Staphylococcus aureus is a contagious pathogen that can cause various diseases in both humans and animals. Antimicrobial-resistant S. aureus is becoming an extremely important global health problem. A cross-sectional study was conducted from December 2019 to May 2020 to assess the occurrence of S. aureus and its antimicrobial susceptibility profiles in milk and traditionally processed dairy products in selected subcities of Addis Ababa. A total of 255 dairy product samples (175 raw milk and 80 traditionally processed dairy products) were collected from farms and retail markets. Samples were cultured for S. aureus according to standard microbiology techniques, and the Kirby–Bauer disk diffusion method was used to assess antimicrobial susceptibility of isolates to a panel of 12 antimicrobials. Susceptibility to methicillin was determined based on the sensitivity of isolates to cefoxitin, and resistant isolates were investigated for the presence of mecA and mecC genes using PCR. Staphylococcus aureus was isolated from 43 (24.6%) of milk, 7 (17.5%) of yogurt, and 2 (5%) of cottage cheese. A significantly higher rate of contamination with S. aureus was recorded among milk samples compared to yogurt and cottage cheese ( p = 0.019 ). Out of 52 S. aureus isolates investigated for susceptibility to 12 antimicrobials, 49 (94.2%) of the isolates were resistant to ampicillin and 42 (80.8%) to amoxicillin+clavulanic acid. Twenty (38.5%) of the isolates were methicillin-resistant S. aureus (MRSA) based on susceptibility to cefoxitin. However, only one of these isolates (5%) was positive for mecA gene, and none of them were positive for the mecC gene. There was no significant difference ( p > 0.05 ) in the rate of occurrence of MRSA among isolates from different sources. In conclusion, this study demonstrated a significant level of contamination of milk and dairy products with S. aureus and most isolates were multidrug resistant. The occurrence of MRSA in raw milk and dairy products signifies a serious public health threat as the practice of consuming raw dairy products in the study area is widespread. The lack of agreement between phenotypic and genotypic detection of MRSA suggests the need for further study to identify the genetic basis for the observed resistance phenotype.

This research aimed to evaluate Staphylococcus aureus contamination from milk and its derivatives available in local dairy sale centers around the Kabul University located in Kabul city. Samples were included caw`s raw and pasteurized milk, cheese, ice cream, yogurt, custard, and dough (native product). The mentioned products were collected and then using the conventional method was tested in the laboratory. 70 samples were tested, from tested only 18 samples were presented Staphylococcus aureus, as these isolates had a positive reaction for coagulase. The disk diffusion method plus 8 antibiotics which are commonly used in the Veterinary section were used for antimicrobial sensitivity reaction. Staphylococcus isolated samples showed resistance against more than 3 antibiotics, maximum resistance was to Nalidixic acid (50%) followed by Ciprofloxacin (44.44%), Norfloxacin (27.77%), Erythromycin and Methicillin (27.77%), and Tetracycline (22.22%). All the isolates susceptibility was high to Polymaxine and resistant to Ampicillin (100%). As we found in our study the biological quality of tested samples sold around Kabul University were notsatisfactory as well as resistance against the mentioned antibiotics except two antibiotics. The contaminated products could be a potential risk to humans .

Prevention of foodborne diseases is a priority for the world health system. In the process of manufacturing milk and dairy products, the most important factor endangering their safety is seeding with a conditionally pathogenic and pathogenic microflora. Salmonella, Escherichia coli, Listeria and other microorganisms that reproduce in dairy products without changing their organoleptic properties are a particular danger. Staphylococcus aureus is an opportunistic, conditionally pathogenic microorganism that often contaminates raw milk and dairy products. The aim of the research presented in this article was to determine the dissemination of S. aureus in milk and milk products of household production in the western regions of Ukraine, to identify the biotypes of S. aureus, production of enterotoxins and the presence of methicillin-resistant strains. S. aureus was isolated on BD Baird-Parker Agar. The biotypes of S. aureus were determined according to Meer. The determination of MRSA was carried out on the chromogenic Agar chromID MRSA ("Biomerioux", Russia). The mecA gene was determined using the LightCycler MRSA Advanced Test with LightCycler 2.0 primer (Roche Molecular Biochemicals, Germany). To determine staphylococcal enterotoxins, the test system RIDASCREENSET A, B, C, D, E (R-Biopharm AG, Darmstadt, Germany) was used. We isolated saprophyte staphylococci from milk of raw and dairy products in western regions of Ukraine in 82.7–97.4% of samples. S. aureus is much more rarely isolated from these dairy products, so it was isolated from sour cream at 62.8 ± 0.9%, from milk at 35.5 ± 1.3% and cottage cheese at 23.0 ± 1.6%. Of the most well0known biotypes of S. aureus present in milk of raw and dairy products of domestic production, two ecological types were distinguished: human and cattle. In this case S. aureus var. hominis was isolated more often than in S. aureus var. bovis. This gives grounds to believe that the main source of contamination with milk staphylococci of raw and dairy products of domestic production is people. Enterotoxin type A, which causes foodborne toxemia, was produced by S. aureus in 40.0 ± 0.5% of cases. Consequently, home-produced dairy products can spread staphylococcal toxicity caused by S. aureus var. hominis. It was found that 17.8 ± 0.6% of S. aureus var. hominis were resistant to methicillin, which is 1.8 times greater than that of S. aureus var. bovis. This gives grounds to consider that there is a risk of MRSA infection to consumers of home-produced dairy products. All methicillin-resistant staphylococci studied produced enterotoxins.

Objectives: To isolate methicillin resistant Staphylococcus aureus (MRSA) from anterior nares of dairy workers and dairy products and assess the antibiotic susceptibility pattern of the isolates. Methods: Swab samples collected from anterior nares of dairy workers and dairy product (butter) were inoculated into mannitol salt agar and incubated at 37ºC for 24 hours. Identification was done based on colony characteristics, Gram's staining, catalase, oxidase and coagulase test. Antibiotic susceptibility testing was done by modified Kirby Bauer disc diffusion method. MRSA was confirmed by using cefoxitin disc. Results: A total of 109 S. aureus (98 from dairy workers and 11 from butter samples) were isolated. Out of them 32 MRSA were isolated from dairy workers and 4 from butter samples. The association between age group and MRSA was found insignificant (p = 0.115). The association of MRSA between male and female workers was found significant (>0.05). About 86% of the MRSA isolates were susceptible to Gentamicin (86.11%) followed by Ciprofloxacin (77.78%). Conclusion: Detection of MRSA among dairy workers and dairy products warrants proper handling and adequate control measures to prevent transmission of MRSA from dairy industry.

Contrary to the safe practices of milk pasteurization or sterilization, which effectively reduce foodborne outbreaks incidence associated with raw milk and dairy products use, outbreaks caused by such products continue to occur. Despite this fact, a worldwide movement advocating for the rights of raw milk and cheese selling and consumption, due to their specific nutritive characteristics, has strengthened significantly in recent years. Traditional agricultural manufacturers from Serbia still sell products related to thermally unprocessed milk, such as cottage cheese and raw cream. In AP Vojvodina during the period of 1981-2010 a total of 179 foodborne outbreaks were reported, where the incriminated cause of the outbreak were milk or diary. In 126 (70.39%) outbreaks, totaling 2276 sick individuals and one casualty, it was confirmed that the incriminated food was from the group of dairy products. In 48 instances (26.82%), bacteriological tests confirmed that milk and dairy products were excluded as the outbreak causes, while in another 5 (2.79%) outbreaks, microbiological analysis of food failed to confirm any relation to the actual epidemiological instances. In some cases, bacteriological testing of incriminated foods was not possible. In the cases of outbreaks associated with the consumption of milk and dairy products, traditional raw milk products were cited as being used. Consumption of unpasteurized milk and cheese represents public health threat. National and international rules ensuring use of safe products for human consumption have to set rules of trade of thermally processed milk and products on the market.

The study was done to isolate Klebsiella spp., Staphylococcus aureus, Escherichia coli & Pseudomonas spp. from raw milk, processed milk (powdered milk & ultra-high-temperature milk) and dairy products (mango milk, and ice cream). During 3 months’ time span of study, a total of ten samples were gathered where eight of the samples powdered milk (2), ultra-high-temperature milk (2), ice cream (2) & mango milk (2) were gathered from different places of Bangladesh & other 2 samples (raw milk) were collected from a cattle farm & individual households in & around Noakhali city. The samples were gathered under aseptic safety measures and were followed by plating on non-selective media (Nutrient broth) & selective media (Mannitol Salt Agar & Macconkey agar). The presumptive Klebsiella spp., Staphylococcus aureus, Escherichia coli & Pseudomonas spp. isolates were identified by biochemical tests. Analysis of the result discovered that out of a total of 10 samples; raw milk (2), processed milk i.e. powdered milk (2), ultra-high-temperature milk (2) and dairy products i.e. mango milk (2) and ice cream (2) resulted in the isolation of 8 isolates (2.5%) of S. aureus, (12.5%) of Klebsiella spp., (2.5%) of Pseudomonas spp. & (2.5%) of Escherichia coli. Results recommended a probability of potential public health threat of Staphylococcus aureus, Klebsiella spp., Escherichia coli & Pseudomonas spp. resulting from contamination of milk and dairy products with pathogenic microorganisms is mainly due to unhygienic practices and conditions in the handling, production, and processing.

A dairy product processing plant was studied for 2.5 years to examine contamination with Staphylococcus aureus and try to correlate the source of contamination. Cultures were submitted to an antibiotic susceptibility test (AST) and characterised by Pulsed-field Gel Electrophoresis (PFGE) analysis. Results showed that 35.2% (19/51) of food handlers were asymptomatic carriers of S. aureus, and that 90.4% (19/21) of raw milk sampled was contaminated. Staphylococcus aureus was isolated from only 10 samples among more than 3200 investigated dairy products. No S. aureus contamination was found on machinery. The AST analysis demonstrated sensitivity of tested S. aureus to oxacillin, cephalothin, vancomycin, gentamicin, and sulfamethoxazole/trimethoprim. AST analysis generated eight different phenotypic profiles, but did not allow us to identify the source of contamination in seven of ten final products. PFGE analysis proved to be a sensitive method as it generated 42 different DNA banding profiles among the 48 S. aureus investigated, demonstrating a lack of predominance of endemic strains in the plant, contrary to suggestions raised by antibiotic resistance typing. Based on PFGE genotyping, S. aureus strains isolated from four contaminated final products were similar to four S. aureus isolated from raw milk. Five final products contained S. aureus different from all other strains collected, and one showed similarity to a strain isolated from a food handler. These results suggest contamination by raw milk as the main source of contamination of the final dairy products.Key words: Staphylococcus aureus, dairy products, antibiotic susceptibility, PFGE.

The article discusses the organizational and economic relations between the participants in the supply chain of milk and dairy products. Generally, organizational and economic relations are a set of relations between the participants of the dairy products chain, which are formed in the process of production, distribution and sale to consumers, as a result of which the economic interests of all its participants are realized. The peculiarities of dairy products of the chain are systematized and the basic principles, conditions for their balanced development are determined, contributes to the coordination of the economic interests of all its participants. It has been proved that economic interest is an important component of organizational and economic relations in the supply chain of milk and dairy products, and its main elements are: prices for raw materials and fnished products, costs of production, processing of raw materials, transportation, terms of mutual settlements, material interest of personnel, norms of fulfllment of contractual obligations, risks and the like. Review of the negative factors of organizational and economic relations in the dairy product chain and their impact on the effectiveness of each of its participants. As a tool to increase the production of raw milk, it is proposed to optimize the raw material zones of dairy processing enterprises, develop strategic programs for the development of the industry and stimulate consumer demand, as well as create industry clusters. The calculations for the distribution of costs and income between participants in the supply chain of milk and milk products prove the need to ensure the equivalence of exchange. In order to improve the efciency of the researcher, it was proposed to increase the volume of milk production and processing, as well as to establish mutually benefcial relations between its participants, connected by a single production cycle. The directions of innovation and investment development of the participants in the dairy products chain, especially the link of dairy cattle breeding, are highlighted. It is highlighted that the innovative renewal of the activities of the chain participants is a process of cyclical activities focused on the production of competitive dairy products of a new generation, which will contribute to an increase in turnover in order to obtain the highest proft of the chain participants and satisfy the demand of consumers with different income levels. Staphylococcal food poisoning is considered one of the leading foodborne illnesses in humans worldwide and is associated with contaminated foods of animal origin, such as milk and dairy products. In this study, we investigated the occurrence of staphylococci and the enterotoxigenic properties of Staphylococcus aureus isolated from raw milk from yaks (Bos mutus) and cattle in Mongolia. Staphylococci were isolated from 72 (74%) of the 97 raw milk samples. Of the samples containing staphylococci, 69% (50 of 72) were from yaks and 30.5% (22 of 72) were from cattle. S. aureus was detected in 10% of yak (7 of 72) and 21% of cattle (15 of 72) milk samples. Staphylococcal enterotoxin C was detected in 23% (5 of 22) of the S. aureus strains investigated, based on the reverse passive latex agglutination technique. Three of the five enterotoxigenic strains were from yaks and two were from cattle. None of the S. aureus strains tested produced staphylococcal enterotoxins A, B, or D. To our knowledge, this is the first report of the occurrence of staphylococci and enterotoxigenic S. aureus in milk from yaks and cattle in Mongolia.

Detailed information on the resistance patterns of Staphylococcus aureus (S. aureus) in milk and cheese is strongly required to facilitate risk assessment analysis in case of food poisoning context and to improve therapeutic approaches used in dairy farms. The present study aimed to perform phenotypic and genotypic antimicrobial characterizations of methicillin, vancomycin, and erythromycin-resistant S. aureus isolated from milk and dairy products through screening mecA, vanA, and ermC using molecular PCR amplification technology. Moreover, the association between each genotypic and its related antibiotic resistance phenotypic features within the isolated S. aureus strains were analyzed. Moreover, the current study aimed to study MRSA's ability to form biofilms. Out of 226 milk and dairy product samples collected from different retailers in Giza Governorate, 69.5% of the samples were positive for the presence of S. aureus. The isolation rate of S. aureus strains from cattle milk, sheep milk, white cheese, flamenco, and mesh samples were 79.7%, 76.5%, 56.0%, 40.0%, and 94.7%, respectively. Multidrug-resistant S. aureus (MDR) was detected in 51% of all isolated S. aureus strains. All tested S. aureus strains were sensitive to trimethoprim-sulfamethoxazole, linezolid, ciprofloxacin, and gentamycin. However, their resistance rates against penicillin, oxacillin, vancomycin, erythromycin, tetracycline, clindamycin and chloramphenicol were 62.4%, 65.0%, 44.6%, 45.9%, 21.0%, 14.0%, and 2.5%, respectively. Of the isolated S. aureus strains, 72.6%, 40.1%, and 48.4% were carriers for mecA, vanA, and ermC genes and the amplified products were at 310, 1030, and 295 bp, respectively. Methicillin-resistant S. aureus isolates were detected in 47.1% of all isolated S. aureus strains. The results indicated that 35.0% of the tested S. aureus strains were genotypic vanA gene carriers and phenotypic resistant to vancomycin representing vancomycin-resistant S. aureus strains. Moreover, 42.7% of all isolated S. aureus strains were carriers for ermC gene and were phenotypic resistant to erythromycin representing erythromycin-resistant S. aureus. The presence of mecA, vanA, and ermC genes in S. aureus was statistically associated with their related phenotypic resistance patterns against both penicillin and oxacillin, vancomycin, and erythromycin, respectively. Moreover, along with an increase in the frequency of mecA, vanA, and ermC genes, their phenotypic antibiotic resistance patterns sharply increased with an odd ratio >1. Of MRSA isolates, 6.8% indicated weak biofilm-formation ability, while 93.2% exhibit no biofilm-forming ability.
