The total oil yield and the fatty acid composition were determined in theAnnona muricataL. fixed oil using organic solvent extraction and GC-FID. The seeds were found to contain about ~21.5% of crude fixed oil on a dry weight basis. The crude oil containing fatty acid was converted into methyl esters and analysed by GC-FID. Fourteen fatty acids were identified using GC-FID. The major monounsaturated and saturated fatty acids were oleic acid (39.2%) and palmitic acid (19.1–19.2%), respectively, whereas theα-linolenic acid (1.2%) and linoleic acid (34.9%) were polyunsaturated fatty acid. The other saturated acids were stearic acid (3.3%), arachidic acid (0.4%), myristic acid (0.1%), heptadecanoic acid (0.1%), behenic acid (0.1%), and lignoceric acid (0.1%). Some of the fatty acids have not been reported earlier from the oil ofAnnona muricataL. Fixed oil exhibited significant free radical scavenging activity which was measured using DPPH and is also known to inhibit the gastrointestinal motility significantly.

The compositions and concentrations of lipid classes, fatty acids, tocopherols were determined in the lipids from blue-green microalga <i>Spirulina platensis</i>. Total lipids (TL) recovered using chloroform: methanol (2:1, v/v) were found to be 163.5 g/kg (on dry weight basis). The level of neutral lipids was the highest, followed by glycolipids and phospholipids, respectively. Among TL and lipid classes, palmitic, &gamma;-linolenic and linoleic acids were the dominating fatty acids. Compared to the neutral lipids, the polar fractions were generally characterised by higher percentages of saturated fatty acids. The recovered lipids were characterised by high percentage of tocopherols, wherein &gamma;-tocopherol constitutes about 73% of total tocopherols present, the rest being &gamma;-tocopherol. <i>Spirulina platensis</i> lipids exhibited a strong radical scavenging activity towards stable DPPH free radicals, whereas 27% of DPPH radicals were quenched after 2 h incubation. TL and lipid classes inhibited the growth of different microorganisms except gram-negative bacteria. At high concentrations, the tested lipids appeared more effective against <i>A. niger</i> (28.3 ± 1.53 mm). The information obtained in the present investigation is useful for lipid characterisation and further chemical and nutritional investigations of <i>Spirulina platensis</i>.

Petroleum reserves have been decreasing in recent years and microalgae are attractive as a potential source of new biomass petroleum. Microalgae are unicellar microscopic algae and most species microalgae produce lipids. In particular, Botryococcus braunii produces large amount of lipids found with nearly 70% on the basis of the dry weight. This chapter reviews high lipid-producing microalgae found from Okinawa area around National Institute of Technology, Okinawa College (NIT, Okinawa). The microalgae collected were isolated on an AF-6 agar plates, and incubated in AF-6 medium. The fatty acids were extracted from the algae, converted into fatty acid methyl esters, and analysed by GC/MS. As a result, two microalgae strains were identified that the produced fatty acids was loaded in the algae with nearly 20% in the dry weight base. In addition, these two microalgae strains produced palmitic acid as nearly 40% of the total produced lipids. Therefore, the two microalga strains isolated are potentially and highly efficient for the organisms applied for the production of biodiesel fuel.

Fatty oils of the seeds of Cleome viscosa accessions from Delhi, Jaipur, Faridabad, Surajkund and Hyderabad were methylated and analyzed by GC and GC-MS. The major fatty acids, identified as their methyl esters, of the oils from these five locations were palmitic acid (10.2-13.4%), stearic acid (7.2-10.2%), oleic acid (16.9-27.1%) and linoleic acid (47.0-61.1%). In addition, palmitoleic acid, octadec-(11 E)-enoicacid, arachidic acid, eicosa-(11 Z)-enoic acid, linolenic acid, heneicosanoic acid, behenic acid, lignoceric acid, pentacosanoic acid, hexacosanoic acid, 12-oxo-stearic acid, and the alkanes tetracosane, pentacosane, hexacosane, heptacosane, octacosane, nonacosane, triocontane, hentriacontane and dotriacontane, were also identified as minor and trace constituents in some of these oils.

Abstract Objectives Saturated branched chain fatty acids (BCFA) terminating with either an isopropyl or sec-butyl group, are common bioactive food components consumed from beef, fish, and dairy products. Little is known about their endogenous metabolism and the enzymes mediating their interconversion. Our main objective was to test the substrate specificity of the fatty acid elongases (ELOVL1–7) towards elongation of representative BCFA, anteiso-15:0 and iso-18:0. We also assessed competition between BCFA and normal saturated fatty acid (n-SFA). Methods MCF7 human breast cancer cells are used for functional studies. PcDNA3.1 expression vector was used to clone seven open reading frames of ELOVL transcripts (ELOVL1–7). MCF7 cells were transiently transfected with specific ELOVL1–7 transgene vector or empty vector (control). After 24 h incubation, the transfected MCF7 cells were treated with BSA-bound substrates. After additional 24 h incubation, cells were harvested by trypsinization, fatty acid methyl esters prepared and analyzed quantitatively by GC-FID. Fatty acid composition was characterized by gas chromatography (GC) –electron ionization mass spectrometry (EIMS) and EIMS/MS for branched carbon chain. Results Transient transfection of ELOVL1–7 into MCF7 cells show that ELOVL6 had highest activity towards elongation of anteiso-15:0→anteiso-17:0, followed by ELOVL5 which showed moderate activity. ELOVL3 was found to be most active ELOVL mediating elongation of iso-18:0→iso-20:0. Our competition results show in ELOVL6 cells anteiso-15:0 competes with n-SFA n-16:0, whereas, in ELOVL3 cells iso-18:0 competes with n-18:0. Conclusions The elongation of anteiso-15:0 and iso-18:0 BCFA is predominantly operated by the ELOVL6 and ELOVL3, respectively that are well known to operate on normal saturated fatty acids, rather than the ELOVL2 and ELOVL5 which operate on lower-melting polyunsaturated fatty acids. The competition between BCFA and n-SFA for ELOVL1–7 mediated elongation may have implications in the skin, sebaceous, and meibomian glands where both normal and BCFA are present at comparable levels. AbstractObjective:The aim of the study was to investigate quantitative and qualitative fatty acid profiles of the gonad and liver tissue of femaleMethods:Total lipids were extracted with 5 mL of chloroform-methanol (2:1 v/v). Samples containing gonad and liver lipid were transesterified with acidified methanol. The fatty acid methyl esters were extracted with hexane. Fatty acids were detected by gas chromatography (GC).Results:The major saturated fatty acids (SFAs) were myristic acid (C14:0), palmitic acid (C16:0) and stearic acid (C18:0) in both gonad and liver tissue. Oleic acid (C18:1 ω-9) and palmitoleic acid (C16:1 ω-7) were the prominent monounsaturated fatty acid (MUFA). The dominant polyunsaturated fatty acids (PUFAs) were linoleic acid (LA, C18:2 ω-6), arachidonic acid (AA, C20:4 ω-6), docosapentaenoic acid (C22:5 ω-3) and docoesahexaenoic acid (DHA, C22:6 ω-3). The ratio of ω-3/ω-6 ranged from 1.55 to 3.44 and 1.18 to 2.71 in the gonad and liver tissue, respectively.Conclusion:The findings of this study will be useful for understanding the seasonal distribution of fatty acid composition in the liver and gonad of spiny eel.

Sorghum and millet are important food staples in semi-arid tropics of Asia and Africa. Sorghum and millet are cereal grains that have prospective to be used as substitute to wheat flour for celiac patients. These are considered as the good source of many important and essential fatty acids. The volatile profiling of these two important crops is comparable to other cereals as well. The present study was an effort to explore biochemical composition of commercially available sorghum and millet varieties with special reference to their fatty acid and volatile profiling. Chemical composition of sorghum and millet was determined according to respective methods. Fatty acid methyl esters were prepared and then subjected to GC-FID for fatty acids analysis. The results indicated that both sorghum and millet oils are rich in essential fatty acids comprising mono and polyunsaturated fatty acids. Main fatty acids that are identified in current study includes palmitic acid, oleic acid, palmitoleic acid, behenic acid, linoleic acid, linoleic acid, stearic acid, myristic acid, etc. On the other hand volatile compounds from sorghum and millet were determined by preparing their respective volatile samples by using calvenger apparatus with suitable volatile extracting solvent. Volatile samples were then subjected to GC-MS analysis and respected results were compared with NIST library. About 30 different volatiles were identified in millet varieties while 35 different compounds were discovered in sorghum varieties belonging to aldehydes, ketones, benzene derivatives, esters, alcohols, sulphur compounds.

Physical properties of commonly grown Sri Lanka groundnuts cultivars and promising accession varied considerably and numbers of kernels, pod beak, reticulation, testa colour, and shell out percentage have differences among groundnuts. However, they showed more similarities for most of the characters. Moisture (5.4-8.4%), crude protein (18.7-28.5%), lipid (43.4-53.0%), ash (4.4-5.8%), carbohydrates (9.3-18.2%) and energy level (565.7-618.2kcal) contents varied considerably. Quality and flavor of edible groundnuts and its products are affected by fatty acid composition of oil. Lipids were mainly composed of mono and polyunsaturated fatty acids (>78% of the total lipids). Fatty acid composition analysis indicated that oleic acid (C18:1) was the main constituent of monounsaturated lipids in all seed samples. With the exception of ANKG1, linoleic acid (C18:2) was the major polyunsaturated fatty acid. The saturated fatty acids (Palmatic, Stearic acid and behenic acid) in different cultivars ranged between 10.2 to 15.6%, 2.5 to 6.3% and 1.1 to 5.3%, respectively. Differences among cultivars for oleic acid exhibited significance which ranged between 38.2 to 47.4%. Similarly, cultivars differed statistically for linoleic acid which showed a range of 23.1 to 38.7%. Oleic to linoleic acid ratio was differed and all the released varieties were below the minimum standard level of 1.6, whereas ICGV 86590 and ICGV 00073 showed higher O/L ratio of 1.94 and 1.75 respectively.

The aims of the present study were to: (1) characterise the metabolome of follicular fluid and serum in dairy cows with similar genetic merit for milk production but with extremes of good (Fert+) or poor (Fert–) genetic merit for fertility; and (2) identify potential biomarkers of dairy cow fertility. Follicular fluid from the first wave dominant follicle and serum were collected on Day 7 of the oestrous cycle. The most pronounced effect of genotype was noted in the serum, where the abundance of total polyunsaturated fatty acids and n-6 polyunsaturated fatty acids was greater in Fert+ cows, and the abundance of total saturated fatty acids was greater in Fert– cows. The abundance of nine fatty acids (arachidic acid, heneicosanoic acid, myristic acid, behenic acid, myristoleic acid, heptadecenoic acid, cis-11-eicosanoic acid, nervonic acid and γ-linolenic acid) in follicular fluid was affected by genotype. Concentrations of cysteine, leucine, ornithine, proline and tyrosine in follicular fluid, and asparagine, creatinine, cysteine, methionine, proline and valine in serum, were also affected by genotype. Receiver operating characteristic curve analysis indicated that the follicular fluid and serum fatty acids and follicular fluid amino acids that were significantly affected by genotype were highly predictive of fertility genotype.

ABSTRACT Background Very-long-chain SFAs (VLCSFAs), such as arachidic acid (20:0), behenic acid (22:0), and lignoceric acid (24:0), have demonstrated inverse associations with cardiometabolic conditions, although more evidence is needed to characterize their relation with risk of type 2 diabetes (T2D). In addition, little is known regarding their potential dietary and lifestyle predictors. Objective We aimed to examine the association of plasma and erythrocyte concentrations of VLCSFAs with incident T2D risk. Methods We used existing measurements of fatty acid concentrations in plasma and erythrocytes among 2854 and 2831 participants in the Nurses’ Health Study (NHS) and Health Professionals Follow-Up Study (HPFS), respectively. VLCSFAs were measured using GLC, and individual fatty acid concentrations were expressed as a percentage of total fatty acids. Incident T2D cases were identified by self-reports and confirmed by a validated supplementary questionnaire. Cox proportional hazards regression was used to evaluate the association between VLCSFAs and T2D, adjusting for demographic, lifestyle, and dietary variables. Results During 39,941 person-years of follow-up, we documented 243 cases of T2D. Intakes of peanuts, peanut butter, vegetable fat, dairy fat, and palmitic/stearic (16:0–18:0) fatty acids were significantly, albeit weakly, correlated with plasma and erythrocyte VLCSFA concentrations (|rs| ≤ 0.19). Comparing the highest with the lowest quartiles of plasma concentrations, pooled HRs (95% CIs) were 0.51 (0.35, 0.75) for arachidic acid, 0.43 (0.28, 0.64) for behenic acid, 0.40 (0.27, 0.61) for lignoceric acid, and 0.41 (0.27, 0.61) for the sum of VLCSFAs, after multivariate adjustments for demographic, lifestyle, and dietary factors. For erythrocyte VLCSFAs, only arachidic acid and behenic acid concentrations were inversely associated with T2D risk. Conclusions Our findings suggest that, in US men and women, higher plasma concentrations of VLCSFAs are associated with lower risk of T2D. More research is needed to understand the mechanistic pathways underlying these associations.
