Enhanced laccase activity of white rot fungi induced by different metal ions under submerged fermentation

Submerged fermentation with single or mixed metal ions as inducers was used for laccase production from white rot fungi. Mixed metal ions were used for the first time as inducers for Pleurotus ostreatus and Flammulina velutipes to enhance laccase activity. The maximum laccase activity from P. ostreatus in basal media, metal ion media 1 containing copper ion, metal ion media 2 containing manganese ion, metal ion media 3 containing manganese and copper ions, metal ion media 4 containing ferrous ion, metal ion media 5 containing manganese and ferrous ions, metal ion media 6 containing ferrous and copper ions, and metal ion media 7 containing manganese, copper and ferrous ions were, respectively, approximately 21.5-fold, 4.7-fold, 14.9-fold, 16.9-fold, 4.0-fold, 11.0-fold, 12.7-fold, and 24.8-fold higher than that from F. velutipes. The combination of copper and manganese ions as inducers was superior to that of a single copper ion or manganese ion. The maximum laccase activity of P. ostreatus rose in media containing manganese and copper ions. The single copper ion as the inducer for enhancing laccase activity was more suitable for F. velutipes. These findings are helpful in selecting the appropriate single metal ion or mixed metal ions to enhance laccase activity.

In this study, pine fruit was used as solid adsorbent for removal of ferrous and copper ions from aqueous solutions through batch equilibrium technique. The influence of contact time, pH of the solution and initial concentration of metal ions on adsorbed amount of metal ions were investigated. 90 minutes of adsorption time was found sufficient to reach equilibrium for ferrous ion and 120 minutes for copper ion. Adsorption of metal ions were pH dependent and the results indicate the optimum pH for the removal of Fe+2was found to be 5.0 and that of Cu+2was 7.0, the highest adsorption capacity was found to be 4.8 and 14.1 mg of metal ion per gram of adsorbent at initial concentration of 22.22 mg/L and 57.6 mg/L of ferrous and copper ions respectively and would be higher with higher initial concentration. Ferrous ion was removed by 96.3 - 97.3% and copper ion by 94.1-96% along the whole range of initial concentrations. Isotherm studies showed that the data were best fitted to the Freundlich isotherm model. The kinetic data corresponded well with the pseudo-second order equation, suggesting that the adsorption process is presumably a chemisorption.

The optical characteristics of copper ion detection, such as the photometric absorbance of specific wavelengths, exhibit significant intensity change upon incident light into the aqueous solutions with different concentrations of metal ions due to the electron transition in the orbit. In this study, we developed a low-cost, small-size and fast-response photoelectric sensing prototype as an optic sensor for copper (Cu) ions detection by utilizing the principle of optical absorption. We quantified the change of optical absorbance from infra-red (IR) light emitting diodes (LEDs) upon different concentrations of copper ions and the transmitted optical signals were transferred to the corresponding output voltage through a phototransistor and circuit integrated in the photoelectric sensing system. The optic sensor for copper (Cu) ions demonstrated not only excellent specificity with other metal ions such as cadmium (Cd), nickel (Ni), iron (Fe) and chloride (Cl) ions in the same aqueous solution but also satisfactory linearity and reproducibility. The sensitivity of the preliminary sensing system for copper ions was 29 mV/ppm from 0 to 1000 ppm. In addition, significant ion-selective characteristics and anti-interference capability were also observed in the experiments by the proposed approach.

Abstract Carbon charcoal was made from rambutan rods and used as an adsorbent. A gram 70/100 mesh size of adsorbent was then used to adsorb 100 ml of copper ion solution with a 70 ppm concentration. In this investigation, the batch procedure was used without shaking (naturally). The charcoal carbon rambutan ability to remove the copper ion was measured by AAS. The percentage result was 48,135% or about 33,694 ppm. SEM and EDX instrument analysis have applied to confirm the presence of copper ions on the adsorbent surface. The copper ion was found at a concentration of 0.09 percent of the total weight. The carbon charcoal adsorbent in rambutan rods has the ability to purify the water contaminated by metal ions.

Peracetic acid can be catalyzed to bleach cotton fibers at temperatures as low as 30°C by incorporating 2,2î-bipyridine in the bleach solution. Treatment of the fibers with HCl prior to bleaching reduces bleaching effectiveness by removing trace transition metal ions from the fibers. Sorption of individual ions (Cr+3 Mn+2, Fe+2, Fe+3 Co+2, Ni+2, Cu+2, and Zn+2) by HCl treated cotton fibers prior to bleaching indicates that the ferrous ion produces the greatest catalytic effect, and it is only effective when the metal ion is in the fiber as opposed to in solution. Ferrous ions in the fibers sorb 2,2î-bipyridine from solution to form the tris-2,2î-bipyridine ferrous ion complex that is associated with the fibers, and it is the trischelate associated with the fibers that catalyzes bleaching. The effects of pH, temperature, and concentrations of 2,2î-bipyridine, sodium lauryl sulfate, and transition metal ions (in the fibers and in solution) on bleaching effectiveness and peracetic acid decomposition have been studied, and a bleaching mechanism is proposed.

SummaryOmphalina sp. a white-rot fungi (WRF)originated from oil palm plantation has abilityto degrade empty fruit bunches of oil palm(EFBOP) so that it is expected to producelaccase with high activity. The ability ofOmphalina sp. to produce laccase enzyme onliquid fermentation will be studied. The enzymewill also be partially purified andcharacterized. The research result showed thatthe highest enzyme activity (1.162 U/mL) wasobtained using glucose malt yeast (GMY)medium at room temperature for four days.The addition of 2,5-xylidine as an inducerproduced laccase earlier i.e two days, but theactivity of laccase was less active afterprolonged incubation compared to that ofcontrol. The laccase produced on mediumcontaining 2% EFBOP reached optimumactivity as much as 0.38 U/mL after 10 th daysof incubation. Partial purification of laccaseon Sephacryl S-200 HR column resulted58.23% of yield recovery with twice purity thanbefore. Different Pleurotus ostreatus and Flammulina velutipes species were compared relative to their ability to produce laccase in submerged fermentation of various lignocellulosic wastes. Fungi cultivation in identical culture conditions revealed wide differences among both species and strains of the same species. The laccase secretion ability of P. ostreatus strains was superior to F. velutipes strains. Maximum laccase production on cottonseed hull, corncob, and poplar wood was secreted by P. ostreatus CY 568, P. ostreatus CCEF 89, and P. ostreatus CY 568, respectively. The nature of lignocellulosic materials played an important role in determining the expression of laccase potential of fungi. The presence of cottonseed hull improved laccase activity and accelerated the rate of enzyme production. Maximum laccase production on cottonseed hull was nearly 1.29-fold and 1.53-fold higher than that on corncob and poplar wood, respectively. Laccase activity was detected in almost all tested strains on cottonseed hull on the first day, while only a few strains on poplar wood and corncob were detected on the first day. These findings will be helpful for selecting the appropriate strain in industrial applications and for optimization of integrated industrial laccase production.

Abstract         Copper and gold mining industry produce large amount of tailings effluent containing heavy metals such as Cu and Hg that will polute  environment and agricultural land, if it is not properly treated.  Effort in reducing pollution level and recovery of metal in tailing could be conducted with biosorption process using microbial cell as an absorbent agent. This study aims to determine the ability of selected white rot fungi (WRF) immobilized on empty fruit bunches of oil palm (EFB)  to absorb Cu (II) metals in tailing. Based on growth rate, ligninolytic enzymes activity, and the absorption capacity of heavy metals, the superior WRF candidate was Omphalina sp. The ability of biomass Omphalina sp. to decrease the con-centrateion of Cu (II) with an initial concentration of 150 ppm, up to 63%. Omphalina sp. that immobilized on TKKS was able to reduce Cu (II) as much as 66-77% at pH 4.0 for 60 minutes. Abstrak                Eksploitasi tambang tembaga dan emas banyak menghasilkan limbah tailing yang masih me-ngandung logam berat seperti Cu (II) dan Hg (II). Limbah tersebut berpotensi mencemari perairan dan lahan pertanian bila tidak ditangani dengan baik.  Usaha untuk mengatasi limbah tailing dan sekaligus memekatkan (recovery) logam di dalam-nya dapat dilakukan dengan proses biosorpsi menggunakan sel mikroba. Penelitian ini bertujuan menetapkan kemampuan biomassa jamur pelapuk putih (JPP) yang diimobilisasi pada tandan kosong kelapa sawit (TKKS) dalam mengabsorpsi ion logam Cu (II). Metode biosorpsi logam yang digunakan dengan sistem batch dengan kadar logam 300 ppm, 200 ppm,dan 150 ppm. Hasil seleksi JPP berdasarkan laju pertumbuhan, aktivitas enzim ligninolitik, dan penyerapan logam berat telah  diperoleh   kandidat   JPP   unggul  yaitu Omphalina sp. Dalam media PDB,  Omphalina sp. toleran terhadap Cu (II) hingga konsentrasi 150 ppm. Kemampuan biomassa Omphalina sp. untuk penurunan konsentrasi Cu (II) dengan konsentrasi awal 150 ppm yaitu mencapai 63%. Omphalina sp. yang diimobilisasi dengan TKKS mampu menurun- kan kandungan Cu (II) sebesar 66-77%  pada pH 4.0 selama 60 menit.
