AbstractTwo Hemicycliophora species, H. lutosa and H. typica, found in samples from fallow soil and sugarcane soil in South Africa, were studied morphologically and, for the first time, molecularly. Diagnostic PCR-IT-rRNA-RFLP profiles generated by five restriction enzymes are provided. Study of phylogenetic relationships using D2-D3 expansion segment of 28S rRNA gene sequences revealed that H. lutosa was related to H. poranga. Hemicycliophora lutosa and H. poranga are compared morphologically. SEM photographs are given for H. typica and for H. lutosa for the first time. The male of H. typica represents a first report for South Africa.

During recent collections in South Africa, two species of Criconemoides were found. Criconemoides brevistylus is described and illustrated from a golf course in KwaZulu-Natal Province. It is compared with several closely related species, and three previously described species (C. helicus, C. onostris and C. paronostris) are regarded as junior synonyms of C. brevistylus. Criconemoides obtusicaudatus is described and illustrated from a maize field in Limpopo Province. The male of the species is described for the first time. Molecular characterisation of C. brevistylus and C. obtusicaudatus using the D2-D3 expansions segments of 28S rRNA and ITS rRNA gene sequences are provided. Phylogenetic relationships of these species with other Criconemoides are discussed.

AbstractCaloosia longicaudata is described from Maui, the Hawaiian Islands, for the first time and both sexes are characterised morphologically using light and scanning electron microscopy. Molecular characterisation of C. longicaudata using the D2-D3 domain of 28S rRNA, partial 18S rRNA and ITS rRNA gene sequences is also provided. The phylogenetic relationships of this species with other representatives of the suborder Criconematina are presented and discussed. A diagnostic PCR-ITS-RFLP profile for C. longicaudata is given together with an identification table for eight species of Caloosia. Caloosia langola n. comb. is transferred to the genus and C. shorai is synonymised with H. psidii.

The reniform nematodes of the genusRotylenchulusare semi-endoparasites of numerous herbaceous and woody plant roots and are mainly distributed in tropical and subtropical regions. In this study, we provide morphological and molecular characterisation of six out of ten presently known valid species ofRotylenchulus:R. clavicaudatus,R. leptus,R. macrodoratus,R. macrosoma,R. reniformisandR. saccharifrom South Africa, USA, Italy and Spain.Rotylenchulus parvuswas only studied morphologically. A new species,R. macrosomoidessp. n., isolated from soil and roots of sugarcane in South Africa, is described. The phylogeny ofRotylenchulus, as inferred from the analyses of D2-D3 of 28S rRNA, ITS rRNA,coxImtDNA andhsp90gene sequences, is presented. The study revealed thatR. reniformisandR. macrosomahave a sister relationship, but that relationships between otherRotylenchulusspecies remain unresolved. The phylogenetic analysis also confirmed the hypothesis that this genus originated from the Afrotropical zoogeographical region. Our study revealed thatR. reniformisandR. macrosomoidessp. n. have two distinct rRNA gene types andR. macrosomahave three rRNA gene types in their genomes. PCR with species-specific primers was developed for rapid diagnostics ofR. reniformis.

Pin nematodes of the genus Paratylenchus are widely distributed across the world and associated with many plant species. Morphological identification of Paratylenchus species is a difficult task because it relies on many characters with a wide range of intraspecific variation. In this study we provide morphological and molecular characterisation of several pin nematodes: Paratylenchus aquaticus, P. dianthus, P. hamatus, P. nanus and P. straeleni, collected in different states of the USA and South Africa. Paratylenchus aquaticus is reported from South Africa and Hawaii and P. nanus is found from South Africa for the first time. Morphological descriptions, morphometrics, light and scanning electron microscopic photos and drawings are given for these species. Molecular characterisation of nematodes using the D2-D3 of 28S rRNA and ITS rRNA gene sequence revealed that samples morphologically identified as P. aquaticus, P. hamatus and P. nanus indeed represent species complexes containing several species. Sequences of the rRNA genes are also provided for several unidentified Paratylenchus. Phylogenetic relationships within the genus Paratylenchus are given as inferred from the analyses of the D2-D3 of 28S rRNA and ITS rRNA gene sequences. We present here the most complete phylogenetic analysis of the genus.

Morphological identification of spiral nematodes of the genus Helicotylenchus is a difficult task because most characters used for their diagnosis vary within species. In this paper we provide morphological and molecular characterisations of several spiral nematodes, H. broadbalkiensis, H. digonicus, H. dihystera, H. microlobus, H. paxilli and H. pseudorobustus, collected in different geographical areas of USA, Switzerland, Italy, New Zealand, Spain, UK, South Korea and Russia. We suggest that H. microlobus and H. pseudorobustus are valid species separated from each other morphologically and molecularly. Seven species with distinct molecular characteristics are also distinguished, but are not ascribed morphologically to any specific taxon because of the low number of specimens available. Phylogenetic relationships of H. pseudorobustus with other Helicotylenchus species are given as inferred from the analyses of 154 sequences of the D2-D3 of 28S rRNA gene and 37 sequences of ITS rRNA gene.

Hemicaloosia vagisclera n. sp. is described from Bermuda grass (Cynodon dactylon) in Florida. This new species is characterised by females with body slightly ventrally arcuate in death, face with a prominent elliptical oral disc, large amphids, slender stylet with mean length 64 μm, lateral field consisting of a single longitudinal line marked by continuous and discontinuous transverse striae, oval and full spermatheca, sclerotised vagina vera and tail annuli width greater than that of remaining body annuli. Diagnostic characters for the males are a C-shaped body, head with 4-5 faint lip annuli and pronounced oval oral disc, lateral field with two longitudinal lines intersected by transverse striae, tail digitate in the distal portion posterior to bursa, distinctly annulated and with a round terminus. Molecular characterisation of H. vagisclera n. sp. using the D2-D3 domain of 28S rRNA, partial 18S rRNA and ITS rRNA gene sequences is also provided. The phylogenetic relationships of this species with other representatives of the suborder Criconematina are presented and indicate that H. vagisclera n. sp. has sister relationships with Caloosia longicaudata supporting the classification of Caloosia together with Hemicaloosia as separate genera in the family Caloosiidae. A diagnostic PCR-ITS-RFLP profile for H. vagisclera n. sp. is also given together with an identification key for seven known species of Hemicaloosia.

AbstractMature worms of Stephanoprora amurensis sp. nov. were obtained in an experimental study of its life cycle. In the Russian southern Far East, this trematode circulates using freshwater snails Parajuga subtegulata, freshwater fish and birds as the first, second intermediate and final hosts, respectively. Stephanoprora amurensis sp. nov. differs from the well-known representatives of Stephanoprora in a number of morphometric indicators of the developmental stages. The validity of the species was also confirmed by nuclear and mitochondrial DNA markers. In addition, new genetic data were obtained for Echinochasmus suifunensis and Echinochasmus milvi. An analysis of phylogenetic relationships within Echinochasmidae based on the 28S rRNA gene and ITS2 region identified two clusters, one of which combines species of Echinochasmus with 20–22 collar spines and short-tailed cercariae, and the other which includes Stephanoprora spp. and a number of representatives of Echinochasmus with 24 collar spines and long-tailed cercariae. The results of phylogenetic analysis based on ITS2 data show interfamily level of differences between the two clusters and intergeneric differentiation between the three subclusters uniting the species of Stephanoprora and Echinochasmus.

The Korean cyst nematode, Heterodera koreana, was recorded for the first time from Japan and characterised morphologically, morphometrically and molecularly. In total, 41 populations were detected from soil samples collected from the rhizosphere of four bamboo species in Japan: 31 populations from moso bamboo (Phyllostachys edulis), seven from madake (P. bambusoides), two from henon bamboo (P. nigra var. henonis) and one from fish pole bamboo (P. aurea). The morphology and morphometrics of the Japanese population were in agreement with those of the original description of H. koreana from South Korea and other subsequent descriptions from China and Iran, with the exception of some minor differences. The results of the phylogenetic analyses of the D2-D3 expansion segments of 28S rRNA gene and ITS rRNA gene sequences confirmed the species identification and phylogenetic relationship of H. koreana with other Heterodera species. The COI mtDNA gene sequences were obtained for the first time for H. koreana. Three COI haplotypes found in Japanese H. koreana populations showed a characteristic geographical distribution in Japan.

The phylogenetic relationships of traditional xylotine genera of tribe Milesiini (Diptera, Syrphidae: Eristalinae) were explored using molecular character of a large fragment of the mitochondrial COI and the D2-3 region of the nuclear ribosomal 28S rRNA gene. Of particular interest was the phylogenetic placement of the enigmatic Neotropical genus Cacoceria. The ingroup included 48 taxa, and Cheilosia illustrata (Rhingiini, Eristalinae) was used as outgroup. Multiple specimens of more common taxa were sequenced for surveying levels of intraspecific variation. The combined dataset was analysed using parsimony and optimisation alignment, using the program POY. Cacoceria was resolved within the Chalcosyrphus clade. Earlier hypotheses placed the taxon in the tribe Myoleptini or Chrysogasterini, or in Xylotini without stating a closer relationship to any particular xylotine taxon. The representatives of traditional xylotine taxa were resolved as ((Hadromyia + (Brachypalpoides + Blera + Lejota) + (Xylota sg. Hovaxylota + Xylota sg. Sterphoides) + (Xylota (including Sterphus)) while Neplas and Brachypalpus were resolved in the Myoleptini albeit with very low support.
