Abstract Lactobacilli are probiotics with Aflatoxin (AF) detoxification ability, found in fermented products, GIT of animals and environment. Purpose of this study was to investigate the ability of broiler isolates of Lactobacillus against Aflatoxin B1 (AFB1). For this purpose, 5 isolates of Lactobacillus from broiler gut were incubated with 100 ppb AFB1 in aqueous environment and effect of different parameters (cell fractions, time, temperature, pH) on detoxification was determined by HPLC. The ameliorative effect of Lactobacillus salivarius (LS) against AFB1 was studied in broiler. The results revealed that LS (CR. 4) showed the best results (in vitro) as compared to other isolates (L. salivarius (CR. 3, CR, 4), L. agilis (CE. 2.1, CE. 3.1) and L. crispatus (CE. 28). Cell debris of CR. 4 showed significantly higher detoxification (P<0.05). Maximum amount of AFB1 was detoxified at 30°C (97%), pH 4.0 (99%) and 6 h (99.97%). In vivo study showed that AFB1 decreased weight gain (1,269 ± 0.04 gm/ bird), feed consumed (2,161 ± 0.08 gm/ bird), serum total protein (2.42 ± 0.34 gm/ dl), serum albumin (0.5 ± 0.2 2 gm/dl) and antibody titer (4.2 ± 0.83). Liver function enzymes were found (alanine transaminase (ALT): 32 ± 10.7 U/L) and aspartate transaminase (AST): 314.8 ± 27 U/L) elevated in AFB1 fed broilers. Treatment with 1% LS not only decreased the toxic effects of AFB1 (group D) but also improved the overall health of broilers due to its probiotic effects (p<0.05) as compared to control negative (group A). The detoxification ability of LS was better than commercial binder (CB) (0.2% Protmyc). It was concluded that detoxification of AFB1 by Lactobacillus was strain, temperature, pH and time dependent. LS has detoxification ability against AFB1 in vivo.

Abstract Immunoglobulin (Ig) G and natural antibody (NAb) IgM are passively transferred to the neonatal calf through bovine colostrum. Maternal IgG provides pathogen- or vaccine-specific protection and comprises about 85 percent of colostral Ig. Natural antibody IgM is less abundant but provides broad and non-specific reactivity, potentially contributing to protection against the dissemination of pathogens in the blood (septicemia) in a calf’s first days of life. In the dairy and beef industries, failure of passive transfer (FPT) of colostral Ig (serum total protein (STP) &lt;5.2 g/dL) is still a common concern. The objectives of this study were to, i) compare colostral IgG concentrations and NAb-IgM titers between dairy and beef cows; ii) assess the effect of beef breed on colostral IgG; iii) compare passive transfer of colostral Ig in dairy and beef calves; and iv) estimate the heritability of colostral IgG and NAb-IgM. Colostrum was collected from Holstein dairy (n=282) and crossbred beef (n=168) cows at the University of Guelph dairy and beef research centres. Colostral IgG was quantified by radial immunodiffusion and NAb-IgM was quantified by an enzyme-linked immunosorbent assay. In dairy (n=308) and beef (n=169) calves, STP was estimated by digital refractometry. Beef cows had significantly greater colostral IgG (146.5 ± 9.5 SEM g/L) than dairy cows (92.4 ± 5.2 g/L, p &lt;0.01). Beef cows with a higher proportion of Angus ancestry had significantly lower colostral IgG (125.5 ± 5.8 g/L) than cows grouped as “Other” (142.5 ± 4.9 g/L, p= 0.02). Using the FPT cut-off, 13% of dairy and 16% of beef calves had FPT; still, beef calves had a significantly larger proportion with excellent passive transfer (STP ≥6.2 g/dL, p &lt;0.01). The heritability of colostral IgG was 0.04 (± 0.14) in dairy and 0.14 (± 0.32) in beef. Colostral NAb-IgM titers in dairy (12.12 ± 0.22, log2 (reciprocal of titer)) and beef cows (12.03 ± 0.19) did not differ significantly (p=0.71). The range of NAb-IgM titers was 9.18 to 14.60, equivalent to a 42-fold range in antibody concentration. The heritability of colostral NAb was 0.24 (± 0.16) in dairy and 0.11 (± 0.19) in beef cows. This study is the first to compare colostral NAb-IgM between dairy and beef cows. Based on the range in NAb-IgM titers and the heritability, selective breeding may improve colostrum quality and protection for neonatal calves in the early days of life.

The study investigated the effect of stem bark hydroalcoholic extract of cocoa (Theobroma cacao) on liver function of phenlyhydrazine (PHZ)-induced forty anaemic (treated) and normal albino rats divided into 8 groups of five rats each in the study. The activities of marker enzymes such as aspartate amino transferase (AST), alanine amino transferase (ALT) and alkaline phosphatase (ALP), were assayed. In addition serum total protein, total bilirubin, albumin were determined. There was a significantly (p<0.05) decrease in AST of anaemic not treated rats compared to normal control rats with the exception of serum total protein, total bilirubin, direct bilirubin, albumin, ALT and ALP where no significant (p>0.005) change was recorded. In the group (anaemic rats) treated with bark extract of Theobroma cacao at the doses of 100, 200, and 500mg/kg, AST was significantly (p<0.05) reduced while there was significant (p>0.05) increase in serum total protein, total bilirubin, albumin, ALT and ALP. In the group (non anaemic rats) treated with graded doses of the extract 200mg, 500mg and 1000mg, there was a significant (p<0.05) of total bilirubin diect bilirubin, and AST while no significant (p>0.05) change was recorded. The study illustrate that the bark extract of Theobroma cacao possess antioxidant and membrane permeability properties. Dose of Theobroma cacao at 500mg does not have antianaemic potentials.

AbstractOutcome of allogeneic stem cell transplantation (alloSCT) is hampered by substantial non-relapse mortality (NRM). Given its impact on organ function and immune response, the nutritional status has been suggested as relevant for NRM. We aimed to evaluate the association of NRM with nutritional status prior to alloSCT and in the post-SCT course. In a retrospective single-center study, we analyzed 128 alloSCTs. Besides standard characteristics, nutrition-associated parameters BMI, serum total protein, and serum albumin were recorded before conditioning and at various time points after alloSCT. Association with NRM was evaluated by univariate and multivariate survival analysis. The cohort comprised patients with a median BMI of 26 kg/m2 (16.7–46.9 kg/m2), median serum total protein of 59 g/l (41–77 g/l), and serum albumin of 36 g/l (22–46 g/l) before SCT. NRM at d+100 was 14.8% and at 1 year 26.6%. Prior to SCT, only serum albumin deficiency was associated with increased NRM (p = .010) in multivariate analysis. After SCT (d+30 and d+100), all nutrition-associated parameters decreased (p < .002), but no association of deteriorating nutritional status with NRM was found. In multivariate analysis, serum albumin (p = .03) and severe albumin deficiency (p = .02) correlated with NRM at d+30 and d+100, while BMI and serum total protein did not. In our study, albumin deficiency, particularly prior to alloSCT, shows a strong correlation with NRM. This finding may add to monitoring, risk evaluation, and counseling of patients and serve as a rational for interventions to improve the nutritional status in patients undergoing SCT.

The study evaluated the haemato-biochemical alterations and acute toxicity of hydro-methanol root extract of Anarcaduim occidentale (AO) in cockerels. Forty chickens randomly assigned into four groups (A, B, C and D) of 10 birds each were used for the experiment. Groups A, B, C and D received 3000, 1500 and 500 mg/kg body weight of the extract and distilled water respectively. Relative weights, haematology, serum biochemistry, gross and histopathological features of the lymphoid organs of chickens were studied. No significant (P>0.05) variation was observed in their body weights. The white blood cell count of group A chicken were significantly (P<0.05) lower than that of groups B and C chickens. Serum biochemistry tests revealed significant (P<0.05) variations at different doses for the alanine aminotransferase, alkaline phosphatase, serum total protein, albumin, cholesterol, triacylglycerol, uric acid, creatinine, calcium and phosphorus activities when compared to the control group.  Gross and histopathological examinations showed no overt alterations in the tissues examined. It was observed that varied doses of root extract of Anarcaduim occidentale caused significant decrease in haematology and serum biochemistry parameters of chickens and no obvious lesions in internal organs.

The aim of this study was to assess and analyze the nutritional status (NS) and immune status of pancreatic cancer (PC) patients. The retrospective analysis included 80 PC patients undergoing curative pancreatic resection in the Department of Digestive Tract Surgery of the Medical University (Katowice, Poland). Patients were divided by the tumor location (proximal vs. distal), age (≤65 years vs. >65 years), Nutritional Risk Score 2002 (NRS 2002) (<3 vs. ≥3), prognostic nutritional index (PNI) (<45 vs. ≥45), and the presence of postoperative complications (no-complication vs. complication) as well as the use of neoadjuvant chemotherapy (no neoadjuvant chemotherapy vs. neoadjuvant chemotherapy) into two subgroups, which were compared. Significantly higher weight loss was related to the proximal tumor location (p = 0.0104). Significantly lower serum total protein (p = 0.0447), albumin (p = 0.0468), hemoglobin (p = 0.0265) levels, and PNI (p = 0.03) were reported in older patients. The higher nutritional risk according to NRS 2002 was significantly associated with higher age (p = 0.0187), higher weight loss (p < 0.01), lower body mass index (BMI) (p = 0.0293), lower total lymphocyte count (p = 0.0292), longer duration of hospitalization (p = 0.020), neoadjuvant chemotherapy (p < 0.01), and preoperative biliary drainage (p = 0.0492). The lower PNI was significantly associated with higher weight loss (p = 0.0407), lower serum total protein and albumin concentration, lymphocyte count (p < 0.01) and higher neutrophil/lymphocyte (NLR), monocyte/lymphocyte (MLR), platelet/lymphocyte (PLR) ratios, and duration of hospitalization (p < 0.01). In the multiple logistic regression analysis, BMI ≥ 30 kg/m2 (OR: 8.62; 95%CI: 1.24–60.04; p = 0.029521) and NRS 2002 ≥ 3 (OR: 2.87; 95%CI: 0.88–9.33; p = 0.048818) predicted postoperative complications. In the multiple linear regression analysis, the higher NRS 2002 score was linked with the longer duration of hospitalization (b = 7.67948; p = 0.043816), and longer duration of postoperative hospitalization was associated with a higher complication rate (b = 0.273183; p = 0.003100). Nutritional impairment correlates with a systemic inflammatory response in PC patients. Obesity (BMI ≥ 30 kg/m2) and malnutrition (NRS 2002 ≥ 3) predict postoperative complications, which are associate with a longer hospital stay. Assessment of nutritional and immune status using basic diagnostic tools and PNI and immune ratio (NLR, MLR, PLR) calculation should be the standard management of PC patients before surgery to improve the postoperative outcome.

Abstract Few data are available on the descriptive characteristics of colostrum quality, passive transfer status and growth performance within a national population in China. This study included 110 dairy operations in 23 provinces, and data were obtained from management software, report system and questionnaires from 2015 to 2019. Data were analyzed using SAS (version 9.0, SAS Institute Inc., Cary, NC, USA). The results showed that colostrum quality was measured in 96.4% of farms and colostrum was pasteurized in 91.8% of farms. 83.6% of colostrum was excellent (Brix &gt; 22%). Colostrum could be fed to calves within 1 hour after birth in 86.4% of farms. Besides, calves that experienced failure of passive transfer (serum total protein &lt; 5.2 g/dl) decreased from 2015 to 2019 and accounted for 0.75% of calves in 2019. Most farms (48.8%) feed 400 kg - 600 kg milk during the preweaning period and whole milk was the main type of liquid diet accounting for 70.6% of farms. Meanwhile, 22.0% of farms chose to feed forage before weaning, and 90.1% of these farms fed oat hay. The average birth weight of calves was 37.8 kg and the average daily gain of preweaning calves was 848.3 g/d. For preweaning calves, the proportion of single housing was more than 60%, whereas postweaning calves were mainly raised in groups. The morbidity of preweaning calves in China decreased annually, from 43.3% in 2015 to 25.4% in 2019. Digestive diseases and respiratory diseases were two main diseases, and accounted for 46.0% and 38.4%, respectively. This study provides an overview on dairy calves raising practices in China.

Six dogs each with open and closed-cervix pyometra (Group I and II) were medically managed with mifepristone @ 2.5mg/kg body weight orally twice daily for five days, followed by cloprostenol @ 5?g/kg body weight subcutaneously on every alternate day after ensuring cervical patency, till complete evacuation of uterus was assessed by ultrasonography. Haematological and serum biochemical values were estimated on the day of presentation. The treatment response was assessed by reviewing both haematological and serum biochemical values further on days 3, 7, 14 and 21 of treatment. The mean total erythrocyte count (TEC), haemoglobin concentration and volume of packed red cells (VPRC) were significantly lower and mean total leucocyte count (TLC) was significantly higher in all animals of both groups during the day of presentation. The values significantly improved after initiation of treatment. Total thrombocyte count, serum total protein, albumin, BUN and serum creatinine levels were within the normal range. TEC, TLC, Hb concentration, VPRC could be used for prognostic markers of treatment evaluation in canine pyometra.

Phytobiotics with a mycotoxin adsorbent were used to mitigate negative effects of multiple mycotoxins in diets fed to pigs. In experiment 1, 120 pigs (11.6 kg body weight; BW) were assigned to five treatments (three pigs/pen) and fed for 28 days. Treatments were CON (control), MTD (CON + 2.5 mg/kg of deoxynivalenol), DP (MTD + phytobiotics at 0.1%), and DPA1 and DPA2 (MTD + phytobiotics and adsorbent at 0.1% and 0.2%, respectively). In experiment 2, 96 pigs (28.5 kg BW) were assigned to four treatments (three pigs/pen) and fed for 26 days. Treatments were CON, MTAF (CON + 0.19 mg/kg of aflatoxin and 8 mg/kg of fumonisins), AFP (MTAF + phytobiotics at 0.1%), and AFPA (MTAF + phytobiotics and adsorbent at 0.1%). Growth performance was measured weekly, and blood was sampled at the end of study to measure hepatic function and inflammatory status (TNF-α). Data were analyzed using the MIXED procedure. In experiment 1, pigs fed MTD, DP, DPA1, and DPA2 had smaller (p < 0.05) BW than CON. Pigs fed DPA2 had greater (p < 0.05) BW than MTD. Pigs fed DP and DPA2 tended to have lower (p < 0.1) serum total protein than CON. Pigs fed MTD and DPA2 tended to have higher (p < 0.1) alanine aminotransferase than CON. Similarly, pigs fed MTD, DP, and DPA2 tended to have higher (p < 0.1) urea nitrogen/creatinine than CON. In experiment 2, pigs fed MTAF, AFP, and AFPA had smaller (p < 0.05) BW than CON. Pigs fed MTAF, AFP, and AFPA had smaller (p < 0.05) ADFI than CON. Pigs fed AFPA had higher (p < 0.05) aspartate aminotransferase than CON and MTAF. Pigs fed AFP and AFPA had higher (p < 0.05) alanine aminotransferase than CON. Pigs fed MTAF, AFP, and AFPA had lower (p < 0.05) urea nitrogen/creatinine than CON. Pigs fed AFPA had higher (p < 0.05) TNF-α than CON and MTAF. In conclusion, feeding an additional 2.5 mg/kg of deoxynivalenol or 0.19 mg/kg of aflatoxin with 8 mg/kg of fumonisins reduced the growth of pigs. Deoxynivalenol compromised the hepatic function of pigs. Phytobiotics with adsorbent could partly overcome the detrimental effects of mycotoxins.

The enzyme activities of ALT, AST, and ALP were significantly reduced by WY, YCW, and YE supplementation. The relative organ weights of the spleen, bursa of Fabricius, and thymus increased significantly in broilers fed with WY, YCW, and YE, and the highest values were observed in the chickens fed with WY. It can be demonstrated that the supplementation of WY or its derivatives in the diet of broiler chickens improves the production performance as well as the physiological and immunological parameters, and consequently produce a healthier chicken
