Chromosomal regions which control sporulation in Bacillus subtilis

Large quantities of sporulation mutants have been isolated with a variety of mutagens. The genetic sites for asporogeny have been localized on the chromosome of Bacillus subtilis through transduction with phage PBSI. Through these procedures specific portions of the chromosome which are associated with sporulation have been identified. Although asporogenic (Sp−) defects were observed to be scattered throughout the four linkage groups of the genetic map of B. subtilis, only three extensive Sp− linkage groups were identified. The first linkage group of Sp− markers is located at the proximal end of the chromosome between the cys A and ery markers. The second cluster of spore genes mapped to the right of ura, and the third linkage group of spore markers mapped to the left of lys-2. Defects within specific regions of the first and third spore gene clusters obstructed some early products of sporogenesis.

Abstract The ease of isolating mutations in zebrafish will contribute to an understanding of a variety of processes common to all vertebrates. To facilitate genetic analysis of such mutations, we have identified DNA polymorphisms closely linked to each of the 25 centromeres of zebrafish, placed centromeres on the linkage map, increased the number of mapped PCR-based markers to 652, and consolidated the number of linkage groups to the number of chromosomes. This work makes possible centromere-linkage analysis, a novel, rapid method to assign mutations to a specific linkage group using half-tetrads.

SUMMARYEight newly isolated 9 + 0 mutants each mapped at one of the four previously known loci. Short flagellum mutants were at three loci, two of which (pf7 and pf8) were closely linked; the third, pf21, was unlinked to these two and mapped on linkage group II. The long flagellum mutants lf1 and lf2 were on linkage groups II and XII respectively. Mutants pf8A and lf1 were both recessive to wild-type. There was no evidence for non-Mendelian flagellum mutants.

ABSTRACT Two dominant suppressors of crossing over have been identified following X-ray treatment of the small nematode C. elegans. They suppress crossing over in linkage group II (LGII) about 100-fold and 50-fold and are both tightly linked to LGII markers. One, called C1, segregates independently of all other linkage groups and is homozygous fertile. The other is a translocation involving LGII and X. The translocation also suppresses rrossing over along the right half of X and is homozygous lethal. CI has been used as a balancer of LGII recessive lethal and sterile mutations induced by EMS. The frequencies of occurrence of lethals and steriles were approximately equal. Fourteen mutations were assigned to complementation groups and mapped. They tended to map in the same region where LGII visibles are clustered.

A small reciprocal translocation T(1;2)1 involving chromosomes 1 and 2 and a paracentric inversion In(1)3 on m chromosome (1) of A. aegypti interact to give peculiar but consistent crossover values. The system is termed COSES and is associated with partial sterility. In females it suppresses crossing over tremendously to the right of bz and enhances crossing over to its left. In the males it enhances crossing over to the right of m (only 3 crossover units away from bz) hut the region to its left remains unaffected. COSES also displays interchromosomal effects by enhancing crossing over in linkage group III. Cytological and genetic evidence for the presence of translocation and inversion are presented. All three pairs of chromosomes are correlated to the three linkage groups.

The inheritance and linkage group assignments were determined for the two enzyme loci, glutamate oxaloacetate transaminase (Got-I) and alcohol dehydrogenase (Adh-I). of Anopheles albimanus Weidemann. Got-I was assigned to the proximal position on the genetic map of the right arm of chromosome 2, and Adh-I was mapped at the distal position on the map of the right arm of chromosome 3. Gene sequences and linkage estimates are now available for 23 mutant and enzyme loci in A. albimanus.Key words: glutamate oxaloacetate transaminase, alcohol dehydrogenase, isozyme, Anopheles albimanus, genetic map.

In Glossina morsitans morsitans Westwood, linkage group III is defined as the autosome carrying the locus Mdh (malic dehydrogenase), the only locus so far identified in this linkage group. The locus αGpd.2 (α-glycerophosphate dehydrogenase) was located 45 map units (MU) to the left of Xo (xanthine oxidase) and the loci Est.1 and Est.2 (loci for two esterases found in the thorax) were mapped approximately 5–10 MU to the right of Ao (aldehyde oxidase) in linkage group II. The location of G6pd (glucose-6-phosphate dehydrogenase) has been confirmed to be approximately 37 MU to the left of oc (ocra body color) in linkage group I and it was shown that this region of the X chromosome does not involve the large paracentric inversion found in the Handeni line. A genetic map for 12 loci in the three linkage groups found in G. m. morsitans is presented.Key words: Diptera, Glossina, mapping, inversion, isozymes.

Objective: Erwinia amylovora causes fire blight disease in Malus. A strong resistance QTL (Mfu10) was previously detected on linkage group 10 of Malus fusca accession MAL0045, using several strains of the bacterium. As no strain capable of breaking the resistance of MAL0045 has been found, it was hypothesized that a second resistance factor contributes to the fire blight resistance of MAL0045. However, to date, no minor locus has been detected with previously published strains of the bacterium. We detected a minor QTL only on a subset of a population following inoculation with strain Ea1038, which heterologously expresses an effector in a derivative of isolate Ea3049. Two genetic maps of MAL0045, one scarce, the other dense with markers, were used for QTL analyses. Results: Mfu10 was detected on LG10 with Ea1038, as was previously with Ea3049. Although no other QTLs of significant LOD was previously detected in other linkage groups with Ea3049, a QTL of significant LOD was detected on LG16 (Mfu16) after inoculation of a subset of 76 individuals with Ea1038, but only using the dense genetic map. Mfu16 improved the effect of Mfu10. However, when the number of individuals inoculated with Ea1038 was increased to 121, Mfu16 was no longer detected in the dense genetic map. We hypothesize some factors, which might be responsible for the instability of this QTL.

A 55-year-old woman was referred because of diffuse pruritic erythematous lesions and an ischemic process of the third finger of her right hand. She was known to have anaemia secondary to hypermenorrhea. She presented six months before admission with a cutaneous infiltration on the left cubital cavity after a paravenous leakage of intravenous iron substitution. She then reported a progressive pruritic erythematous swelling of her left arm and lower extremities and trunk. Skin biopsy of a lesion on the right leg revealed a fibrillar, small-vessel vasculitis containing many eosinophils.Two months later she reported Raynaud symptoms in both hands, with a persistent violaceous coloration of the skin and cold sensation of her third digit of the right hand. A round 1.5 cm well-delimited swelling on the medial site of the left elbow was noted. The third digit of her right hand was cold and of violet colour. Eosinophilia (19 % of total leucocytes) was present. Doppler-duplex arterial examination of the upper extremities showed an occlusion of the cubital artery down to the palmar arcade on the right arm. Selective angiography of the right subclavian and brachial arteries showed diffuse alteration of the blood flow in the cubital artery and hand, with fine collateral circulation in the carpal region. Neither secondary causes of hypereosinophilia nor a myeloproliferative process was found. Considering the skin biopsy results and having excluded other causes of eosinophilia, we assumed the diagnosis of an eosinophilic vasculitis. Treatment with tacrolimus and high dose steroids was started, the latter tapered within 12 months and then stopped, but a dramatic flare-up of the vasculitis with Raynaud phenomenon occurred. A new immunosupressive approach with steroids and methotrexate was then introduced. This case of aggressive eosinophilic vasculitis is difficult to classify into the usual forms of vasculitis and constitutes a therapeutic challenge given the resistance to current immunosuppressive regimens.

Choice of profession is a social phenomenon that every person has to face in life. Numerous studies convince us that not only the well-being of a person depends on the chosen work, but also his attitude to himself and life in general, therefore, the right and timely professional choice is very important. Research about factors of career self-determination of students of higher education institutions in Ukraine shows that self-determination is an important factor in the socialization of young person, and the factors that determine students' career choices become an actual problem of nowadays. The present study involved full-time and part-time students of Institute of Philology and Mass Communications of Open International University of Human Development "Ukraine" in order to examine the factors of career self-determination of students of higher education institutions (N=189). Diagnostic factors of career self-determination of students studying in the third and fourth year were carried out using the author's questionnaire. Processing of obtained data was carried out using the Excel 2010 program; factorial and comparative analysis were applied. Results of the study showed that initial stage of career self-determination falls down on the third and fourth studying year at the university, when an image of future career and career orientations begin to form. At the same time, the content of career self-determination in this period is contradictory and uncertain, therefore, the implementation of pedagogical support of this process among students is effective.

ABSTRACT Parental perception will affect the fulfillment of children’s participation rights. Fullfilment of children’s participation rights will be fulfilled optimally if parents pay anttention to opinions while providing opportunities for children to make and make decisions about the child’s goals and self-interest. The subjects studied consisted of 5 subjects consisting of father and mother who had children aged 9- 12 years. This study uses qualitative research methods, with data retrieval tools in teh form of interviews, observation and documentation. From the research found data were the subjects do not fulfill the right of participation of children up to the maximum ladder where children’s participation rights range from the first ladder to the third ladder. The first ladder to the third ladder is actually a non- participating ladder. This means that children is manipulated, dominated by parents, there is direct communation and the severity of the parent. The children felt disappointed, sad, and angry with the parents but they still tried to hear and obey the parent’s decision. Children from third and fourth subjects experienced excessive fear to speak to their parent (father). Suggestions for parents to be more caring and fulfill the rights of children’s participation so as not to affect the growth and development of children.
