The effect of interferon on the ESR spectra of erythrocytes treated with ascorbic acid has been investigated. This model system has been chosen since it represents identically the spectra obtained in cases with acute lymphatic leukemia. The data obtained show that small interferon concentrations increase, while larger concentrations decrease the effect produced by ascorbic acid resulting, finally, in the original erythrocyte ESR spectrum. Atomic absorption studies reveal the presence of copper which might be part of the active principle

Abstract The effect of 1 g of ascorbic acid, administered intravenously to healthy male and female volunteers, on blood and its constituents was investigated by means of electron spin resonance (ESR) spectroscopy and by differential hematologic examinations. The native blood ESR spectrum exhibits 2 min after injection of vitamin C a considerable increase in spin concentration and a new signal at about g = 2.005 which we previously had found to correlate to the semidehydroascorbate radical. This spectrum is identical to that obtained in acute lymphatic leukemia. While it prevails in untreated leukemic patients, spin concentration and shape of the spectrum obtained return to normal within several hours to one day in healthy individuals. Since neither in erythrocytes nor in plasma modifications could be observed, the ESR changes detected must have their origin in the leukocytes. Hematologic studies of the peripheral blood show that the number of granulocytes, lymphocytes, and thrombocytes was not drastically affected by the vitamin C injection.

Abstract The electron spin resonance (ESR) spectrum of erythrocytes of a healthy male volunteer exhibited 2 min after intravenous administration of 1 g of ascorbic acid a considerable increase in spin concentration and a new signal at about g = 2.005 which we previously had found to correlate to the semidehydroascorbate (SDA) radical and which is not identical with the O j radical. Moreover, the vitamin C concentration in erythrocytes and plasma was considerably higher than in comparable samples of other volunteers treated identically. In the latter cases, the ESR spectrum of the erythrocytes was not modified at all. These findings suggest that there must be a substance which reacts with ascorbic acid specifically. It can be assumed that the enzyme ascorbate oxidase plays this decisive role in the ascorbic acid metabolism. For this reason, different amounts of ascorbate oxidase have been added to healthy erythrocytes treated in vitro with ascorbic acid and to tissue samples of lung cancer. As expected, the vitamin C effect as expressed by the appearance of the SDA signal and the increase in spin concentration could be reversed. It is suggested, therefore, that in special types of cancer, such as acute lymphatic leukemia and lung cancer, the concentration of ascorbate oxidase or of an enzyme acting like it is, primarily, diminished, while in other types it might be enlarged.

The ESR spectra of peroxidase systems of methaemoglobin-ascorbic acid-hydrogen peroxide and methaemoglobin-haptoglobin complex-ascorbic acid-hydrogen peroxide have been measured in the acetate buffer of pH 4.5. For the system with methaemoglobin an asymmetrical signal with g ~ 2 has been observed which is interpreted as the perpendicular region of anisotropic spectrum of superoxide radical. On the other hand, for the system with methaemoglobin-haptoglobin complex the observed signal with g ~ 2 is symmetrical and is interpreted as a signal of delocalized electron. After realization of three repeatedly induced peroxidase processes the ESR signal of the perpendicular part of anisotropic spectrum of superoxide radical is distinctly diminished, whereas the signal of delocalized electron remains practically unchanged. An amino acid analysis of methaemoglobin along with results of the ESR measurements make it possible to derive a hypothesis about the role of haptoglobin in increasing of the peroxidase activity of methaemoglobin.

Lymphomatoid granulomatosis, a tumor-like process of unknown etiology, produced progressively destructive disease of the larynx, trachea and bronchi in an eight-year-old girl with acute lymphoblastic leukemia of five years duration. The leukemia had been in remission for 4½ years at the clinical onset of the lymphomatoid granulomatosis. Whether this occurrence suggests that lymphomatoid granulomatosis is a type of neoplasm, or is associated with immunologic depression, cannot be stated. Fortunately rare, and of poor prognosis, the disorder may become more frequent with improved survival rate of patients with leukemia and other neoplasms.

A 30-year-old housewife with past history of acute lymphoblastic leukemia 12 years back, still in remission, was admitted with polyarthritis of 2 months duration. She was evaluated and found to have SLE with positive ANA and Anti ds DNA which were strongly positive.

Arsenic speciation, besides total arsenic content determination, is very important in analysis of water, foodstuffs, and environmental samples, because of varying degrees of toxicity of different species. For such purpose hydride generation atomic absorption spectrometry can be used based on the generation of certain types of hydride, depending on the pH value and pretreatment in different reaction media. In this study, we have investigated the effect of L-ascorbic acid as the reaction medium as well as the pre-reducing agent in speciation of arsenic by hydride generation-atomic absorption spectrometry in order to determine monomethyl arsonic acid (MMA) in the presence of inorganic forms of arsenic.
