A highly sensitive radioimmunoassay for the determination of thyroxine in blood spotted on filter paper is described. The radioimmunoassay developed by Weeke and Ørskov for determination of serum thyroxine was simplified and shortened, and its cost was decreased, for use in programmes for mass screening of thyroid function. The method permits the use of small (2 mm diameter) dried blood dots, thus avoiding interference by binding proteins. Polyethylene glycol 6000 is used for the final separation of antibody bound labelled thyroxine, instead of the expensive and time-consuming wick chromatography used in the reference method. Serum thyroxine concentrations obtained using dried blood by the modified procedure correlate well with those obtained using serum by the reference method. The distribution of serum thyroxine concentration for a population of 2134 babies is given.

Capecitabine is a 'pro-drug' to the cytotoxic agent 5-fluorouracil (5-FU) intended to administered orally. Capecitabine is generally used as first line monotherapy for advanced colon cancer. Simple, rapid, accurate UV spectrophotometric methods were developed in the present study and validated for the estimation of Capecitabine in bulk and its formulations as per ICH guidelines. Three solvent systems viz., 0.1N NaOH, 0.1N HCl and Methanol: Water (1:3) was tried. The results suggest that the developed method shows linearity over the range of concentration 2-24μg/ml and a correlation coefficient of 0.9999. Accuracy, precision, linearity, robustness, and ruggedness were statistically validated as per ICH guidelines for all the developed methods. The % RSD values for validated methods were found to be less than 1.5 and methods will find application in routine analysis of drug formulations containing Capecitabine.

Abstract A highly sensitive method with specificity for the peptide chain backbone was developed for determination of total urinary protein. Interfering substances are removed by gel filtration and cupric ions are stoichiometrically bound to the peptide bonds of protein by the biuret reaction. Ion-exchange characteristics of the gel are neutralized, allowing protein—copper complex to be separated from excess cupric ions by a second gel filtration step. Copper bound to peptide bonds is colorimetrically determined by use of sodium diethyldithiocarbamate. Nonprotein substances do not interfere unless they simultaneously bind to protein and chelate copper. As little as 1 mg of total protein per deciliter can be determined in heterogeneous biological fluids such as urine.

A study of circulating immune complexes was carried out using a reaction with polyethylene glycol. The method turned out to be simple, highly sensitive and affordable for any clinical laboratory with a photoelectric colorimeter. Analysis of the survey data of 115 healthy donors, 63 patients with rheumatoid arthritis and 16 patients with systemic lupus erythematosus made it possible to establish the level of circulating immune complexes in health and disease. The circulating immune complexes were studied in patients with rheumatism and chronic tonsillitis. To assess the results of the reaction, human aggregated gamma globulin (manufactured by Kazan NIIEM) was used.
