Redescription of Bursaphelenchus parapinasteri (Tylenchina: Aphelenchoididae) isolated from Pinus thunbergii in China with a key to the hofmanni-group

Bursaphelenchus parapinasteriis redescribed morphologically and with new molecular characterisation. It was isolated from a deadPinus thunbergiion Changgang Mountain, Zhoushan Islands, Zhejiang Province, China. Detailed morphology of the spicule, female gonad, hemizonid position, arrangement of male caudal papillae, and female tail terminus shape are documented. The ITS-RFLP patterns and the ITS1/2, partial 28S D2-D3 and partial 18S gene sequences were characterised. The phylogenetic analyses revealed thatB. parapinasteribelongs to thehofmanni-groupsensuBraasch and is close toB. anamurius,B. hofmanni,B. mazandaranense,B. paracorneolus,B. pinasteriandB. ulmophilus. A diagnostic key to species of thehofmanni-group is presented.

Pseudaphelenchus zhoushanensis n. sp. was isolated from a dead Pinus thunbergii at Changgang Mountain, Zhoushan Islands, Zhejiang Province, China. It is characterised by the small to medium length body, cuticle slightly annulated, presence of three lateral lines, stylet 9.0-10.7 μm with small but conspicuous basal knobs, excretory pore located from same level as the metacorpus to slightly anterior to metacorpus, true bursa surrounding entire tail but inconspicuous, male tail conical with a single mucron, spicule with distinct condylus and rostrum strongly arcuate to a pointed end, female tail conical with annulation, strongly ventrally bent in distal part of tail, with terminus bluntly pointed or finely mucronate. Phylogenetic analyses using sequences of the18S and 28S D2-D3 regions of rDNA confirmed the status of P. zhoushanensis n. sp. as a new species. Combining the molecular phylogenetic analyses, morphology and biology of P. zhoushanensis n. sp. and Tylaphelenchus jiaae indicates that T. jiaae is a member of Pseudaphelenchus to which it is herein transferred as P. jiaae n. comb. (= T. jiaae).

Summary Ektaphelenchoides compsi is redescribed morphologically with new molecular characterisation. It was isolated from a dead Pinus massoniana tree in Ningde City, Fujian Province, China. Detailed morphology of the spicule, female gonad, hemizonid position, arrangement of male caudal papillae and female tail terminus shape are documented. It is characterised by a lateral field with three lines (forming two bands), tripartite stylet 17.8 (17.0-19.4) μm long without basal thickenings, metacorpus rectangular with anterior 40% granular and posterior part weakly muscular, metacorpal valve slightly posterior to middle of metacorpus, excretory pore at level of nerve ring, vagina with thickened walls and strongly developed muscular bundles, vulval lips slightly protuberant, vulval flap absent, distal region of post-vulval uterine sac appearing as a weakly developed oogonia, anus and rectum indistinct, female posterior part (‘tail’) dorsally convex, conical, terminal region contracted into a bluntly pointed tip. The spicules are arcuate, 15.6 (14.3-16.3) μm along the chord, lamina smoothly curved to distal end, capitulum slightly concave, condylus well-developed with broadly rounded tip and slightly depressed at dorsal end, rostrum triangular with finely rounded tip, cucullus absent, and with seven caudal papillae present. The near full length 18S and 28S D2-D3 regions of rRNA genes sequences were characterised. The phylogenetic analyses revealed that the Fujian population of E. compsi grouped with the Zhejiang population of E. compsi, both being morphologically identical.

Summary Helicotylenchus ciceri n. sp. and H. scoticus are described and illustrated based on morphological, morphometric and molecular characters. The new species is characterised by a conical and truncated lip region with five or six distinct annuli, stylet 32-37 μm long with anteriorly concave knobs, secretory-excretory pore posterior to the pharyngo-intestinal valve, dorsally convex-conoid tail with a terminal projection, phasmids 14 (7-20) annuli anterior to the level of anus, empty spermatheca and absence of males. Intraspecific variation of 16 populations of H. scoticus, collected from chickpea and lentil fields in Kermanshah province, western Iran, is discussed. The results of the phylogenetic analyses based on the sequences of the partial 18S rRNA, D2-D3 expansion segments of 28S rRNA and ITS rRNA genes are provided for the studied species, confirming their differences from each other and determining the position of them and their relationships with closely related species.

Summary Laimaphelenchus pannocaudus, isolated in Ningbo port from Picea gluaca, is redescribed based upon characteristic morphological details elucidated by light and scanning electron microscopy photographs. The recovered population is characterised by 907 (771-1024) μm long females with 11.2 (8.9-12.2) μm long stylet, four incisures in the lateral field, excretory pore located at the same level as, or slightly anterior to the nerve ring, vulva lacking a flap and located at 68.7 (63.9-73.4)% of the body, elongate post-vulval uterine sac 78.5 (55.6-101.1) μm long, and tail 40.6 (33.4-45.6) μm long. Males are common and characterised by 16.5 (15.7-17.8) μm long spicules (chord) with their condylus slightly recurved, squared to rounded in shape with a blunt rounded tip and rostrum triangular with a bluntly pointed tip, and six visible caudal papillae. The female tail is ventrally curved, conoid and bears a stalk-like terminus with 4-8 pedunculate projections. Phylogenetic analyses using partial 18S and 28S rDNA D2-D3 data revealed that L. pannocaudus formed a sister relationship with L. suberensis in both phylogenies.

During a survey on pin nematodes in western Iran, two populations of Paratylenchus audriellus and Paratylenchus tenuicaudatus were collected and subsequently analysed morphologically and molecularly. Paratylenchus audriellus is characterised by the long stylet (48-61 μm) and the typical female tail with a characteristic claw-like process with sharply pointed terminus. To our knowledge, the Iranian population of P. tenuicaudatus is the first record from Iran. The molecular characterisation of P. audriellus nematodes using the D2-D3 of 28S rRNA and the partial 18S rRNA gene sequences revealed that this species is clearly separated from P. straeleni and should be considered as a valid taxon.

Bursaphelenchus andrassyi sp. n., found in conifer wood samples from Romania and Turkey, is characterised morphologically and genetically. Bursaphelenchus andrassyi sp. n. clearly belongs to the sexdentati group, having a terminal bursa, four lateral lines, a very small female ‘vulval flap’, strongly arcuate spicules and the typical position of caudal papillae of males. It is morphologically most similar to B. vallesianus and B. sexdentati. It can be differentiated from B. vallesianus by the usually subcylindrical female tail with rounded or wedge-shaped terminus vs conical female tail with a more or less rounded terminus, slightly different shape of spicules (low square condylus, lacking a distinct cucullus, pointed rostrum) and from B. sexdentati by lacking a distinct post-vulval constriction, shorter stylet and shorter spicules. The species status is supported by ITS-RFLP patterns and sequencing results of both partial 18S and 28S rDNA regions.

Species within Gennadas differ from each other largely only in male (petasma) and female (thelycum) copulatory characters, which were restudied in scanning electron microscopy and used as a basis for phylogenetic analyses. Twenty-six petasma characters and 49 thelycum characters were identified. All 16 recognized species of Gennadas and Aristaeomorpha foliacea (outgroup) were included as terminals. Four robust monophyletic clades were retrieved, described and diagnosed as new species groups. The thelycum characters had greater impact on tree topology and supported deeper nodes than did the petasma characters. We hypothesize that features of the thelycum evolved first followed by aspects of the petasma. Relatively more conservative characters include parts of the sternites of the thelycum and of the petasma, while the scuti and protuberances on the thelycum and the shape and subdivisions of the petasma lobes are evolutionarily plastic. We identified two groups of copulatory characters, which are likely coupled functionally and interlinked evolutionarily: (i) the external part of the petasma and the posterior part of the thelycum and (ii) the internal part of the petasma and anterior part of the thelycum. We reconstruct possible mating position during copulation for each of the new species groups presented here. We also present an updated key to genera of Benthesicymidae and key to species of Gennadas .

A Gram-stain-negative, rod-shaped and facultatively aerobic bacterial strain, designated F7430T, was isolated from coastal sediment collected at Jingzi Wharf in Weihai, PR China. Cells of strain F7430T were 0.3–0.4 µm wide, 2.0–2.6 µm long, non-flagellated, non-motile and formed pale-beige colonies. Growth was observed at 4–40 °C (optimum, 30 °C), pH 6.0–9.0 (optimum, pH 7.5–8.0) and at NaCl concentrations of 1.0–10.0 % (w/v; optimum, 1.0 %). The sole respiratory quinone of strain F7430T was ubiquinone 8 and the predominant cellular fatty acids were summed feature 8 (C18 : 1  ω7c / C18 : 1  ω6c; 60.7 %), summed feature 3 (C16 : 1  ω7c/C16 : 1  ω6c; 30.2 %) and C15 : 0 iso (13.9 %). The polar lipids of strain F7430T consisted of diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol, phosphatidylcholine, one unidentified phospholipid and three unidentified lipids. Results of 16S rRNA gene sequences analyses indicated that this strain belonged to the family Halieaceae and had high sequence similarities to Parahaliea aestuarii JCM 51547T (95.3 %) and Halioglobus pacificus DSM 27932T (95.2 %) followed by 92.9–95.0 % sequence similarities to other type species within the aforementioned family. The rpoB gene sequences analyses indicated that the novel strain had the highest sequence similarities to Parahaliea aestuarii JCM 51547T (82.2 %) and Parahaliea mediterranea DSM 21924T (82.2 %) followed by 75.2–80.5 % sequence similarities to other type species within this family. Phylogenetic analyses showed that strain F7430T constituted a monophyletic branch clearly separated from the other genera of family Halieaceae . Whole-genome sequencing of strain F7430T revealed a 3.3 Mbp genome size with a DNA G+C content of 52.6 mol%. The genome encoded diverse metabolic pathways including the Entner–Doudoroff pathway, assimilatory sulphate reduction and biosynthesis of dTDP-l-rhamnose. Based on results from the current polyphasic study, strain F7430T is proposed to represent a novel species of a new genus within the family Halieaceae , for which the name Sediminihaliea albiluteola gen. nov., sp. nov. is proposed. The type strain of the type species is F7430T (=KCTC 72873T=MCCC 1H00420T).
