Abstract Background Imidacloprid, a neonicotinoid insecticide, has been associated to severe reproductive toxicity in mammals. Although some preventive measures have been reported, curative strategies are yet to be explored. The present study was designed to investigate the alleviating effects of Lannea acida on the reproductive toxicity of colibri®, a commercial formulation of imidacloprid, in adult male rats. Materials and methods Seventy rats were orally administered with colibri® (22.5 mg/kg, 10 mL/kg) for 14 days and treated for other 14 or 28 days with either aqueous or methanol extracts of L. acida (170 or 340 mg/kg). Control animals were similarly treated with clomiphene citrate or vitamin E. Sexual organ weights, spermatozoa characteristics, sexual hormones, stress markers and testis histology were evaluated at the end of each treatment period. Results Colibri® exposition induced reproductive toxicity marked by a decrease in sex organ weights, spermatozoa count, motility and viability. Colibri® also decreased testosterone, luteinizing hormone, follicle stimulating hormone concentrations and increased testicular oxidative stress. Spermatozoa morphology and testis histology were also severely altered. Similar to clomiphene citrate and vitamin E, treatment with L. acida extracts significantly (p ≤ 0.05–0.001) reversed the above-mentioned damages, especially after 28 days of treatment with aqueous (340 mg/kg) and methanol (170 mg/kg) extracts. Conclusion Present results indicate that L. acida exerts curative effects against colibri®-induced male reproductive toxicity. These results justify the use of this plant as fertility enhancer and suggest that it could be an alternative in the management of pesticide-derived male infertility.

The present study investigated the protective effect of kolaviron, a biflavonoid from the seed of Garcinia kola, on ethylene glycol monoethyl ether (EGEE)–induced reproductive toxicity in male rats. The protective effect of kolaviron was validated using vitamin E, a standard antioxidant. EGEE was administered at a dose of 200 mg/kg. Other groups of rats were simultaneously treated with kolaviron (100 and 200 mg/kg) and vitamin E (50 mg/kg) for 14 days. EGEE treatment resulted in significant decrease in glutathione (GSH) level, superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) activities but markedly increased the glutathione-S-transferase (GST) and lactate dehydrogenase (LDH) activities in the testes. In the spermatozoa, administration of EGEE caused significant decrease in the activities of CAT, GPx, GST and LDH as well as in the level of GSH but significantly increased SOD activity with concomitant increase in hydrogen peroxide and malondialdehyde levels in both testes and spermatozoa. EGEE-exposed rats showed marked testicular degeneration with concomitant decrease in spermatozoa quantity and quality. Overall, EGEE causes reproductive dysfunction in rats by altering antioxidant systems in the testes and spermatozoa. Kolaviron or vitamin E exhibited protective effects against EGEE-induced male reproductive toxicity by enhancement of antioxidant status and improvement in spermatozoa quantity and quality.

Crude oil intoxication is a major threat among people and animals living around the crude oil producing regions of the world, hence the search for ameliorating agents. Forty-four male Wistar rats assigned into three groups were used to investigate the effects of vitamin E supplementation on crude oil-induced reprotoxicity (reproductive toxicity) in male rats. Group A represented the unexposed control, whereas groups B and C were exposed orally to 0.15 and 0.3 ml of crude oil respectively every other day for 56 days. Both the low dose and high dose oral administration of crude oil caused a significant reduction in the serum testosterone level (STL) and cauda epididymal sperm reserve (CESR) of the exposed rats when compared to the control. Crude oil withdrawal and vitamin E supplementation significantly improved the cauda epididymal sperm reserve (CESR) in all the subgroups. The serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities of the control and low dose group were significantly lower than those of the high dose group. The high dose crude oil administration significantly decreased the mean serum total protein (STP) and sodium ions (Na+) concentration. The mean serum total cholesterol (STC) value of the low dose group was significantly higher than those of the control and high dose group. However, crude oil withdrawal and vitamin E supplementation did not significantly alter the mean serum total protein (STP) and mean serum total cholesterol (STC) values in all the subgroups. Vitamin E supplementation following low dose crude oil withdrawal enhanced the mean serum Chloride ions (Cl-)concentration. The present findings revealed that Nigerian Qua Iboe Brent crude oil induced serious reprotoxic effects in male rats which vitamin E administration within 28 days did not completely reverse.

Monosodium glutamate (MSG) is one of the most common food additives extensively used as a flavor enhancer. MSG induced lipid peroxidation and inflammation. The present study aimed to assess the neurotoxicity, testicular impairment, inflammation and apoptosis induced by MSG. Thirty adult male Wistar rats, weighing about 180-200 g were assigned equally into five groups, each consists of six rats. Animals of Group I are controls and they received distilled water, whereas animals of Groups II, III, IV and V were given oral daily doses of MSG 0.8, 1, 2 and 3 g/kg body weight, respectively for consecutive 70 days. Administration of different doses of MSG revealed a significant elevation in the levels of malondialdehyde (MDA), nitric oxide (NO), β amyloid 1-42, proinflammatory cytokines (IL-6, TNF-α), cholesterol and sperm abnormality while it showed reduction in the level of GSH and SOD, CAT and GST antioxidant enzymes activities, sperm count and sperm motility. MSG led to disruption in neurotransmitter levels; serotonin, norepinephrine, glutamate and GABA, also disorders in sexual hormones; testosterone, FSH and LH. The present results were confirmed by histological and immunohistochemical observations that obviously designate the neurodegeneration and reproductive toxicity. In conclusion, administration of low and high doses of MSG provoke deleterious effects on oxidant/ antioxidant markers, neurotransmitters, inflammatory cytokines, sexual hormones, brain and testes structures. Prominence hazards of lasting exposure to low and high doses of MSG on neuronal and testicular health. Therefore, its use should be restricted.

Obesity is associated with renal lipomatosis, inflammation and oxidative stress. Vitamin E is a liposoluble antioxidant vitamin and PLGA (polylactic-CO-glycolic acid) nanoparticles (NPs) represent a delivery carrier of this vitamin to the abdominal viscera. The aim of this study is to evaluate the renal and systemic effects of PLGA- NPs loaded with vitamin E in high fat diet Wistar rats. Young heathy Wistar male rats (3months old, n=20) were fed for 4 weeks either with standard diet or with high caloric/fat diet. These two experimental groups were divided in two subgroups and for the following 21 days, one group received PLGA-NPs loaded with vitamin E (1 mg/kg) body and the other didn�t. Metabolic serum parameters, oxidative stress markers ( malondialdehyde, MDA and total thiols) from kidney homogenates were measured by spectrophotometric methods. Hematoxylin-eosin stain histopathologic examination of kidneys was performed. The obesogenic diet increased the serum levels for triglycerides (p[0.001) and renal function markers: urea (p[0.02), creatinine (p[0.02) and cystatin (p[0.01). The treatment reduced the kidney inflammation, prevented tubulonephrosis damage and improved the renal oxidative stress markers (p[0.05) in the high fat diet group. In conclusion, PLGA-NPs loaded with vitamin E had renoprotective effect in high fat diet Wistar rats.

Varicocele is a disease characterized by an abnormal dilation of the pampiniform plexus that drains the testis. The main objective of this work was to evaluate the curative effects of aqueous and ethanolic extracts of Dracaena arborea on some reproductive and antioxidant markers in rats with experimental varicocele. Following varicocele induction, rats (5 per group) were randomly partitioned into untreated varicocele, vitamin E-treated (150 mg/kg), aqueous extract-treated (500 mg/kg), and ethanolic extract-treated (100 mg/kg) animals. Two other groups served as normal and sham-operated. After 2 or 4 weeks of treatments, body and sex organ weights, spermatozoa characteristics, antioxidant status, NO level, sex hormones, and testis histology were measured. Animals with 3 weeks of varicocele showed a significant ( p  < 0.05–0.001) decrease in body and sex organ weights, total proteins, sperm characteristics, testosterone concentration, SOD, catalase, and total peroxidase activities. An increase in the plasmatic FSH, LH, and testicular MDA and NO concentrations was also recorded. Moreover, marked disorganization of the testicular architecture was observed. Treatment with D. arborea significantly reversed these impairments due to varicocele. For instance, after 4 weeks, treatment with aqueous extract of D. arborea significantly ( p  < 0.05–0.001) increased testes and epididymis weights, sperm viability (89.12 ± 1.09 vs 68.22 ± 1.42), sperm density (148.50 ± 2.59 vs 110.25 ± 2.51), and sperm motility (68.16 ± 2.39 vs 55.88 ± 3.20) in the left side, compared with varicocele-untreated rats. The extract also significantly ( p  < 0.05–0.001) decreased malondialdehyde level (2.19 ± 0.04 vs 3.50 ± 0.13) but elevated catalase (0.97 ± 0.03 vs 0.55 ± 0.03), SOD (0.5 ± 0.03 vs 0.15 ± 0.03), and peroxidase (65.80 ± 2.9 vs 40.95 ± 2.44) activities. Present results showed that D. arborea extracts possess antioxidant effects and improve sperm quality in male rats with an existing varicocele.
