Determination of the composition and the allelic state of disease and pest resistance genes in potato parental lines using DNA markers

A combination of recombinant-inbred lines of the F6 generation from the cross B-16 ќ AR 7086 between lines with two wheat-rye translocations, 1BL/1RS from the Petkus and 1AL/1RS from the rye Insave, was developed. Using gliadin and secalin loci as genetic markers we identified recombinant arm 1RS in positions 1A and 1B in about 10% of lines. The rest of lines with the rye material may also carry recombinant 1RS, which can be identified with DNA markers. Lines with recombinant arm 1RS may serve as a source of new combination of rye genes for disease and pest resistance.

Abstract Sugarcane brown rust, caused by Puccinia melanocephala, is one of the main diseases of sugarcane in China. The identification and discovery of new resistance genes have important theoretical and practical significance for preventing outbreaks of brown rust and ensuring the sustainable production of sugarcane. To screen for polymorphic simple-sequence repeat (SSR) molecular markers for localization of brown rust resistance genes, we used two populations that are suitable for genetic linkage map construction and mapping of new resistance genes to construct resistant and susceptible genetic pools. We then screened 449 pairs of primers to identify polymorphic SSR markers in the parental lines and the resistant/susceptible genetic pools. The results showed that 25 pairs of primers directed amplification of polymorphic DNA fragments between the parents of the cross combination ‘Yuetang 03-393’ × ‘ROC 24’, and 16 pairs of primers amplified polymorphic fragments between the parents of the cross combination ‘Liucheng 03-1137’ × ‘Dezhe 93-88’. Four pairs of primers (SMC236CG, SCESSR0928, SCESSR0636 and SCESSR2551) amplified polymorphic DNA fragments between the parental lines and the resistant/susceptible genetic pools in ‘Yuetang 03-393’ × ‘ROC 24’. The results of this study will establish a solid foundation for the mapping of new brown rust resistance genes, genetic linkage map construction and the development of closely-associated molecular markers in sugarcane.

Detection of carbapenem resistance genes is a critical issue for hospitals due to possible recommendations for infection control and targeted therapy. The Cepheid Xpert instrument, a Carba-R test for the detection and differentiation of five common carbapenemase genes, was tested from September 2020 to February 2021. As part of the approbation, 20 tests were provided. This review presents the results of the approbation of a relatively regular sensitivity study on Siemens WalkAway‑96 plus. Cepheid Xpert Carba-R analysis has been shown to be an accurate and fast tool for detecting colonization by carbapenem-resistant gram-negative bacteria, which can help limit the spread of these organisms in hospitals.

The purpose of this study was the determination of antiseptic resistance genes (qacA/B and qacC) from staphylococcal mastitis in cattle in various regions of Turkey. In total, 283 isolates (Burdur: 36, Hatay: 47 and Van: 200) were studied, and the antiseptic resistance genes were detected using simplex PCR. The distribution of the qacA/B and qacC genes, mediating resistance against quaternary ammonium compounds, was found to vary among the different isolates. The qacA/B genes were found in three of the Burdur isolates, six of the Hatay isolates and seven of the Van isolates. The qacC gene was found in two of the Burdur isolates, none of the Hatay isolates and two of the Van isolates. The presence of these genes and transmission among Staphylococcus spp. strains may pose risks in the control of mastitis, as well as to public health.

This study was conducted to determine the inheritance of common bunt resistance in twelve bread wheat varieties and their half-diallel hybrids in Turkey. The disease ratings were performed on the F2 generations of the hybrids in field conditions. The obtained data were analysed by the χ2 test to determine the effective gene numbers and inheritance type in the disease resistance. In addition, the data were evaluated according to the Jinks-Hayman diallel analyses. In conclusion, it was found that of the twelve wheat parents, four contained three resistance genes and four of them contain two resistance genes. The dominant genes were prominent in the population and complete dominance was present. Therefore, the selection for disease resistance should be delayed until the following generations.
