Alveolar osteitis (AO) is one of the most common infectious complications after dental extractions. The data on the species composition of AO pathogens and their susceptibility to antimicrobial drugs can be the basis for their empirical use in case of inflammatory process aggravation. Objective. To determine the species composition and susceptibility to the antimicrobial agents of microorganisms, which were detected in patients with AO, who sought medical help in the oral surgery department of the dental medical center of Bogomolets National Medical University. Methods. Throughout 2018–2021, microbiological examination of tooth sockets from 30 patients with AO and 20 patients without AO was performed. The studied biological material was plated on appropriate nutrient media for isolation of aerobic, facultative and obligate-anaerobic microorganisms. Anaerobic conditions were achieved in GENbox 7.0 L and GENbox 2.5 L aerostats using GENbox anaerobic packages (“Biomerieux”, France). The genus and species identity of the bacteria were determined according to Bergey. Antibiotic susceptibility of the isolated strains was determined by disk diffusion method. Results. It was found that most commonly microorganisms from tooth sockets in case of AO are: Staphylococcus aureus, Staphylococcus epidermidis, Streptococcus spp., Escherichia coli, Bacteroides spp., Clostridium spp., and Candida species, as well as their mixed cultures of 3–5 species of microorganisms. These aerobic and facultative anaerobic bacteria were susceptible to amoxicillin, ceftriaxone and ciprofloxacin in 92.6–100% of cases. The growth of anaerobic bacteria in 100% of cases was inhibited by colistin and meropenem. Conclusions. AO developing is caused by pathological colonization of socket of the extracted tooth by representatives of endogenous microbiota, namely Staphylococcus aureus, Staphylococcus epidermidis, Streptococcus spp., Escherichia coli, which are present mainly in the mixed cultures with Candida albicans. For empirical antibiotic therapy of complicated forms of AO, amoxicillin or ceftriaxone or ciprofloxacin in complex with colistin or meropenem should be used, since these drugs suppress the growth of 92.6–100% of strains of aerobic, facultative and obligate anaerobic microorganisms, which are potential pathogens of the purulent forms of AO.

Among urgent surgical diseases of abdominal cavity, an acute intestinal obstruction is the most difficult to be diagnosed and treated. Leading factor, determining the development of pathophysiological processes is considered to be the progressive manifestations of enteric insufficiency syndrome, resulting in intestinal barrier impairment, negative changes in ecology of intestinal flora, increased endotoxins. To identify the small intestine microflora in acute intestinal obstruction and determine the role of dysbiotic disorders in clinical manifestations of main pathological process, a study was conducted in 60 patients with mechanical intestinal obstruction. The small intestine has a relatively rare microflora, consisting mainly of gram−positive facultative aerobic microorganisms, streptococci, lactobacilli. The distal ileum in nearly 30−55 % of healthy people contains scanty microflora, and yet the flora of this area differs from the microbial population of the higher gastrointestinal tract due to higher concentration of gram−negative bacteria. Optional−anaerobic coliform bacilli, anaerobic bifidobacteria and fusobacteria, bacteroids, the number of which starts exceeding the one of gram−positive species, are presented in significant quantities. Distal to the ileocecal valve there are significant changes in the microflora quantitative and species composition. Obligatory anaerobic bacteria become the predominant part of microflora, exceeding the number of aerobic and facultative anaerobic bacteria. The bacterial flora in different parts of gastrointestinal tract has its own specifics and is quite constant, as a result of the interaction of many factors, regulating the bacterial population in small intestine. The most important among them are: acidity of gastric juice, normal peristaltic activity of the intestine, bacterial interactions and immune mechanisms. Disorders of the intestine motor and evacuation function with its obstruction lead to slow passage of the chyme and contamination of the upper gastrointestinal tract with new types of microbes. There is a syndrome of small intestine excessive colonization, which means an increased concentration of bacterial populations in it, similar in species composition to the colon microflora. Pathological intra−intestinal contents become a source of endogenous infection and re−infection of the patient, leads to internal digestive disorders, which is manifested by syndrome of malabsorption of proteins, carbohydrates and vitamins. An imbalance in the colonic microbiota might underlie many human diseases, but the mechanisms that maintain homeostasis remain elusive. Recent insights suggest that colonocyte metabolism functions as a control switch, mediating a shift between homeostatic and dysbiotic communities. During homeostasis, colonocyte metabolism is directed toward oxidative phosphorylation, resulting in high epithelial oxygen consumption. The consequent epithelial hypoxia helps to maintain a microbial community dominated by obligate anaerobic bacteria, which provide benefit by converting fiber into fermentation products absorbed by the host. Conditions that alter the metabolism of the colonic epithelium increase epithelial oxygenation, thereby driving an expansion of facultative anaerobic bacteria, a hallmark of dysbiosis in the colon. Enteric pathogens subvert colonocyte metabolism to escape niche protection conferred by the gut microbiota. The reverse strategy, a metabolic reprogramming to restore colonocyte hypoxia, represents a promising new therapeutic approach for rebalancing the colonic microbiota in a broad spectrum of human diseases.

RESUMO O uso de plantas medicinais no tratamento de doenças é uma estratégia antiga utilizada por praticamente todas as populações do mundo, e, embora novos antibióticos tenham sido desenvolvidos para o controle de micro-organismos infecciosos, às vezes são ineficazes. Diversos extratos de plantas medicinais têm efeitos antimicrobianos, principalmente quando associados à antibióticos de uso clínico, representando alternativa terapêutica para doenças infecciosas. Montrichardia linifera, conhecida popularmente como aninga, é espécie macrófita, aquática emergente de hábito herbáceo, pertencente a família Araceae e ocorre em áreas alagáveis. A utilidade farmacológica desta espécie é diversificada tendo sido relatada como cicatrizante, antirreumático, antidiurético e expectorante. Devido à relevância no campo etnofarmacológico, ampla utilização na medicina popular e escassez de trabalhos relacionados à atividade antibacteriana desta espécie, objetivou-se com este trabalho avaliar a atividade antibacteriana de extratos alcoólicos de folhas de Montrichardia linifera, coletadas na margem do rio Igaraçu, Parnaíba-PI. O extrato foi testado em oito cepas de bactérias: Staphylococcus aureus, Staphylococcus epidermidis, Enterococcus faecalis, Escherichia coli, Klebsiella pneumoniae e Pseudomonas aeruginosa. Foram utilizadas as técnicas de verificação da formação de halos de inibição e determinação das concentrações inibitórias e bactericidas mínimas. Os testes antibacterianos evidenciaram como principais resultados que o extrato metanólico seco (EMS), extrato metanólico fresco (EMF), e o extrato etanólico seco (EES), apresentaram ação antibacteriana, enquanto o extrato etanólico fresco (EEF) não apresentou atividade para as bactérias testadas. O EMS foi o mais eficiente, inibindo o crescimento bacteriano na concentração de 200 μg/mL para E. faecalis, 400 μg/mL para S. aureus, 400 μg/mL para S. epidermidis e 2.000 μg/mL para P. aeruginosa. O EMF obteve CIM de 2.000 μg/mL para E. faecalis e EES obteve CIM de 250 μg/mL para E. faecalis. Os resultados demonstraram que M. linifera constitui fonte eficiente de compostos bioativos antibacterianos. Os estudos sobre as propriedades farmacológicas de plantas da família Araceae são escassos, e os resultados deste trabalho são pioneiros em relação a atividade antibacteriana desta espécie.

ABSTRACTTreatment with streptomycin enhances the growth of human commensalEscherichia coliisolates in the mouse intestine, suggesting that the resident microbial community (microbiota) can inhibit the growth of invading microbes, a phenomenon known as “colonization resistance.” However, the precise mechanisms by which streptomycin treatment lowers colonization resistance remain obscure. Here we show that streptomycin treatment rendered mice more susceptible to the development of chemically induced colitis, raising the possibility that the antibiotic might lower colonization resistance by changing mucosal immune responses rather than by preventing microbe-microbe interactions. Investigation of the underlying mechanism revealed a mild inflammatory infiltrate in the cecal mucosa of streptomycin-treated mice, which was accompanied by elevated expression ofNos2, the gene that encodes inducible nitric oxide synthase. In turn, this inflammatory response enhanced the luminal growth ofE. coliby nitrate respiration in aNos2-dependent fashion. These data identify low-level intestinal inflammation as one of the factors responsible for the loss of resistance toE. colicolonization after streptomycin treatment.IMPORTANCEOur intestine is host to a complex microbial community that confers benefits by educating the immune system and providing niche protection. Perturbation of intestinal communities by streptomycin treatment lowers “colonization resistance” through unknown mechanisms. Here we show that streptomycin increases the inflammatory tone of the intestinal mucosa, thereby making the bowel more susceptible to dextran sulfate sodium treatment and boosting theNos2-dependent growth of commensalEscherichia coliby nitrate respiration. These data point to the generation of alternative electron acceptors as a by-product of the inflammatory host response as an important factor responsible for lowering resistance to colonization by facultative anaerobic bacteria such asE. coli.

Objetivo: Describir el perfil microbiológico y de resistencia antibiótica en el servicio de pediatría de la Clínica Jesús del Norte, 2016. Material y métodos: Se diseñó un estudio descriptivo de serie de casos. El estudio se desarrolló en el Servicio de Pediatría de la Clínica Jesús del Norte. Resultados: Se analizaron 1264 muestras de pacienres <15 años con cultivos positivos de orina, heces, sangre y secreción faríngea. El 67,7% fue en <5 años y predomino en mujeres con 59,22%. El germen aislado en mayor frecuencia fue Escherichia coli con 46,68%. Por tipo de muestra el más frecuente fueron: En Coprocultivo: Escherichia coli enteropatógena, Urocultivo: Escherichia coli, Hemocultivo: Staphylococcus epidermidis y en secreción faríngea: Staphylococcus aureus. Llamó la atención la presencia de 105 casos (7,27%) de E. coli BLEE en urocultivos. Conclusiones: Los cultivos positivos fueron más frecuentes en mujeres, en menores de 5 años, atendidos en emergencia y de muestra de heces. Apreciándose alta frecuencia de resistencia a fármacos de primera y segunda línea en diversos agentes etiológicos aislados en las muestras.

Οι μέθοδοι ανίχνευσης και προσδιορισμού ειδικών αλληλουχιών νουκλεϊκών οξέων βασίζονται στην υβριδοποίηση (μοριακή αναγνώριση) του αναλύτη με συμπληρωματικό μόριο DNA/RNA, το οποίο είναι συνδεδεμένο, άμεσα ή έμμεσα, με κατάλληλο ιχνηθέτη για παραγωγή σήματος. Το φάσμα εφαρμογών των μεθόδων υβριδοποίησης συνεχώς διευρύνεται, κυρίως ως αποτέλεσμα της προόδου των προγραμμάτων προσδιορισμού της αλληλουχίας του γονιδιώματος του ανθρώπου και άλλων οργανισμών. Σκοπός της παρούσας διατριβής είναι αφενός η ανάπτυξη μεθόδων υβριδοποίησης που παρέχουν τη δυνατότητα ταυτόχρονου προσδιορισμού πολλών αλληλουχιών νουκλεϊκών οξέων και αφετέρου η απλοποίηση των μεθόδων υβριδοποίησης με τη χρήση βιοαισθητήρα DNA. Στο Κεφάλαιο 1 της διατριβής περιγράφεται η αρχή της αλυσιδωτής αντίδρασης πολυμεράσης (PCR) και η αντίστροφη μεταγραφή-PCR (RT-PCR) ως οι πιο σημαντικές τεχνικές εκθετικού πολλαπλασιασμού ειδικών αλληλουχιών DNA και RNA, οι οποίες αποτελούν αναπόσπαστο μέρος κάθε σύγχρονης μεθόδου ανάλυσης νουκλεϊκών οξέων. Παρουσιάζεται επίσης η τεχνική της ποσοτικής PCR και διάφορες μέθοδοι προσδιορισμού των προϊόντων της PCR. Στο Κεφάλαιο 2 γίνεται παρουσίαση των δοκιμασιών υβριδοποίησης νουκλεϊκών οξέων, οι οποίες αποτελούν και τον ακρογωνιαίο λίθο της αναλυτικής χημείας του DNA και RNA. Περιγράφεται η εξέλιξη των ιχνηθετών από τα ραδιοϊσότοπα μέχρι τους πιο σύγχρονους μη ραδιενεργούς ιχνηθέτες που παρέχουν υψηλή ανιχνευσιμότητα και δυνατότητες αυτοματοποίησης. Παρουσιάζονται οι ομογενείς και ετερογενείς δοκιμασίες υβριδοποίησης και διάφορες παραλλαγές που διευκολύνουν την εκτέλεση τους. Στο Κεφάλαιο 3 παρέχεται σύντομη περιγραφή της αρχής λειτουργίας και της οργανολογίας της κυτταρομετρίας ροής, η οποία αποτελεί ένα ισχυρό αναλυτικό εργαλείο για τη διερεύνηση ιδιοτήτων μικροσωματιδίων (π.χ. κυττάρων αλλά και συνθετικών μικροσφαιριδίων). Αντικείμενο του Κεφαλαίου 4 είναι η ανάπτυξη πολλαπλών φθορισμομετρικών δοκιμασιών υβριδοποίησης DNA και RNA σε εναιώρημα φασματικά διακριτών μικροσφαιριδίων πολυστυρενίου. Κάθε μικροσφαιρίδιο περιέχει δύο φθορίζουσες χρωστικές σε συγκεκριμένη αναλογία και με αυτό τον τρόπο έχει καταστεί ένα φασματικά κωδικοποιημένο στερεό υπόστρωμα για την εκτέλεση δοκιμασιών υβριδοποίησης. Ειδικά ολιγονουκλεοτίδια-ανιχνευτές προσδένονται ομοιοπολικά στην επιφάνεια κάθε μικροσφαιριδίου. Η αλληλουχία του DNA- ή RNA-στόχου (αναλύτης) υβριδοποιείται με τα ακινητοποιημένα ολιγονουκλεοτίδια και τα υβρίδια ποσοτικοποιούνται με άλλη φθορίζουσα ουσία. Το εναιώρημα αναλύεται με κυτταρομετρητή ροής με ανιχνευτή φθορισμού επαγώμενου με δύο πηγές λέιζερ. Μελετήθηκε ο προσδιορισμός μονόκλωνου DNA, δίκλωνου DNA και RNA και πραγματοποιήθηκαν πειράματα ανάλυσης γονιδιακής έκφρασης σε ασθενείς με λευχαιμία. Στο Κεφάλαιο 5 παρουσιάζεται ο πρώτος βιοαισθητήρας DNA τύπου εμβαπτιζόμενης ταινίας ξηρών αντιδραστηρίων για την ανίχνευση 7 χρωμοσωμικών αλληλομεταθέσεων, οι οποίες σχετίζονται άμεσα με την οξεία ή τη χρόνια λευχαιμία. Ο βιοαισθητήρας βασίζεται σε ολιγονουκλεοτίδια συζευγμένα με νανοσωματίδια χρυσού και επιτρέπει την ταυτοποίηση των αλληλομεταθέσεων δια γυμνού οφθαλμού, έπειτα από πολλαπλασιασμό με RT-PCR. Σε αντίθεση με τις μέχρι τώρα χρησιμοποιούμενες μεθόδους ανίχνευσης αλληλομεταθέσεων, η ανάλυση με το βιοαισθητήρα είναι απλή, δεν απαιτεί ειδικά όργανα και αποφεύγει πολλά στάδια επωάσεων και εκπλύσεων. Αντικείμενο του Κεφαλαίου 6 της διατριβής είναι η ανάπτυξη μεθόδου υβριδοποίησης DNA για την ταχεία ανίχνευση βακτηριακής επιμόλυνσης σε αρθροπλαστικές επεμβάσεις, ένα σημαντικό πρόβλημα στην ορθοπεδική. Απομονώνεται DNA από δείγματα αρθρικού υγρού, ακολουθεί πολλαπλασιασμός με PCR χρησιμοποιώντας καθολικούς εκκινητές και ανίχνευση των προϊόντων με βιοαισθητήρα DNA. Ανιχνεύονται οι μικροοργανισμοί Escherichia coli, Staphylococcus aureus, Staphylococcus epidermidis, Haemophilus influenza, Enterococcus faesium και Streptococcus pneumoniae. Στο Κεφάλαιο 7 περιγράφεται η εφαρμογή του βιοαισθητήρα DNA για την ανίχνευση γενετικά τροποποιημένων οργανισμών (GMO) σε τρόφιμα. Αν και η παρουσία GMO μπορεί να διαπιστωθεί με ανίχνευση είτε της νέας πρωτεΐνης που συνθέτει το διαγονιδιακό φυτό ή της αλληλουχίας DNA που έχει εισαχθεί σ’ αυτό, το DNA είναι ο προτιμότερος αναλύτης λόγω της μεγαλύτερης σταθερότητάς του, ιδιαίτερα σε επεξεργασμένα τρόφιμα. Χαρακτηριστικές αλληλουχίες DNA που περιέχονται στα περισσότερα GMO, δηλαδή ο προαγωγέας 35S και ο τερματιστής NOS, πολλαπλασιάζονται με PCR και τα προϊόντα ανιχνεύονται με δοκιμασία υβριδοποίησης στο βιοαισθητήρα DNA. Η παρούσα εργασία εισάγει, για πρώτη φορά στο πεδίο της ανάλυσης GMO, έναν βιοαισθητήρα DNA τύπου εμβαπτιζόμενης ταινίας ξηρών αντιδραστηρίων.

Combination effects between Cymbopogon flexuosus and Cymbopogon nardus essential oils were studied to determine whether the combination could emerge as better and more powerful antimicrobial agents against six selected bacteria includes Bacillus subtilis, Escherichia coli, Salmonella typhimurium, Serratia marcescens, Staphylococcus aureus, and Staphylococcus epidermidis. This combination study exhibited 40.67% additive, 28.67% antagonistic, 16.00% indifferent and 14.66% synergistic effects. C. flexuosus and C. nardus essential oils in combination showed a high inhibitory activity against S. aureus with 16% synergistic, 64% additive and 20% indifferent effects.

Abstract: A cesarean delivery increases the risk of wound infection it should be prevented by using antibiotics. This study was to determine the pattern of bacterial sensitivity in surgical wounds of cesarean section patients for selected antibiotics, i.e ceftriaxone, cefixime, sulbactam ampicillin, ciprofloxacin, clindamycin and gentamicin.This observational study was conducted at the Ulin Public Hospital in Banjarmasin from July to September 2019. Using a cross sectional approach Samples of bacteria were identidied from 36 that were pusposively sampled, i.e., Staphylococcus aureus, Staphylococcus epidermidis and Escherichia coli. Antibiotic sensitivity test showed that Staphylococcus aureus was sensitive against gentamicin (100%),whereas Staphylococcus epidermidis and Escherichia coli. A espécie Ptychopetalum olacoides Bentham, popularmente conhecida como “marapuama” ou “muirapuama”, é uma Olacaceae nativa da região norte do Brasil, há muito conhecida e utilizada por suas propriedades estimulantes e afrodisíacas, sendo inclusive exportada para diversos países. O estudo fitoquímico do lenho desta árvore, neste trabalho representado pelos galhos da planta, revelou a presença majoritária de vários ácidos graxos, esteróides e xantinas, sendo eles: ácido palmítico, ácido esteárico, -sitosterol, estigmasterol, lupeol, glutinol, a-amirina, cafeína, teobromina e adenina, sendo que as três últimas não haviam ainda sido reportadas na espécie. O estudo das atividades antimicrobianas revelou que os extratos da planta não apresentam atividade inibitória sobre o desenvolvimento de cepas de Escherichia coli, Staphylococcus aureus ou Staphylococcus epidermidis. Porém, o resíduo do extrato alcoólico apresenta uma significativa ação inibitória do crescimento micelial de Colletotrichum acutatum, e ação menos pronunciada sobre o crescimento de Fusarium oxysporum. PHYTOCHEMICAL STUDY AND ANTIMICROBIAL ACTIVITY OF Ptychopetalumolacoides Bentham Abstract Ptychopetalum olacoides Bentham, popularly known as “marapuama” or “muirapuama”, is an Olacaceae native from north Brazil, known and used for its stimulating and aphrodisiac properties for a long time, being also exported to several countries all over the world. The phytochemical study of the wood of this tree, represented by its stems, revealed the presence of several fatty acids, sterols and xanthines, including palmitic acid, stearic acid, -sitosterol, stigmasterol, lupeol, glutinol, a-amirin, caffeine, theobromine and adenine, and the latest three substances have not been reported in this specie yet. The study of antimicrobial properties revealed that the plant extracts don’t have any inhibitory activity against strains of Escherichia coli, Staphylococcus aureus or Staphylococcus epidermidis. However, the alcoholic extract residue does have a remarkable inhibitory action on the mycelial growth of Colletotrichum acutatum and Fusarium oxysporum.26

Se evaluó la actividad antibacteriana in vitro de aceites esenciales de diferentes especiesdel género Citrus frente a cepas ATCC de Staphylococcus aureus, Staphylococcus epidermidis,Klebsiella pneumoniae, Pseudomonas aeruginosa y Escherichia coli, determinandola concentración mínima inhibitoria (CMI) y la concentración mínima bactericida(CMB). Las bacterias se replicaron en medios de agar y caldos específicos. Se determinóel momento de máxima densidad óptica (DO620) para emplearlo como tiempode incubación; luego se hicieron pruebas de evaluación de sensibilidad con la exposiciónde las cepas a concentraciones a 1000 g/mL del extracto en caldo. Para solubilizarse empleó dimetilsulfóxido (DMSO) al 1%. Posteriormente, se le determinó laconcentración mínima inhibitoria mediante metodologías de microdilución en caldoy la concentración mínima bactericida. Encontrándose una actividad de los aceitesesenciales del género Citrus, con valores de CMI ≥ 600 mg/mL frente a S. aureus,S. epidermidis, K. pneumoniae, P. aeruginosa y E. coli. En función a los resultados obtenidos,se concluye que las diferentes especies del género Citrus son consideradas comopromisorias para el control del componente bacteriano.
