Small-scale photobioreactors (PBRs) in the inoculum stage were designed with internal (red or green) and external white LED light as an initial step of a larger-scale installation aimed at fulfilling the integral biorefinery concept for maximum utilization of microalgal biomass in a multifunctional laboratory. The specific growth rate of Scenedesmus obliquus (Turpin) Kützing biomass for given cultural conditions was analyzed by using MAPLE software. For the determination of total polyphenols, flavonoids, chlorophyll “a” and “b”, carotenoids and lipids, UHPLC-HRMS, ISO-20776/1, ISO-10993-5 and CUPRAC tests were carried out. Under red light growing, a higher content of polyphenols was found, while the green light favoured the flavonoid accumulation in the biomass. Chlorophylls, carotenoids and lipids were in the same order of magnitude in both samples. The dichloromethane extracts obtained from the biomass of each PBR synergistically potentiated at low concentrations (0.01–0.05 mg/mL) the antibacterial activity of penicillin, fluoroquinolones or oregano essential oil against the selected food-borne pathogens (Staphylococcus aureus, Escherichia coli and Salmonella typhimurium) without showing any in vitro cytotoxicity. Both extracts exhibited good cupric ion-reducing antioxidant capacity at concentrations above 0.042–0.08 mg/mL. The UHPLC-HRMS analysis revealed that both extracts contained long chain fatty acids and carotenoids thus explaining their antibacterial and antioxidant potential. The applied engineering approach showed a great potential to modify microalgae metabolism for the synthesis of target compounds by S. obliquus with capacity for the development of health-promoting nutraceuticals for poultry farming.

In this work, nitrogen dioxide (NO2) gas sensors based on zinc oxide nanorods (ZnO NRs) decorated with gold nanoparticles (Au NPs) working under visible-light illumination with different wavelengths at room temperature are presented. The contribution of localized surface plasmon resonant (LSPR) by Au NPs attached to the ZnO NRs is demonstrated. According to our results, the presence of LSPR not only extends the functionality of ZnO NRs towards longer wavelengths (green light) but also increases the response at shorter wavelengths (blue light) by providing new inter-band gap energetic states. Finally, the sensing mechanism based on LSPR Au NPs is proposed.

Abstract Background Cyanobacteria are engineered via heterologous biosynthetic pathways to produce value-added chemicals via photosynthesis. Various chemicals have been successfully produced in engineered cyanobacteria. Chemical inducer-dependent promoters are used to induce the expression of target biosynthetic pathway genes. A chemical inducer is not ideal for large-scale reactions owing to its high cost; therefore, it is important to develop scaling-up methods to avoid their use. In this study, we designed a green light-inducible alcohol production system using the CcaS/CcaR green light gene expression system in the cyanobacterium Synechocystis sp. PCC 6803 (PCC 6803). Results To establish the green light-inducible production of isobutanol and 3-methyl-1-butanol (3MB) in PCC 6803, keto-acid decarboxylase (kdc) and alcohol dehydrogenase (adh) were expressed under the control of the CcaS/CcaR system. Increases in the transcription level were induced by irradiation with red and green light without severe effects on host cell growth. We found that the production of isobutanol and 3MB from carbon dioxide (CO2) was induced under red and green light illumination and was substantially repressed under red light illumination alone. Finally, production titers of isobutanol and 3MB reached 238 mg L−1 and 75 mg L−1, respectively, in 5 days under red and green light illumination, and these values are comparable to those reported in previous studies using chemical inducers. Conclusion A green light-induced alcohol production system was successfully integrated into cyanobacteria to produce value-added chemicals without using expensive chemical inducers. The green light-regulated production of isobutanol and 3MB from CO2 is eco-friendly and cost-effective. This study demonstrates that light regulation is a potential tool for producing chemicals and increases the feasibility of cyanobacterial bioprocesses. Graphical Abstract

Abstract The paper aims to explore the possibilities of improving road traffic in the central area of cities characterized by a longitudinal arrangement of the street network, with application for the case of Pitesti, where the road network in the central area consists of two main roads arranged longitudinally, having one-way regulated traffic, interconnected by several streets. A special traffic problem is reported in the city center: on the main road connecting the two boulevards, the vehicle storage space is insufficient - because the distance between the two road intersections is too small and there is no correlation between the Green phases of traffic lights in the two intersections. The research, based on traffic measurements performed with DataFromSky software and micro-simulation traffic analyses performed with Vissim PTV software, indicated that the best solution is the partial or total correlation of the green time from the traffic light intersections that delimit the connecting road artery. As, almost exclusively, the works dedicated to the correlation of green light of traffic lights treat the problem only along the road arteries, this paper raises a special issue and reveals the possibility of simple solutions, by correlating the traffic lights at the intersections connecting the main arteries.

After the fall of communism and the wars that accompanied the disintegration of Yugoslavia, countries in the region turned towards the European Union. Upon signing the SAA, the countries of the Western Balkans, such as Albania, have had a green light to move forward in the EU's pre-accession process. As Albania works toward membership, how do domestic political actors in Albania (not) change their with agendas according to EU requirements? This chapter will address the puzzle of the transformative power of EU's conditionality in main political parties in Albania.

Kiwifruit (Actinidia deliciosa [A. Chev.], C.V. Liang & A. R. Ferguson, 1984) is a perennial plant, with morphologically hermaphroditic and functionally dioecious flowers. Fruits of this species are berries of great commercial and nutritional importance. Nevertheless, few studies have analyzed the molecular mechanisms involved in sexual differentiation in this species. To determine these mechanisms, we performed RNA-seq in floral tissue at stage 60 on the BBCH scale in cultivar ‘Hayward’ (H, female) and a seedling from ‘Green Light’ × ‘Tomuri’ (G × T, male). From these analyses, we obtained expression profiles of 24,888 (H) and 27,027 (G × T) genes, of which 6413 showed differential transcript abundance. Genetic ontology (GO) and KEGG analysis revealed activation of pathways associated with the translation of hormonal signals, plant-pathogen interaction, metabolism of hormones, sugars, and nucleotides. The analysis of the protein-protein interaction network showed that the genes ERL1, AG, AGL8, LFY, WUS, AP2, WRKY, and CO, are crucial elements in the regulation of the hormonal response for the formation and development of anatomical reproductive structures and gametophytes. On the other hand, genes encoding four Putative S-adenosyl-L-methionine-dependent methyltransferases (Achn201401, Achn281971, Achn047771 and Achn231981) were identified, which were up-regulated mainly in the male flowers. Moreover, the expression profiles of 15 selected genes through RT-qPCR were consistent with the results of RNA-seq. Finally, this work provides gene expression-based interactions between transcription factors and effector genes from hormonal signaling pathways, development of floral organs, biological and metabolic processes or even epigenetic mechanisms which could be involved in the kiwi sex-determination. Thus, in order to decode the nature of these interactions, it could be helpful to propose new models of flower development and sex determination in the Actinidia genus.

In recent years, play has been shown to be a powerful means to enhance learning and brain development. It is also known that through play children enhance their executive function (EF) skills. Furthermore, well-developed EF in preschoolers has been shown to be an important predictor for later academic and life success. Armed with this information a program, Building Brains and Futures (BBF), for developing EF through play was designed for 3–5-year-old. The program consisted of 10 simple, fun, and interactive games selected to enhance various facets of EF. The 10 games included were: dimensional change card sort, lips and ears, block building, musical freeze, opposites, pretend play, red light/green light, shared project, Simon says, and wait for it. The program was implemented with a group of children shown to have challenges with respect to kindergarten readiness. The approach was first, to build adult capability by sharing knowledge of brain development, EF, and the importance of play with educators, caregivers, and parents. Second, to build skills in delivering the program in the school setting. Children engaged with the program of games for a minimum of 6 weeks. Their performance on a battery of direct measures of EF, language, and motor skills, were recorded before and after the program. The results showed improvement in all three domains. In addition, adopters of the BBF program reported it was easily and successfully integrated into their existing preschool curricula. The importance of intentional adult directed play in building developmental learning, including EF, is discussed.

The Colorado River is a critical source of water supply for 40 million people in nine states spanning two nations in western North America. Overallocated in the 20th century, its problems have been compounded by climate change in the 21st century. We review the basin’s hydrologic and water management history in order to identify opportunities for adaptive governance to respond to the challenge of reduced system flows and distill the ingredients of past successes. While significant advances have been made in the first two decades of the 21st century, these past actions have not been sufficient to halt the declines in the basin’s reservoirs. We find that the mix of federal, state, and local responsibility creates challenges for adaptation but that progress can be made through a combination of detailed policy option development followed by quick action at hydrologically driven moments of opportunity. The role of directives and deadlines from federal authorities in facilitating difficult compromises is noted. The current state of dramatically decreased overall flows has opened a window of opportunity for the adoption of water management actions that move the river system toward sustainability. Specific measures, based on the existing institutional framework and on policy proposals that have circulated within the Colorado River community, are suggested.
