Abstract Background Euphorbia helioscopia, conventionally known as sun spurge, has been used as a traditional medicine to treat different diseases owing to its reported antitumor, antiviral and antioxidant activities. Methods The current research was formulated to assess the in-vitro antioxidant and antidiabetic ability of Euphorbia helioscopia subsequent to the phytochemical analysis of its various extracts. For this purpose, methanol, ethanol and aqueous extracts were prepared using the whole dried plant. Phytochemical analysis of the extracts was done to evaluate the total flavonoid components (TFC) and total phenolic components (TPC) in the extracts. A total of seven phenolic and three flavonoid contents were documented and quantified using HPLC. Antioxidant values were found by DPPH● assay, FRAP and ABTS assays. The antidiabetic potential of the extracts was evaluated by measuring the inhibition ability of the activity of enzymes α amylase and α glucosidase. Results After analyzing statistically, the results showed that methanolic extract possesses the highest TFC and TPC values while aqueous extract encompassed the lowest level of these contents. Invitro results showed that methanolic extract of the Euphorbia helioscopia has the maximum antioxidant capability since it showed the highest scavenging ability towards the DPPH● (IC50 value = 0.06 ± 0.02 mg/ml), FRAP (758.9 ± 25.1 μMFe+ 2/g), and ABTS (689 ± 25.94 μMTEq/g) due to the presence of high TPC (24.77 ± 0.35 mgGAEq/g) and TFC (17.95 ± 0.32 mgQEq/g) values. Antidiabetic activity in terms of inhibition potential of α amylase and α glucosidase activity was also observed maximum in methanolic extract having lowest IC50 value (0.4 ± 0.01 mg/ml and 0.45 ± 0.01 mg/ml respectively) and minimum in the aqueous extract (IC50 value = 0.57 ± 0.02 mg/ml and 0.76 ± 0.1 mg/ml respectively). Conclusion The experiment outcomes have shown that Euphorbia helioscopia extracts used in the current study contain antioxidant and antidiabetic activities; however, it is highest in its methanolic extract. The presence of the same trend towards the highest antidiabetic activity of the methanolic extract in terms of maximum inhibiting activity of α amylase and α glucosidase enzymes suggests a close association of TFC and TPC in minimizing diabetes.

Objective: To investigate the presence of phytochemical components and to evaluate the in vitro antioxidant activity of pet. ether, chloroform, methanol and aqueous extracts of Grewia heterotricha mast leaves.Methods: The leaves of Grewia heterotricha mast were dried and extracts were prepared using a pet. ether, chloroform, methanol by soxhlet extraction method. The aqueous extract was prepared using distilled water by cold extraction method. The preliminary phytochemical analysis was carried out on aqueous, methanol, chloroform and pet. either leaf extracts of the plant using standard qualitative procedures. The total phenolic content (TPC) was estimated using modified Folin-Ciocalteau method, tannin content by Folin-Denis method and total flavonoids by aluminum chloride method. In vitro, antioxidant activities were evaluated by 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, H2O2 scavenging activity and FRAP assay.Results: The preliminary phytochemical analysis revealed the presence of complex bioactive constituents like phenols, tannins, alkaloids, terpenoids, flavonoids, saponins, steroids, glycosides, coumarins, proteins and carbohydrates. Methanolic extract showed highest total phenolic content (87.58±2.52 mg CE/g) than aqueous extract (78.46±5.36 mg CE/g). Higher tannin content was found in the aqueous extract (148.0±8.96 mg TAE/g). Total flavonoids were highest in chloroform extract (314.9±25.06 mg QE/g) followed by aqueous (242.98±32.42 mg QE/g) and methanolic extract (217.0±18.32 mg QE/g) and lowest in a pet. ether extract (188.86±23.35 mg QE/g). The methanolic extract had shown very significant DPPH radical scavenging activity (IC50 98.95 µg/ml) and H2O2 scavenging activity (IC50110.1µg/ml) compared to the standard ascorbic acid. Higher reducing ability was observed in methanol extract (131.8±11.67 mg AE/g).Conclusion: The results obtained reveal that the leaves of Grewia heterotricha mast have potent antioxidant property. The observed activity may be associated with bioactive components like phenolics, flavonoids present in the leaf extracts and could have greater importance as therapeutic agents in oxidative stress-related degenerative diseases. Further studies are needed in order to purify bioactive compounds responsible for the antioxidant property.

Abstract Background Cleome viscosa is considered as an important medicinal plant extensively used in India, China, Bangladesh, and a few countries in Africa. In the present study, in vitro anti-radical and antidiabetic potential of isolated fractions of methanolic extract of C. viscosa whole plant (MeCV) has been investigated. The identification of polyphenols and their related functional groups in the bioactive fraction was categorized by using HPLC and FT-IR. Results The total phenolic and flavonoid contents of F-D were higher than those of F-A, F-B, and F-C. The F-D exhibited superior antioxidant capacity when compared with the remaining three fractions. However, the F-D showed the highest glucose diffusion activity over the 30 min–27 h incubation period and also inhibited both α-glucosidase and α-amylase enzyme activity. HPLC analysis revealed the presence of the two known compounds (protocatechuic acid hexoside, rutin) and six unknown compounds in the F-D. FTIR spectrum confirmed the presence of phenol group. Conclusion The isolated F-D obtained from MeCV displayed superior antioxidant and antidiabetic activity which indicate the presence of polyphenols in the fraction. The data findings of the present study support the traditional uses of the whole plant of C. viscosa as a promising natural source of biological medicines for oxidative stress and diabetes.

In traditional systems of medicine, the bark of Dolichandroneatrovirens has been used to treat various disorders. The main aim of this study was to determine the anti-inflammatory effect of D. atrovirens bark through the inhibition of nitric oxide (NO) production in lipopolysaccharide-stimulated RAW 264.7 cells.For this purpose, preliminary phytochemical composition and in vitro antioxidant activities of various solvent extracts of D. atrovirens bark were evaluated to select the most effective extract.The methanol extract of D.atrovirens registered the highest amount of total phenolics (476.2 mg GAE/g) and flavonoid (129.0 mg RE/g)contents witha strong antioxidant activity as measured in DPPH (IC50 of 19.52 μg/mL) and ABTS (IC50of 10.82 μg/mL) scavenging activities.Hence, the methanol extract was selected for cell line study. Further, the methanol extract of D. atrovirens effectively inhibited the production of NO in RAW 264.7 cells induced by LPS (13.1 μM at the concentration of 80 μg/mL). It could be concluded that the presence of higher level of total phenolic components in the methanol extract of D. atrovirens bark might be responsible for reducing the NO level in cells.

Oxidative stress plays a major role in diabetic complications. The study aim was to investigate the in-vitro antidiabetic and antioxidant activities of methanolic extract of Canthium coromandelicum leaves. The plant material was extracted with methanol and the methanolic extract was screened for in-vitro antioxidant activity using 1, 1-diphenyl-2-picryl hydrazyl (DPPH) assay. The efficiency of the antidiabetic activity of the plant extract was evaluated against α-amylase and α-glucosidase digestive enzymes. The study revealed that the C. coromandelicum extract exhibited significant α-amylase and α-glucosidase inhibitory activities with an IC50 value of 31.52 ± 0.42 and 41.49 ± 0.28 µg/mL respectively and compared with standard acarbose drug. The extract efficiently scavenging DPPH radical with IC50 values of 65.46 ± 0.50 µg/ml. Therefore, the extract could be a promising therapeutic in management of diabetic complications.

Objective: To standardise the protocol for rapid callogenesis in Mussaenda frondosa L. using leaf explants. Qualitative and quantitative phytochemical analysis of leaf, stem and callus cultures.Methods: The leaf explants were inoculated onto MS medium supplemented with varying concentrations of growth regulators such as 2, 4 - D, NAA, BAP, Kn for the induction of callus. Qualitative and quantitative analysis of total phenol, flavonoids and alkaloids contents of leaf, stem and callus were tested by standard methods.  The antioxidant activities were investigated using DPPH radical scavenging method and reducing power assay. The anti - inflammatory activity was evaluated by membrane stabilizing activity.Results: Pale green, healthy, friable and fast growing callus was obtained on the medium enriched with NAA (2mg/l) + Kn (4mg/l). Quantitative determination showed the highest concentration of total phenolics in the methanolic extract of in vitro grown callus (10 ± 1.1 mg of GA/g of extract), flavonoids in methanolic stem extract (137±1.6 mg of Quercitin/g of extract) and alkaloids in methanolic extract of leaf (118.3±1.5 mg/10g of extract). The methanolic leaf extract exhibited highest free radical scavenging activity with IC50 value of 40.6±10.06 μg/ml. The highest membrane stabilizing activity was shown by chloroform extract of the leaf (66.02%).Conclusion: The present preliminary phytochemical and pharmacological analysis may form the basis for drug development in future using callus cultures of M. frondosa.

Introduction: High blood pressure is a major risk factor for cardiovascular diseases which is one of the leading causes of death worldwide. Piper sarmentosum (PS) leaves have been widely used in traditional medicine and it has been proven for its antihypertensive and antioxidant effects. This study aims to evaluate the antihypertensive potential of PS aqueous extract (PSAE) and to investigate the factors modulating nitric oxide (NO) production through its anti-oxidant activities. Materials and Methods: PS leaves were extracted with distilled water and freezedried. The PSAE was examined to quantify their antioxidant activities through DPPH and FRAP test. It is also screened for total phenolic and flavonoids content. The antihypertensive effect of PSAE in SHR was evaluated using four different groups (n=6); C group (negative control), K group (PSAE 500mg/kg), P group (3 mg/kg perindopril) and M group (PSAE + 1.5 mg/kg perindopril). PSAE and other treatment were given via oral gavage for 28 consecutive days. The blood pressure and heart rate were determined using the non-invasive blood pressure monitoring (NIBPM) tail cuff technique and recorded weekly for four weeks. SHR’s blood was collected for the determination of serum NO level using Griess assay. Asymmetric dimethylarginine (ADMA) and arginine levels were determined using HPLC. Results: PSAE shows good in-vitro antioxidant activities with significant reduction of BP and decreased serum ADMA level. Conclusion: PSAE enhances the clearance of ADMA that maintains arginine level, thus increasing the serum NO level, which ameliorate the blood pressure of SHR.

AbstractTaraxacum officinale F.H. Wigg (Asteraceae) root is traditionally used to treat diabetes, dyspepsia, heartburn, anorexia and hepatitis. In this work, petroleum ether, chloroform, methanol and aqueous extracts of T. officinale root were evaluated for their antidiabetic activity in normoglycemic and alloxan-induced diabetic mice at two concentrations (200 and 400 mg/kg) using antidiabetic and subcutaneous glucose tolerance tests. Herein, in vitro glucose uptake assay was performed using HepG2 and 2-NDBG, while LC-MS/MS was employed for the phytochemical study of the main active constituents in the active extract. In the experiments, T. officinale root aqueous extract (400 mg/kg) showed a significant decrement in blood glucose level (62.33%, p ≤0.05), while other extracts (p >0.05) showed insignificant activity – in alloxan-induced diabetic mice with no apparent effect on the normoglycemic model. The extracts also showed an insignificant reduction in glucose levels (p >0.05) in the subcutaneous glucose tolerance test. However, a significant glucose uptake enhancement (149.6724%, p ≤0.05) was exhibited by the aqueous extract. Phytochemical study of the aqueous extract showed higher total phenolic than total flavonoid content, in which chlorogenic acid, protocatechuic acid, and luteolin-7-glucoside were identified.

The phytochemical contents of the acetone, ethanol, and boiled and cold aqueous whole-plant extracts of Helichrysum petiolare were determined using standard phytochemical reaction methods. ABTS, DPPH, NO and TAC assays were used to evaluate their antioxidant properties. This study reported the highest total phenolic content (212,963 mg/g) in the boiled aqueous extract, while the ethanol had the highest flavonoid (172.393 mg/g) and proanthocyanidin contents (65.855 mg/g). Alkaloids, flavonols, and saponin were highest in the acetone extract, while the cold aqueous extract had the lowest phytochemical content. Among the extracts, the boiled aqueous extract had the highest DPPH•+ (IC50 0.02 mg/mL) and ABTS•+ (IC50 0.07) inhibition capacities, while the ethanol extract exhibited the highest NO• Inhibition (IC50 0.41 mg/mL) and TAC (IC50 0.19 mg/mL). These findings justify the use of H. petiolare in traditional medicine and further recommend the ethanol and boiled aqueous extracts of the plant as more effective extracts for medicinal treatment.

BACKGROUND: The popular commercially cultivated Pleurotus ostreatus mushroom contains very high nutrients and bioactive compounds with high antioxidant activity. The ethanolic extract seems to be the most active in preparation. AIM: This study has an aim to compare the phytochemical analysis of a fresh, dry and ethanolic extract of Pleurotus ostreatus, to measure the total phenolic content and antioxidant activities in vitro of ethanolic extracts of Pleurotus ostreatus. METHODS: The fresh plant's materials (FPM), dry plants materials (DPM), ethanolic extracts were macerated with 70% (EE70) and 96% ethanol (EE96) of Pleurotus ostreatus which were used for phytochemical analysis, and EE96 was used for antioxidant activity in vitro. The phytochemical analysis was conducted using the Dragendorf and Meyer, FeCl3 test, Salkowsky method, Lieberman method, amyl alcohol, foam test and the NaOH reagent. The total phenol test was carried out using the Follin-Ciocalteu method. The antioxidant activity was tested using the ABTS and H2O2 essay. RESULTS: The phytochemical screening showed that the flavonoid, phenolic compounds, tannin, saponin, alkaloids, and steroids were detected in the FPM, DPM, EE70 and also the EE96. The alkaloid, however, was not identified by the Meyer Reagent in the FPM and DPM. The DPM and EE70 seemed to have the highest amount of saponin based on the foam that was formed. Meanwhile, steroids and flavonoids were detected at a higher level in the EE96, based on the strength of visible colour. However, triterpenoid and quinones could not be identified. In the total phenol test, there was an amount of 6.67 μg phenol in a 1 mg extract sample which was equivalent to 1 mg of Gallic Acid. The EE96 has an IC50 of 108.07 μg/mL for ABTS and an IC50 reduction of 229.17 μg/mL. The process of Pleurotus ostreatus drying did not reduce the content of active substances. The polar active substances seem to be more soluble in the EE70 than the EE96. CONCLUSION: The higher the bioactive substances in the preparation, the more significant the bio-therapeutic effects. Ethanolic extract of Pleurotus ostreatus has a phenol content and a good antioxidant action.

A freshwater macroalga, Spirogyra spp., were analyzed for its phytochemical composition, antioxidant activity and total phenolic content (Folin−Ciocalteu method). Phytochemical analysis of Spirogyra neglecta revealed presence of phenolics, tannins, glycosides and saponins. The crude extraction of Spirogyra spp. was carried out using two solvents via, methanol (methanolic extract) and water (aqueous extract). The total phenolic contents of crude extracts were shown at 346.58±1.61 and 589.77±1.65 mg gallic acid equivalents/g extract in aqueous and methanolic extracts, respectively. The antioxidant capacity of aqueous and methanolic extract was estimated by two different methods; ABTS assay, and DPPH assay. The antioxidant activity of two extracts is affected by the extracting solvent and different assay. In the DPPH scavenging assay and ABTS assay, both extracts showed high antioxidant activity. In addition, the high correlation between results of all antioxidant activities and total phenolic content was found. The rapid TLC assay in methanolic extract is considered as the rapid test to evaluate the antioxidant activity of natural compounds. The compounds showing four bands at Rf = 0.25, 0.35, 0.42, 0.64. This study showed that Spirogyra spp. might constitute an important source of natural antioxidants.
