Purpose: To investigate the hepatoprotective effect of Ipomoea batatas extract against alcohol-induced liver damage in mice. Methods: Male C57BL/6 mice were randomly divided into 4 experimental groups (n = 10). Normal Group: The animals received distilled water 5 ml/kg for 7 days; Alcohol Group: The animals received alcohol 5 ml/kg of 40 % w/v alcohol for 7 days; Alcohol + Purple sweet potato leaf extract (PSPE) Group: PSPE 400 mg/kg was for 7 days. The animals received alcohol 5 ml/kg of 40 % w/v alcohol for 7 days; Alcohol + Hovenia dulcis Thunb extract (HDE) Group: HDE 400 mg/kg was for 7 days. To confirmed to the liver protection effect of PSPE, it was calculated, and the levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), triglyceride (TG), and total cholesterol (TC) in serum were detected. To evaluate changes of histological in alcohol-fed mice, liver tissue was determined by H&E staining. Results: Blood alcohol concentration in purple sweet potato leaf extract (PSPE) 200 mg/kg and Hovenia dulcis (H. dulcis) extract (HDE) 200 mg/kg treated group significantly decreased compared to - alcohol with water treated group (p < 0.05). Serum ALT (alanine aminotransferase) and AST (aspartate aminotransferase) were markedly reduced. Liver sections in mice stained with H&E (hematoxylin and eosin) stain to displayed the physiological changes in the liver tissue. Furthermore, the results showed that inflammatory cells increased in the alcohol group compared to the normal group, but spontaneously decreased in the PSPE or HDE-treated group. Conclusion: These results demonstrate that Ipomoea batatas may be therapeutically effective in protecting the liver from alcohol-induced hepatotoxicity and fatty liver.

AbstractAdequate integrated management of potato leaf diseases can have an influence on pathogens which are challenging to control and cause main potato tuber diseases. Deriving from this hypothesis, an experiment was designed to evaluate the efficacy of foliar application of twelve fungicides on late blight in tubers, common scab, black scurf and silver scurf. In the period of 2013 to 2017, the fungicides were equally applied six times in registered doses during the growing seasons. Eight tuber disease indicators and eight yield characteristics were then evaluated in harvested tuber samples. Significant differences were found among the fungicides used. In comparison to the negative control (n. c.), the application of Infinito, Ranman Top and Revus Top reduced the quantity of tubers infected by late blight by 72% on average. The incidence of common scab increased by 65% of n. c. under the effect of copper oxychloride, while an average reduction by 19% of n. c. was observed with the use of Acrobat MZ WG, Curzate Gold and Consento. The occurrence of silver scurf was significantly enhanced by Curzate Gold and Consento (105% of n. c.), while it was suppressed by Revus Top and Revus (85% of n. c.). All fungicides were ineffective on black scurf yet had a positive influence on the potato yield and quality, with the choice of fungicide having a great impact on the quality of tubers. The application of any safe fungicide is important, especially in years similar to 2015, which was the most devastating recent year for potato production in terms of structural changes in yield of tubers.

The paper presents the results of enzyme immunoassay (ELISA) of the presence of potato viruses S (PVS), M (PVM), Y (PVY), X (PVX), and Potato Leaf Roll Virus (PLRV) in tubers of various potato varieties grown in 2019 in the Republic of Bashkortostan. PVS, PVM, PVY, PVX were detected, the VSLK virus was not detected. The greatest infection with PVS, PVM, and PVX was observed in samples of tubers of early-maturing varieties. Tubers of mid-early and late-maturing varieties were more often affected by PVY than early-maturing ones, and PVX was not detected in plants of those varieties. Ribonuclease (RNase) activity in potato tubers of 14 varieties depended on the earliness of the variety, regardless of the soil and climatic conditions. A statistically significant positive correlation (P<0.05) was found between the abundance of PVS and PVM viruses and RNase activity in tubers of medium-early and late-maturing varieties, whereas, on the contrary, a negative correlation between PVM and RNase activity was revealed when analyzing tubers of early-maturing varieties. It is concluded that the prevalence of potato viruses in the territory of the Republic of Bashkortostan and the activity of RNase in tuber seedlings depend on the potato variety and the type of viruses.

Potato is an important crop in Shanxi province located in north-central China. During 2019-2020, 319 potato leaf samples were collected from eight locations distributed in three major potato production areas in Shanxi. Bio-chip detection kit revealed the presence of several potato viruses, and among them potato virus Y (PVY) was the most common one, reaching the incidence of 87.8% of all symptomatic samples. The immuno-captured multiplex reverse transcription (RT)-PCR was used to identify strains for all 280 PVY-positive samples, unveiling 242 samples infected with a single strain of PVY (86.4%) and 38 (13.6%) with a mixed infection. Of samples with a single-strain infection, PVY -SYR-II accounted for 102 (42.1%), followed by PVYN-Wi (33, 13.6%) , PVY -SYR-I (28, 11.6%), 261-4 (22, 9.1%), PVYNTNa (20, 8.3%), PVYNTNb (19, 7.9%), and PVY -SYR-III (18, 7.4%). Seven isolates representing different recombinants were selected for whole genome sequencing. Phylogenetic and recombination analyses confirmed the RT-PCR based strain typing for all seven strains of PVY found in Shanxi. SXKL-12 is the first SYR-III strain from potato reported from China. However, unlike that in other known SYR-III isolates, the region positioned from 1,764 to1,902 nt in SXKL-12 shared the highest sequence identity of 82.2% with an uncharacterized PVY isolate, JL-23, from China. Interestingly, the PVYN-Wi isolate SXZY-40 also possessed a more divergent sequence for the region positioned from 6,156 to 6,276 nt than other N-Wi isolates known to date, sharing the highest identity of 86.6% with an uncharacterized Chinese PVY isolate, JL-11. Pathogenicity analysis of dominant strains PVY -SYR-II and PVYN-Wi in six local popular potato cultivars revealed that Kexin 13, Helan 15 and Jizhangshu 12 were susceptible to these two strains with mild mottling or mosaic symptoms expression, while three cultivars, Jinshu 16, Qingshu 9, Xisen 6 were found fully resistant.

The miniaturization of hyperspectral cameras has opened a new path to capture spectral information. One such camera, called the hybrid linescan camera, requires accurate control of its movement. Contrary to classical linescan cameras, where one line is available for every band in one shot, the latter asks for multiple shots to fill a line with multiple bands. Unfortunately, the reconstruction is corrupted by a parallax effect, which affects each band differently. In this article, we propose a two-step procedure, which first reconstructs an approximate datacube in two different ways, and second, performs a corrective warping on each band based on a multiple homography framework. The second step combines different stitching methods to perform this reconstruction. A complete synthetic and experimental comparison is performed by using geometric indicators of reference points. It appears throughout the course of our experimentation that misalignment is significantly reduced but remains non-negligible at the potato leaf scale.

Plant-based transient expression systems have recognized potential for use as rapid and cost-effective alternatives to expression systems based on bacteria, yeast, insect, or mammalian cells. The free-floating aquatic plants of the Lemnaceae family (duckweed) have compact architecture and can be vegetatively propagated on low-cost nutrient solutions in aseptic conditions. These features provide an economically feasible opportunity for duckweed-based production of high-value products via transient expression of recombinant products in fully contained, controlled, aseptic and bio-safe conditions in accordance with the requirements for pharmaceutical manufacturing and environmental biosafety. Here, we demonstrated Agrobacterium-mediated high-yield transient expression of a reporter green fluorescent protein using deconstructed vectors based on potato virus X and sweet potato leaf curl virus, as well as conventional binary vectors, in two representatives of the Lemnaceae (Spirodela polyrhiza and Landoltia punctata). Aseptically cultivated duckweed populations yielded reporter protein accumulation of &gt;1 mg/g fresh biomass, when the protein was expressed from a deconstructed potato virus X-based vector, which is capable of replication and cell-to-cell movement of the replicons in duckweed. The expression efficiency demonstrated here places duckweed among the most efficient host organisms for plant-based transient expression systems, with the additional benefits of easy scale-up and full containment.
