Seven extracts were prepared from Mentha x piperita (peppermint) leaves in sequence using a Soxhlet apparatus, viz. (40-60°) light petroleum (PE), dichloromethane (CH2Cl2), acetonitrile (ACN), ethyl acetate (EtOAc), methanol (MeOH), n-butanol and water (H2O) extracts. The phenolic and flavonoid content of each extract were estimated using spectrophotometric methods whilst a qualitative-quantitative analysis was made by reverse-phase high performance liquid chromatography coupled with photodiode array detection (HPLC-PDA). Each extract was assessed in a battery of six antioxidant-related assays so as to determine their iron(III) reductive, iron(II) chelating and free radical scavenging abilities. The MeOH-soluble extract contained the greatest content of total phenols and flavonoids based upon the Folin-Ciocalteu and 2-aminoethyl diphenylborate reagent data and HPLC-PDA analysis. Based upon the chromatographic and UV-spectral data, the leaves principally contained the cinnamic acid caffeic acid, the depside rosmarinic acid and flavonoids (flavones and flavanones). Eriocitrin (383.3 ± 2.2 mg/g extract) and rosmarinic acid (381.2 ± 1.9 mg/g extract) were the most abundant components identified within the leaves, whilst naringenin-7- O-glucoside (0.8 ± 0.01 mg/g extract) was the least abundant component identified being found only in the EtOAc-soluble extract. The EtOAc, ACN and H2O-soluble extracts demonstrated the most potent iron(III) reductive and 1,1′-diphenyl-2-picrylhydrayl, 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonate) and hydroxyl free radical scavenging properties; however, the H2O and CH2Cl2-soluble extracts were the most potent extracts in the β-carotene-linoleic acid bleaching inhibition assay. In terms of iron(II) chelation – an important antioxidant property - the PE, MeOH and H2O extracts demonstrated moderate iron(II) chelating activity.

Lipid profile is a blood test that measures the amount of lipids, or fats, in your blood. The lipids measured are usually total cholesterol, HDL-C, LDL-C and triglycerides. When levels of these lipids are abnormal, there is an increased risk of heart attack and stroke. In our study, we use methanol crude extract for lipid lowering activity on rabbit and found that lipid profile was reduced by 14.0, 1.0, 4.0 and 15.0 mg/dl for total cholesterol, HDL-C, LDL-C and triglyceride respectively after using the plant extract (dose 500mg/rabbit for 10 days); where atorvastatin (0.005mg/rabbit) was used as standard lipid lowering agent. We also focused on the antioxidant property of crude methanol extract. Here we also carried out free radical-scavenging activity study and found the IC50 value for C. tamala is 6.00 μg/ml where the standard antioxidant (ascorbic acid) gave the value of 3.21μg/ml. Flavones and flavonoids are known to have potent antioxidant activity due to intracellular free radical scavenging capacities. Flavonoids are found ubiquitously in plants as a member of polyphenolic compounds which share diverse chemical structure and properties. Quercetin is among the most efficient antioxidants of the flavonoids. The antioxidant property of quercetin has been highlighted in this review. These compounds have pivotal role in treatment of diabetes, cancers and some cardiovascular diseases.

The fruit of Capsicum annuum var. cerasiforme belonging to the Family Solanaceae is a widely used spice in Sikkim, a North Eastern state of India. The fruit of Capsicum annuum var. cerasiforme locally called ‘Dalle Khorsani’ is a valuable cash crop of Sikkim and also possess medicinal properties. Heracleum nepalense D. Don locally called ‘Chimphing’ belongsto the Family Umbillifereae and is extensively used in folk medicine. The present study evaluated the antimicrobial property of fruits of Capsicum annuum var. cerasiforme (Mill.) Irish and Heracleum nepalense D. Don using agar well diffusion method against Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa, Proteus vulgaris, Staphylococcus aureus, Bacillus cereus and Bacillus subtilis. The antioxidant property of methanol extracts of fruits of these test plants was also investigated. The antioxidant property was evaluated by 2, 2-diphenyl-1-picryl hydrazyl (DPPH) free radical scavenging activity assay and Ferric reducing power assay. The total phenolic content was determined by using Gallic acid as a standard. The methanol extract of both the plants inhibited the growth of mostly Gram positive bacteria and Escherichia coli among Gram negative bacteria. However the acetone extract of the selected plants did not exhibit antimicrobial activity. The methanol extracts of these plants exhibited significant (p>0.05) concentration dependent increase in DPPH free radical scavenging activity. The extract of Capsicum annuum var. cerasiforme (Mill.) Irish also exhibited significantly higher (p>0.05) ferric reducing ability thereby showing higher antioxidant activity. The phytochemical analyses of the plant extracts revealed the presence of phenol, tannin, flavonoid, alkaloid, steroid and saponin. The results of the present study indicate that Capsicum annuum var. cerasiforme (Mill.) Irish and Heracleum nepalense D. Don can be a potential source of antimicrobial and antioxidant agents and can be explored further for its therapeutic use.

Objective: To investigate the photochemical constituents present in methanol extract of martynia annua seeds using Gas Chromatography-Mass Spectroscopy(GC-MS), High-Performance Thin–Layer Chromatography(HPTLC) analysis and study antioxidant activity. Methods: Methanol extract of Martynia annua seeds were subjected to GC-MS and HPTLC analysis. HPTLC analysis was carried out using GAMAG system with a linomate5 applicator, system mobile phase (Toluene: Chloroform: Ethanol (4:4:1 V/V/V)), two different volume of extract was applied 2 µl and 5 µl. GC-MS analysis was carried out on JEOL GC MATE ΙΙ, column HP 5 MS and Quadruple double focusing mass analyzer. Antioxidant activity was determined by DPPH assay. Results: GC-MS analysis provided 17 peaks indicating the presence of seventeen different phytochemicals in methanol extract of martynia annua seeds. HPTLC fingerprint showed 6 peaks at both size 2 µl and 5 µl at 254 nm whereas 4 peaks, 9 peaks were detected at 366 nm for 2 µl and 5 µl respectively. After derivatization with 10 % methanolic sulphuric acid, 8 peaks, 11 peaks were detected for 2 µl and 5 µl respectively when the derivatized plate was scanned at 540 nm. DPPH free radical scavenging result showed EC50 value of 44.1±1.1 µg/ml. Conclusion: The GC-MS analysis showed the presence of fatty acids, ester, aldehydes and ketones whereas in HPTLC different peaks at different UV-lights before and after derivatization were observed. Maximum percentage inhibition using DPPH assay was found 74 at concentration of 50 µg/ml.

Isolation and free-radical-scavenging properties of cyanidin 3-O-glycosides from the fruits of Ribes biebersteinii Berl. The reversed-phase preparative high performance liquid chromatographic purification of the methanol extract of the fruits of Ribes biebersteinii Berl. (Grossulariaceae) afforded five cyanidin glycosides, 3-O-sambubiosyl-5-O-glucosyl cyanidin (1), cyanidin 3-O-sambubioside (2), cyanidin 3-O-glucoside (3), cyanidin 3-O-(2G-xylosyl)-rutinoside (4) and cyanidin 3-O-rutinoside (5). They showed considerable free-radical-scavenging properties in the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay with the RC50 values of 9.29 × 10-6, 9.33 × 10-6, 8.31 × 10-6, 8.96 × 10-6 and 9.55 × 10-6 mol L-1, respectively. The structures of these compounds were elucidated by various chemical hydrolyses and spectroscopic means. The total anthocyanin content was 1.9 g per 100 g dried fruits on cyanidin 3-glucoside basis.

Objective: The objective of this study was to evaluate the free radical scavenging potential and high-performance thin-layer chromatography (HPTLC) fingerprinting of the ethanolic extract of south Indian orthodox black tea (OBT).Methods: Phytochemical analysis was carried out using standard methods, and free radical scavenging activity of the extract was determined using 2,2-diphenyl-1-picrylhydrazyl (DPPH), nitric oxide (NO), superoxide anion (SOD), and hydroxyl radical scavenging capacities. The ethanolic extract of OBT was loaded in the pre-coated HPTLC plates (silica gel 60 F 254) E-MERCK KGaA. HPTLC was carried out with toluene: ethyl acetate: diethylamine (7:2:1), chloroform: methanol:formic acid (8.5:1.0:0.5), and butanol: isopropyl alcohol (1:1) as mobile phase for alkaloids, flavonoids, and terpenoids, respectively.Results: HPTLC results confirmed that the extract contained several potential active components such as flavonoids, alkaloids, and terpenoids as the slides revealed multicolored bands of varying intensities. Extract of OBT reliably showed the total phenolics 132.27 mg/g, flavonoids 72.52 mg/g, and alkaloids 66.01 mg/g of dry matter. The IC50 value of OBT for DPPH was found to be 372.22 μg/ml, SOD 311.93 μg/ml, NO 362.17 μg/ml, hydroxyl radical 342.14 μg/ml, and reducing power 178.54 μg/ml.Conclusion: The HPTLC fingerprinting profile developed for ethanolic extract will help in proper identification and quantification of marker compounds. The ethanolic extract of OBT was found to possess a wide range of phytochemicals with excellent antioxidant properties. This information may help to choose the best beverage to be consumed in the future.

This study aimed to investigate the extract with high antioxidant activity of Tetrastigma hemsleyanum Diels et Gilg and identify the antioxidant components in vitro. α, α-Diphenyl-β-picrylhydrazyl (DPPH) radical assay, Trolox equivalent antioxidant capacity (TEAC) assay, ferric reducing antioxidant power (FRAP), and hydroxyl radical scavenging method were used to screen the extract with high antioxidant activity. The antioxidant capacity of the extracts was evaluated by the free radical scavenging ability of DPPH. The ability of extracts to scavenge 2, 2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) free radical was evaluated by TEAC assay. The FRAP method was used to evaluate the ability of extracts to reduce Fe3+. The ability to scavenge hydroxyl radicals produced by the interaction of hydrogen peroxide and Fe2+ was measured by monitoring the change in the absorbance of the reaction mixture at 536 nm. Then, high-performance liquid chromatography-DPPH (HPLC-DPPH) and HPLC-hydroxyl radical scavenging methods were used to screen the antioxidant components in the extract. The molecular weight of the above antioxidant components was investigated using the qualitative analytical method of high-performance liquid chromatography coupled with quadrupole time-of-flight tandem mass spectrometry (HPLC-Q-TOF LC/MS). Based on the concentrations of the samples (0.2–4 mg/mL), the DPPH free radical scavenging ability, ABTS+ free radical scavenging ability, hydroxyl free radical scavenging ability, and Fe3+ reducing ability of the ethyl acetate extract (EAE) were stronger than that of the crude extract (CE), petroleum ether extract (PEE), and n-butanol extract (BE). The EAE has higher antioxidant activity than CE, PEE, and BE. Six antioxidant components, rutin, quercetin, isoquercetin, astragalin, kaempferol, and kaempferol-3-o-rutoside, were identified in the EAE.

Pathological aging due to harmful free radicals and oxidative stress has been a serious threat to human health (Kensella & He, 2008). Thus, the study aimed to evaluate the free radical scavenging activity, antioxidants, essential vitamins, and physicochemical properties of Hydrocotyle vulgaris L. (Pennywort) which could serve as basis in developing a biocosmetic antidote for pathological aging. H.vulgaris L. plant was harvested, air dried and then extracted via soxhlet. UV-vis spectroscopy was used for scavenging activity and antioxidants of the plant while High Performance Liquid Chromatography (HPLC) was applied for the identification of essential vitamins. Phytochemicals, pH and spreadability values of the plant were also tested. Results revealed that H.vulgaris L. has an abundant presence of flavonoid and alkaloid while traced presence of tannin. Good trend for scavenging activity was also observed exhibiting low absorbance of possible free radicals with IC50 of 29.75 and antioxidant activity of 158.13 (Total Phenolics as gallic acid in %w/w). Meanwhile, β-carotene (10.4 mg/kg), Riboflavin (4.08mg/kg), and Vitamins C (70.2mg/kg) and E (26.9mg/kg) were also found in the plant. Non-irritating pH levels of 5.7 and 7.14 for 50% and 75% concentrations of the extracts with good spreadability value of 17.51 g∙cm/sec (for 75% concentration) were also obtained. Based on the findings of the study, Pennywort has a very promising therapeutic characteristics with its good scavenging activity and antioxidants. Thus, the development of a biocosmetic product that could serve as an antidote for pathological aging from Pennywort appeared to be very apparent.
