One hundred and fifty-seven Chinese hamsters ( Cricetulus griseus) from the Lovelace Foundation Inhalation Toxicology Research Institute colony had pulmonary granulomas, nodular hyperplasia of the liver, granulocytic bone marrow hyperplasias and myelogenous leukemia, nephrosclerosis and uterine adenocarcinomas. These Chinese hamsters had a median life span of 1 075 days. The long life span and information on naturally-occurring diseases suggest that this species might be more widely used for toxicological and biomedical research.

The frequent closure of roads for the sake of maintenance and refurbishment is an increasing problem for the road administrators and users, because of induced high costs, restrictions and influence they impose, traffic jam, and the many other disruptions such interventions have on traffic. In these circumstances, carriageways with long life span, with adapted tar, have much to offer, especially if they can exhibit optimum performances without requiring major repairs for over 30 years. On high traffic roads, research have shown that binders with elevated modules are a reliable and credible alternative to render their life span more sustainable and durable. In this context, the advantages presented by the absence of important repairs and refurbishing can be enough to justify the initial high costs of these advanced carriage way coatings. Studies on the different binders to be used were done, which have permitted in accordance with the performance of the layers, the required tests and the different study levels. Also, this has permitted us to analyze the obtained results and conclude that due to the expected needs, the LA and MDE tests, the PSV tests are the criteria that determine the selection of aggregates; The complexes modular methods, fatigue resistance (level IV), seem to be necessary for the forecasting of the mechanical behavior of bituminous materials.

ABSTRACT A gene encoding a protein that shows sequence similarity with the histone H1 family only was cloned in Ascobolus immersus. The deduced peptide sequence presents the characteristic three-domain structure of metazoan linker histones, with a central globular region, an N-terminal tail, and a long positively charged C-terminal tail. By constructing an artificial duplication of this gene, namedH1, it was possible to methylate and silence it by the MIP (methylation induced premeiotically) process. This resulted in the complete loss of the Ascobolus H1 histone. Mutant strains lacking H1 displayed normal methylation-associated gene silencing, underwent MIP, and showed the same methylation-associated chromatin modifications as did wild-type strains. However, they displayed an increased accessibility of micrococcal nuclease to chromatin, whether DNA was methylated or not, and exhibited a hypermethylation of the methylated genome compartment. These features are taken to imply thatAscobolus H1 histone is a ubiquitous component of chromatin which plays no role in methylation-associated gene silencing. Mutant strains lacking histone H1 reproduced normally through sexual crosses and displayed normal early vegetative growth. However, between 6 and 13 days after germination, they abruptly and consistently stopped growing, indicating that Ascobolus H1 histone is necessary for long life span. This constitutes the first observation of a physiologically important phenotype associated with the loss of H1.

Abstract Guinea pigs, in which cells with long life span were selectively labeled (3H-thymidine), were joined in parabiosis to nonlabeled syngeneic litter mates at a time when label reutilization detectable by radioautography could be excluded. The distribution of labeled cells was investigated quantitatively using radioautography and liquid scintillation counting in the marrow and blood at the time of establishment of parabiosis and again at its termination 2 wk later, when the thoracic duct lymph, lymph nodes, spleen, and thymus were also examined. Single animals labeled in the same manner served as controls. Of all cells with a slow rate of turnover and long life span, only small lymphocytes entered the circulation and crossed the anastomosis in detectable numbers. As indicated by the similar percentages of labeling in respective tissues, a complete intermixing of long-lived lymphocytes occurred in the bone marrow, lymph, lymph nodes, and spleen of the parabionts. The sum of the per cent labeled lymphocytes in two parabionts was in agreement with the extent of labeling in respective tissues of single controls. The presence of a minor population of lymphocytes with a long life span was confirmed in the marrow. Ten to 30 times as many labeled long-lived lymphocytes migrated into the bone marrow of initially unlabeled animals as were found in an equal volume of blood. The majority, if not all long-lived lymphocytes migrate to the marrow from the blood, and they also reenter the blood. They have a similar life span and in parabionts equilibrate in a similar manner as recirculating long-lived lymphocytes.
