Abstract Background A simple and sensitive spectrophotometric method was developed for the quantitative measurement of dolutegravir in pure form and pharmaceutical formulation. The present method was based on redox reaction between dolutegravir and ferric chloride, which upon complexation with 1,10-phenanthroline formed an orange-colored complex that showed absorption maximum at 520.0 nm. Results The developed method obeyed linearity in the concentration range of 40.00–140.00 μg/mL. The method was also validated as per International Council for Harmonization guidelines and the results were within acceptance values. The validated method was employed for the determination of dolutegravir in pharmaceutical dosage form and the percentage assay value was found to be 102.5, which is in agreement with its label claimed. Conclusion The developed redox-based colorimetric method could be used in the routine quality control analysis of dolutegravir present in various pharmaceutical dosage forms.

Simple sensitive and accurate extractive colorimetric method was developed for the estimation of favipiravir in Pharmaceutical dosage forms. The method was based on the formation of colored ion pair complexes by the drugs with thiocynate ions. These ion pair complexes were quantitatively extracted under the experimental condition in chloroform. The absorbance values were measured at 618 respectively. The proposed method was validated statistically. A recovery of method was carried out by standard addition methods. The Beer’s law ranges were found to be 1-12μg/ml, respectively. The low values of standard deviation and percentage RSD indicate high precision of method. Hence the method is useful for routine estimation of favipiravir in tablets respectively.

A simple, precise and accurate spectrophotometric method was developed for analysis of the osteoporesis drug alendronate sodium (ALS). The method is based on reaction of the drug with sodium-1,2-naphthoquinone-4-sulphonate (NQS) in presence of alkali to form a brown colored complex giving absorption maximum at 525 nm. The drug obeyed Beer’s law in the range of 5-70 µg/ml with a correlation coefficient of 0.999. The LOD and LOQ values are 1.7 µg/ml and 5.0 µg/ml, respectively. The average recoveries for recovery study were found to be in the range of 99.37%-100.46%. The R.S.D. values for intraday and inter-day precision were found to be 0.48 and 0.62, respectively. The optimized assay conditions were applied successfully for determination of ALS in pharmaceutical dosage forms. No interference was observed from the excipients present in the dosage form. The method is statistically validated as per the ICH requirements.

A simple, rapid, economic, sensitive and precise HPLC method has been developed for the simultaneous determination of Sulphadoxine and Pyrimethamine in pharmaceutical dosage form by taking Tolterodine as an internal standard. The method was carried out using Phenomenex C18 (4.6ID × 250mm; 5µm) column and mobile phase comprised of methanol and Phosphate Buffer in proportion of ratio 60:40 v/v. The flow rate was 1.0mL/min and detection was carried out at 276nm. The retention time of Sulphadoxine, Pyrimethamine and Tolterodine were found to be 2.967, 4.058 and 6.908 respectively. Linearity of Sulphadoxine and Pyrimethamine in the range of 2 to 12μg/mL and 4 to 24μg/mL respectively. The % recoveries of Sulphadoxine and Pyrimethamine were found to be in between 99.93% to 99. 96 % respectively. The proposed method is suitable for the routine quality control analysis for simultaneous determination of Sulphadoxine and Pyrimethamine was in bulk and pharmaceutical dosage form.

Abstract A colorimetric method is proposed for determination of terbutaline sulfate, orciprenaline sulfate, and their dosage forms. The suggested method depends on nitrosation of the 2 drugs by using sodium nitrite and hydrochloric acid. Addition of sodium hydroxide increases the intensity of the color developed. The difference between absorption values measured in acid and alkaline media is taken as a measure of concentration. Variables were carefully studied and optimized. Results for both compounds adhered to Beer's law over the range 2- 28 μg/mL. The method has proved to be accurate and precise for analysis of pharmaceutical dosage forms.

Abstract A method is presented for extracting and determining the starch content of tobacco. Dried tobacco is ground to pass a 60 mesh screen, interfering colored materials are removed from the tobacco by a methanol-water extraction, and the starch is extracted from the tobacco by a perchloric acid treatment. Final quantitative measurement of the starch is made on a colored starch–iodine complex. The absorbance of this colored complex at 600 nm is proportional to the concentration of the starch in solution and can be related to the starch content of the original tobacco sample. Also included is a procedure for the isolation of pure tobacco starch. Such material was prepared for use as a calibration standard needed for the calculation of starch content of tobacco samples from the colorimetric data.

Abstract A colorimetric method is described for the determination of certain mono-substituted acetylenic hypnotic drugs—ethinamate, methyl pentynol carbamate, and ethchlorvynol. Silver acetylide is formed and extracted into isobutyl methyl ketone. After acidification, an equivalent amount of silver ions is liberated which is assayed colorimetrically as silver dithizonate (λ max. 472 nm). About 10–50 μg compound could be assayed with good accuracy. The procedure is reproducible and precise (standard deviation 1.5–2.19%). The procedure was successfully applied to different pharmaceutical dosage forms.

A simple, reliable, precise and sensitive UV-spectrophotometric method was developed and validated for the estimation of azithromycin in pharmaceutical dosage form and compared with official USP 2010 method. The proposed method utilizes the oxidation of azithromycin with potassium permanganate to liberate formaldehyde. This formaldehyde reacts with acetone-ammonium reagent and produces yellow colored chromogen 3,5-diacetyl-2,6-dihydrolutidine. The colored solution exhibited a maximum absorption at 412 nm which can be detected with UVspectrophotometer. The method was found linear over the concentration range 80% to 120% of the working concentration (R2=0.999). The intra- and inter-day RSD (n = 6) was ? 2.0%. The developed method was validated according to ICH guidelines and values of accuracy, precision and other statistical analysis were found to be in good accordance with the prescribed values. Abstract A method has been investigated for the determination of bismuth in pharmaceutical dosage forms by conventional dc polarography. The method is applicable to tablets, emulsions, suspensions, injectables, and bulk powders. A collaborative study has shown the method to be superior in accuracy and precision to the present official colorimetric method, 36.300- 36.304. The results from 16 determinations performed by 8 collaborators were: 100.0±1.5% recovery on a synthetic powder mix, a relative standard deviation of 1.9% on a commercial tablet sample, and a relative standard deviation of 2.1% on a magma sample. By comparison, the collaborative data reported for the present official method gave relative standard deviations of 3.4 and 3.2% on 2 samples of powder mixtures. It is recommended that the polarographic method be adopted as official first action.

Objective: The objective of the study was to develop a new, simple, accurate, precise and reproducible RP-UPLC method for the estimation of bumetanide in bulk and pharmaceutical dosage form.Methods: Acquity SB C18, 2 x 100 mm, 1.8 µmm, 5m particle size column with the mobile phase consisting of water: acetonitrile in the ratio of 30:70 v/v were used. The effluents were moniRTat 254 nm and the flow rate was 1.0 ml/minute.Results: The retention time was 0.852 min. Quantitative linearity was obeyed in the concentration range of 12.5 to 75 μg/ml. The correlation coefficient for bumetanide was found to be 0.999. Recovery and assay studies of bumetanide were within 99 to 102%, indicating that the proposed method can be adaptable for quality control analysis of bumetanide. The % RSD for precision and accuracy of the method was found to be less than 2%. Bumetanide was subjected to stress environment of degradation in aqueous solutions including oxidation, hydrolysis, thermal and photolysis degradation.Conclusion: Proposed method was found to be simple, accurate, precise, and quick and can be used for regular analysis. This condition was applied to tablet dosage form. The statistical parameters and recovery studies were reported.

A simple, accurate, rapid and precise method was developed for the simultaneous estimation of Ibuprofen and Diphenhydramine in Pharmaceutical dosage form. Chromatogram was run through Inertsil ODS (250x4.6mm) 5µ. Mobile phase used was Acetonitrile and Phosphate buffer (45:55) at a flow rate of 1.0 ml/min and detection wavelength was found to be 260 nm. The retention time was found to be 2.32 min and 2.93 min for Ibuprofen and Diphenhydramine respectively. The accuracy and reliability of the method was assessed by evaluation of linearity, precision (intra-day and inter-day % RSD >2), accuracy (98-102%), specificity, LOD, LOQ values in accordance with ICH guidelines. The developed method is applicable for routine quality control analysis of selected combined dosage forms.
