Late Quaternary history of the Lake O'Hara region, British Columbia – an evaluation of sedimentation rates and bulk amino acid ratios in lacustrine records

Lake O'Hara (subalpine) and Opabin Lake (alpine) are situated directly adjacent to a high section of the Continental Divide in the central Canadian Rocky Mountains. Core samples recovered from the lakes show a consistent stratigraphy comprising gyttja and underlying inorganic clastic sediments. The gyttja contains Bridge River (2350 years BP) and Mazama (6800 years BP) tephras and is separated from the lower clastic sediments by a sharp, conformable contact. Radiocarbon dates obtained from conifer needles, extracted from directly above the contact, indicate that deglaciation had proceeded upvalley from the O'Hara basin priorto ca. 10 100 years BP. Preliminary palaeobotanical and macrofossil data suggest that a Pinus–Abies forest with lesser Picea was established in the vicinity of Lake O'Hara by this time. Consequently, the minimum age of moraine systems situated downvalley from Lake O'Hara is Late Wisconsinan.Mean annual sedimentation rates were derived from sediment thickness data from 14 Lake O'Hara and 2 Opabin Lake cores. Averaged total sedimentation rate values from the Lake O'Hara cores are 0.13 mm/year (post-Bridge River), 0.13 mm/year (Mazama – Bridge River) and 0.05 mm/year (11 000 years BP – Mazama). Averaged total sedimentation rate values from the Opabin Lake cores are 0.19 mm/year (post-Bridge River), 0.07 mm/year (Mazama – Bridge River), and 0.06 mm/year (8530 years BP – Mazama). Higher total sedimentation rates in post-Bridge River sediments of Opabin Lake are presumably related to climatic conditions associated with more extensive upvalley ice during the last ca. 2300 years. Highly variable sedimentation rate data obtained from the Lake O'Hara cores suggest that the use of sedimentation rate data as a proxy record of upvalley glacial activity is inappropriate in the Lake O'Hara setting where inflowing glacial stream systems are interrupted by upvalley lake basins.Aspartic acid D/L ratios were derived from bulk gyttja samples of known age from seven Lake O'Hara and one Opabin Lake core. In all but two cases, aspartic acid D/L ratios increase consistently with respect to sediment age. The increasing downcore trends in the aspartic acid D/L ratios suggest the possibility of using amino acid data from bulk gyttja samples as a check for reworking in cases where chronostratigraphic markers are absent.

ABSTRACT A sequence of surface and buried paleosols from the slopes of Mount Kenya, East Africa, has been identified and dated by radiocarbon and amino acid dating techniques in order to elucidate the Quaternary history of the area. Buried paleosols vary in radiocarbon age from 900 to > 40,000 yrs BP. They have developed in glacial and periglacial deposits of variable texture, consisting of a high percentage of clasts of phonolite, basalt and syenite. All but two paleosols are located in the Afroalpine zone (above 3200 m). D/L ratios of amino acids in Ab horizons were determined in order to establish their reliability for relative age dating. Alanine, aspartic acid, glutamic acid, leucine, valine, and phenylalanine were routinely analyzed. Aspartic acid, as in other cases, proved reliable yielding remarkably consistent results, with higher ratios corresponding to increasing age. Other acids analyzed showed distinct trends, although not as convincing as aspartic acid. In most cases, the aspartic acid ratio/ age relationships were supported by radiocarbon dates. D/L ratios of aspartic acid varied from approximately 0.07 for modern samples, to approximately 0.45 in samples > 40,000 years old.

AbstractThis study examines a sediment core (SR-63) from a mangrove ecosystem along the Sibun River in Belize, which is subject to both changes in sea-level and in the characteristics of the river's drainage basin. Radiocarbon dates from the core show a decreased sedimentation rate from ~ 6 ka to 1 cal ka BP and a marked change in lithology from primarily mangrove peat to fluvial-derived material at ~ 2.5 cal ka BP. Changes in the sedimentation rates observed in mangrove ecosystems offshore have previously been attributed to changes in relative sea-level and the rate of sea-level rise. Pollen analyses show a decreased abundance of Rhizophora (red mangrove) pollen and an increased abundance of Avicennia (black mangrove) pollen and non-mangrove pollen coeval with the decreased sedimentation rates. Elemental ratios ([N:C]a) and stable isotope analyses (δ15N and δ13C) show that changes in the composition of the organic material are also coeval with the change in lithology. The decrease in sedimentation rate at the site of core SR-63 and at offshore sites supports the idea that regional changes in hydrology occurred during the Holocene in Belize, influencing both mainland and offshore mangrove ecosystems.

Five accelerator (TAMS system, University of Arizona) radiocarbon dates on small (30–50 mg) paired bivalves or shell fragments have been obtained on two piston cores. These dates are compared with conventional and accelerator 14C dates on the total organic fraction (< 2 μm size fraction) from similar levels in the two cores. The shell dates are always younger by several thousands of years. We argue that the shell dates are the more reliable; for example, we demonstrate that pre-Quaternary pollen is abundant in the lowermost 6 m in core HU77-159 from Frobisher Bay. Since the occurrence of shells is rare in piston cores, we have developed an empirical relation that enables us to change dates on organic matter to a more probable "true" age. If this correction is applied to dates on the total organic fraction that we obtained on an additional five cores from fiord and shelf sites, a consistent pattern of sedimentation rate variations is shown. This indicates that sedimentation rates were highest between ca. 9000 and 6500 years BP, a time when fiord glaciers in Baffin Island were undergoing substantial retreat.

AbstractThe genomic DNA sequence of a 2.4-kb region of the X-linked developmental gene fused was determined in 15 Drosophila virilis strains. One common replacement polymorphism is observed, where a negatively charged aspartic amino acid is replaced by the noncharged amino acid alanine. This replacement variant is located within the serine/threonine kinase domain of the fused gene and is present in ~50% of the sequences in our sample. Significant linkage disequilibrium is detected around this replacement site, although the fused gene is located in a region of the D. virilis X chromosome that seems to experience normal levels of recombination. In a 600-bp region around the replacement site, all eight alanine sequences are identical; of the six aspartic acid sequences, three are also identical. The occurrence of little or no variation within the aspartic acid and alanine haplotypes, coupled with the presence of several differences between them, is very unlikely under the usual equilibrium neutral model. Our results suggest that the fused alanine haplotypes have recently increased in frequency in the D. virilis population.

The 210Pb dating method has been applied successfully to the determination of recent sedimentation rates at four sites distributed among the three major sedimentary basins (Niagara, Mississauga and Rochester) of Lake Ontario. Following correction for effects due to compaction of the sediments, mean sedimentation rates ranging from 0.02 cm/year at the periphery of the Mississauga basin to 0.11 cm/year in the Niagara and Rochester basins were determined. Allowance for compaction reduced the non-compaction-corrected sedimentation rates by 20–35%. Neither 210Pb nor fallout 137Cs profiles indicated surface mixing of sediment sufficient to noticeably affect the calculated sedimentation rates. At all four sites, the sedimentation rate seems to have remained constant during the last 100–150 years.

To study the relationship between the primary structure of transforming growth factor alpha (TGF-alpha) and some of its functional properties (competition with epidermal growth factor (EGF) for binding to the EGF receptor and induction of anchorage-independent growth), we introduced single amino acid mutations into the sequence for the fully processed, 50-amino-acid human TGF-alpha. The wild-type and mutant proteins were expressed in a vector by using a yeast alpha mating pheromone promoter. Mutations of two amino acids that are conserved in the family of the EGF-like peptides and are located in the carboxy-terminal part of TGF-alpha resulted in different biological effects. When aspartic acid 47 was mutated to alanine or asparagine, biological activity was retained; in contrast, substitutions of this residue with serine or glutamic acid generated mutants with reduced binding and colony-forming capacities. When leucine 48 was mutated to alanine, a complete loss of binding and colony-forming abilities resulted; mutation of leucine 48 to isoleucine or methionine resulted in very low activities. Our data suggest that these two adjacent conserved amino acids in positions 47 and 48 play different roles in defining the structure and/or biological activity of TGF-alpha and that the carboxy terminus of TGF-alpha is involved in interactions with cellular TGF-alpha receptors. The side chain of leucine 48 appears to be crucial either indirectly in determining the biologically active conformation of TGF-alpha or directly in the molecular recognition of TGF-alpha by its receptor.

Abstract Florunner peanut seeds contained five trypsin isoinhibitors. Amino acid profiles of the trypsin inhibitors fraction showed high levels of aspartic acid, half-cystine and serine and low levels of histidine and tyrosine. The molecular weight of the inhibitor was 8.3 KDa. The presence of multiforms of this inhibitor, its low molecular weight and the high amount of half-cystine indicate that peanut trypsin inhibitor is of the Bowman-Birk type.

Environmental conditions of the lowland tropical forests during the last glacial maximum (LGM) between ca 20,000 and 18,000 14C yr B.P., are reevaluated in terms of dating control and lithology analyzed in seven pollen records from South America. The reevaluation shows that probably in none of the published records are LGM sediments present or abundant. This conclusion is based on the occurrence of abrupt lithologic changes coupled with changes in sedimentation rate interpolated from radiocarbon dates. These findings suggest that the LGM was represented probably by a hiatus of several thousand years, indicative of drier climates than before or after.

ABSTRACT Amino acid substitutions in Escherichia coliς70 were generated and characterized in an analysis of the role of region 1.1 in transcription initiation. Several acidic and conserved residues are tolerant of substitution. However, replacement of aspartic acid 61 with alanine results in inactivity caused by structural and functional thermolability.
