Changes in hair growth characteristics following the wounding of vibrissa follicles in the hooded rat

The effect on hair growth of wounding the lower region of whisker follicles, and in particular the dermal papilla, with sharply pointed tungsten needles was studied in adult hooded rats. Following injury hair growth ceased, but was subsequently resumed. While it might have been anticipated that follicle wounding would have a negative effect on whisker length, regular postoperative length measurements revealed that in follicles where cellular material was not displaced from the follicle by the original manipulation, 50 % of the subsequent hairs produced were longer than their counterparts on the opposite side of the face, with 25 % shorter and 25 % with their length unchanged. In every case increased hair length was achieved by a prolongation of the g that the factors which control the duration of the hair cycle and fibre growth rate are independent in vibrissa follicles. Since removal of most of the epidermal component by plucking of the hair just prior to injury produced equivalent hair length increases, this implicated the proximal dermal components as being mainly responsible for the observed changes.

Hair loss is becoming increasingly prevalent as dietary and living habits change. The search for natural products to limit hair loss has led to tapping into traditional cosmetic knowledge. We studied three plants of the Polynesian cosmetopoeia, Bidens pilosa, Calophyllum inophyllum and Fagraea berteroana, to determine their ability to promote hair growth. Their chemical content was characterized by liquid chromatography coupled to mass spectrometry (LC-MS). Their proliferative activity on dermal papilla cells (DPCs) was assessed via MTT assay and molecular targets were evaluated by RT-qPCR analysis of seven factors involved in the modulation of the hair cycle, CCND1, LEF1, DKK1, WNT5A PPARD, TGFΒ1, PPARD and RSPO2. Our results show that our extracts significantly increased proliferation of dermal papilla cells. Furthermore, LC-MS/MS analysis revealed a diversity of molecules, flavonoids, iridoids and organic acids, some known for hair-inducing properties. Finally, specific extracts and fractions of all three plants either upregulated CCND1, LEF1 and PPARD involved in stimulating hair follicle proliferation and/or lowered the gene expression levels of hair growth inhibiting factors, DKK1 and TGFB1. Our findings suggest that extracts from B. pilosa, C. inophyllum and F. berteroana are interesting candidates to stimulate hair growth.

Rab27a/b are known to play an important role in the transport of melanosomes, with their knockout causing silvery gray hair. However, the relationship between Rab27a/b and hair growth is not well known. To evaluate the role of Rab27a/b in hair cycle, we investigated the expression of Rab27a/b during hair cycling and human outer root sheath (hORS) cells. The expression of Rab27a in ORS cells was mainly detected at the anagen, whereas expression of Rab27b in ORS, and epidermal cells was strongly expressed at the telogen. Additionally, Rab27a/b were expressed in the Golgi of hORS cells. To evaluate the role of Rab27a/b in hair growth, telogen-to-anagen transition animal and vibrissae hair follicles (HFs) organ culture models were assayed using Rab27a/b siRNAs. The knockdown of Rab27a or Rab27b suppressed or promoted hair growth, respectively. These results were also confirmed in human dermal papilla cells (hDPCs) and hORS cells, showing the opposite mitogenic effects. Moreover, Rab27b knockdown increased the expression levels of various growth factors in the hDPCs and hORS cells. Overall, the opposite temporal expression patterns during hair cycling and roles for hair growth of Rab27a/b suggested that Rab27a/b might regulate the hair cycle. Therefore, our study may provide a novel solution for the development of hair loss treatment by regulating Rab27a/b levels.

The progression in the hair follicle cycle from the telogen to the anagen phase is the key to regulating hair regrowth. Dermal papilla (DP) cells support hair growth and regulate the hair cycle. However, they gradually lose key inductive properties upon culture. DP cells can partially restore their capacity to promote hair regrowth after being subjected to spheroid culture. In this study, results revealed that DP spheroids are effective at inducing the progression of the hair follicle cycle from telogen to anagen compared with just DP cell or minoxidil treatment. Because of the importance of paracrine signaling in this process, secretome and exosomes were isolated from DP cell culture, and their therapeutic efficacies were investigated. We demonstrated that miR-218-5p was notably up-regulated in DP spheroid–derived exosomes. Western blot and immunofluorescence imaging were used to demonstrate that DP spheroid–derived exosomes up-regulated β-catenin, promoting the development of hair follicles.

Hair follicles are essentially composed of two tissues. The inner epidermal component, which gives rise to, among other products, the keratinized hair shaft, is confluent with the surface epidermis and is ensheathed by the dermal component which is confluent with the pars papillaris of the dermis. A specialization of the dermal component is the dermal papilla which, in follicles producing hair, is enclosed by the epidermal matrix of the hair bulb and is connected to the dermal sheath by the papilla stalk. Many authorities have considered that the dermal papilla is an essential component of the hair follicle (reviews: Cohen, 1965; Oliver, 1969). It has been suggested that the dermal papilla may be involved in both the induction of follicle lengthening and hair growth during the proanagen phase (Chase, 1965) of the hair cycle, a concept now justified by direct experimentation in the vibrissa follicle at least (Oliver, 1967b), and perhaps also in determining the nature of the hair produced by a follicle.

The term cosmetopoeia refers to the use of plants in folks’ cosmetics. The aerial parts of Bidens pilosa L., the leaves of Calophyllum inophyllum L. and the fruits of Fagraea berteroana A.Gray ex Benth are traditionally used in French Polynesia for hair and skin care. During the hair cycle, dermal papilla cells and their interaction with epithelial cells are essential to promote hair follicle elongation. The aim of our investigations was the identification of metabolites from these three plants and chemical families responsible for their hair growth activity. A bioactivity-based molecular network was produced by mapping the correlation between features obtained from LC-MS/MS data and dermal papilla cell proliferation, using the Pearson correlation coefficient. The analyses pointed out glycosylated flavonols and phenolic acids from B. pilosa and C. inophyllum, along with C-flavonoids, iridoids and secoiridoids from F. berteroana, as potential bioactive molecules involved in the proliferation of hair follicle dermal papilla cells. Our results highlight the metabolites of the plant species potentially involved in the induction of hair follicle growth and support the traditional uses of these plants in hair care.

SUMMARY Hypophysectomy of, or administration of thyroxine to, male or female rats advances the activity of hair follicles in the ensuing wave of growth; administration of propylthiouracil retards it. The effects seem to be more marked in the dorsal and head regions than in the flank or belly. Propylthiouracil does not prevent the activation of the follicle induced by plucking of the club hair. It is suggested that the hormonal effects may be produced indirectly through the dermis. Hypophysectomy increased definitive hair length in females but not in males; thyroxine also produced a small increase and propylthiouracil a decrease which proved to be significant in females, but not in males.

AbstractHair loss is a disorder or disorder of hair apart from the scalp or body skin so that it interferes with various biological functions of hair on the body. Types of hair care cosmetic that is effective in dealing with hair loss are hair tonic. Hair tonic is a preparation for the treatment of the scalp and hair. Bandotan (Ageratum conyzoides L.) is used by the public as medicine and hair care. The purpose of this study was to determine the formulation of hair tonic preparations from the ethanol extract of bandotan leaves according to the requirements and hair growth activity of local male rabbits. The extraction method used the maceration method with 96% ethanol and applied hair tonic extract of bandotan leaves on the skin of local male rabbits (Oryctolagus cuniculus) which were treated as blanks without smearing, positive control (Natur), negative control hair tonic base, concentration bandotan leaf extract. (5%, 10%, 15% and 20%) for 21 days. The data obtained in the form of hair length, and hair weight. Analysis of the data obtained for hair growth activity was seen from the results of the ANOVA test and continued with the BNT test (Least Significance Different). The results of the research for evaluating hair tonic preparations included organoleptic tests, pH tests, viscosity tests, specific gravity tests, and cycling tests. Hair tonic ethanol extract of bandotan leaves has hair growth activity. Hair tonic ethanol extract of bandotan leaves with a concentration of 20% had the highest hair length and hair weight. From the results of one way ANOVA, a significant value was obtained 0.000 < 0.005, so it was concluded that there was a significant difference between treatments, and continued with the BNT test (Least Significance Different) positive control was not significantly different from hair tonic concentration of 20%.Keywords: Bandotan leaves, Hair tonic, Hair growth AbstrakKerontokan rambut adalah suatu gangguan atau kelainan rambut terlepas dari kulit kepala atau kulit tubuh sehingga mengganggu berbagai fungsi biologis rambut terhadap tubuh. Jenis kosmetik perawatan rambut yang efektif mengatasi rambut rontok adalah hair tonic. Hair tonic adalah sediaan untuk perawatan kulit kepala dan rambut. Bandotan (Ageratum conyzoides L.) digunakan oleh masyarakat sebagai obat dan perawatan rambut. Tujuan dari penelitian ini mengetahui formulasi sediaan hair tonic dari ekstrak etanol daun bandotan yang sesuai dengan persyaratan dan aktivitas pertumbuhan rambut pada kelinci jantan lokal. Metode ekstraksi menggunakan metode maserasi dengan etanol 96% dan dilakukan pengolesan hair tonic ekstrak daun bandotan pada kulit kelinci jantan lokal (Oryctolagus cuniculus) yang diberikan perlakuan sebagai blanko tanpa pengolesan, kontrol positif (Natur), kontrol negatif basis hair tonic, ekstrak daun bandotan konsentrasi (5%,10%,15% dan 20%) selama 21 hari. Data yang diperoleh berupa panjang rambut, dan bobot rambut. Analisa data yang diperoleh untuk aktivitas pertumbuhan rambut dilihat dari hasil uji ANOVA dan dilanjutkan dengan uji BNT (Beda Nyata Terkecil). Hasil penelitian untuk evaluasi sediaan hair tonic antara lain uji organoleptis, uji pH, uji viskositas, uji bobot jenis, dan uji cycling test. Hair tonic ekstrak etanol daun bandotan memiliki aktivitas pertumbuhan rambut. Hair tonic ekstrak etanol daun bandotan dengan konsentrasi 20% memiliki panjang rambut dan bobot rambut tertinggi. Dari hasil one way ANOVA didapatkan nilai signifikan 0,000 < 0,005 sehingga disimpulkan adanya perbedaan signifikan antar perlakuan, dan dilanjutkan uji BNT (Beda Nyata Terkecil) didapatkan kontrol positif tidak berbeda nyata dengan hair tonic konsentrasi 20%.Kata kunci : Daun bandotan; Hair tonic; Pertumbuhan rambut

The capacity of lower follicle dermal sheath to restore hair growth was tested by removing the lower halves of follicles, and then immediately implanting material containing dermal sheath cells from these bases, into the remaining upper epidermal follicle cavity. Over 60% of recipient follicles produced stout emergent vibrissa fibres and some operations resulted in multiple hair production from a single follicle. Histological examination revealed new dermal papillae within large bulb structures which were sited below the level of amputation--a feature that indicated that the new dermal papilla was derived from implanted material. For many follicles, the failure to produce emergent fibres could be accounted for after histological examination. These results provide clear evidence that lower follicle dermal sheath cells are capable of replacing those of the dermal papilla and it shows that they can do so in the context of the upper follicle. However, because elements of lower follicle epidermis were present in the implant material, the interactive sequence of events cannot be established. Dermal sheath cells have immense potential for papilla cell replacement: questions remain as to whether the distinction between sheath and papilla cells is one of context, or whether the transition requires specific external influences.
