Despite the possible influence of tannins on the nutritional value of the forages from Caatinga vegetation, there are few studies that evaluated their tannin concentration. This study was conducted to characterize condensed tannins present in the legumes species Mimosa hostilis (Jurema Preta), Mimosa caesalpinifolia (Sabiá) and Bauhinia cheilantha (Mororó), at three stages of their phenological cycle. The concentration of soluble tannin (ST), bound tannin (BT) and total tannin (TT) were determined using the butanol-HCL method; astringency was by the radial diffusion method, and the monomeric composition of purified tannins by a high-performance liquid chromatograph with delphinidin, cyanidin and pelargonidin as standards. Concentration and astringency of purified condensed tannins, as well as their monomeric composition varied between species, and in some cases among phenological cycles. The values observed were always above the limits considered beneficial for ruminal digestion (i.e. 5%). Jurema Preta presented the highest values (30.98% TT and 22% astringency at full growth stage), and Mororó the lowest (10.38% TT and 14% astringency during fructification). Jurema Preta presented a mean relationship prodelfinidin (PD): procyanidin (PC) of 97:3, which did not vary during the phenological cycle, showing the high astringent capacity of these tannins. Sabiá presented a relationship of 90:20 during full growth and flowering stages, decreasing to 40:50 at fructification. In Mororó the PD:PC relationship was more equilibrated, around 40:50 during full growth and flowering stages, decreasing to 35:60 During fructification. Propelargonidin was not detected or was present at low concentration in the three species.

Relative to non-tannin-containing fresh forages, condensed tannins in Lotus pedunculatus increased duodenal N flow and calculated absorption of amino acids from the small intestine, but depressed ruminal digestion of soluble carbohydrate and hemicellulose. Objective To evaluate the relationship of glucose and glycated haemoglobin (HbA1C) in type 1 diabetes model induced by streptozotocin. Research Design and Methods Induction of diabetes mellitus was achieved through the intraperitoneal injection of 70mg/kg body weight of streptozotocin dissolved in 1m citrate buffer pH 4.5 twice daily for 2 days. A total number of thirty rats were used selected among those that have exceeded glucose threshold (>10.0mmol/l) 2 weeks after streptozotocin induction. All rats weighed between 240-300g. Samples for fasting plasma glucose and glycated haemoglobin were collected at the tail vein. Glucose was determined by the glucose oxidase method and HbA1C was determined by High Performance Liquid Chromatograph (HPLCEsi/ ms) with uv detection. Data was analysed by one way and two way analysis of variance using SPSS version. Results Significant linear relationship was demonstrated between plasma glucose level and glycated haemoglobin which could be predictive of risk of developing diabetes. Control samples had values within reference range, glucose (3.5-6.5 mmol/l) and glycated hemoglobin (4.3-7%). However diabetic test rats elicited values that varied significantly with time. Test result confirms the fact that higher mean values of plasma glucose in diabetic (positive) controls were due to the effect of streptozotocin. Conclusion Plasma glucose and glycated hemoglobin show positively mutual relationship and can be used in early diagnosis of diabetes mellitus. Using correlation coefficient and regression enhances measurement of the strength of the bivariate association and is predictive. Pain is defined as a disagreeable sensory and emotional experience related to a tissue or potential lesion. Paracetamol (Acetaminophen) is the most used non-morphine analgesic. For the determination of paracetamol we developed and validated the high performance liquid chromatography (HPLC) analysis using a Dionex Ultimate 3000 liquid chromatograph equipped with a multidimensional detector. After determining the optimum conditions of analysis (80/20 water / acetonitrile mobile phase, flow rate 1.0 mL / min, detection wavelength 245 nm) we validated the method following the following parameters: linearity of response function, linearity of results, limit (LD = 0.66 mg / mL) and quantification limit (LQ = 2.00 mg / mL), and precision. The method of determining paracetamol by HPLC was applied to 30 samples of serum collected from patients who had pain and were treated with paracetamol.

Mushrooms produce a variety of bioactive compounds that are known to have anti-pathogenic properties with safer and effective therapeutic effects in human disease prognosis. The antibacterial activity of ethanol and methanol extracts of Pleurotus opuntiae were checked against pathogenic microorganisms viz. Pseudomonas aeruginosa ATCC 27853, Proteus mirabilis NCIM 2300, Proteus vulgaris NCIM 5266, Serratia marcescens NCIM 2078, Shigella flexeneri NCIM 5265, Moraxella sp. NCIM 2795, Staphylococcus aureus ATCC 25923 by agar well diffusion method at different concentrations of the extracts. Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) of the extracts was determined by INT (Iodonitrotetrazolium chloride) colorimetric assay. Extracts were standardized by thin layer chromatography (TLC) in different solvent systems. The Retention factors (Rf) of different compounds were calculated by high performance TLC (HPTLC) fingerprinting at UV 254, 366, and 540 nm before and after derivatization. The ethanol and methanol extracts of P. opuntiae showed bactericidal activity against all the test pathogens at MIC values of 15.6 to 52.08 mg/mL and 20.81 to 52.08 mg/mL respectively. Whereas the MBC values for ethanol and methanol extract of P. opuntiae against all pathogens were recorded as 26.03 to 62.5 mg/mL and 125 mg/mL respectively. Preliminary mycochemical screening of both the extracts revealed high contents of bioactive compounds. Amongst all the solvent systems used in TLC, the best result was given by chloroform + hexane (8:2) which eluted out 5 different compounds (spots). HPTLC results revealed spots with different Rf values for all the 24 compounds present. Thus, it can be inferred from the present investigation that the mycoconstituents could be an alternative medication regimen and could play a role in new drug discoveries against different infections. Further, the antimicrobial components of these mushrooms can be transformed to bioengineered antimicrobial coatings for surfaces, drug and other hybrid systems for public health implications in combating persistent infections.

Ultraviolet (UV) radiation is one of the primary factors responsible for disturbances in human skin cells phospholipid metabolism. Natural compounds that are commonly used to protect skin, due to their lipophilic or hydrophilic nature, show only a narrow range of cytoprotective activity, which prompts research on their combined application. Therefore, the aim of this study was to examine the effect of ascorbic acid and rutin on the phospholipid and ceramide profiles in UV-irradiated fibroblasts cultured in a three-dimensional system that approximates the culture conditions to the dermis. An ultra-high-performance liquid chromatograph coupled with a quadrupole time-of-flight mass spectrometer was used for phospholipid and ceramide profiling. As a result of UVA and UVB cells irradiation, upregulation of phosphatidylcholines, ceramides, and downregulation of sphingomyelins were observed, while treatment with ascorbic acid and rutin of UVA/UVB-irradiated fibroblast promoted these changes to provide cells a stronger response to stress. Moreover, an upregulation of phosphatidylserines in cells exposed to UVB and treated with both antioxidants suggests the stimulation of UV-damaged cells apoptosis. Our findings provide new insight into action of rutin and ascorbic acid on regulation of phospholipid metabolism, which improves dermis fibroblast membrane properties.

ABSTRACT Aim This study evaluated the in vitro antibacterial effects of the formulated Punica granatum (PG) gel against Streptococcus mutans, Streptococcus sanguinis, and Lactobacillus casei. Materials and methods The PG extract was dissolved in water at 500 mg/mL. High performance liquid chromatography (HPLC) was used for identification and quantification of chemical marker punicalagin. Minimum bactericidal concentration (MBC) and time-kill assay (TKA) were investigated. Antibacterial activities of the formulated PG gel, 2% chlorhexidine (CHX) gel and blank gel were tested by measuring the zones of inhibition through agar well diffusion method. Results The HPLC results showed presence of punicalagin at 2023.58 ± 25.29 μg/mL in the aqueous PG extract and at 0.234% (w/w) in the formulated PG gel. The MBC for S. mutans, S. Sanguinis, and L. casei were 250, 125, and 500 mg/mL respectively. The TKA of 500 mg/mL aqueous PG extract showed total inhibition of S. mutans, S. Sanguinis, and L. casei at 6, 1, and 24 hours contact time respectively. Agar well diffusion revealed that for S. mutans, CHX gel > PG gel > blank gel; for S. sanguinis, CHX gel = PG gel > blank gel; for L. casei, CHX gel > PG gel = blank gel. Comparison of the PG gel potency showed that S. sanguinis = S. mutans > L. casei. Conclusion The PG gel equivalent to 0.234% punicalagin (w/w) inhibited S. mutans and S. sanguinis but not L. casei within 24 hours incubation period and has the potential to be used for caries prevention. Background/ Aims: Little is known about the effect of P450 oxidoreductase (POR) gene polymorphisms on the activities of CYPs with multiple genotypes. Methods: We genotyped 102 human livers for 18 known POR single nucleotide polymorphisms (SNPs) with allelic frequencies greater than 1% as well as for 27 known SNPs in 10 CYPs. CYP enzyme activities in microsomes prepared from these livers were determined by measuring probe substrate metabolism by high performance liquid chromatograph. Results: We found that the effects of the 18 POR SNPs on 10 CYP activities were CYP genotype-dependent. The POR mutations were significantly associated with decreased overall Km for CYP2B6 and 2E1, and specific genotypes within CYP1A2, 2A6, 2B6, 2C8, 2D6 and 2E1 were identified as being affected by these POR SNPs. Notably, the effect of a specific POR mutation on the activity of a CYP genotype could not be predicted from other CYP genotypes of even the same CYP. When combining one POR SNP with other POR SNPs, a hitherto unrecognized effect of multiple-site POR gene polymorphisms (MSGP) on CYP activity was uncovered, which was not necessarily consistent with the effect of either single POR SNP. Conclusions: The effects of POR SNPs on CYP activities were not only CYP-dependent, but more importantly, CYP genotype-dependent. Moreover, the effect of a POR SNP alone and in combination with other POR SNPs (MSGP) was not always consistent, nor predictable. Understanding the impact of POR gene polymorphisms on drug metabolism necessitates knowing the complete SNP complement of POR and the genotype of the relevant CYPs.
