Currently there is an active search for medicines increasing the stability of liver to pathological effects, reinforcing its neutralizing function, promoting its functional recovery after various injuries, including poisoning by industrial poisons. Therefore, the study of opportunities to reduce toxic liver damage by carbon tetrachloride introduction of synthetic and natural antioxidants is of interest. In experimental conditions the possibility to correct free radical lipid oxidation of rats’ organism membranes was studied with the introduction of the succinate containing drug called Reamberin (Polysan, St.Petersburg) and licorice syrup. The animals were divided into 4 groups and each of them had 10 rats: the group with intact animals which were held in standard conditions of vivarium; the control group in which rats were given carbon tetrachloride during 3 days daily; the experimental group in which before the introduction of carbon tetrachloride animals had a daily intra-abdominal intake of the Reamberin in a dose of 100 mg/kg (20 ml/kg); the experimental group in which before the introduction of carbon tetrachloride animals had a daily oral intake of the licorice syrup in a dose of 5 ml/kg. It was found out that in the blood and in the liver of experimental animals an introduction of carbon tetrachloride during 3 days contributes to the increase of lipid hydroperoxides level (by 24-43%), of diene conjugate (by 19-47%), and of malonic dialdehyde (by 61-81%) against the decrease of antioxidant system activity in the blood of intact animals. The introduction of the succinate containing drug to rats in the conditions of oxidative stress contributes to the decrease in the blood and in the liver of lipid hydroperoxides by 13-26%, of diene conjugates by 11-28%, and of malonic dialdehyde by 30-34% in comparison with the rats of the control group. Adding of the licorice syrup to rats contributes to the reliable decrease in the blood and in the liver of lipid hydroperoxides by 6-23%, of diene conjugates by 9-24%, and of malonic dialdehyde by 25-30% in comparison with the rats of the control group. While analyzing the effect of the antioxidants on the activity of the components of antioxidant system it was shown that the level of ceruloplasmin in the blood and in the liver of animals was reliably higher by 10-47%, of vitamin E by 13-42% in comparison with the same parameters of the rats of the control group. So, the application of the synthetic and natural antioxidants in the conditions of introduction of carbon tetrachloride of the organism of animals under experiment leads to the stabilization of the processes of peroxidation against the increase of antioxidant system activity.

The search and development of methods for correction of oxidative stress in conditions of exposure to adverse environmental factors is an actual problem of modern medicine. In experimental conditions the possibility to correct free radical lipid oxidation of rats’ organism membranes was studied with the introduction of the succinic acid and of the succinate containing medication called Reamberin (Polysan, St. Petersburg). The animals were divided into 4 groups and each of them had 20 rats: the group with intact animals which were held in standard conditions of vivarium; the control group in which rats were exposed to ultraviolet radiation during three minutes daily; the experimental group in which before ultraviolet radiation animals had a daily intraabdominal intake of the succinic acid in a dose of 100 mg/kg; the experimental group in which before ultraviolet radiation animals had a daily intraabdominal intake of Reamberin in a dose of 100 mg/kg (20 ml/kg). It was found out that in the blood of experimental animals a daily ultraviolet radiation during three minutes contributes to the increase of lipid hydroperoxides level (by 48-53%), of diene conjugate (by 43-48%), and of malonic dialdehyde (by 48-61%) against the decrease of antioxidant system activity in the blood of intact animals. The introduction of the succinic acid to rats contributes to the significant decrease in the blood of lipid hydroperoxides by 15-16%, of diene conjugates by 9-16%, and of malonic dialdehyde by 15% in comparison with the rats of the control group. The introduction of the succinate containing Reamberin to rats in the conditions of oxidative stress contributes to the significant decrease of lipid hydroperoxides in the blood by 27-28%, of diene conjugates by 23-28%, and of malonic dialdehyde by 26-29% in comparison with the rats of the control group. While analyzing the effect of the succinate containing drugs on the activity of the components of antioxidant system it was shown that the level of ceruloplasmin in the blood of animals was significantly higher by 25-32%, of vitamin E by 28-33% in comparison with the rats of the control group. So, the application of the succinate containing antioxidants in the conditions of ultraviolet radiation under experiment leads to the stabilization of the processes of peroxidation against the increase of antioxidant system activity. The intraabdominal introduction in laboratory animals of Reamberin in a dose of 100 mg/kg of succinate prevents the accumulation of lipoperoxidation products and increases the activity of main components of the antioxidant system in rats’ blood plasma, which indirectly exceeds similar effect of succinic acid in a dose of 100 mg/kg in the conditions of oxidative stress.

Modern environmental conditions dramatically increased the level of radical forming processes in the body. Heat exposure stimulates the generation of reactive oxygen species inducing peroxidation of lipids as a result of the development of hypoxia. In experimental conditions the possibility to correct free radical lipid oxidation of rats’ organism membranes was studied with the oral introduction of the phytoadaptogens that contain the complex of natural antioxidants. The animals were divided into 5 groups and each of them had 30 rats: intact animals (1) which were held in standard conditions of vivarium; the control group (2) in which rats were exposed to heat during forty-five minutes daily; the experimental groups (3, 4, 5) in which before the effects of heat animals had a daily oral intake of the extracts of Eleutherococcus, Rhodiola, liquorice in a dose of 1 ml/kg. It was found out that in the liver tissue of experimental animals a daily heat exposure during forty-five minutes contributes to the increase of lipid hydroperoxides level (by 34-41%), of diene conjugate (by 45-50%), and of malonic dialdehyde (by 62-74%) against the decrease of antioxidant system activity in the liver of intact animals. The introduction of phytoadaptogens to rats in the conditions of heat exposure contributes to the reliable decrease in the liver of lipid hydroperoxides by 22-28%, of diene conjugates by 22-32%, and of malonic dialdehyde by 30-37% in comparison with the rats of the control group. While analyzing the effect of the phytoadaptogens on the activity of the components of antioxidant system it was shown that the level of ceruloplasmin in the liver of animals was reliably higher by 68-82%, of vitamin E by 42-65% in comparison with the same parameters of the rats of the control group. So, the application of phytoadaptogens in the conditions of long heat exposure of the organism of animals under experiment leads to the stabilization of the processes of peroxidation against the increase of antioxidant system activity.

The animals were divided into 4 groups and each of them had 20 rats: intact animals which were held in standard conditions of vivarium; the control group in which rats were exposed to ultraviolet radiation during three minutes daily; the experimental group in which before ultraviolet radiation animals had a daily external application of the ointment of herb convolvulus; the experimental group in which before ultraviolet radiation animals had a daily external application of the ointment of herb chickweed. It was found out that in the blood of experimental animals a daily ultraviolet radiation during three minutes contributes to the increase of lipid hydroperoxides level (by 15-19%), of diene conjugate (by 14-16%), and of malonic dialdehyde (by 40-66%) against the decrease of antioxidant system activity in the blood of intact animals. The introduction of the ointment of herb convolvulus and of the ointment of herb chickweed to rats in the conditions of oxidative stress contributes to the reliable decrease in the blood of lipid hydroperoxides by 9-13%, of diene conjugates by 8-13%, malonic dialdehyde by 15-28% in comparison with the rats of the control group. While analyzing the effect of the ointment on the activity of the components of antioxidant system it was shown that the level of ceruloplasmin in the blood of animals was significantly higher by 13-20%, of vitamin E by 8-14%, of catalase by 13-28% in comparison with the same parameter of the rats of the control group. So, the application of the ointment of herb convolvulus and of the ointment of herb chickweed in the conditions of oxidative stress induced by the influence of ultraviolet rays leads to the stabilization of the processes of peroxidation against the increase of antioxidant system activity.

The aim of this study was to study the effect of combined MMSC and HSC transplantation on liver regeneration under conditions of toxic carbon tetrachloride damage. Materials and methods. The study was performed on white male mice with toxic liver damage by intraperitoneal administration of carbon tetrachloride at a dose of 50 µl per mouse once. An hour after modeling liver damage, placental MMSCs and HSCs were administered intravenously at a dose of 4 million cells/kg and 330 thousand cells/kg, respectively, suspended in 0.2 ml of 0.9% NaCl solution. Control group animals were given 0.9% NaCl solution-0.2 ml intravenously. On days 1, 3, and 7 after cell transplantation, changes in inflammatory activity in the liver were evaluated, and mitotic and apoptotic indices were determined. On the 7th day after the introduction of cells, the activity of DNA repair enzymes of the PARP family was analyzed. Results. Combined MMSC and HSC transplantation leads to a decrease in the index of inflammatory activity in the liver due to a decrease in necrosis, hepatocyte dystrophy, and a decrease in infiltration. As a result of the study, an increase in the activity of PARP repair enzymes was found, which led to a decrease in programmed cell death. Also, cotransplantation of MMSCs and HSCs was accompanied by increased mitotic activity of hepatocytes. Conclusion. Cotransplantation of MMSCs and HSCs under conditions of toxic liver damage reduces the inflammatory response, stimulates the mitotic activity of hepatocytes, and increases the activity of enzymes of the DNA repair system. Activation of the liver's reparative system, in turn, reduces the programmed death of hepatocytes.

Introduction. In Russia, the problem of toxic hepatitis is of great importance and relevance. The etiology of toxic hepatitis (industrial toxicants, drugs, ethanol) and, as a consequence, pathogenesis may have significant differences at the molecular genetic level. The aim of the study was to analyze the expression of genes involved in the response to toxic liver damage of various etiologies. Material and methods. Toxic hepatitis was modulated in male albino mongrel rats weighing 180-200 grams assigned to four groups (control group, carbon tetrachloride, paracetamol, ethanol). After 24 and 72 hours of paracetamol administration, rats were anesthetized and the mRNA levels of the Chek1, Gclc, Gstm1, Gstp1, Gstt1, Nfe2l2, Nqo1, Ripk1 genes in the liver homogenate were examined. Results. As a result of the analysis of the genes expression studied, the expression profile was found TO be differed depending on the etiology of toxic hepatitis. With carbon tetrachloride poisoning, an increase in the expression of the Nqo1 genes (p = 0.001), Gstm1 (p = 0.037) and a decrease in the expression of the Nfe2l2 genes (p = 0.004), Ripk1 (p = 0.004) was observed. With the liver damage by paracetamol and its metabolites, opposite to the expression of the Gstm1 gene (p = 0.001) decreased, and the expression of the Nfe2l2 (p = 0.009), Gclc (p = 0.001), Chek1 (p = 0.011) genes increased. During alcohol intoxication, there were no statistically significant changes in the expression profiles of the genes studied. Conclusion. the results obtained may indicate the involvement of various molecular genetic mechanisms in the process of response to toxic liver damage, depending on the etiology.

Among the most common pathologies of parasitic etiology in ruminants, trematode diseases, namely fasciolosis, occupy a prominent place in terms of the degree of damage and economic damage caused. Activation of lipid peroxidation processes plays a significant role in the mechanisms of the development of fasciolosis in animals. The research aimed to investigate the level of lipid peroxidation products in cows' blood in experimental fasciolosis. For the test, 12 cows of 4–5 years of age, black-spotted breed, were selected, from which 2 groups were formed, six animals in each. Animals in the control group were clinically healthy. Animals of the experimental group were experimentally infected with adolescents. During the research, the rules of compulsory experiments were followed – selecting and keeping analog animals in groups. The cows' diet was balanced in terms of nutrients and minerals. Based on the studies, it was found that after the experimental infestation of cows of the experimental group with the causative agent of fasciolosis, an increase in the level of diene conjugates of their blood on the 15th day of the experiment by 30.3 %, and on the 25th day – by 50.4 % relative to the control group. There was also an increased level of lipid hydroperoxides in their blood, where, respectively, on the 10th and 15th day of the experiment, this figure increased by 18.3 and 32.9 % compared with the control group. On the 25th day of the experiment, the level of lipid hydroperoxides in the blood of cows of the experimental group was the highest. Compared with the control group, it increased by 46.6%, respectively. In the study of the final products of lipid peroxidation, it was found that on the 10th day of the test, the level of TBA-active products in infected cows of the experimental group increased by 9.5 %. On the 15th and 20th days of the experiment, the level of end products of LPO in the experimental group's blood of animals was higher by 20.3 and 26.0 % relative to the control group. The increase in TBA-active products' level reflects the activation of peroxide processes in animals with experimental fasciolosis. Therefore, in the clinical manifestation of fasciolosis, parasites secrete metabolic products that promote the formation of free radicals, which in turn enhance the initiation of lipid peroxidation processes.

The purpose of this search was to investigate the influence of “Thireomagnile” and “Thyrioton” on the antioxidant status of cows in the development of endotoxicosis. The experimental part of the work was performed at the Department of Pharmacology and Toxicology of Stepan Gzhytskyj Lviv National University of Veterinary Medicine and Biotechnologies, in the laboratory of Pharmacology and Toxicology of the State Scientific Research Institute of Veterinary Preparation and Feed Additives (Lviv) and in the Ltd. “Barkom”, village Yampil of Pustomyty district, Lviv region. To solve the tasks of the research, three experimental groups of 10 pregnant cows of the Ukrainian Black-Spotted Dairy breed were formed in each: control and two experimental. Cows of the control group (K) had characteristic clinical signs of endotoxicosis. Animals of the first experimental group (E1) were given 10 mL per animal of “Thyroid magnet” at the eighth and ninth months of gestation. The animals of the second experimental group (E2) were given Thyrioton at a dose of 10 mL per animal in the eighth and ninth months of gestation. As a result of the conducted researches it is found out that in pregnant cows with clinical manifestation of endotoxicosis the inhibition of the activity of the antioxidant protection system and intensification of processes of lipid peroxidation occurs. The use of “Thireomagnile” and “Thyrioton” in cows by the development of endotoxicosis contributed to the increased activity of the antioxidant system, which is indicated by an increase in the activity of glutathione peroxidase, glutathione reductase and glucose-6-phosphate dehydrogenase. The use of of “Thireomagnile” and “Thyrioton” to cows in the development of endotoxicosis contributed to the suppression of lipid peroxidation processes, which is indicated by the low level of primary and final products of lipid peroxidation, namely: lipid hydroperoxides and TBA-active products. Better normalizing action on the equilibrium on the activity of the antioxidant system and the intensity of lipid peroxidation was shown by the introduction of “Thyroid magnet” at a dose of 10 cm3 per animal.

Aim. Study of the activity of lipid peroxidation and state of antioxidant system in children with a nephrotic form of chronic glomerulonephritis. Methods. 104 children with a nephrotic form of glomerulonephritis were examined. The average age of children was 10.18±4.03 years. 46.2% of children were in remission, 32.7% of children had 1st degree of activity, 14.4% had 2nd degree, and 6.7% had 3d degree of nephrotic syndrome. Control group included 30 children without chronic glomerulonephritis. The levels of diene conjugates, malonic dialdehyde, catalase, reduced glutathione, glutathione peroxidase, and glutathione reductase in blood plasma and erythrocytes were determined. Results. In children with chronic glomerulonephritis the concentration of diene conjugates and malonic dialdehyde was elevated in blood plasma and erythrocytes. In erythrocytes their concentration was statistically significantly higher at all degrees of activity of nephrotic syndrome than in control group. We have studied the interdependence of the intensity of oxidative processes/antioxidant level and the thyroid status parameters in a group of students aged 18–24 who lived for a long time in the territory of enhanced radioecological control (density of soil contamination by isotopes (137Cs 3.7 ∙ 104 – 18.5 ∙ 104 Bq/m2). We examined 50 people from relatively environmentally friendly areas (control group) and 50 people from IV radiation zone (experimental group). In the experimental group, there were no individuals with clinical manifestations of thyroid pathology. However, subgroups with signs of hyperthyroidism and hypothyroidism were identified. We evaluated the level of cortisol, thyrotrophic hormone (TSH), triiodothyronine (T3), thyroxine (T4), malonic dialdehyde (MDA), ceruloplasmin (CP), transferrin (Tf), sulfhydryl groups (SH); we calculated the oxidative stress index (OSI). The research was conducted one month before the examination time and also during the exams as a factor in increased emotional stress. A lowered CP level was found in the subgroup with signs of hypothyroidism; SH groups – in all subgroups, separated by thyroid status. The oxidative stress index was higher in all students examined of the experimental group, compared with the control. The growth of MDA level is marked in the experimental group – it is the most strongly pronounced in conditions of additional emotional load in people with signs of hyperthyroidism and hypothyroidism. CP level significantly decreased in the subgroup of hyperthyroidism on the background of T3 decrease. OSI increased in all students examined from the experimental group. In the subgroup of hypothyroidism it became significantly higher than in the subgroup of euthyroidism. A positive correlation between the levels of CP and T3 was found. The highest values of the correlation coefficients were noted for subgroups with signs of hyperthyroidism and hypothyroidism, with the coefficient significance increasing under conditions of emotional stress. The index of oxidative stress in the experimental group positively correlated with the level of TSH – in terms of emotional stress, the statistical significance of the coefficients disappeared. In the subgroups divided by thyroid status, variability of interactions between OSI and T3 was observed but it was not statistically significant. It was found that the participation of thyroid status in supporting redox homeostasis in people aged 18–24 who suffered from chronic small-doze radiation exposure was realized mainly by the influence on the antioxidant system. The ability of thyroid hormones to maintain a proper antioxidant state was suppressed in this group. The unbalanced relationship between thyroid hormones and oxidative stress indicators is strongly manifested under conditions of additional emotional stress.

A highly relevant problem of modern veterinary science is the study of features and mechanisms of combined action of the most common heavy metals – cadmium and plumbum and their influence on the body of humans and animals in the regions of Ukraine under technogenic pollution. The purpose of the work was to study the influence of heavy metals on the state of the antioxidant protection system of cows, in particular on the content of lipid peroxidation products (malonic dialdehyde, lipid hydroperoxides and diene conjugates), and activity of antioxidant enzymes (glutathione peroxidase and superoxide dismutase), depending on the distance to the heaps of mines in the coal basin. The study objects were cows of black-and-white breed at the age of 3–7 years. It was established that this parameter in the place with the highest concentration of diene conjugates in the blood of cows was by 25.8 % higher compared to the place of low concentration and 12.1 % higher compared to the place with medium concentration. In the place with the highest content of lipid hydroperoxides in the blood of cows the parameter was 23.7 % higher compared to the cows from the place with the low content. The concentration of lipid hydroperoxides in the blood of cows from the place with the medium content was 16.7% higher compared to the cows from the place with the low content. The parameter from the place with the lowest content of lipid hydroperoxides in the blood of cows was 12.1% lower compared to the place with the highest content. The level of malonic dialdehyde in the blood of cows from the technogenic pollution zone in the place with the largest amount was higher by 36.2; 34.0 and 18.8 % – compared to places with medium and low levels, respectively. The activity of superoxide dismutase in the blood of cows in the place with its highest activity was 0.284 ± 0.0099 % block. reac/g Hb, and in the place with the lowest activity – 0.23 ± 0.0051 % block. reac/g Hb. The activity of glutathione peroxidase in the blood of cows in farms of the technogenic pollution zone depended on the distance to the mine. These researches will further develop effective methods of treating cows under the influence of heavy metals, in particular regarding the antioxidant system.
