Abstract Background Kiwifruit (Actinidia chinensis var. Chinensis) is abundant with vitamin C and is a rapidly developing crop in China, New Zealand, and other countries. It has been widely used as a raw material for food and kiwifruit wine. Among these, A. chinensis var. chinensis and A. chinensis var. deliciosa are the most valuable kiwifruit in production. Kiwifruit is a typical dioecious plant and its female and male plants have different economic values. Therefore, sex identification, especially at the seedling stage, has important implications for the scientific planning of its production and economic benefits. However, the kiwifruit sex regulation mechanism is very complex and molecular studies are in the initial stages. Currently, there is not a universal and effective sex identification method for A. chinensis. Methods In this study, we used a label-free quantitative proteomics approach to investigate differentially accumulated proteins, including their presence/absence and significantly different levels of abundances during A. chinensis var. chinensis male and female flower bud development. Results A total of 6485 proteins were identified, among which, 203 were identified in male buds, which were mainly associated with phenylalanine metabolism, tyrosine metabolism, and plant hormone signal transduction. In female buds, 241 were identified, which were mainly associated with the ErbB signaling pathway, growth hormone synthesis, secretion and action, and mRNA surveillance pathway. A total of 373 proteins were significantly differentially accumulated proteins (fold change > 2; P < 0.05), of which, 168 were upregulated and 205 were downregulated. Significant differences between proteins involved 13 signaling pathways, most of which were involved in flavonoid biosynthesis, phenylpropanoid biosynthesis, and starch and sucrose metabolism. Protein interaction analysis showed that enriched protein nodes included cell division cycle 5-like protein, 40S ribosomal protein S8, ribosomal protein, and 40S ribosomal protein like, which interact with 35, 25, 22, and 22 proteins, respectively. Conclusions This study provide valuable information for cloning key genes that control sex traits and functionally analyze their roles, which lay a foundation to the development of molecular markers for male and female kiwifruit identification.

The endoparasitic wasp Cotesia vestalis is an important biological agent for controlling the population of Plutella xylostella, a major pest of cruciferous crops worldwide. Though the genome of C. vestalis has recently been reported, molecular mechanisms associated with sexual development have not been comprehensively studied. Here, we combined PacBio Iso-Seq and Illumina RNA-Seq to perform genome-wide profiling of pharate adult and adult development of male and female C. vestalis. Taking advantage of Iso-Seq full-length reads, we identified 14,466 novel transcripts as well as 8770 lncRNAs, with many lncRNAs showing a sex- and stage-specific expression pattern. The differentially expressed gene (DEG) analyses showed 2125 stage-specific and 326 sex-specific expressed genes. We also found that 4819 genes showed 11,856 alternative splicing events through combining the Iso-Seq and RNA-Seq data. The results of comparative analyses showed that most genes were alternatively spliced across developmental stages, and alternative splicing (AS) events were more prevalent in females than in males. Furthermore, we identified six sex-determining genes in this parasitic wasp and verified their sex-specific alternative splicing profiles. Specifically, the characterization of feminizer and doublesex splicing between male and female implies a conserved regulation mechanism of sexual development in parasitic wasps.

We report the identification of two SNPs in Cannabis sativa that are associated with female and male plant sex phenotypes, and are located on the top arm of the X chromosome. High Resolution Melt analysis was used to develop and validate a novel, rapid method for sex identification in medical/recreational cannabis as well as in hemp. This method can distinguish between dioecious male (XY) and dioecious female (XX) cannabis plants with 100% accuracy, and can also be used to differentiate between male and female Humulus lupulus (hop) plants.

Leucocoryne, a native to Chile, has violet, blue, or white flowers and is increasing in popularity as a cut flower. The effects of storage temperature and duration on flower bud development, shoot emergence, and anthesis were investigated. Bulbs stored at 20 to 30 °C for 22 weeks produced 3.4 flower stems per bulb between March and April. Bulbs stored at 20 °C flowered earliest, followed by those stored at 25 °C. Bulbs stored at 30 °C flowered last. After 16 weeks of storage at 20 °C, a further 2 weeks dry storage at 15 °C before planting resulted in 1 month earlier flowering with no reduction of the number of flowering stems. As dry storage at 20 °C increased to 11 months, the time to emergence and flowering decreased. After dry storage at 20 °C for 12 months, the primary flower stems aborted and secondary stems then developed. Secondary and tertiary flower stems tend to commence flower bud development after the flower bud on the primary flower stem has reached the gynoecium or anther and ovule stage of initiation.

Biomass has a large share in the energy generation matrix, due to the regional economic benefits. This work has as main objective to evaluate the parameters used in the manufacture of briquettes produced with forest residues and the economic engineering for the manufacturer. The forest residues were: wood chips and chips of Eucalyptus spp. and barks of Pinus taeda. The evaluations were the chemical characterization of forest residues and the costs involved in the briquetting process. The forest residues presented extractive chemical composition and lignin. The production costs of the briquettes were affected mainly by the equipment, being the Pinus taeda barks the one that presented higher production cost. However, the production costs obtained in this study are approximately 20% lower than the production costs using traditional methods for forest firewood. The energy value from biomass allows the reduction of the dependence of energy, which can be used for the generation of steam or electricity, for subsistence. A fact for energy conversion is to evaluate the material moisture. It is suitable to burn residues with moisture between 45 to 55%. The consumed electric energy is important to evaluate the total costs. The energy required depends on the quality of raw material and the system employed. Forest residues is an important source for eco firewood production, contributing to energy generation and decreasing of the solid waste stored at the company. In this way, the new parameters for briquetting biomass forest wastes shown in this work, which is an important stage of the process, make economically viable and environmentally suitable the eco firewood production.

This program is conducted to assist Siponjot Village while empowering the people of Siponjot Village to be able to utilize and maximize the benefit of the village forest. Village forest might support the availability of raw materials required for making musical instruments, such as high quality of wood, including how to process wood waste from making musical instruments to be used as creative souvenirs and improve the economic value. The raw material for production determined its results of the production of wood-based musical instruments. Thus, in order to produce a high quality tanginang, hasapi, and gondang, which previously began to be produced by arts crafts in the Sitangkubang area of ​​Siponjot Village, a high quality of raw materials is needed. The community service team surveyed the location of planting seeds for village forest restoration, provided socialization related to the importance of village forest cultivation and the suitability of the Siponjot Village area for the cultivation of these plants. Village forest restoration aims to maintain the beauty and beauty of the village. Implementation of village reforestation activities starting from socializing forest tourism and the strength of village forests to the community, followed by a discussion about village forest management and its economic benefits. Handover of a thousand units of forest plant seedlings given to the villagers of Siponjot as part of the forest restoration program in the area.

Abstract Background: Hepatocellular carcinoma (HCC) poses a severe threat to human health. The NET-1 protein has been proved to be strongly associated with HCC proliferation and metastasis in our previous study. Methods: Here, we developed a label-free proteome mass spectrometry workflow to analyze formalin-fixed and paraffin-embedded HCC xenograft samples collected in our previous study. Results: The result showed that 78 proteins were differentially expressed after NET-1 protein inhibited. Among them, the expression of 61 proteins up-regulated and the expression of 17 proteins were significantly down-regulated. Of the differentially expressed proteins, the vast majority of Gene Ontology enrichment terms belong to the biological process. The KEGG pathway enrichment analysis showed that the 78 differentially expressed proteins significantly enriched in 45 pathways. We concluded that the function of the NET-1 gene is not only to regulate HCC but also to participate in a variety of biochemical metabolic pathways in the human body. Furthermore, the protein-protein interaction analysis indicated that the interactions of differentially expressed proteins are incredibly sophisticated. All the protein-protein interactions happened after the NET-1 gene has been silenced. Conclusions: Finally, our study also provides a useful proposal for targeted therapy based on tetraspanin proteins to treat HCC, and further mechanism investigations are needed to reveal a more detailed mechanism of action for NET-1 protein regulation of HCC.
