Abstract Serological screening of 199 serum samples from Dromedary camels—from different cities in Saudi Arabia—was performed using enzyme-linked immunosorbent assay for detecting antibodies against two cyst-forming coccidian parasites, namely Toxoplasma gondii and Neospora caninum. Antibodies against T. gondii were detected in 68 (34.2%) samples, while those against N. caninum were present in 33 (16.6%) samples. The highest seroprevalence of T. gondii antibodies was reported in samples from Taif (51.2%), while the lowest seroprevalence was reported in samples from Riyadh and Hofuf (15.1%). The highest seroprevalence of N. caninum antibodies was reported in samples from Jizan (35.9%) while the lowest was reported in samples from Taif (2.4%). A total of 47 male and 21 female camels exhibited antibodies against T. gondii , while 19 male and 14 female camels showed antibodies against N. caninum . Concurrent detection of both T. gondii and N. caninum antibodies was observed in 18 camels. It has been demonstrated that T. gondii and N. caninum antibodies are prevalent in camels from different cities of the Kingdom of Saudi Arabia.

Toxoplasma gondii is zoonotic protozoan parasite that causes infections in many vertebrate species. The present study determined the seroprevalence of T. gondii and N. caninum in goats from the Czech Republic. Serum samples were collected from 251 healthy adult goats in the Czech Republic during the years 2006 to 2009. Sera samples were tested for serum antibodies to Toxoplasma gondii by an enzyme-linked immunosorbent assay with cut off equal to or higher than 50% S/P. The same samples were tested for serum antibodies to Neospora caninum by a competitive-inhibition enzyme-linked immunosorbent assay with cut off equal to or higher than 30% inhibition; positive sera were confirmed by an indirect fluorescent antibody test with cut-off titre equal to or higher than 40. Sera positive in both tests were marked as positive. In total, 166 (66%) and 15 (6%) goat sera reacted positively for T. gondii and N. caninum antibodies, respectively. All sera positive for N. caninum antibodies were simultaneously positive for T. gondii antibodies. This is the first detection of N. caninum antibodies in goats in the Czech Republic. Our findings indicate that goats in the Czech Republic are frequently exposed to T. gondii, but less frequently to N. caninum.

ABSTRACT An immunochromatographic test (ICT) with recombinant surface antigen 1 of Neospora caninum (NcSAG1) was developed for the rapid detection of antibodies to N. caninum in cattle. The ICT was used to clearly discriminate between immunofluorescent-antibody test (IFAT)-positive bovine sera and IFAT-negative bovine sera. Serum samples collected from cattle in Yanbian, China, were examined by the ICT. Of the 96 serum samples, 23 (24.0%) were positive by the ICT, and 19 (19.8%) samples were positive by a previously developed enzyme-linked immunosorbent assay (ELISA). Eighteen of 19 ELISA-positive samples were positive according to the ICT. A good agreement was found between the results of the ICT and the ELISA. The results presented here suggest that the ICT with recombinant truncated NcSAG1 fused to glutathione S-transferase is a useful and reliable method for the detection of antibodies to N. caninum in cattle.

ABSTRACTDense granule antigen proteins derived fromToxoplasma gondii(TgGRAs) are potential antigens for the development of diagnostic tools. TgGRA7 and TgGRA14 were detected in the peritoneal fluid ofT. gondii-infected mice, suggesting that TgGRAs may be highly antigenic proteins. Here, TgGRA7 and TgGRA14 were evaluated as candidates for the development of a marker for a rapid diagnostic test. The specificity and sensitivity of purified recombinant proteins of TgGRA7 and TgGRA14 were compared in an indirect enzyme-linked immunosorbent assay (iELISA) using a series of serum samples fromT. gondii-experimentally infected mice and using recombinantT. gondiimajor surface antigen 2 (TgSAG2) as a reference control. The iELISA with TgGRA7 showed the greatest diagnostic accuracy and could detect anti-TgGRA7 antibody in acute and chronic infections. A total of 59 field samples from pigs were also examined by the iELISAs, and the results compared with those of the latex agglutination test (LAT). Among the three recombinant antigens, TgGRA7 had the highest rates of positivity, with significant concordance (88.14) and kappa value (0.76) in comparison with the results using LAT. Furthermore, an immunochromatographic test (ICT) based on recombinant TgGRA7 was developed for rapid detection of antibodies to the infection. The ICT differentiated clearly between sera fromT. gondii-infected mice and uninfected orNeospora caninum-infected mice. Pig sera were examined with the ICT, and the results compared favorably with those of LAT and iELISA for TgGRA7, with kappa values of 0.66 and 0.70 to 0.79, respectively. These data suggest that the ICT based on TgGRA7 is a promising diagnostic tool for routine testing in the clinic and mass screening of samples in the field.

ABSTRACT A baculovirus carrying the SAG2 gene of Toxoplasma gondii was constructed, and recombinant SAG2 protein (S-rSAG2) was expressed in insect cells. S-rSAG2 was recognized by sera from cats and pigs infected with T. gondii. Mice immunized with S-rSAG2 produced high titers of specific immunoglobulin G2a (IgG2a) and IgG1 antibodies. In an indirect fluorescent antibody test, all mouse antisera against S-rSAG2 reacted strongly to the natural parasites, but those against rSAG2 expressed in Escherichia coli (E-rSAG2) only showed very weak reaction, although no markedly difference was found in the reaction to denatured antigen, T. gondii lysate, in Western blot analysis. The results suggest that S-rSAG2 is better than E-rSAG2 in both antigenicity and immunogenicity. Enzyme-linked immunosorbent assay (ELISA) with S-rSAG2 could differentiate clearly between sera from 30 specific-pathogen-free cats and 4 experimentally infected cats. Serum samples from domestic cats in Japan were tested by the ELISA and compared with a latex agglutination test (LAT) and ELISA with E-rSAG2. Of 187 samples, all 35 LAT-positive sera had strong reactions to S-rSAG2 and E-rSAG2. Of the 152 LAT-negative sera, 18 were positive in the ELISA with S-rSAG2, whereas only 2 were positive in the ELISA with E-rSAG2. Although there were significant correlations among the three methods, the ELISA with S-rSAG2 was more sensitive than the others, which could be attributed to the fact that S-rSAG2 shares some common conformational structure with the native antigen. The results suggest that S-rSAG2 would be a useful reagent for the detection of T. gondii infection in cats.

ABSTRACT To investigate whether the production of an antigen-specific antibody is associated with Neospora caninum-induced bovine abortion, 62 serum samples were tested with an enzyme-linked immunosorbent assay using the recombinant antigens NcSAG1, NcSRS2, and NcGRA7. Our study suggested that NcGRA7 would be a new marker for the serodiagnosis of N. caninum infection resulting in abortion.

Toxoplasmosis is a zoonotic disease caused by infection with the parasite called Toxoplasma gondii (T. gondii). The health and social impacts of the infection are enormous, including miscarriage, hydrocephalus, blindness, and mental retardation. The occurrence of toxoplasmosis in maternal women cannot be separated from cats around their houses. This study aimed to determine the seroprevalence of the parasite in cats found in the human carriers residences and identified the risk factors of toxoplasmosis in maternal women in Badung District, Bali Province, Indonesia. A total of 80 cat serum samples were obtained from two residential groups, 40 from the housing where the maternal women were infected and another 40 from where there were no identified sufferers of the disease. All the samples were examined using the enzyme-linked immunosorbent assay (ELISA) method to detect the presence of antibodies T. gondii in feral cat serum. The results showed that 47.5% of the examined subjects had the said antibodies. As much as 65% came from housing with cases of toxoplasmosis in maternal mothers, and 30% came from residences with none. The presence of feral cats is a major risk factor for the transmission of T. gondii to humans.

Aim: The study aimed to evaluate the occurrence of anti-Toxoplasma gondii and anti-Neospora caninum antibodies in sheep breeding in a particular husbandry system called "Elevage en case" in Dakar, Senegal. Materials and Methods: Blood samples were collected from 278 sheep. Serum was harvested and used for analysis. For the detection of T. gondii antibodies, 278 sera were analyzed using the modified agglutination test, while the enzyme linked-immunosorbent assay was used on 174 sheep sera to look for N. caninum antibodies. Results: This study showed that 29.4±6.8% of sheep carried both T. gondii and N. caninum antibodies. The overall prevalence was 60.1±5.7% and 41.9±7.3% for toxoplasmosis and neosporosis, respectively. For toxoplasmosis, the prevalence was higher in Gueule Tapée (63.3%) than in Medina (58.9%), but the variation was not significant (p=0.45). Regarding the age of animals, the prevalence was significantly higher in animals over 2 years old compared to those under 2 years old (p=0.002). For neosporosis, the prevalence was significantly higher in Medina (48.67%) than Gueule Tapée (16.7%) (p=0.001), but non-significant variation was noted according to animal age. Conclusion: The study showed that sheep reared in households have carried antibodies of T. gondii and N. caninum. The prevalence was high and it means that consumption of meat from these animals is risky if the meat is eaten undercooked.

Toxoplasmosis is a disease that can infect animals and zoonotic in humans. In pregnant women (congenital toxoplasmosis), the risk and severity of symptoms of congenital toxoplasmosis are more severe if infection occurs early in pregnancy and asymptomatic. The only way to determine disease is by serological screening. Serological diagnosis techniques for toxoplasmosis include ELISA (Enzyme-linked Immunosorbent Assay) and ICT (Immunochromatography Assay Test). However, until now, research on toxoplasmosis with the ICT method is still minimal. This study aims to analyze the results of the IgM antibody examination Toxoplasma gondii using the ICT method with the ELISA method and calculate the sensitivity and specificity of the ICT method and the ELISA method in pregnant women. The research design was used analytic (cross-sectional), with the number of samples is 33 serums. Data from the ELISA method obtained from secondary medical record data and the remaining serum samples were examined by the ICT method by researchers at the Harapan Kita Hospital Laboratory. In the ICT method, two samples (6.1%) were positive, and 31 samples (93.9%) were negative. In the ELISA method, results obtained similar to the ICT method, two samples (6.1%) positive and 31 samples (93.9%) with negative results. Toxoplasmosis examination can be done using the ICT method as the initial serological screening stage, but it not recommended as a diagnosis enforcer.

SUMMARYIn the Czech Republic, sera from 551 clinically healthy adult slaughtered pigs (females, 6–8 months old) were collected during the first half of June in 2010. Sera were tested for Toxoplasma gondii-specific IgG antibodies by an enzyme-linked immunosorbent assay; samples with more than 50% S/P were considered as positive. The same samples were also analysed for Neospora caninum antibodies using a commercial competitive-inhibition enzyme-linked immunosorbent assay; samples with more than 30% inhibition were considered as positive. Antibodies against T. gondii were found in 198 pigs (36%) in all districts with prevalences ranging from 18% to 75%. Antibodies against N. caninum were found in 16 pigs (3%); positive animals were found in 4 districts with prevalences ranging from 1% to 20%. Indication of mixed infections (concurrent presence of both N. caninum and T. gondii antibodies) was found in 8 (1·5%) pigs. The results of our study indicate that pigs in the Czech Republic have a relatively high seroprevalence for T. gondii, while they have only a low seroprevalence for N. caninum. Therefore, natural infection with T. gondii seems to be very common in Czech pigs. It is the first evidence of N. caninum antibodies in pigs in the Czech Republic. These results complete data about N. caninum infection in pigs in Europe.

SUMMARYToxoplasma gondii and Neospora caninum are closely related protozoan parasites which cause lowered production and increased abortion in dairy cows. The aim of the present study was to determine the seroprevalence of T. gondii and N. caninum infection in dairy cows in the Guangxi Zhuang Autonomous Region (GZAR), subtropical southern China. In total, 875 serum samples were collected from the tail veins of dairy cows in 6 main dairy cow-rearing districts of 4 administrative cities in GZAR. The samples were surveyed for T. gondii antibody using the Indirect Haemagglutination Test (IHA), and 365 of the serum samples were examined for N. caninum antibody by indirect Enzyme-Linked Immunosorbent Assay (ELISA). The overall seroprevalence of T. gondii in dairy cows was 13·71% (120/875), and the average seroprevalence of N. caninum was 15·07% (55/365). There were significant differences in the seroprevalence of N. caninum infection between different districts (P = 0·002, χ2 = 9·261). The highest prevalences of T. gondii and N. caninum were found in cows older than 8 years and those that had completed 5–6 pregnancies. Five cows (1·37%) presented antibodies against both T. gondii and N. caninum, and dairy cows with both T. gondii and N. caninum antibodies had higher abortion rates. The present results indicate widespread exposure of dairy cows to T. gondii and N. caninum in GZAR, subtropical southern China.
