Tempeh and red ginger flour for hypercholesterolemic rats

Purpose This study aims to evaluate the effect of the combination of tempeh and red ginger flour in hypercholesterolemic rats. Design/methodology/approach Sprague Dawley male rats (n = 30; body weight 150-200 g) were randomly divided by five groups (n = 6), consist of negative control (K−) group: group fed by normal diet (laboratory standard diet laboratory, 4.35 kcal/g, 0% cholesterol); positive control (K+) group: group fed by high cholesterol diet (5.28 kcal/g, 12.1% cholesterol); Treatment 1 (P1) group: group fed by high cholesterol diet and treated by tempeh flour (TF) 1.9 g/200 g body weight; Treatment 2 (P2) group: group fed by high cholesterol diet and treated by red ginger flour (RGF) 0.036 g/200 g body weight; and Treatment 3 (P3) group: group fed by high cholesterol diet and treated by a combination TF 0.95 g/200 g body weight and red ginger 0.018 g/200 g body weight. The lipid profiles and malondialdehyde (MDA) were assessed. Findings The combination of tempeh and red ginger has successfully exerted the total cholesterol (121.9 ± 3.41 mg/dL; p < 0.01), triglycerides (TG) (89.3 ± 2.94 mg/dL; p < 0.01), low-density lipoprotein (39.1 ± 1.59 mg/dL) and MDA (2.0 ± 0.24 mg/dL; p < 0.01) lower than the other treatments. Also, the combination of tempeh and red ginger treatment appeared the highest high-density lipoprotein concentration (63.4 ± 2.26 mg/dL; p < 0.01) compared to the other treatment groups. Total cholesterol and TG were predicted as the increasing of MDA concentration. Originality/value The combination treatments successfully improved the lipid profiles and MDA. Furthermore, the combination of tempeh and red ginger could be effective in supporting therapy as the hypolipidemic condition.

Obesity has been linked with several comorbidities like cardiovascular disorders, diabetes, and hypertension. Weight reduction is directly associated with the consumption of green tea. So, with this premise of background, we evaluated the potential of green tea consumption on obesity and serum lipid profiles in both normo- and hyper-cholesterol emic rabbits. Sixty New Zealand white rabbits (age: 6-8 months; weight 1.5-2 kg) were used for this experiment. Rabbits were randomized and equally divided into three different groups. Group I (control group) received 10 ml/kg of distilled water twice daily, Group II received high cholesterol diet twice daily, and Group III received high cholesterol diet along with green tea solution 10 ml/kg, twice daily for 90 days. Weight measurements and serum analysis were conducted on Day 0 and day 90. For the obtained data, SPSS analysis was performed (SPSS 21). A significant increment of body weight was observed in control (1.97 ± 0.12 vs 2.87 ± 0.21, p<0.0001) and high cholesterol groups (1.91 ± 0.23 vs 3.13 ± 0.31, p<0.0001) at Day 90 when compared with Day 0. Though the results were not significant, there was a reduction in the body weight in the green tea group (2.20 ± 1.47 vs 1.72 ± 0.22) at Day 90 when compared with Day 0. Consumption of green tea for 90 days resulted in weight loss and inhibition of the increase in the levels of TC, LDL, and VLDL. So, green tea can act as a natural food supplement for the management of obesity and dyslipidemia.

Garlic has been widely recognized as herbal medicine for prevention and treatment of cardiovascular disease, atherosclerosis, and hyperlipidemia. The main bioactive compound, allicin, can influence cholesterol metabolism, reduce total cholesterol and increase HDL cholesterol. The purpose of this research was to find out the effect of garlic extract on the reduction of total cholesterol and HDL cholesterol ratio in hypercholesterolemic rats. This research was an experimental research with pre and post-test control group design. The research used male Wistar rats (Rattus norvegicus) with ±3 mounts old and ±200 grams of body weight. The rats were divided into two groups, group I and II; each group consisted of 15 rats. Before the treatment, all rats were induced by a high-cholesterol diet for two weeks, to get a hypercholesterolemic condition. Group I, as a control group, were induced by a high-cholesterol diet, while group II, treatment group, were also induced by a high-cholesterol diet orally by 0.432 grams garlic extract /200 grams body weight/day. The treatment occurred for 25 days. The data were taken before the treatment and after the last treatment, by taking the blood from orbital sine of rats which been fasted for 12 hours, and the level of serum cholesterol total and the level of serum HDL cholesterol were checked from both groups, then the results were analyzed with t-test. Based on the results, it could be concluded that the oral induction of 0.432 grams garlic extract /200 grams body weight/day for 25 days on rats, could not reduce total cholesterol, therefore the serum total cholesterol and HDL cholesterol ratio was not significant (p>0.05). Meanwhile, the decrease of total cholesterol in treatment group and the decrease of HDL cholesterol in both groups were significant (p<0.05).

Objective: The present study is to determine the effect of liquid of sodium alginate and powder on the prevention of raising cholesterol levels and atherosclerosis in rats (preventive effect).Method: Experimental animals were divided into prevention groups. Preventive groups used were 24 normal rats and then divided into four groups, each group consisted of 6 animals: Group 1 (fed a regular diet), Group 2 (fed high cholesterol diet), Group 3 (fed high cholesterol diet and alginate liquid 2% diet), and Group 4 (fed high cholesterol and alginate powder). Rats are given with cholesterol food, alginate liquid, alginate powder a dose of 1 ml/100 g body weight once a day. Parameters measured were cholesterol level, low-density lipoprotein (LDL) levels, high-density lipoprotein (HDL) levels, triglyceride (TG) level, body weight every week, and histopathologically of the aorta for foam cells examined at the end of the experiment. Analysis of data was performed using one-way ANOVA.Results: The lipid profiles in the preventive group showed alginate liquid and powder which have the effect of preventing increase total cholesterol, LDL, TG, number of foam cells, and decrease HDL. Diet using alginate powder better than alginate liquid, but the effect was not different significantly (p<0.05) so do the regular diet, but higher than given with high cholesterol diet significantly (p>0.05).Conclusion: This study showed that the alginate liquid and alginate powder have an anti-hypercholesterolemia property which has been proved by preventing increase LDL, TG, and increased HDL levels.

Introduction: Lipid metabolism disorder or hyperlipidemia is known as a risk factor for cardiovascular disease, the increase in serum homocysteine and leptin are associated with atherosclerotic disease. The purpose of the present study was to examine the effects of bovine lactoferrin (bLF) on serum homocysteine (Hcy), apolipoproteinA-I (ApoA-I) and B (ApoB), leptin and lipid profile changes in high-cholesterol-diet (HCD) fed rats. Methods: The Healthy Adult Sprague-Dawley (SD) male rats were randomly assigned into three experimental groups. Each group consisted of eleven male rats including control group, HCD rats and hypercholesterolemic rats, which were treated with bLF (HCD+bLF). bLF was given by gavage (200 mg/kg/d). After 4 weeks of feeding and overnight fasting, total blood samples were collected. Results: The results showed the elevated level of Hcy, leptin, total cholesterol, low density lipoprotein cholesterol (LDL-C), ApoB and decrease in ApoA-I in non-treated HCD group compared to the control rats. Administration of bLF significantly ameliorated the Hcy and leptin levels with decrease in LDL-C and total cholesterol in rats fed with a high-cholesterol diet. bLF also tended to increase low serum concentration of ApoA-I and high density lipoprotein cholesterol (HDL-C) in HCD rats. Meanwhile, upon bLF-treated rats, there was a significant decrease in ApoB in HCD group. Conclusion: The findings indicated that bLF can improve the alteration of serum Hcy, leptin, apolipproteins and lipid changes in male rats fed with high-cholesterol diet. So, bLF can counteract with HCD elicited hyper-homocysteinemia and hyper-leptinemia, suggesting it to have the useful therapeutic potential in patients with atherosclerosis and lipid disorder.

Excessive fat consumption can increase blood cholesterol level. Phytosterol composition in the plant can decrease blood cholesterol level. One of the plants that contain phytosterol is beans. This study was to prove the effect of antihypercholesterol the ethanol extract of the beans (Phaseolus vulgaris L.) with variety of doses of 50 mg/Kg BW; 100 mg/Kg BW and 150 mg/Kg BW in male Wistar rats with high fat level condition. Subjects of this study were 24 Wistar rats divided into 6 groups randomly with posttest control group study design. First group as negative control, second group as positive control, and the third (high cholesterol diet and ethanol extract in dose of 50 mg/kg,bw), fourth ( high cholesterol diet and ethanol extractin dose of 100 mg/kg,bw), fifth ( high cholesterol diet and ethanol extract in dose of 150 mg/kg,bw),  sixth (high cholesterol diet and simvastatin drug). The data was analyzed with ANOVA Tukey test. The result of this study shows total cholesterol levels decrease 23,88%; 30,14%; 35,82%. Cholesterol LDL levels decrease 38,09%; 52,38%; 61,35%. Cholesterol HDL levels increase 21,86%; 56,56%; 63,84%. Dose variant on this study giving the best result to decrease total cholesterol levels and LDL was 150 mg/Kg BW. Furthermore, 100 mg/Kg BW dose gave the best result to increase HDL cholesterol level. Based on the results it can be suggested that beans ethanol extract is able to decrease total cholesterol level, LDL and increase HDL cholesterol levels.

In this work, hypercholesterolemia was induced in rabbits by feeding them a high cholesterol diet (CD, 350 mg/d) and the effects of supplementing this diet with 0.5 g/d Spirulina platensis was evaluated by measuring the levels of serum total-cholesterol (TC), triacylglycerols (TAG) and high-density lipoprotein (HDL-cholesterol) at the start of the experiment and after 30 d and 60 d. It was found that the levels of serum cholesterol decreased from 1,054±101 mg.dL-1 in the rabbits fed a CD without S. platensis to 516±163 mg.dL-1 to those fed with a high cholesterol diet supplemented with S. platensis (significant at p <0.0001). The addition of Spirulina to the cholesterolemic diet did not cause significative decrease on the levels of triacylglycerols in the animals. The levels of serum high-density lipoprotein (HDL-cholesterol) was 73±31 mg.dL-1 for rabbits fed a CD without S. platensis as compared to 91.0±15.7 mg.dL-1 in those fed a CD supplemented with S. platensis (significant at p=0.1533).

Nutritional strategies to reduce hyperlipidemia and the risk of cardiovascular disease are gaining more public favor and medical professionals’ attention. The authors of this study explored the effect of sweet potato leaf powder (SPLP) feeding on the parameters of plasma lipids, reactive oxygen species, and time to thrombosis formation in Syrian hamsters fed with high-cholesterol diets. The animals were separated into six groups: a feeding control diet, a control diet containing 0.1% cholesterol, a control diet containing 0.2% cholesterol, a control diet containing 0.1% cholesterol plus 2.5% SPLP, a control diet containing 0.1% cholesterol plus 5% SPLP, and a control diet containing 0.2% cholesterol plus 5% SPLP for six weeks. The levels of serum total cholesterol (51% increase), low-density lipoprotein cholesterol (70.6% increase), very-low-density lipoprotein cholesterol (51.3% increase), and the triglyceride and atherogenic index (LDL-C/HDL-C) significantly increased in the high-cholesterol diet groups. Concomitant 5% sweet potato leaf powder ingestion significantly decreased the lipid profiles, with a 20.6% total cholesterol reduction in the 0.1% cholesterol diet groups, a 17.2% reduction in the 0.2% group, a 48.7% LDL reduction in the 0.1% cholesterol group, and a 30.3% reduction in the 0.2% group, with a consequent decrease in the atherogenic index. SPLP feeding was found to be associated with increased fecal sterol contents, with a 188.6% increase in the 0.1% cholesterol-fed group and a 177.3% increase in the 0.2% group. The SPLP-fed groups had depressed ROS levels, elongated FeCl3-induced times to thrombosis formation, and increased liver superoxide dismutase contents and SREBP-1 protein expression. Sweet potato leaf intake could reduce plasma total cholesterol, LDL, and oxidative stress. We suggest sweet potato leaf intake as a choice of nutritional strategy for hyperlipidemia and cardiovascular disease prevention.

Abstract BackgroundEmodin (EM) is one of bioactive components extracted from Rheum palmatum L. (Dahuang), which possesses numerous pharmacological activities including hypolipidemic effect. However, the potential action of EM on hyperlipidemia (HLP) remains unclear. Here, the theraputic effect of EM against HLP were investigated.MethodsIn this study, the hypolipidemic properties of EM were evaluated using high-cholesterol diet (HCD)-stimulated zebrafish larvae model. The body weight, body length and body mass index (BMI) was measured. The total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C) and high-density lipoprotein cholesterol (HDL-C) as well as the activities of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were detected by corresponding assay kits. Tg (flil: eGFP) zebrafish were utilized to observe vascular cholesterol accumulation and Tg (mpx: eGFP) zebrafish to visualize and quantify neutrophil inflammation. The hepatic lipid deposition and hepatic histopathology were analyzed by Oil red O staining and H&E staining, respectively. Finally, the underlying mechanism of EM were investigated using real-time quantitative PCR (RT-qPCR) analysis to assess the gene levels of adenosine monophosphate-activated protein kinase alpha (AMPKα), sterol regulatory element binding protein 2 (SREBP-2), proprotein convertase subtilisin kexin 9 (PCSK9), low-density lipoprotein receptor (LDLR), 3-hydroxy-3-methyl-glutaryl-CoA reductase (HMGCR), adenosine triphosphate binding cassette transporter A1 (ABCA1) and adenosine triphosphate binding cassette transporter G1 (ABCG1).ResultsOur data indicated that EM reduced obesity of zebrafish as evidenced by the decrease in body weight, body length and BMI. EM significantly reduced TC, TG, and LDL-C, and increased HDL-C contents. Moreover, it displayed a prominent inhibitory effect on blood cholesterol accumulation, hepatic lipid accumulation, and neutrophil inflammation in vascular site. Additionally, EM improved the liver function through decreasing ALT and AST levels of zebrafish with HCD-induced hepatosteatosis. Further investigation showed that EM treatment attenuated lipid accumulation via upregulating the expression of AMPKα, LDLR, ABCA1 and ABCG1, and downregulating the expression of SREBP-2, PCSK9 and HMGCR.ConclusionTo conclude, EM alleviated lipid metabolism disorder symptoms caused by HCD via modulating AMPK/SREBP-2/PCSK9/LDLR pathway in larvae, suggesting that EM may be developed into hypolipidmic agent for treating lipid metabolism related diseases.

Chemical screening of the leaves and flowers of Crataegus aronia resulted in the isolation of hyperoside, quercetin, rutin and β-sitosterol for the first time from this plant. The effects of the hydroethanolic extract of C. aronia (CAHE) on hypercholesterolemic rats were investigated. The rats, treated orally for four weeks with 400 mg/kg/day CAHE, exhibited significant decreases in serum total cholesterol (TC) and low-density lipoprotein (LDL). The results were compared with those obtained after oral administration of atorvastatin (10 mg/kg/day). Furthermore, 10-week daily co-administration of a high cholesterol diet and CAHE (200 mg/kg/day) prevented the increase in TC and LDL. These observations indicate that CAHE has a hypocholesterolemic effect.
