The antenna model used for correcting the influence of the antenna pattern on synthetic aperture radar (SAR) images requires on-ground validation and in-flight verification. A methodology for the in-flight verification that is based upon the measurement of azimuth antenna patterns using ground receivers has been successfully demonstrated for the operational SAR modes of the TerraSAR-X (TSX) and TanDEM-X (TDX) missions. Recently, the novel (terrain observation by progressive scans) TOPS mode was for the first time implemented as an experimental mode on TerraSAR-X to demonstrate its feasibility in support of its implementation on ESA's Sentinel-1 mission. In this mode, besides scanning in elevation, the antenna beam is steered in flight direction from aft to the front at a constant rate to achieve an enhanced radiometric image performance. This paper discusses the methodology and presents results of the first in-flight antenna characterization of a SAR instrument operating in TOPS mode, in this case TerraSAR-X, using ground receivers. The results demonstrate that the TOPS one-way azimuth antenna pattern can be accurately modeled by the TSX antenna model indicating the general suitability of this approach for the in-flight antenna model verification during TOPS mode operations.

ABSTRACT Borna disease virus (BDV), the causative agent of severe meningoencephalitis in a wide variety of animal species, has been considered to be genetically invariable and to form a single type within the genus Bornavirus of the familyBornaviridae. BDV infections are of particular interest, because for the first time a virus infection appears to be linked to human psychiatric disorders. We now describe a new subtype of BDV isolated from a horse which was euthanatized due to severe, incurable neurological disease. The nucleotide sequence of this new strain, named No/98, differs from the reference strains by more than 15%, and the subtype is difficult to detect by standard reverse transcriptase PCR protocols. The nucleotide exchanges of the novel BDV isolate have surprisingly little effect on the primary structures of most viral proteins, with the notable exception of the X protein (p10), which is only 81% identical to its counterpart in reference strains. Our data indicate that the genome of BDV is far more variable than previously assumed and that naturally occurring subtypes may escape detection by currently used diagnostic assays.

Proper mitochondrial genome inheritance is important for eukaryotic cell survival. Trypanosoma brucei, a protozoan parasite, contains a singular mitochondrial genome, the kDNA. The kDNA is anchored to the basal body via the tripartite attachment complex (TAC) to ensure proper segregation. Several components of the TAC have been described. However, the connection of the TAC to the kDNA remains elusive. Here, we characterize the TAC associated protein TAP110. Depletion as well as overexpression of TAP110 leads to a delay in the separation of the replicated kDNA networks. Proteome analysis after TAP110 overexpression identified several kDNA associated proteins including a TEX-like protein that dually localizes to the nucleus and the kDNA potentially linking replication/segregation in the two compartments. The assembly of TAP110 into the TAC region seems to require the TAC but not the kDNA itself, however once TAP110 has been assembled it also interacts with the kDNA. Finally, for the first time we use ultrastructure expansion microscopy in trypanosomes to reveal the precise position of TAP110 between TAC102 and the kDNA, showcasing the potential of this approach.

AbstractProper mitochondrial genome inheritance is key for eukaryotic cell survival, however little is known about the molecular mechanism controlling this process. Trypanosoma brucei, a protozoan parasite, contains a singular mitochondrial genome aka kinetoplast DNA (kDNA). kDNA segregation requires anchoring of the genome to the basal body via the tripartite attachment complex (TAC). Several components of the TAC as well as their assembly have been described, it however remains elusive how the TAC connects to the kDNA. Here, we characterize the TAC associated protein TAP110 and for the first time use ultrastructure expansion microscopy in trypanosomes to reveal that TAP110 is the currently most proximal kDNA segregation factor. The kDNA proximal positioning is also supported by RNAi depletion of TAC102, which leads to loss of TAP110 at the TAC. Overexpression of TAP110 leads to expression level changes of several mitochondrial proteins and a delay in the separation of the replicated kDNA networks. In contrast to other kDNA segregation factors TAP110 remains only partially attached to the flagellum after DNAse and detergent treatment and can only be solubilized in dyskinetoplastic cells, suggesting that interaction with the kDNA might be important for stability of the TAC association. Furthermore, we demonstrate that the TAC, but not the kDNA, is required for correct TAP110 localization in vivo and suggest that TAP110 might interact with other proteins to form a >669 kDa complex.Summary StatementTAP110 is a novel mitochondrial genome segregation factor in Trypanosoma brucei that associates with the previously described TAC component TAC102. Ultrastructure expansion microscopy reveals its proximal position to the kDNA.

The roots, leaves and stems of Christia vespertilionis were separately and successively extracted with methanol and aqueous-methanol (1:4, v/v) and were evaluated in vitro for their antiplasmodial potential against Plasmodium falciparum NF-54. The aqueous-methanolic stem (AS) extract was the most active (IC50 7.5 μg/mL) followed by the methanolic leaf (ML) extract (IC50 32.0 μg/mL). The in vivo antimalarial activity of the combined plant extract of C. vespertilionis was also assessed in P. berghei infected mice, which showed 87.8% suppression of parasitaemia as compared with complete suppression by chloroquine on day 8. Finally, detailed chemical investigation of C. vespertilionis resulted in the isolation and characterization of fifteen compounds (1–15), of which two (1 and 4) are being reported for the first time from nature. The novel compound 1 possesses potent antiplasmodial activity (IC50 = 9.0 μg/mL).

This work represents an innovative study that, for the first time, explores the possibility to use waste flours to produce thermoplastic polymeric bio-films. To the best of our knowledge, this is the first time that waste flours, derived from bakeries, pizzerias or pasta factories, have been proposed for the production of bio-polymers, as a replacement of neat starch. To this aim, durum waste flour derived from a pasta factory, soft waste flour derived from pizzerias and neat maize starch used as control material were firstly analyzed from dimensional, morphological and chemical points of view. Afterwards, waste flour films were produced by the addition of a nature-based plasticizer, glycerol. Mechanical characterization of the plasticized thermoplastic films, produced by compression molding, evidenced low performances, even in the case of the neat maize starch. In order to improve the mechanical properties, the possibility to include polylactic acid and cardanol-based plasticizer was also investigated. Mass transport properties of all the produced bio-films were investigated by measuring their water vapor permeability and hygroscopic absorption. The durability properties of the bio-films were assessed by accelerated ageing tests, while the bio-degradability of the waste-based films was evaluated by measuring the solubility and the degradation in water. The physicochemical analyses of the novel bio-films evidenced good mechanical properties; specifically, the waste-based films showed a lower hygroscopic absorption and water solubility than those of the blends containing neat starch.

Rotavirus A (RVA) has been considered the main cause of diarrheal disease in children under five years in emergency services in both developed and developing countries. RVA belongs to the Reoviridae family, which comprises 11 segments of double-stranded RNA (dsRNA) as a genomic constellation that encodes for six structural and five to six nonstructural proteins. RVA has been classified in a binary system with Gx[Px] based on the spike protein (VP4) and the major outer capsid glycoprotein (VP7), respectively. The emerging equine-like G3P[8] DS-1-like strains reported worldwide in humans have arisen an important concern. Here, we carry out the complete genome characterization of a previously reported G3P[8] strain in order to recognize the genetic diversity of RVA circulating among infants in Colombia. A near-full genome phylogenetic analysis was done, confirming the presence of the novel equine-like G3P[8] with a Wa-like backbone for the first time in Colombia. This study demonstrated the importance of surveillance of emerging viruses in the Colombian population; furthermore, additional studies must focus on the understanding of the spread and transmission dynamic of this important RVA strain in different areas of the country.

The article is dedicated to the analysis of the genres' eclecticism in the novel by Viktoria Hranetska "The body©". The purpose of an article is to find out the specifics of the genre on the base of the novel, the studying of the plot and compositional elements according to the postmodern discourse, the finding out the correlation between the different directions of the fantastic genre in the structure of the novel. Research methods In the context of studying the literary work, we used the basic principles of mythological, comparative-historical, psychological and typological methods. Results. The article deals with the genre and plot-compositional features of the novel "The body©", found out its leading motives, ways of representing them in the text and also postmodern features. We were able to identify the genres in which the text was created, and how they manifest themselves on the plot-compositional level. Scientific novelty. The author explores the genesis of science fiction and its kinds on the material of the novel and also gives the characterization of the genres of science fiction, fantasy and horror. The author studies deeply the leading motifs of the horror literature – the motives the mettempsychosis with all its modifications on the example of the works of fiction of Ukrainian and world literatures for the first time. The article thoroughly analyzes the novel "The body©" as a postmodernist work of fiction wich have all features of postmodernism, in particular eclectic genres. Practical significance. The main principles of research can be used for a deeper understanding of the characteristic features of the fantastic sub-genres, the ability to distinguish them from the general canvas of science fiction.

Retrieval of Sun-Induced Chlorophyll Fluorescence (F) spectrum is one of the challenging perspectives for further advancing F studies towards a better characterization of vegetation structure and functioning. In this study, a simplified Spectral Fitting retrieval algorithm suitable for retrieving the F spectrum with a limited number of parameters is proposed (two parameters for F). The novel algorithm is developed and tested on a set of radiative transfer simulations obtained by coupling SCOPE and MODTRAN5 codes, considering different chlorophyll content, leaf area index and noise levels to produce a large variability in fluorescence and reflectance spectra. The retrieval accuracy is quantified based on several metrics derived from the F spectrum (i.e., red and far-red peaks, O2 bands and spectrally-integrated values). Further, the algorithm is employed to process experimental field spectroscopy measurements collected over different crops during a long-lasting field campaign. The reliability of the retrieval algorithm on experimental measurements is evaluated by cross-comparison with F values computed by an independent retrieval method (i.e., SFM at O2 bands). For the first time, the evolution of the F spectrum along the entire growing season for a forage crop is analyzed and three diverse F spectra are identified at different growing stages. The results show that red F is larger for young canopy; while red and far-red F have similar intensity in an intermediate stage; finally, far-red F is significantly larger for the rest of the season.
