Introduction:Infertility resulting in men has evoked considerable medical and social interest. The dermatoglyphic study has been proven to be a useful diagnostic tool for different diseases. It is easy due to its cost-effectiveness and non-invasive procedure. Multiple aetiological factors are responsible for azoospermia and oligozoospermia. This study was done to know thedermatoglyphic patterns among male patients suffering from azoospermia and oligozoospermia and compare the findings with the general population. Methods:Total seventy-six cases suffering from azoospermia and oligozoospermia were taken with an equal number of individuals in the control group. The fingerprint patterns and ridge counts were compared with healthy age-matched control subjects. The fingerprint patterns were collected by standard ink method. The data was collected and subjected to Chi-square and analysis of variance at a 95 % confidence interval. Results:The overall frequency was loop was higher, followed by whorls and arches in men with azoospermia and oligozoospermia. It was observed that comparing the fingertip ridge patterns and their frequencies were significant (p<.05) inazoospermia and oligozoospermia as compared to controls.The increase in the total number of arches wasobserved in men with oligozoospermia as compared with respect to the control group. Thedifferencesin cases and control groups with respect to parameters such as Total finger ridge count (TFRC)as well as Absolute finger ridge count (AFRC) were found statistically insignificant. Conclusions:There is a substantial correlation between fingerprint patterns of men withazoospermia and oligozoospermiamales attending the In Vitro fertilisation Centre. These correlations may have an essential role in the early diagnosis of reproductive dysfunction in the future.

In order to reduce fetal losses of bovine clones the replacement of trophectoderm (TE) by microinjection of immunosurgically isolated inner cell mass (ICM) was proposed. It was reported that those techniques could damage both ICM and TE. An alternative to decrease this problem would be the technique of cell aggregation by approximation. Currently it has been used only in pre-compaction embryos. The aims of this study were to evaluate the effectiveness to aggregate post-compaction embryos and to verify the feasibility of reconstructed blastocysts production by ICM and TE approximation. Mice (Swiss Webster, SW; and C57BL/6/EGFP, EGFP) were superovulated to obtain embryos 2.5 to 3.5 days post-coitus. In both control groups (CG; exp. 1 and 2), whole embryos from 8 cells to early morula stages and without zona pellucida were aggregated. In all experiments, bisection was performed with a blade controlled by micromanipulator to produce two fragments according to the requirements of each experiment (i.e. demi embryos in Exp. 1 and Exp. 2 or TE fragment and whole ICM in Exp. 3). The joined fragments were in vitro cultured in microwell (300 µm diameter and based on well of the well system) filled with 400 µL of KSOMaa, covered with mineral oil by 24 h (incubation 37°C, 5% of CO2 and saturated humidity). Embryonic aggregation rate (AR) was evaluated by detection of a single and cohesive cell mass (CG from Exp. 1 and Exp. 2) or by the presence of a typical morphologically blastocyst (all the remaining groups). Only to validate AR evaluation, 25% of the aggregation attempts were performed with distinct phenotype embryos (EGFP and SW) and they could be tracked under epifluorescence. In Exp. 1, the aggregation by approximation was tested to evaluate embryonic stages: 2 demi-blastocysts (2DB; n = 28), demi-morula and demi-blastocyst (DMDB; n = 20) and control group (CG; n = 25). In Exp. 2, an adhesive agent (phytohemagglutinin; PHA) and fragment increasing were used in the groups: 2DB (n = 24), 4 demi-blastocysts (4DB, that is four half-blastocyst) and CG (n = 22). After aggregation validation (Exp. 1 and Exp. 2), blastocyst reconstruction by the approximation of ICM and TE fragments (with PHA presence) was tested on groups: 1 TE fragment and 1 ICM (TI; n = 48) and 2 TE fragments and 1 ICM (2T1I; n = 17). The rates were analyzed by chi-square and Fisher’s exact tests (significance of 5%). In experiments 1 (3.6; 15.0; and 60.0%, respectively to 2DB, DMDB, and CG) and 2 (8.3; 36.4; and 77.3%, respectively to 2DB, 4DB, and CG), the AR differed among groups with exception of 2DB and DMDB (exp. 1; P > 0.05). In Exp. 3, there was no difference on the AR between TI and 2T1I (27.1 and 29.4%, respectively). Despite the low adhesion potential of the trophectoderm cells, is feasible to produce chimeric embryos by aggregation. We infer that the use of an adhesive agent could increase AR maintaining the contact of the embryonic fragments. The proposed technique was practical and effective for blastocyst reconstruction. Additionally, the present model established in mice is about to be tested in cattle for further validation. Financial support: FAPESP.

Introduction: Dermatoglyphics is an advancing branch of medical science in which the dermal ridge patterns are studied and used in prediction of genetic disorders. Objective: To assess the usefulness of dermatoglyphics, as a non-invasive early predicator in RAS, since genetics plays a role in both. Materials and methods: This case control study comprised of 40 patients with recurrent aphthous stomatitis compared to 30 control subjects of similar age and sex. All subjects were investigated for their dermatoglyphic patterns of both hands, which were obtained by smearing the ink uniformly over the palm and fingers and pressing the hands firmly against the good quality paper. Qualitative analysis of prints was done for arches, loops and whorls and quantitative analysis included Total Finger Ridge Count (TFRC) and atd angle. Fisher’s Exact Tests were used to evaluate the significance between recurrent aphthous stomatitis and the dermatoglyphic features. Results: Among various dermatoglyphic parameters analyzed, recurrent aphthous stomatitis patients showed 9% arches, 62% loops and 29% whorls as the finger ridge configuration. In comparing the results between the two groups, the frequency of arches was observed more in study group while composite whorl and the nar pattern were more frequent in control group, the results were statistically significant (p < 0.05). In addition to this, study group patients demonstrated higher frequency of total finger ridge count, and control group showed higher frequency for atd angle on both hands, statistically significant difference is observed (p < 0.05). Conclusion: The study found significant correlation between palmar dermatoglyphics and recurrent aphthous stomatitis, suggesting that genetics is one of the host risk factor associated with the latter, and could aid in early detection of the disease.

Abstract α-tocopherol is known to be a powerful antioxidant, in this regard, it was added to bovine oocyte in vitro maturation media to evaluate its effect on oocyte maturation. Oocytes (n = 624) aspirated from ovaries of slaughtered cows were classified by quality and divided in four categories according to cytoplasm appearance and cumulus cells layers. Oocytes were washed in TCM-199 supplemented with fetal bovine serum (FBS) and FSH, then distributed in maturation media (TCM-199 supplemented with FBS, FSH and gentamicin). Three experimental groups of α-tocopherol (50, 100 and 200 mM) and a control group without α-tocopherol were used. Maturation was carried 22 h at 38.5°C in a 5% CO2 atmosphere. Oocytes were examined to determine cumulus expansion as categorical data (expansion or no expansion), as well as cumulus expansion Index (CEI). For CEI determination oocytes were graded 0 to 4, being 0 those with null expansion and 4 those with a noticeable cell expansion, then the number of oocytes were multiplied by the grade given and a sum of the totals was obtained, the new total was divided by the total of oocytes in the group and the result obtained corresponded to the CEI of the group. Results were analyzed with Chi Square test (for maturation rates) and an ANOVA (for the CEI) using the SAS system, data are presented as mean ± standard error. There was no statistical difference between control and α-tocopherol groups (P &gt;0.05). Numerically, the control group showed a higher maturation rate (100%) and obtained a higher CEI (2.44±0.20), followed by the 50 mM group (98.16%; 2.39±0.13), the groups 200 mM (97.40%; 2.00±0.14) and 100 mM (96.25%; 2.06±0.24) were the lowest. The addition of the minimum concentration (50 mM) of α-tocopherol to the maturation media could improve maturation rates without exposing oocytes to toxic effects.

AbstractEndometriosis is an inflammatory disease which diagnostics is difficult and often invasive, therefore non-invasive diagnostics methods and parameters are needed for endometriosis detection. The aim of our study was to analyse the glycosylation of native serum IgG and IgG isolated from sera of women classified as: with endometriosis, without endometriosis but with some benign ginecological disease, and control group of healthy women, in context of its utility for differentiation of advanced endometriosis from the group of healthy women. IgG sialylation and galactosylation/agalactosylation degree was determined using specific lectins: MAA and SNA detecting sialic acid α2,3- and α2,6-linked, respectively, RCA-I and GSL-II specific to terminal Gal and terminal GlcNAc, respectively. The results of ROC and cluster analysis showed that the serum IgG MAA-reactivity, sialylation and agalactosylation factor may be used as supplementary parameters for endometriosis diagnostics and could be taken into account as a useful clinical tool to elucidate women with high risk of endometriosis development. Additionally, we have shown that the analysis of native serum IgG glycosylation, without the prior time-consuming and expensive isolation of the protein, is sufficient to differentiation endometriosis from a group of healthy women.

Background. Retained hemothorax (RH) is a common problem in cardiothoracic and trauma surgery. We aimed to determine the optimum agitation technique to enhance thrombus dissolution and drainage and to apply the technique to a porcine-retained hemothorax. Methods. Three agitation techniques were tested: flush irrigation, ultrasound, and vibration. We used the techniques in a benchtop model with tissue plasminogen activator (tPA) and pig hemothorax with tPA. We used the most promising technique vibration in a pig hemothorax without tPA. Statistics. We used 2-sample t tests for each comparison and Cohen d tests to calculate effect size (ES). Results. In the benchtop model, mean drainages in the agitation group and control group and the ES were flush irrigation, 42%, 28%, and 2.91 ( P = .10); ultrasound, 35%, 27%, and .76 ( P = .30); and vibration, 28%, 19%, and 1.14 ( P = .04). In the pig hemothorax with tPA, mean drainages and the ES of each agitation technique compared with control (58%) were flush irrigation, 80% and 1.14 ( P = .37); ultrasound, 80% and 2.11 ( P = .17); and vibration, 95% and 3.98 ( P = .06). In the pig hemothorax model without tPA, mean drainages of the vibration technique and control group were 50% and 43% (ES = .29; P = .65). Discussion. In vitro studies suggested flush irrigation had the greatest effect, whereas only vibration was significantly different vs the respective controls. In vivo with tPA, vibration showed promising but not statistically significant results. Results of in vivo experiments without tPA were negative. Conclusion. Agitation techniques, in combination with tPA, may enhance drainage of hemothorax.

AbstractBackgroundAnxiety and fear of labor pain has led to elevated cesarean section rate in some countries. This study was conducted to investigate the effect of yoga in pregnancy on anxiety, labor pain and length of labor stages.MethodsThis clinical trial study was performed on 84 nulliparous women who were at least 18 years old and were randomly divided into two groups of yoga and control groups. Pregnancy Yoga Program consisting of 6 60-min training sessions was started every 2 weeks from week 26 of pregnancy and continued until 37 weeks of gestation. Anxiety severity at maternal admission to labor was measured by the Spielbergers State-Trait Anxiety Inventory, and labor pain was measured by Visual Analogue Scale (VAS) at dilatation (4–5 cm) and 2 h after the first measurement. Data were analyzed using Chi-Square and t-test.ResultsIntervention group reported less pain at dilatation (4–5 cm) (p=0.001) and 2 h after the first measurement (p=0.001) than the control group. Stat anxiety was also lower in intervention group than the control group (p=0.003) at the entrance to labor room. Subjects in the control group required more induction compared to intervention group (p=0.003). Women in intervention group experienced shorter duration of the first phase of the labor than the control group (p=0.002). Also, the total duration of two stages of labor was shorter in intervention group than the control group (p=0.003).ConclusionsPracticing yoga during pregnancy may reduce women’s anxiety during labor; shorten labor stages, and lower labor pain.

SUMMARY Uptake of tritiated lysine vasopressin ([3H]LVP) was studied in halved neural lobes of rats (which had been given either tap water (control group) or 2% (w/v) NaCl solution as drinking water for 4 days) as well as in slices of pig neural lobe. Uptake of radioactivity into the neural lobes was shown but analysis of the extracts of incubated lobes of both species by ion exchange chromatography showed that very little of it remained in the tissue as hormone. In addition, some radioactivity was associated with trichloroacetic acid-insoluble proteins. After 90 min of incubation, and after correction for the breakdown, the uptake of unchanged [3H]LVP, expressed as a tissue: medium ratio, was 0·14 ± 0·04 and 0·09 ± 0·03 (mean ± s.e.m.) for the saline-treated and control rats respectively, while the tissue: medium ratios for the breakdown product(s) were 6·47 ± 0·45 and 5·50 ± 0·36. The results suggest uptake of [3H]LVP into the cell with almost complete intracellular breakdown of the hormone.

AbstractAssessment of the endometrium often necessitates a biopsy, which currently involves an invasive, transcervical procedure. Here, we present an alternative technique based on deriving organoids from menstrual flow. We demonstrate that organoids can be derived from gland fragments recovered from menstrual flow. To confirm they faithfully reflect the in vivo state we compared organoids derived from paired scratch biopsies and ensuing menstrual flow from patients undergoing in vitro fertilisation (IVF). We demonstrate that the two sets of organoids share the same transcriptome signature, derivation efficiency and proliferation rate. Furthermore, they respond similarly to sex steroids and early-pregnancy hormones, with changes in morphology, receptor expression, and production of ‘uterine milk’ proteins that mimic those during the late-secretory phase and early pregnancy. This technique has wide-ranging impact for non-invasive investigation and personalised approaches to treatment of common gynaecological conditions, such as endometriosis, and reproductive disorders, including failed implantation after IVF and recurrent miscarriage.

Abstract Background To compare the aqueous humor (AH) and the serum clusterin levels of patients with pseudoexfoliation syndrome (PEX), pseudoexfoliation glaucoma (PEXG), and primary open-angle glaucoma (POAG) with each other and with an age- and sex-matched control group. Methods This prospective, cross-sectionalstudy evaluated 92 eyes from 92 adult cases of uncomplicated phacoemulsification and posterior chamber intraocular lens (IOL) implantation. The cases were divided into PEX, PEXG, POAG, and control groups. Serum samples were taken from the antecubital vein just before the surgery, and the AH samples were aspirated at the beginning of the surgery. Kruskal-Wallis H, One-way ANOVA, Mann-Whitney U with Bonferroni correction and Chi-Square tests were used for statistical analysis. Results The serum clusterin levels were the highest in the PEXG group, but no statistically significant differences were observed between the groups (p=0.633). The mean AH clusterin levels were 286.79±29.64 μg/mL in the PEXG group, 263.92±31.70 μg/mL in the PEX group, 272.59±49.71 μg/mL in the POAG group, and 193.50±62.38 μg/mL in the control group (p< 0.001). This came out to be 1.48 times increase for the PEXG group, 1.36 for the PEX group, and 1.41 for the POAG group when compared with the control subjects. Conclusions A higher level of clusterin in the anterior chamber was found to be associated with PEX and PEXG. In addition, a high level of anterior chamber clusterin in POAG, which is a new finding, showed that this molecule might be important not only in pseudoexfoliation, but also other types of glaucoma like POAG.

Objective: To evaluate the effectiveness of placing a resorbable collagen barrier in impeding the extrusion of obturation material in primary molars undergoing resorption. Study design: All the 94 canals in 47 mandibular molars were allocated to 2 groups- Group ‘A’- 47 canals with collagen barrier (Test group) and Group ‘B’- 47 canals without collagen barrier (Control group) based on randomization protocol. Pulpectomy was performed and obturation of both test and control canals were radiographically assessed. Pearson’s chi – square test was applied to analyze the results. The significance level was predetermined at p &lt; 0.05. Results: Among the test group, 93.6% of the canals showed no extrusion while, 6.4% showed visible extrusion of the material outside the apex. In the control group, 83% showed no extrusion whereas 17% of the canals showed visible extrusion outside the apex. But no significant difference was noted (p&gt;0.05). Conclusion: The placement of resorbable collagen barrier in the apical third of the canal prevented the extrusion of obturating material beyond the apex in resorbing primary molars.
