DNA barcoding revels first records of three rare coleopteran genera in Northern lakes of Egypt.

Abstract One aquatic coleopteran species from family Dytiscidae and two aquatic coleopteran genera from family Hydrophilidae were recorded in the summer period and represent first records in the Egyptian lakes. Beetles were collected from two northern lakes, Lake Idku and Lake Burullus. They were identified by morphological characteristics as well as the mtDNA barcoding method. A molecular phylogenetic approach was used to determine the genetic identity of the collected samples based on the mitochondrial cytochrome oxidase I (COI). Prodaticus servillianus (Dytiscidae) from Egypt showed no significant difference in the COI region and they are highly similar to P. servillianus from Madagascar. The phylogenetic analysis revealed that the other two coleopteran genera belong to family Hydrophilidae. Based on COI only, there is no clear evidence for their genetic identity at the species level. So, we defined them to the closest taxon and denoted them as Cymbiodyta type A and B. The results indicated that resolving the molecular identity of the aquatic beetles from northern lakes of Egypt need more considerations in the field of biological conservation. We concluded that utilization of COI as a barcoding region for identifying some coleopteran species is not sufficient and additional molecular markers are required to uncover the molecular taxonomy at deep levels.

Due to active human intervention in natural genetic and population processes, survival and distribution areas of honey bees have been negatively affected. Preservation of the gene pool of aboriginal bees that are well adapted to local environmental conditions is an urgent problem that cannot be solved without the use of molecular methods to monitor the genetic composition of local populations. This type of research requires the use of state-of-the-art approaches based on application of molecular markers. In the molecular taxonomy of insects, mitochondrial cytochrome oxidase genes are widely used to identify closely related forms. The peculiarity of mtDNA is that it is maternally inherited, and the mitochondrial molecular markers are not separated by recombination. The СоІІ gene encoding the second subunit of cytochrome oxidase is widely used to descriminate the subspecies of honey bee. Therefore, to assess the distribution of different subspecies / breeds of Apis mellifera in Ukraine, the 5′-region of the СоІІ gene was sequenced and compared for bees from different geographical regions. In the CoII gene, base substitutions were found, which makes it possible to distinguish between honey bees of the Carpathian and Ukrainian steppe breeds, which are widespread in Ukraine. It has been revealed that the distribution of these breeds does not always correspond to the official zoning. The widespread practice in Ukraine of transporting the genetic material of Apis mellifera from different regions leads to uncontrolled hybridization and poses a threat to the preservation of aboriginal breeds of honey bees.

Species in the genus Tomentella are distributed throughout the temperate and tropical regions worldwide, but few studies associated with the taxonomy and phylogeny of this genus had been reported from Northwest China. In this paper, molecular phylogenetic analyses of the nuclear ribosomal ITS (internal transcribed spacer: ITS1-5.8S-ITS2) and LSU (large subunit: 28S) sequences combined with morphological characteristics identified three new species from Xinjiang Autonomous Region in Northwest China, which were named T. aurantispora, T. kanasensis, and T. schrenkiana. Similar macromorphological and anatomical characteristics are shared by these new species: arachnoid basidiocarps; byssoid sterile margins; utriform basidia with a clamp connection at the base; the absence of rhizomorphs and cystidia; and slightly thick-walled, subglobose to globose basidiospores. Among these new species, the color of the hymenophoral surface, the size of the basidiospores, and some other features can be used for species delimitation. The new species and closely related species in the phylogenetic tree were discussed, and a key to the identified species of Tomentella from China was provided.

In this study, four new dextral camaenid from China are reported, based on shell morphology, reproductive system anatomy, and molecular phylogenetic analyses: Camaena funingensis Zhou, Wang & Lin, sp. nov., Camaena gaolongensis Zhou, Wang & Lin, sp. nov., Camaena maguanensis Zhou, Wang & Hu, sp. nov., and Camaena yulinensis Zhou, Wang & Hu, sp. nov. Detailed descriptions of the morphological characteristics including shells and genitalia, DNA sequences, and living environments of the four new species are provided, with further comparisons with congeners.

Barley is a small grain cereal, tolerant to high temperatures and drought. Due to this characteristic, it can be regarded as a promising crop for production in dry conditions. Research on five spring landraces of two-rowed barley lasted two experimental years (2011 and 2012) in agro-ecological conditions of Banja Luka. In the experiment, the standard cropping practices were applied, without irrigation. The experiment was set as a complete randomized design with four replications. All measurements were performed in 10 plants per replication. Morphological characteristics included plant height (cm), spike length (cm), grain number per spike and grain weight per spike (g). Obtained data were subjected to two-way analysis of variance with landrace and growing season as main factors. Significant treatment or interaction effects were further analysed by Fisher's least significant difference (LSD) test, often utilized for pairwise comparisons among arithmetic means. In all tested traits the maximum average values were obtained in barley landrace AM2, i.e. the plant height (86.89 cm), the spike length (8.90 cm), the grain number per spike (24.74) and the grain weight per spike (1.17 g). Due to these facts, two-rowed barley landrace AM2 can be marked as the most productive in this research, bearing a potential for different crop breeding practices.

Brown algae of the genus Cystoseira sensu lato form the most diverse and productive marine ecosystems throughout the Mediterranean Sea and have equal roles also in the Black Sea where they have been decreasing in the recent years. The taxonomy of Cystoseira s.l. taxa from the Black Sea is still not well understood, and questions arise when related taxa have to be delimited. In addition to morphological descriptions, this study provides for the first time molecular data of the Black Sea Cystoseira s.l. distinct morphologies as an additional tool to clarify their identities and phylogenetic affinities. The analysis of two mitochondrial markers (cytochrome oxidase subunit 1—COI, and 23S-tRNAVal intergenic spacer—mt-spacer) showed that Cystoseira s.l. specimens from the Black Sea belong to two recently resurrected genera, namely Gongolaria and Ericaria. Molecular data confirm the morphological identification of G. barbata, which is characterized by high morphological plasticity in the Black Sea. The morphological data presented in this study support the transition of G. barbata to the genus Gongolaria, which was previously proposed based solely on genetic data. For the Black Sea endemic taxon C. bosphorica, sequence divergence suggests conspecificity with Mediterranean Sea species E. crinita and E. barbatula. However, considering original morphological characteristics of the taxon, its geographical isolation, and endemism, the new combination Ericaria crinita f. bosphorica comb. nov. is proposed.

This study presents one telamonioid species new to science based on morphological characteristics and molecular phylogenetic analyses. Cortinarius khinganensis was collected from the Greater Khingan Mountains, Northeast China and it is characterized by hygrophanous, vivid brownish red and striate pileus, white universal veil, and subglobose spores. According to phylogenetic analyses results, C. khinganensis belongs to the section Illumini, which is a lineage distantly related from subgenus Telamonia sensu stricto. Detailed descriptions of the new species and the comparisons with morphologically similar species are provided. The phylogenetic relationships within the section Illumini are also discussed.

During a faunal survey in southern Xizang, we collected 27 specimens of voles that could not be identified as any knownspecies in the Arvicolinae. These specimens shared the following morphological characteristics, not corresponding withany other arvicoline species: the first lower molar possessed five closed triangles, the third upper molar exhibited eitherfour or three inner angles, and the tails of all specimens measured 30% of the body length. Their proximal baculum of theglans was very sturdy and trumpet-shaped, the distal baculum was tongue-like and sturdy, and the lateral bacula were veryshort. Molecular phylogenetic analyses based on nucleotide sequences of the mitochondrial cytochrome b (cyt b) geneclustered these specimens as a distinct lineage within the genus Neodon. According to the morphological and moleculardata, we described them as a new species, Neodon linzhiensis. Our phylogenetic analysis strongly supported that Lasio-podomys fuscus, Phaiomys leucurus, Neodon sikimensis, N. irene and the new species formed a monophyletic group, notincluding N. juldaschi. We suggested that L. fuscus and P. leucurus should be transferred to Neodon and that N. juldaschishould be removed from this genus. Following our new delineation of Neodon, we proposed a redefinition of the morphological diagnostic characters of the genus.

We report five alien ascidians with some distinct features that were investigated from August 2009 to October 2011 in Korea, among which Ascidiella aspersa (Müller, 1776) belonging to family Ascidiidae of order Phlebobranchia and Molgula manhattensis (De Kay, 1843) belonging to family Molgulidae of order Stolidobranchia are newly discovered invasive species. These ascidians were identified and their phylogenetic relationships were clarified through molecular analysis using about 680 bp of nuclear 18S rDNA and about 670 bp of mt-COI genes along with detailed morphological characteristics, and reported for the first time in Korea. It was discovered that A. aspersa was widespread three coastlines of Korea except Jeju Island, and M. manhattensis first found in Mokpo, Gunsan, and Incheon in June 2010 extended into Busan of Korea Strait in 2011.

AbstractA revision of the North American members of the Leptogium saturninum group (i.e. species with long lower-surface hairs, isidia, and usually smooth upper surface) is presented based on molecular phylogenetic analyses of mtSSU and nrITS sequence data, together with an extensive morphological study. Three species supported by both molecular and morphological characteristics are recognized: L. acadiense sp. nov. (distinguished by granular saturninum-type isidia, medulla composed of irregularly arranged or perpendicular hyphae), L. cookii sp. nov. (distinguished by cylindrical saturninum-type isidia) and L. hirsutum (distinguished by hirsutum-type isidia and medulla composed of loosely intertwined hyphae). One species supported by morphological characteristics, but for which no molecular data could be generated, is also recognized: L. compactum sp. nov. (distinguished by hirsutum-type isidia and medulla composed of tightly packed hyphae). Finally, L. saturninum (distinguished by granular saturninum-type isidia and medulla composed of perpendicular and parallel hyphae) is supported by morphological characteristics but molecular data from geographically diverse populations, including those near the type locality, indicate that the morphologically defined species is paraphyletic. Leptogium burnetiae is excluded from North American based on morphological study of the type. The species are described and illustrated in detail, and are distinguished morphologically by their isidium development, morphology of mature isidia, and pattern of hyphae in the medulla in transverse sections near lobe margins. A key to the members of the L. saturninum group and related species is also presented.

From historical strain records, Penicillium citreonigrum NBRC 4692 was originally isolated as the toxigenic fungus responsible for the yellow rice incident in Japan in 1937. The fungus was named Penicillium toxicarium by I. Miyake, which was considered invalid due to the lack of a Latin diagnosis. Initially, it was named Penicillium sp. by Miyake et al. in 1940. Subsequently, P. toxicarium, which was erroneously cited as synonym of P. citreo-viride, was validated by C. Ramírez in 1982 with a Latin diagnosis and type designation (CBS 351.51). Later, CBS 351.51 was assigned to Penicillium trzebinskii by Houbraken et al. in 2014. In 2016, P. toxicarium was treated as a synonym of Penicillium citreosulfuratum based on a conclusion of molecular phylogenetic analysis. Recently, we discovered the taxonomic and nomenclatural short communication (in Japanese) by I. Miyake in 1947 on P. toxicarium sp. nov. with its Latin description and four illustrations but lacking the type designation. In this paper, we re-examined strain NBRC 4692 for its current taxonomic position based on morphological characteristics and molecular phylogenetic analysis. NBRC 4692 (received from I. Miyake in 1951) has been found to have the same morphological characteristics as P. toxicarium, as was shown in Miyake's diagnosis in 1947. Phylogenetic analysis revealed that the NBRC strain belongs to a unique clade, different from the clade comprising P. citreosulfuratum strains. As a conclusion, herein, P. toxicarium I. Miyake (1947) is reinstated as a correct name with the lectotype designation.
