Optimal embryonic development plays a major role in the health of an individual beyond the developmental stage. Nutritional perturbation during development is associated with cardiovascular and metabolic disease later in life. With both nutritional uptake and overall growth being risk factors for eventual health, it is necessary to understand not only the behavior of the processes during development but also their interactions. In this study, we used differential equations, image analyses, curve fittings, parameter estimation and laboratory experiments to quantify the rate of yolk absorption and its effect on early development of a vertebrate model (Danio rerio). Findings from this study establish a nonlinear functional relationship between nutrient absorption and early fish growth. We found that the rate of change in fish length and yolk utilization is logistic, that is the yolk decays rapidly for a period of time before leveling out. An interesting finding from this study is that yolk utilization reaches its maximum at 84 hours post fertilization. We validated our mathematical models against experimental observations, making them powerful tools for replication and future simulations.

The aim of this study was to investigate the long-term effects of subchronic exposure to sublethal levels of nitrite, ranging from 15 to 130 mg l-1 NO2-, on growth in aquarium fish Danio rerio. The juvenile growth test according to OECD 215 was used in the experiments. Fish weight was measured at the beginning of the experiment and then using the same method, fish weight was observed 28 days after fish stocking. Compared to the control, growth suppression was detected from the concentration of 73 mg l-1 NO2- (P < 0.05) and a significant inhibition of fish body growth was shown from 130 mg l-1 NO2- (P < 0.01). An exponential relationship between nitrite concentrations and specific growth rate (R2 = 0.896) was detected.

Excessive production of melanin implicates hyperpigmentation disorders. Flavokawain A (FLA) and flavokawain B (FLB) have been reported with anti-melanogenic activity, but their melanogenic inhibition and toxicity effects on the vertebrate model of zebrafish are still unknown. In the present study, cytotoxic as well as melanogenic effects of FLA and FLB on cellular melanin content and tyrosinase activity were evaluated in α-MSH-induced B16/F10 cells. Master regulator of microphthalmia-associated transcription factor (Mitf) and the other downstream melanogenic-related genes were verified via quantitative real time PCR (qPCR). Toxicity assessment and melanogenesis inhibition on zebrafish model was further observed. FLA and FLB significantly reduced the specific cellular melanin content by 4.3-fold and 9.6-fold decrement, respectively in α-MSH-induced B16/F10 cells. Concomitantly, FLA significantly reduced the specific cellular tyrosinase activity by 7-fold whilst FLB by 9-fold. The decrement of melanin production and tyrosinase activity were correlated with the mRNA suppression of Mitf which in turn down-regulate Tyr, Trp-1 and Trp-2. FLA and FLB exhibited non-toxic effects on the zebrafish model at 25 and 6.25 µM, respectively. Further experiments on the zebrafish model demonstrated successful phenotype-based depigmenting activity of FLA and FLB under induced melanogenesis. To sum up, our findings provide an important first key step for both of the chalcone derivatives to be further studied and developed as potent depigmenting agents.

We assessed the fish length – otolith length relationship (FL–OL) in Dolly Varden (Salvelinus malma malma) to verify proportional growth. A decoupling was detected during first ocean migration where fish growth was occurring at a greater rate than otolith growth. Because of this decoupling, the application of traditional back-calculation models overestimated the size-at-age in premigratory char. We developed modified back-calculation equations from existing traditional models to account for this decoupling based on discontinuous piecewise regressions. The new biological intercept breakpoint method (BI–BP) provided the most accurate representation of fish size-at-age throughout all life history stages when compared with known size-at-capture values in fish. The decoupling indicates that factors other than somatic growth are important for otolith accretion. Physiological changes during smoltification likely alter calcium uptake and thereby affect calcium deposition rates on otoliths during this short but biologically critical time period of life history. It is probable that species exhibiting similar complex ontogenetic shifts in life history will likely exhibit decoupling to some extent in the FL–OL relationship.

In this paper an automatic parameter identification technique is presented which can be used to generate a nonlinear functional relationship of dynamic draft gear characteristics using experimental data.

The paper proposed a modified LMS algorithm of variable step size based on a brief analysis of traditional LMS,variable step size LMS algorithm and its improved algorithm.The novel algorithm based on nonlinear functional relationship between the step-size and the error ,increases adaptively at the beginning of the algorithm or when the channel is varying with time ,and it would be smaller during the steady state.So the algorithm has the excellences of faster constringency,little steady error ,tracking the change of the system and avoiding the effects of the noise. The theoretical analysis and computer simulation prove that the algorithm is better than traditional LMS algorithm.

ABSTRACTMemory in animals is labile in the early phase post-training. Memory in the early phase has been shown to be disrupted by treatments such as electroconvulsive shock or cold-shock (Quinn and Dudai, 1976). Using hypothermic shock and other pharmacological interventions, the various underlying memory pathways in Drosophila can be identified as an immediate short-lasting anesthesia sensitive memory and a delayed anesthesia resistant memory which is followed by a more stable protein synthesis dependent long-term memory (Margulies et al., 2005). In another ectothermic animal, Danio rerio, a popular vertebrate model, we ask if such a memory component exists which is sensitive to hypothermic disruption. To test this, we developed a fear conditioning assay with a green light at the bottom of the tank as the conditioned stimulus (CS) and electric shock as the unconditioned stimulus (US). We also standardized a cold anesthesia protocol in adult zebrafish to induce stage V anesthesia. The learning/memory was found to be NMDA-R mediated. Cold anesthesia as well as tricaine mediated anesthesia did not significantly affect the early-acting memory trace induced by a fear-conditioning protocol in adult zebrafish. We suggest future directions to tease out the underlying memory components in the early phase of memory in zebrafish.
