OBJECTIVE: The aim of this study was to follow changes in body composition, estimated by dual-energy X-ray absorptiometry (DXA), in relation to changes in leptin during the first year of GH therapy in order to test the hypothesis that leptin is a metabolic signal involved in the regulation of GH secretion in children. DESIGN AND METHODS: In total, 33 prepubertal children were investigated. Their mean (S.D.) chronological age at the start of GH treatment was 11.5 (1.6) years, and their mean height was -2.33 (0.38) S.D. scores (SDS). GH was administered subcutaneously at a daily dose of 0.1 (n=26) or 0.2 (n=7) IU/kg body weight. Ten children were in the Swedish National Registry for children with GH deficiency, and twenty-three children were involved in trials of GH treatment for idiopathic short stature. Spontaneous 24-h GH secretion was studied in 32 of the children. In the 24-h GH profiles, the maximum level of GH was determined and the secretion rate estimated by deconvolution analysis (GHt). Serum leptin levels were measured at the start of GH treatment and after 10 and 30 days and 3, 6 and 12 months of treatment. Body composition measurements, by DXA, were performed at baseline and 12 months after the onset of GH treatment. RESULTS: After 12 months of GH treatment, mean height increased from -2.33 to -1.73 SDS and total body fat decreased significantly by 3.0 (3.3)%. Serum leptin levels were decreased significantly at all time points studied compared with baseline. There was a significant correlation between the change in total body fat and the change in serum leptin levels during the 12 months of GH treatment, whereas the leptin concentration per unit fat mass did not change. In a multiple stepwise linear regression analysis with 12 month change in leptin levels as the dependent variable, the percentage change in fat over 12 months, the baseline fat mass (%) of body mass and GHt accounted for 24.0%, 11.5% and 12.2% of the variability respectively. CONCLUSIONS: There are significant correlations between changes in leptin and fat and endogenous GH secretion in short children with various GH secretory capacities. Leptin may be the messenger by which the adipose tissue affects hypothalamic regulation of GH secretion.

Abstract Context: Polymorphisms in the ESR1 gene encoding estrogen receptor (ER)-α may be associated with fat mass in adults. Objectives: The objective of the study was to establish whether ESR1 polymorphisms influence fat mass in childhood. Design: This was a cross-sectional analysis after genotyping of rs9340799, rs2234693, and rs7757956 ESR1 polymorphisms. Setting: The Avon Longitudinal Study of Parents and Children (ALSPAC) was a population-based prospective study. Participants: Participants included 3097 11-yr-old children with results for ESR1 genotyping, puberty measures, and dual-energy x-ray absorptiometry results. Outcomes: Relationships between ESR1 polymorphisms and indices of body composition were measured. Results: The rs7757956 polymorphism was associated with fat mass (P = 0.002). Total body fat mass (adjusted for height) was reduced by 6% in children with TA/AA genotypes, and risk of being overweight (≥85th centile of fat mass) was decreased by 20%. This genetic effect appeared to interact with puberty in girls (P = 0.05 for interaction): in those with the TT genotype, total body fat mass (adjusted for height) was 18% higher in Tanner stages 3–5 vs. stages 1–2; the equivalent difference was 7% in those with TA/AA genotypes. Furthermore, the risk of being overweight was 36% lower in girls with TA/AA genotypes in Tanner stages 3–5, but no reduction was seen in those in stages 1–2. Neither rs9340799 nor rs2234693 polymorphisms were associated with body composition measures. Conclusions: Fat mass in 11-yr-old children was related to the rs7757956 ESR1 polymorphism. This association was strongest in girls in more advanced puberty, in whom the risk of being overweight was reduced by 36% in those with the TA/AA genotype.

OBJECTIVE To analyze the effects of a 20-week concurrent training on the variables of body composition, lipid profile, and fatty liver diagnosis in obese adolescents. METHODS An open clinical trial was carried out with 34 obese adolescents aged between 12 and 15 years. Total body fat, trunk fat mass, total cholesterol and its fractions (HDL, LDL and VLDL), and triglycerides were analyzed; an upper abdominal ultrasound was performed in order to diagnose fatty liver. The participants underwent concurrent training (association of weight training with aerobic training) three times per week, lasting one hour for 20 weeks. Statistical analysis included paired Studentâ€(tm)s t-test and frequency analysis in order to verify the relative and absolute reductions of fatty liver diagnosis, being significant p<0.05. RESULTS The studied adolescents showed statistically significant improvement in body composition, with a decrease of total body fat percentage, total fat mass, trunk fat, and an a increase in the lean body mass. They also presented reduced size of liver lobes, decrease in total cholesterol and in LDL-cholesterol, with a lower prevalence of fatty liver. CONCLUSIONS The concurrent training was effective for promoting significant improvements in body fat composition and lipid profile variables, besides reducing fatty liver prevalence rate.

Abstract. The purpose of this study was to evaluate the use of a cross-sectional scan as an alternative to the total body DXA scan for predicting the body composition of pigs in vivo. A total of 212 pigs (56 to 138 kg live body weight) were scanned by DXA. The DXA scans were analyzed for percentage fat and lean in the total body and in 14 cross-sections (57.6 mm wide): 5 in the front leg/thoracic region, 4 in the abdominal region, and 5 in the back leg region. Regression analysis was used to compare total body and cross-sectional DXA results and chemical analysis of total body fat, protein and water. The relation (R2) between the percentage fat in individual slices and the percentage of total body fat measured by DXA ranged from 0.78 to 0.97 and by chemical analysis from 0.71 to 0.85, respectively. The relation between the percentage of lean in the individual slices and chemical analysis for percentage of total body protein and water ranged from 0.48 to 0.60 and 0.56 to 0.76, respectively. These results indicate that total body composition of the pig can be predicted (accurately) by performing a time-saving single-pass cross-sectional scan.

<p>This study was aimed to evaluate the physiological status, blood profiles and body composition of sheep fed with Ca-saponified lemuru oil coated by herbs. Twenty fat-tailed sheep (av. BW 23±1.2 kg) were used in this experiment by using Completely Randomized Design with five treatments ration and four replications of each. Sheep fed with concentrate containing 3% Ca-saponified lemuru oil and king grass (1:1) ad libitum. Treatments were control diet without herbs (R1); Ca-saponified lemuru oil coated by curcumae domestica (R2); coated by Zingiber officinale Rosc (R3); coated by Eugenia polyantha (R4) and coated by Pluchea indica Less (R5). Data of physiological parameters were measured three times a day, in the morning, at noon and afternoon. Blood samples were collected at the end of the experiment through jugular vein, together with zero sample for Urea Space measurement. The variables observed were physiological data (heart rate, respiration rate, and rectal temperature), blood profiles (erythrocyte, hemoglobin, packed cell volume (PCV), leucocytes) and body composition (water, protein and fat) measured using Urea Space technique. Results of the study showed that sheep fed with Ca-saponified lemuru oil coated by herbs was not significantly different on heart rate, respiration rate and rectal temperature among treatments. Meanwhile, total leucocytes, neutrophil cell, and lymphocytes significantly increased (P&lt;0.05) by the treatment. Body composition percentage (water, protein and fat) were same in all treatments, except the total body fat and energy retained. In conclusion, supplementation of 3% Ca-saponified lemuru oil coated by Curcumae domestica, Zingiber officinale Rosc, Eugenia polyantha and Pluchea indica Less in fat-tailed sheep had no effect on physiological parameters, but improved the leucocyte and neutrophil cells. The regulation of leptin production in humans is poorly understood but appears to depend on total body fat, changes in energy intake and insulin levels. Since growth hormone (GH) is an important regulator of both lipid metabolism and insulin secretion and action, we tested whether GH status directly or indirectly regulates leptin secretion. Circadian serum leptin concentrations were measured in GH-deficient patients in two different protocols involving different modes of acute and prolonged GH exposure. In study I, eight GH-deficient patients all underwent three 4 week study periods in random order: (1) evening (2000 h) s.c. GH injections (2 IU); (2) morning (0800 h) s.c. GH injections (2 IU); (3) no GH administration. At the end of each period the patients were admitted to hospital for 24-h measurements of hormones and metabolites. For comparison, 10 age- and sex-matched healthy untreated subjects were hospitalised under identical conditions. In study II, six GH-deficient patients were hospitalised for 44 h on three occasions, separated by at least 4 weeks without GH treatment. On each occasion they received 2 IU GH, administered i.v. as (1) two boluses (at 2000 and 0200 h), (2) eight boluses (at 3 h intervals starting at 2000 h) or (3) a continuous (2000-2000 h) infusion. In both studies, serum leptin levels peaked between midnight and early morning followed by low day-time levels (P < 0.01). The mode of GH treatment or previous discontinuation did not affect the leptin level (P > 0.05), but the patients had significantly higher leptin levels than the controls (P < 0.01). The diurnal variation in leptin was compared with changes in GH, insulin, non-esterified fatty acids, 3-hydroxybutyrate, insulin-like growth factor I and glucose, but no robust cross-correlations could be demonstrated. The following conclusions were made. (1) The circadian pattern of serum leptin is not influenced by either experimental or spontaneous changes in serum GH concentrations. (2) GH deficiency is associated with elevated leptin levels which most likely reflects increased fat mass in these patients.

Growth, body composition and distribution of carcass tissues were compared in Omani sheep and goats. Animals had ad libitum access to Rhodes-grass hay (8 % CP) and a concentrate diet (16% CP) from weaning until slaughter. The two species had similar birth weights but sheep had higher preweaning (181 g/day), postweaning (175 g/day) and overall (179 g/day) growth rates than goats (120, 102 and 111 g/day, respectively) and thus they reached slaughter weights earlier. Sheep had higher slaughter weight (22·26 kg), empty body weight (20·39 kg), hot carcass weight (12·48 kg) and dressing out percentage (55·94%) than goats (21·17, 18·82, 11·48 kg and 53·97%, respectively). Sheep also had higher proportions of skin, liver and lungs and trachea (P<0·01) than goats, which had higher proportions of head, feet and gut contents. As proportions of carcass weight, sheep had higher fat (25·08%) but lower muscle content (57·24%) than goats (15·72 and 65·88%, respectively). There were no significant differences between the two species in proportion of carcass bone (13·76 and 14·17%). These effects resulted in sheep having a lower muscle: bone ratio (4·19 and 4·68) and higher fat: muscle ratio (0·44 and 0·24). Sheep had higher proportions of non-carcass, carcass and total body fat in the empty body weight (EBW) than goats. However, sheep had less non-carcass but more carcass fat than goats when fats were expressed as proportions of total body fat. Sheep had higher proportions of muscles in the proximal hind limb, distal hind limb (P<0·01), around the spinal column, connecting forelimb to thorax and high-priced muscle group (P<0·05), but lower proportions of muscles in the abdominal wall, proximal forelimb (P<0·05), distal forelimb (P<0·01), connecting neck to forelimb, intrinsic muscles of neck and thorax (P<0·05) and total forequarter muscles (P<0·01) than goats. As proportions in carcass bone, sheep had higher axial skeleton (P<0·05) but lower forelimb than goats. Among species/sex/slaughter weight groups, castrated male and female goats had the lowest growth rates. Castrates and female sheep, particularly at heavier liveweights, had higher carcass and non-carcass fat contents than intact males and goats of all sexes. Although Omani goats produced leaner carcasses and had higher proportions of some non-carcass offals than Omani sheep, they had slower growth rates and a less attractive muscle distribution. This may negatively affect their potential for large scale meat production under Omani conditions.
