Two samples of Ligularia kanaitzensis collected in the Shizhongshan Mountain region of Yunnan Province, as well as one sample collected in Sichuan Province, were analyzed with regard to root constituents and neutral DNA sequences. The two Shizhongshan samples were found to be different from each other and from other samples analyzed previously, indicating that the intra-specific diversity in the species was highly complex.

Abstract Background Accumulating evidence suggest that compromised CYP2D6 enzyme activity caused by gene mutation could contribute to primaquine failure for the radical cure of vivax malaria. The current study aims to preliminarily reveal the association between the recurrence of vivax malaria in Yunnan Province and CYP2D6 gene mutation by analysing polymorphisms in the entire coding region of human CYP2D6 gene. Methods Blood samples were collected from patients with vivax malaria, who received "chloroquine and 8-day course of primaquine therapy" in Yunnan Province. The suspected relapsed cases were determined by epidemiological approaches and gene sequence alignment. PCR was conducted to amplify the CYP2D6 gene in the human genome, and the amplified products were then sequenced to compare with the non-mutation “reference” sequence, so as to ensure correct sequencing results and to determine 9 exon regions. Subsequently, the DNA sequences of 9 exons were spliced into the coding DNA sequence (CDS), which, by default, is known as maternal CDS. The paternal CDS was obtained by adjusting the bases according to the sequencing peaks. The mutation loci, haplotypes (star alleles), genotypes and odds ratios (OR) of all the CDSs were analysed. Results Of the119 maternal CDS chains in total with 1491 bp in length, 12 mutation sites in the 238 maternal and paternal CDS chains were detected. The c.408G > C mutation was most frequently detected in both suspected relapsed group (SR) and non-relapsed group (NR), reaching 85.2% (75/88) and 76.0% (114/150), respectively. The c.886C > T mutation was most closely related to the recurrence of vivax malaria (OR = 2.167, 95% CI 1.104–4.252, P < 0.05). Among the 23 haplotypes (Hap_1 ~ Hap_23), Hap_3 was non-mutant, and the sequence structure of Hap_9 was the most complicated one. Five star alleles, including *1, *2, *4, *10 and *39, were confirmed by comparison, and CYP2D6*10 allele accounted for the largest percentage (45.4%, 108/238). The frequency of CYP2D6*2 allele in the SR group was significantly higher than that in the NR group (Χ2 = 16.177, P < 0.05). Of the defined 24 genotypes, 8 genotypes, including *4/*4, *4/*o, *2/*39, *39/*m, *39/*x, *1/*r, *1/*n, and *v/*10, were detected only in the SR group. Conclusion Mutation of CYP2D6*10 allele accounts for the highest proportion of vivax malaria cases in Yunnan Province. The mutations of c. 886C > T and CYP2D6*2 allele, which correspond to impaired PQ metabolizer phenotype, are most closely related to the relapse of vivax malaria. In addition, the genotype *4/*4 with null CYP2D6 enzyme function was only detected in the SR group. These results reveal the risk of defected CYP2D6 enzyme activity that diminishes the therapeutic effect of primaquine on vivax malaria.

Abstract Background Failed diagnoses of some falciparum malaria cases by RDTs are constantly reported in recent years. Plasmodium falciparum histidine-rich protein 2 (pfhpr2) gene deficiency has been found to be the major reason of RDTs failure in many countries. This article analysed the deletion of pfhpr2 gene of falciparum malaria cases isolated in Yunnan Province, China. Methods Blood samples from falciparum malaria cases diagnosed in Yunnan Province were collected. Plasmodium genomic DNA was extracted and the pfhrp2 gene exon2 region was amplified via nested PCR. The haplotype of the DNA sequence, the nucleic acid diversity index (PI) and expected heterozygosity (He) were analyzed. Count PfHRP2 amino acid peptide sequence repeat and its times, and predict the properties of PfHRP2 peptide chain reaction to RDTs testing. Results A total of 306 blood samples were collected, 84.9% (259/306) from which pfhrp2 PCR amplification products (gene exon2) were obtained, while the remaining 47 samples were false amplification. The length of the 250 DNA sequences ranged from 345 - 927 bp, with 151 haplotypes, with PI and He values of 0.169 and 0.983, respectively. The length of the PfHRP2 peptide chain translated from 250 DNA sequences ranged from 115 to 309 aa. All peptide chains had more than an amino acid codon deletion. All 250 PfHRP2 strands ended with a type 12 amino acid repeat, 98.0% (245/250) started with a type 1 repetition and 2.0% (5/250) with a type 2 repetition. The detection rate for type 2 duplicates was 100% (250/250). Prediction of RDT sensitivity of PfHRP2 peptide chains based on type 2 and type 7 repeats showed that 9.60% (24/250), 50.0% (125/250), 13.20% (33/250) and 27.20.5% (68/250) of the 250 peptide chains were very sensitive, sensitive, borderline and non-sensitive, respectively. Conclusion The diversified polymorphism of the pfhrp2 gene deletion from different infection sources in the Yunnan province are extremely complex. The cause of the failure of pfhrp2 exon2 amplification is still to be investigated. The results of this study appeal to Yunnan Province for a timely evaluation of the effectiveness and applicability of RDTs in the diagnosis of malaria.

Two new riffle beetles of the genus Zaitzevia Champion, 1923: Z. chenzhitengi sp. nov. from Sichuan Province and Z. xiongzichuni sp. nov. from Yunnan Province are described from China. Habitus and other diagnostic features of the new species are described and illustrated. Habitat information and collection data of the new species are also provided.

A new orchid species, Cymbidium jiangchengense, from Yunnan Province, China, is described and illustrated. Its distinctiveness is evaluated with morphology and molecular analyses. A detailed comparison between the newly discovered orchid and other members of Cymbidium was performed. The new plant was characterized by stem-like pseudobulbs, narrowly oblong leaves, coriaceous leaves with an acute apex, a 2-flowered inflorescence, a purplish pink flower, narrowly elliptic sepals, petals, a obovate-lanceolate lip with a cordate midlobe, a yellow central callus, and a disc with a trough shape longitudinal lamella from the base extending to the base of the midlobe and a lamellae apex inflated to form two calluses that are not confluent apically. These features distinguish this new orchid from all other known species of Cymbidium. A molecular study based on nuclear ribosomal ITS and plastid matK and rbcL DNA sequence data indicates that C. jiangchengense is a distinct species that sister to C. wadae and a member of section Eburnea, subgenus Cyperorchis.

As an important part of farmland, the slope farmland is widely distributed in the central and western plateau mountain region in China. It is necessary to scientifically evaluate the slope farmland quality (SFQ) and analyze the spatial structure characteristics of SFQ to ensure reasonable utilization and partition protection of slope farmland resources. This paper takes the typical plateau mountain region—Yunnan Province in China—as an example and systematically identifies the leading factors of SFQ. The sloping integrated fertility index (SIFI) is adopted to reflect the SFQ. The evaluation system is built to quantitatively evaluate the SFQ and the spatial structure characteristics of SFQ were analyzed by a geostatistical model, autocorrelation analysis and spatial cold–hot spot analysis. The results show that the SFQ indexes in Yunnan Province are between 0.36 and 0.81, with a mean of 0.59. The SFQ grade is based on sixth-class, fifth-class, seventh-class and fourth-class land. The SFQ indexes present a normal spatial distribution, and the Gaussian model fits well with the semi-variance function of the spatial distribution of SFQ indexes. Furthermore, the spatial distribution of SFQ indexes is moderately autocorrelated. The structural factors play a major role in the spatial heterogeneity of SFQ indexes, but the influence of random factors should not be ignored. The spatial distribution of SFQ grades has a significant spatial aggregation characteristic, and the types of local indicators of spatial association (LISA) are based on high–high (HH) aggregation and low–low (LL) aggregation. The cold spot and hot spot distributions of SFQ grades display the significant spatial difference. The hot spot area is mainly distributed in Central Yunnan and the Southern Fringe, while the cold spot area mainly distributes in the Northeastern Yunnan, Northwestern Yunnan and Southwestern Yunnan. This study could provide a scientific basis for SFQ management and ecological environment protection in the plateau mountain region.

Chilli veinal mottle virus (ChiVMV), a potyvirus, is widespread over the world. In China, it was first reported in chili pepper (Capsicum annuum) in Hainan Province (south China) in 2006 (2). Subsequently, it was reported in tobacco (Nicotiana tabacum) in Yunnan Province (southwest China) in 2011 (1). Sichuan Province is one of the largest vegetable producing areas of China. In May 2012, tomatoes with leaves displaying virus-infected symptoms like mottling, mosaic, narrowing, or curling were observed in several fields of Chengdu, eastern Sichuan Province, southwest China. Of the 20 fields we investigated, four fields with 90% tomato plants were infected. During 2012 and 2013, six samples were collected from symptomatic tomato leaves based on different symptoms and locations. All six samples were assayed by western blotting using polyclonal antisera (Cucumber mosaic virus [CMV], Tobacco mosaic virus [TMV]) obtained from Agdia (Elkhart) and one antiserum to ChiVMV obtained from Yunnan Academy of Agricultural Science (China). Two samples from Pengzhou and one sample from Shuangliu exhibiting mosaic leaves were positive for TMV, one sample from Pixian exhibiting narrowing leaves was positive for CMV, and the other two samples from Shuangliu exhibiting mottle and leaf distortion were positive for ChiVMV. Total RNAs was extracted from all six samples and healthy tomato leaves using Trizol reagent (Invitrogen), First-strand cDNA synthesis primed with oligo(dT) by SuperScript III Reverse Transcriptase (Invitrogen). RT-PCR was performed using primer pairs ChiVMV-CP F (5′-GCAGGAGAGAGTGTTGATGCTG-3′) and ChiVMV–CP R (5′-(T)16AACGCCAACTATTG-3′), which were designed to direct the amplification of the entire capsid protein (CP) gene and 3′ untranslated region (3′-UTR) of ChiVMV (GenBank Accession No. KC711055). The expected 1,166-bp DNA fragment was amplified from the two tomato samples from Shuangliu that were positive for ChiVMV in the western blot tests, but not from the others. The obtained fragments were purified and cloned into the PMD18-T vector (TaKaRa) and sequenced. The sequencing results showed that the two ChiVMV isolates from tomato in Shuangliu were identical (KF738253). Nucleotide BLAST analysis revealed that this ChiVMV isolate shared ~84 to 99% nucleotide identities with other ChiVMV isolates available in GenBank (KC711055 to KF220408). To fulfill Koch's postulates, we isolated this virus by three cycle single lesion isolation in N. tabacum, and mechanically inoculated it onto tomato leaves. The same mottle and leaf distortion symptoms in systemic leaves were observed. Subsequent RT-PCR, fragment clone, and sequence determination tests were repeated and the results were the same. All the evidence from these tests revealed that the two tomato plants were infected by ChiVMV. To our knowledge, this is the first report of ChiVMV naturally infecting tomato in China. It shows that ChiVMV is spreading in China and is naturally infecting a new solanaceous crop in the southwest area, and the spread of the virus may affect tomato crop yields in China. ABSTRACT The Botryosphaeriaceae accommodates many important pathogens of woody plants, including Eucalyptus. Recently, Botryosphaeriaceae were isolated from diseased plant parts from surveys of Eucalyptus plantations in the YunNan Province, China. The aims of this study were to identify these Botryosphaeriaceae isolates and to evaluate their pathogenicity to Eucalyptus. A total of 166 isolates of Botryosphaeriaceae were obtained from six regions in the YunNan Province, of which 76 were from Eucalyptus urophylla × E. grandis hybrids, 49 from E. globulus trees, and 41 isolates were from other unknown Eucalyptus species or hybrids. Isolates were identified by comparing DNA sequences of the internal transcribed spacer ribosomal RNA locus (ITS), partial translation elongation factor 1-alpha (tef1), β-tubulin 2 (tub2) and DNA-directed RNA polymerase II subunit (rpb2) genes, and combined with their morphological characteristics. Eleven species were identified, including Botryosphaeria fusispora, B. wangensis, Lasiodiplodia pseudotheobromae, Neofusicoccum kwambonambiense, N. parvum, and six novel species described as B. puerensis, N. dianense, N. magniconidium, N. ningerense, N. parviconidium and N. yunnanense. The dominant species across the regions were N. yunnanense, N. parvum and B. wangensis, representing 31.3, 25.3 and 19.9% of the total isolates, respectively. Species diversity and composition changed across the different climatic zones, despite their relatively close geographic proximity and the fact that some of the species have a global distribution. All the Botryosphaeriaceae species were pathogenic to one-year-old plants of an E. urophylla × E. grandis clone and E. globulus seed-derived plants, but showed significant inter- and intra-species variation in aggressiveness amongst isolates. The study provides a foundation for monitoring and management of Botryosphaeriaceae through selection and breeding of Eucalyptus in the YunNan Province of southwestern China.

Constraining the duration of magmatism is of vital importance to the understanding of the magmatic-hydrothermal mineral system. The Bozhushan batholith, located in the middle section of the southeastern Yunnan ore district, mainly consists of biotite monzogranite and monzogranite. Many Sn–W–polymetallic deposits are developed around the Bozhushan batholith, but their temporal and genetic relationships remain controversial. LA-ICP-MS U–Pb zircon and monazite dating were respectively conducted on the same two samples, yielding weighted mean 206Pb/238U zircon ages of 85.1 ± 0.7 and 85.6 ± 0.9 Ma, and weighted mean 206Pb/238U monazite ages of 87.1 ± 0.9 and 88.1 ± 1.1 Ma. The crystallization ages of S-type granites obtained from the zircon U–Th–Pb system and monazite U–Th–Pb system are consistent within the analytical errors. After combining the new ages obtained in this study with recently published U–Pb zircon and cassiterite ages from the giant Baniuchang Ag–Sn–Pb–Zn deposit in the north, and U–Pb zircon and Re-Os molybdenite ages from the large Guanfang W deposit in the south, a temporal framework of magmatism-mineralization in the Bozhushan region has been established. The duration of magmatic activity at Bozhushan is about 7 Ma, with W mineralization occurring at ca. 92 Ma and Sn mineralization at 88–87 Ma.

Geohazards occur in south west mountain area of China each year frequently. Therefore, selecting a typical region to establish a geohazard early warning system and to research the mechanisms of geohazard can greatly improve the ability of geohazard prevention in that mountain area, and then a geohazard early warning system is established at Huaying Mountain region. This article not only presents the construction of that system, but also elaborately analyses mechanism of some geohazards in that mountain region.
