Reproductive activity and morphometric assessment of three commercial species of sea cucumber (Echinodermata) from Karimunjawa National Park, Indonesia. Biodiversitas 22: 3333-3341. Sea cucumbers are benthic animals that have economic value and play an important role in the environment. Exploitation on large scale to fulfill high market demand, caused overexploitation. The number of sea cucumbers in Karimunjawa decreased as a result of limited management. There is no study conducted on the commercial sea cucumbers (Holothuria edulis, Pearsonothuria graeffei, and Bohadschia vitensis) in Nyamuk Island, Karimunjawa. The objectives of the research were to determine the morphometric assessment and the reproductive cycle of the three species. The morphometric assessment of weight showed that the weight of H. edulis ranges 247.3-2539g, P. graeffei 415.3-428.1g, and B. vitiensis 672.4-678.1g, respectively. The first maturity size of gutted body weight (GWB50) for H. edulis was 127.84g, P. graeffei 357.61g, and B. vitiensis 279.34g. The gonad somatic index (GSI) range 13.60-15.60% for H. edulis, 14.75-12.25% for P. graeffei and 10.83-14.57% for B. vitiensis. The result of gonad somatic index with a value above 10% means that it has entered the spawning phase. The above data indicated that the three commercial species spawned monthly, during new moon. It is likely that the spawning peak occurred when the temperature was increased. However, the pattern of reproductive activity in this study was limited at Nyamuk Island. Future studies are needed to observe whether there is a correlation between the increasing water temperature and the reproductive activity of the three sea cucumbers.

Frondoside C (1) is a new sulfated nonholostane triterpene glycoside obtained (with the glycosides closed by aglycone structure as impurities) from the sea cucumber Cucumaria frondosa. Its structure has been elucidated on the basis of spectral data (NMR and MS) of compound 1 and of its desulfated derivative (2) obtained by solvolysis. Frondoside C (1) is just the seventh glycoside reported from sea cucumbers, having a lanostane-type aglycone devoid of the typical 18(20)- lactone ring.Key words: Cucumaria frondosa, sea cucumbers, frondosides, triterepene glycosides, antitumor activity.

Frondosides A2-4 (1), A2-7 (2), and A2-8 (3) are new monosulfated triterpene glycosides isolated from the sea cucumber Cucumaria frondosa. Their structures have been elucidated on the basis of spectral data (2D NMR and MS). Frondosides A2-7 (2) and A2-8 (3) are isomers and differ from each other only by the position of a double bond in their non-holostane-type aglycones.Key words: Cucumaria frondosa, sea cucumbers, frondosides, triterpene glycosides.

A reliable and harmless mark–recapture method provides valuable information for the management of commercial sea cucumber species. Nevertheless, marking and tracking sea cucumbers is notoriously difficult and represents a serious challenge. In this study, we tested one external and one internal tag in the sea cucumber Holothuria grisea. A passive integrated transponder (PIT) tag was inserted into the coelomic cavity in one treatment, whereas a T-bar (external tag) was attached in the upper surface of the body wall in the other treatment; sea cucumbers were then followed for 17 weeks. The tagging procedure caused no evisceration in the experimental groups, nor was a significant difference in growth rate found between treatments. The retention of the PIT tag was low, with 100% lost by Week 9. Retention of T-bars was higher, with 90% still attached by the end of the Week 8, but retention decreased thereafter as T-bar absorption increased. No relationship was found between initial weight and the number of weeks PIT tags (r=–0.173, P=0.781) or T-bars (r=–0.220, P=0.652) were retained. Neither the T-bar nor the PIT tags fulfilled the requirements of high retention required for long-term studies. However, we do recommend the use of T-bars for short-term studies for H. grisea under laboratory conditions or in a habitat with low substrate complexity.

Frondosides A2-1 (1), A2-2 (2), A2-3 (3), and A2-6 (4) are new monosulfated holostane pentaosides isolated from the sea cucumber Cucumaria frondosa. Their structures have been elucidated on the basis of spectral data (2D NMR and MS). Frondoside A2-2 (2) contains an α,β-unsaturated ketone fragment in the side chain of the aglycon, unique for triterpene glycosides of sea cucumbers.Key words: Cucumaria frondosa, sea cucumbers, frondosides, triterpene glycosides.

Holothuroids (sea cucumbers) are one of the most ubiquitous groups of benthic animals found across diverse marine ecosystems. As echinoderms, they also occupy an important place in the evolutionary hierarchy, sitting close to vertebrates in the deuterostome clade, making them valuable multidisciplinary model organisms. Apart from being ecologically and phylogenetically important, many species are commercially exploited for luxury seafood markets. With the global rise of aquaculture and fisheries, management and protection of these valuable species relies on a better understanding of how their immune systems respond to environmental and anthropogenic stressors. Here, the cellular, hormonal and behavioral indicators of stress in the North Atlantic sea cucumber Cucumaria frondosa were examined. The immediate and carry-over (post recovery) effects of a 1-hour exposure to low salinities or to emersion (at two temperatures) highlighted that morphoplasticity in C. frondosa was accompanied by shifts in all monitored indicators. From baseline levels measured in controls, densities of free coelomocytes increased, showing successions of specific cell types and subsequent coelomocyte aggregations, combined with a rise in cortisol levels. These responses mirrored increased fluctuations in cloacal opening rates, decreased force of attachment to the substrate, and enhanced movements and active buoyancy adjustment with increasingly severe stressors. The findings suggest that many systems of sea cucumbers are impacted by stresses that can be associated with harvesting and handling methods, with likely implications for the quality of the processed products. Gaining a deeper understanding of immune and hormonal responses of sea cucumbers is not only of broad ecological and evolutionary value, but also helpful for the development of sustainable fisheries and aquaculture practices, and conservation programs.

Orange-footed Sea Cucumber (Cucumaria frondosa; Echinodermata: Holothuroidea) is a dark-brown species that is broadly distributed in North Atlantic and Arctic waters. Here, we document the rare occurrence of colour morphs showing various degrees of albinism, from totally white to faint orange pigmentation. These unusually coloured individuals were found across a broad distribution range in eastern Canada and northeastern United States, with their occurrence in Newfoundland samples ranging from 0.2% to 0.5%. Two fully albino individuals were noticeably smaller than other colour morphs. The occurrence of rare, unusually coloured sea cucumbers is important from an ecological standpoint and may also have commercial implications.

Sea cucumbers (Cucumaria frondosa) (Echinodermata: Holothuroidea) held in flow-through tanks were exposed to different chloropigment concentrations by manipulating the seston concentration via the use of water filters in the inflow lines and the addition of cultured algae or natural plankton to tanks. The percentage of sea cucumbers open and feeding increased asymptotically as the seston chloropigment concentration increased. Below 50 µg·L-1 the percentage of sea cucumbers with tentacles extended and feeding decreased. The rate at which sea cucumbers inserted their tentacles into their mouths (0.96 ± 0.34 insertion/min (mean ± SD)) increased with increasing seston quality. Cucumaria frondosa fed at faster rates at the higher concentrations. There was a significant positive relationship between the stomach content (amount of chloropigment) of sea cucumbers and the seston chloropigment concentration to which they were exposed (r2 = 0.200, p < 0.001).

Seven highly polar trisulfated triterpene glycosides belonging to the frondoside A7 group have been isolated from the North Atlantic sea cucumber Cucumaria frondosa. The structures of five new glycosides, frondosides A7-1 (1), A7-2 (2), A7-3 (3), A7-4 (4), and isofrondoside C (5) were elucidated, three of which contained lanostane aglycons without a lactone-ring. Three pairs of the glycosides proved to be isomers by virture of the positions of double bonds in the aglycons nuclei; two pairs of the glycosides differed from each other by the character of the oxygen functionality at C-22. The results from these studies provided the basis for postulating the biosynthetic pathways of norlanostane glycosides in the sea cucumber.Key words: Cucumaria frondosa, sea cucumbers, frondosides, triterpene glycosides.

<strong>The Utilization of Sea Cucumber in Kaimana Regency, West Papua Province.</strong>Sea cucumbers are one of marine commodities that are traded at local, national and international scale. Kaimana Regency, West Papua Province is one of the sea cucumber producers in Indonesia. The exploitation of sea cucumber in this regency has been carried out for long times, applying the<em>sasi</em>system. The purpose of this study is to determine the exploitation of sea cucumbers with the <em>sasi</em> system. Data were collected in April 2018 in two location, namely Kayumerah Island and Kambala Village. Focus Group Discussion (FGD) was perfomed out using structured questions to gather information of the sea cucumber exploitation. The snowball technique was used to locate the sea cucumber collectors. This study results showed 22 commercial species of sea cucumber were found in Kayumerah Island and 33 species in Kambala Village. Duration of open-closed sasi in Kayumerah Island was 2 to 3 years for 14 days, while in Kambala Village was 2 years for 6 months. The average catched of sea cucumbers was 3.57 kg dry weight/person/ day in Kayumerah Island and 0.09 kg dry weight/person/day in Kambala Village. These commodities were sold to the city nearby. Sasi system at the study site seems to have changed due to an auction system or fee for collecting of sea cucumbers and a changes in capture methods, through diving supported by compressor
