present work reproduce the experimental data better that the full quantal mechanical calcautions.

The earlier proposed general approach for description of the non-ideal mixer is coupled with corresponding boundary conditions for the closed system. Some simplifications in this procedure result in relations which are in agreement with experimental data.

Natural flavor and fragrance market size is expected to grow steadily due to the rising consumer demand of natural ingredients. This market request is guided by the general opinion that the production of natural compounds leads to a reduction of pollution, with inherent advantages for the environment and people’s health. The biotransformation reactions have gained high relevance in the production of natural products. In this context, few pieces of research have described the role of microalgae in the oxidation of terpenoids. In this present study, we questioned the role of microalgal based oxidation in the synthesis of high-value flavors and fragrances. This study investigated the role of three different microalgae strains, Chlorella sp. (211.8b and 211.8p) and Chlorococcum sp. (JB3), in the oxidation of different terpenoid substrates: α-ionone, β-ionone, theaspirane and valencene. Unfortunately, the experimental data showed that the microalgal strains used are not responsible for the substrate oxidation. In fact, our experiments demonstrate that the transformation of the four starting compounds is a photochemical reaction that involves the oxygen as oxidant. Even though these findings cast a shadow on the use of these microorganisms for an industrial purpose, they open a new possible strategy to easily obtain nootkatone in a natural way by just using an aqueous medium, oxygen and light.

The adaptive significance of adjusting behavioral activities to the right time of the day seems obvious. Laboratory studies implicated an important role of circadian clocks in behavioral timing and rhythmicity. Yet, recent studies on clock-mutant animals questioned this importance under more naturalistic settings, as various clock mutants showed nearly normal diel activity rhythms under seminatural zeitgeber conditions. We here report evidence that proper timing of eclosion, a vital behavior of the fruit fly Drosophila melanogaster, requires a functional molecular clock under quasi-natural conditions. In contrast to wild-type flies, period01 mutants with a defective molecular clock showed impaired rhythmicity and gating in a temperate environment even in the presence of a full complement of abiotic zeitgebers. Although period01 mutants still eclosed during a certain time window during the day, this time window was much broader and loosely defined, and rhythmicity was lower or lost as classified by various statistical measures. Moreover, peak eclosion time became more susceptible to variable day-to-day changes of light. In contrast, flies with impaired peptidergic interclock signaling ( Pdf01 and han5304 PDF receptor mutants) eclosed mostly rhythmically with normal gate sizes, similar to wild-type controls. Our results suggest that the presence of natural zeitgebers is not sufficient, and a functional molecular clock is required to induce stable temporal eclosion patterns in flies under temperate conditions with considerable day-to-day variation in light intensity and temperature. Temperate zeitgebers are, however, sufficient to functionally rescue a loss of PDF-mediated clock-internal and -output signaling
