Protective role of Sterculia tragacantha aqueous extract on pancreatic gene expression and oxidative stress parameters in streptozotocin-induced diabetic rats

Abstract Objectives The current study evaluates the protective role of aqueous extract of Sterculia tragacantha leaf (AESTL) on pancreatic gene expressions (insulin, PCNA, PDX-1, KI-67 and GLP-1R) and oxidative stress parameters in streptozotocin-induced diabetic rats. Methods Diabetes mellitus was induced into the experimental Wistar animals via intraperitoneal (IP) injection of streptozotocin (35 mg/kg body weight) and 5% glucose water was given to the rats for 24 h after induction. The animals were categorized into five groups of 10 rats each as follows normal control, diabetic control, diabetic rats administered AESTL (150 and 300 mg/kg body weight) and diabetic rats administered metformin (200 mg/kg) orally for two weeks. Thereafter, the animals were euthanized, blood sample collected, pancreas harvested and some pancreatic gene expressions (such as insulin, PCNA, PDX-1, KI-67, and GLP-1R)s as well as oxidative stress parameters were analyzed. Results The results revealed that AESTL significantly (p<0.05) reduced fasting blood glucose level, food and water intake, and lipid peroxidation in diabetic rats. Diabetic rats administered different doses of AESTL showed a substantial upsurge in body weight, antioxidant enzyme activities, and pancreatic gene expressions (insulin, PCNA, PDX-1, KI-67, and GLP-1R). Conclusions It can therefore be concluded that AESTL has the ability to protect the pancreas during diabetes mellitus conditions.

Background: The present study evaluated the effect of phenolic aqueous leaf extract of Vitex doniana on body weight, serum glucose and oxidative stress parameters in diabetes mellitus (DM) rats. Methods: DM was induced in rats by intraperitoneal injection of 100 mg/kg alloxan monohydrate in phosphate buffered saline (PBS; pH = 7.4). A total of 36 adult male Wister albino rats were divided into 6 groups of 6 rats. The groups consist of normal untreated rats, untreated DM rats, DM rats treated with 500 mg/kg dimethylguanide (MetforminTM) and DM rats treated with 100, 200 and 400 mg/kg body weight of phenolic aqueous leaf extract of Vitex doniana. The rats were treated for 28 days. Serum glucose, malondialdehyde (MDA), glutathione (GSH) and ascorbic acid concentrations, and serum superoxide dismutase (SOD) and catalase (CAT) activities were measured using standard methods. The changes in body weight was also measured. Results: The results showed a significant reduction (p < 0.05) in serum glucose and MDA concentration, whereas serum SOD, CAT activities, as well as GSH and ascorbic acid concentration were significantly elevated (p < 0.05) in treated DM groups in a dose dependent manner. Conclusion: The results of the present study indicated that phenolic aqueous leaf extract of V. doniana promoted increased body weight, ameliorated DM and alleviated reduced antioxidant activities in alloxan-induced DM rats. Phenolic aqueous leaf extract of V. doniana could serve as a potential natural and safe remedy for the management of DM.

Bridelia atroviridis Müll. Arg. (B. atroviridis) is a plant used in Cameroonian traditional medicine to manage diabetes. The effects of hydroethanolic barks extract from B. atroviridis were evaluated on diabetes disorders including hematology, inflammatory, and oxidative stress parameters. The in vitro antioxidant capacity of the hydroethanolic bark extract (70 : 30) was evaluated. Nicotinamide-/streptozotocin-induced diabetic rats were daily treated with the B. atroviridis extract for fifteen days. Glycemia were evaluated every 5 days, insulin sensibility test was performed, and haematological, inflammatory, and oxidative stress parameters were analysed. Histomorphometry of the pancreas was realized. The extract was able to scavenge free radicals in vitro and decrease significantly the blood glucose levels. The treatment resulted in a significant alleviation of insulin resistance, anemia, leukocytopenia, and thrombocytopenia observed in untreated diabetic rats. The extract significantly decreased proinflammatory cytokines TNF-α, IL-1β, and IL-10. The rate of reduced glutathione was increased in the pancreas, whereas the catalase activity and nitrite concentration were decreased. Diabetic control showed a reduced size of Langerhans islet, whereas the size of islets was large in treated groups. The hydroethanolic extract of B. atroviridis was able to improve glycemia and alleviate haematological and inflammatory parameters disorders observed in diabetic conditions, probably due to its antidiabetic, anti-inflammatory, and antioxidant capacities.

The aim of present study was to unravel the protective role of vitamin C on oxidative stress parameters in lung homogenates of bifenthrin intoxicated rats. Rats were divided into four groups. Group I served as control while group II animals were treated with bifenthrin @ 5.8mg/Kg/day. In group III, vitamin C was orally administered @ 60mg/Kg/day where as group IV received both vitamin C and bifenthrin @ 60mg/Kg/day and 5.8mg/Kg/day respectively. After 30th day of treatment, lung samples were taken and analysed for oxidative stress parameters. Significant (P<0.05) increase in lipid peroxidation was observed from control value of 4.80±0.39 to 7.90±0.50 in bifenthrin treated animals. Mean control values of SOD, GSH-Px and CAT were 0.55±0.05, 0.98±0.03 and 138.70±6.01 which were significantly (P<0.05) decreased to values of 0.27±0.0, 0.53±0.05 and 91.10±9.70 respectively in bifenthrin treated animals. The value of GST increased significantly (p<0.05) to 1.05±0.06 in bifenthrin administered animals from control value of 0.70±0.08. Pre-treatment with vitamin C in ameliorative group IV significantly restored the normal values of lipid peroxidation, SOD, GST and CAT but could not reverse the decreased values of GSH-Px. The present research is first of its type where in free radical generation due to bifenthrin –a commonly used insecticide was evaluated in lung homogenates when given orally which might be due to residues present in the lung. Besides it will be helpful in better understanding of toxicological profile of pyrethoids, the most commonly used insecticides.

In this research, to determine toxicity of mercury (Hg) and whether selenium (Se) has any role in protection of this toxicity, it was investigated the alterations in oxidative stress parameters in tissues of Oreochromis niloticus. For this purpose, fish were exposed to 0.01 and 0.05 ppm Hg and 0.01 ppm Hg+0.01 ppm Se and 0.05 ppm Hg+0.05 ppm Se for 4 and 21 days and activities of superoxide dismutase (SOD) and catalase (CAT) and levels of malondialdehyde (MDA) in gill and liver were determined. It was observed significant changes in all analyzed parameters due to tissue, medium concentration and exposure period in the exposure of mercury alone and Hg+Se mixtures. In Hg alone, and in combination with Se especially in their higher medium concentrations, activities of SOD and CAT in the gill and liver significantly increased at 4 days (P˂0.05), while they significantly decreased at 21 days (P˂0.05). In the exposure of Hg and Hg+Se and in the both tissues, it not determined significant alteration in the MDA levels at 4 days (P˃0.05), while they elevated in their higher concentrations at 21 days (P˂0.05). In conclusion, it was determined the increases or decreases in activities of SOD and CAT and levels of MDA in O. niloticus were higher in the Hg alone than Hg+Se mixtures and selenium has a protective effect on oxidative toxicity of mercury.
