An Analysis of the Antimicrobial Activity of Ten Root Canal Sealers - A Duration Based in vitro Evaluation

Aim and Design: Sealers with prolonged antimicrobial activity are needed by the fact that microorganisms are commonly recovered from teeth with failed endodontic therapy. The aim of this study was to evaluate the duration of antimicrobial activity of ten root canal sealers (Apexit plus, Tubli Seal Xpress, Endoflas FS,Endomethasone, Endomethasone N, AH Plus, Epiphany, EndoRez, Ketac Endo, Roeko Seal) against Enterococcus faecalis and Candida albicans by the agar diffusion test. The zones of inhibition were examined immediately and after 24, 48, 72 hours, 5 and 7 days. Statistical analysis was done using Kruskal - Wallis test and Mann Whitney U test. Results and conclusions: Against Enterococcus faecalis, Endoflas FS showed the largest inhibitory zones immediately and 24 hours after manipulation, whereas, there was no significant difference between Endoflas FS and Endomethasone after 48 hours. Against Candida albicans, Endoflas FS performed better than the other sealers. All the sealers (except AH Plus, Epiphany and Roeko Seal) demonstrated higher antimicrobial action in the first 24 hours after manipulation The antimicrobial action of all the sealers (except AH Plus and Roeko Seal which showed no antimicrobial activity in any studied time and Epiphany which ceased to show any antimicrobial action after 24 hours) decreased significantly with time.

Abstract Aim The purpose of this in vitro study was to analyze the antimicrobial activity of root canal sealers by using the agar diffusion test (ADT). Methods and Materials Three categories of root canal sealers were included in the study: resin-based sealers (4), zinc oxide-based sealers (3), and calcium hydroxide-based sealers (2). The microbial strains used were: S. aureus (2 strains), C. albicans (2 strains), and E. faecalis (1 strain). Statistical analysis was conducted using a one-way analysis of variance (ANOVA). Tests of differences were analyzed using the Tukey's test with a value of p < 0.05 considered statistically significant. Results The antimicrobial activity of root canal sealers was ranked in descending order as follows: Sealite Regular, Cortisemol, Dentalis KEZ, AH26, Sealapex, Acroseal/Topseal, and Endorez/AH plus. Conclusion Root canal sealers showed different inhibitory effects depending on their types and the bacterial strains tested. Root canal sealers containing formaldehyde and eugenol proved to be effective against the microorganisms studied. Clinical Significance The incorporation of antimicrobial components into root canal sealers may become an essential factor in preventing the re-growth of residual bacteria and control of bacteria re-entry into the root canal system. The aim of the present study was to analyze the antimicrobial properties of five endodontic sealers: Endo Fill, Endomethasone, Endomethasone N, Sealer 26 and AH-Plus, against the following microorganisms: Candida albicans, Staphylococcus aureus, Enterococcus faecalis, Streptococcus sanguis and Actinomyces naeslundii. The sealers were tested immediately, 24 h, 48 h and 7 days after manipulation.The direct contact method through the observation of the microbial growth in liquid medium and the agar diffusion test were used to evaluate the antimicrobial properties of the sealers. The results, in both methodologies used, showed that immediately after manipulation, Endo-Fill and Endomethasone demonstrated the highest antimicrobial activity, with no statistically significant difference between them. Sealer 26 demonstrated the lowest antimicrobial activity. At all other times after manipulation, there were no statistically significant differences among all the sealers tested. In conclusion, none of the sealers totally inhibited the growth of the microorganisms. Furthermore, the antimicrobial activity of each sealer decreased with time and was dependent upon the microbial susceptibility to them.

ABSTRACT Aim This study aims to compare the marginal adaptation and sealing ability [mineral trioxide aggregate (MTA)-Fillapex, AH Plus, Endofill sealers] of root canal sealers. Materials and methods In the present study, the inclusion criteria include 45 single-rooted extracted mandibular premolar teeth, with single canal and complete root formation. The sectioning of the samples was done at the cementoenamel junction using a low-speed diamond disc. Step-back technique was used to prepare root canals manually. The MTA-Fillapex, AH Plus, and Endofill sealers were the three experimental sealer groups to which 45 teeth were distributed. Under scanning electron microscope (SEM), marginal gap at sealer and root dentin interface were examined at coronal and apical halves of root canal. Results Among the three maximum marginal adaptations were seen with AH Plus sealer (4.10 ± 0.10) which is followed by Endofill sealer (1.44 ± 0.18) and MTA-Fillapex sealer (0.80 ± 0.22). Between the coronal and apical marginal adaptation, significant statistical difference (p = 0.001) was seen in AH Plus sealer. When a Mann–Whitney U-test was done on MTA-Fillapex sealer vs AH Plus sealer and AH Plus sealer vs Endofill sealer, there was a statistically significant difference (p < 0.05) found between the above two groups at coronal and apical third. Conclusion The present study proves that AH Plus sealer has a better marginal adaptation when compared with other sealers used. Clinical significance For sealing space of crown wall and main cone in root canal treatment, sealers play an important role. The other advantages of sealers are that they are used to fill voids and irregularities in root channel, secondary, lateral channels, and space between applied gutta-percha cones and also act as tripper during filling. ABSTRACT Aim The purpose of this in vitro study was to evaluate the antimicrobial activity of 2% chlorhexidine gel (CHX) as auxiliary chemical substance and intracanal medications on Candida albicans, Candida albicans, Enterococcus faecalis, Escherichia coli, and their endotoxins in the root canals. Materials and methods The study was conducted on 48 single-rooted human teeth divided into four groups (n = 12), according to intracanal medications used: (1) Calcium hydroxide + apyrogenic saline solution (Ca(OH)2 + SS), (2) 20% ginger glycolic extract (GEN), (3) calcium hydroxide + 20% ginger glycolic extract (Ca(OH)2 + GEN), (4) apyrogenic SS (control). Collections were made from the root canal content before preparation (baseline–S1), immediately after instrumentation (S2), 7 days after instrumentation (S3), after 14 days the action of intracanal medication (S4), and 7 days after removal of the intracanal medication (S5). The antimicrobial activity and endotoxin content were analyzed for all collections. The results were statistically analyzed by the Kruskal–Wallis and Dunn tests at a significance level of 5%. Results After instrumentation with CHX, there was complete elimination of E. coli and C. albicans, except for E. faecalis, which was significantly reduced and then completely eliminated after intracanal medication. There was significant reduction of endotoxin after instrumentation. Comparison of collection after instrumentation and intracanal medication revealed reduction of endotoxins in all groups; this reduction was greater in group Ca(OH)2 followed by the group GEN. Conclusion It was concluded that the instrumentation using CHX and intracanal medication used were able to eliminate the microorganisms from the root canal; the endotoxins were reduced, yet not completely eliminated. Clinical significance This study is important and relevant for searching alternatives during endodontic therapy, since it aims to study the effect of Zingiber officinale on microorganisms and endotoxins present in root canals. ABSTRACT Introduction: The aim of this in vitro study was to evaluate the bond strength of Photo-Activated Disinfection (PAD) system to dentin with different root canal sealers by using a push-out test design. Materials and Methods: A total of 30 extracted mandibular premolar teeth with single and straight roots were used. The crowns were removed and the root canals were prepared by using ProTaper rotary files. The smear layer was removed and the roots were randomly divided into two groups (n = 15) according to the use of PAD system as the final disinfecting agent. Each group was then divided into 6 (n = 5) subgroups and obturated with gutta-percha and 3 different root canal sealers. The groups were Group 1: Sodium hypochlorite (NaOCl) + ethylenediaminetetraacetic acid (EDTA)-AH Plus sealer; Group 2: NaOCl + EDTA + PAD-AH Plus; Group 3: NaOCl + EDTA-Sealapex; Group 4: NaOCl + EDTA + PAD-Sealapex; Group 5: NaOCl + EDTA-mineral trioxide aggregate (MTA)-Fiallapex; and Group 6: NaOCl + EDTA + PAD-MTA-Fillapex. 1-mm thickness horizontal sections (n: 5 × 4 = 20) were sliced for the push-out bond strength measurement. Results: Group 3 and 4 showed significantly lower bond strengths compared with all the other groups (P < 0.05). No statistically significant difference was found among Groups 1, 2 and 5, but there was statistically significant difference between Group 5 and 6 (P < 0.05). Conclusion: This in vitro study indicated that the PAD system adversely affected the bond strength of the MTA Fillapex root canal sealer.

Abstract Aim The aim of this study was to evaluate the antimicrobial efficiency of different root canal sealers on Enterococcus faecalis (E. Faecalis) at different time intervals. Methods and Materials All sealers used were mixed according to the manufacturers’ instructions then 75 mg of each sealer was added to different sterile tubes and evaluated at 20 minutes, 24 hours, 7 days, and 30 days. A time-kill assay (TKA) was used to determine the antimicrobial efficiency of the sealers. Results AH Plus and MCS were found to be bactericidal at 20 minutes and 24 hours, but only MCS was bactericidal at the seventh and thirtieth days. Epiphany and Sealapex were found to be bacteriostatic at the seventh and thirtieth days but indifferent at 20 minutes and 24-hours. MCS and AH Plus were both found to be bactericidal in freshly mixed samples, but only MCS was bactericidal at longer time periods. Epiphany Sealer and Sealapex were found to be bacteriostatic at longer time periods but indifferent at 20 minutes and 24 hours. Conclusion The antibacterial effect of MCS was greater than the other sealers evaluated.
