Relevance. Summer savory (Satureja hortensis L.) is an annual herbaceous plant whose essential oil and extracts are used in medicine, perfumery and the food industry.Methods. The article presents the results of studying the effect of the ratio of blue (BL) and red light (RL) in the total spectrum of LED lamps on the morphometric parameters of plants of summer savory varieties Gnom and Perechny aromat in vitro. For in vitro culture, seeds were sterilized with 5% NaCl solution for 10 minutes, placed in Petri dishes with Murashige and Skoog culture medium (MS). Aseptic seedlings at the age of 4-5 days were transplanted into tubes and transferred to racks with LED lamps (for 28 days): (1) from 1st day to 14th day 100% RL, from the 15th to the 28th day 60% RL, 40% BL; (2) 90% RL, 10% BL; (3) 60% RL and 40% BL; (4) 32% RL and 68% BL. The total illumination intensity in all variants was 175 μmol/m2 s.Results. It was found that a change in the RL:BL ratio in the total spectrum affects the morphometric parameters of the savory plants in an in vitro culture. When cultivated under 100% RL, tall plants with long internodes and small leaves are formed, a reduction in the share of RL to 60...32% results in more short-growing plants with large leaves. The results of this work can be used to develop the technology of clonal micropropagation of valuable breeding forms, as well as to create source material for breeding by in vitro culture methods.

Human adipose-derived stromal/stem cells (hASC) are widely used for in vitro modeling of physiologically relevant human adipose tissue. These models are useful for the development of tissue constructs for soft tissue regeneration and 3-dimensional (3D) microphysiological systems (MPS) for drug discovery. In this systematic review, we report on the current state of hASC culture and assessment methods for adipose tissue engineering using 3D MPS. Our search efforts resulted in the identification of 184 independent records, of which 27 were determined to be most relevant to the goals of the present review. Our results demonstrate a lack of consensus on methods for hASC culture and assessment for the production of physiologically relevant in vitro models of human adipose tissue. Few studies have assessed the impact of different 3D culture conditions on hASC adipogenesis. Additionally, there has been a limited use of assays for characterizing the functionality of adipose tissue in vitro. Results from this study suggest the need for more standardized culture methods and further analysis on in vitro tissue functionality. These will be necessary to validate the utility of 3D MPS as an in vitro model to reduce, refine, and replace in vivo experiments in the drug discovery regulatory process.

Garut using in vitro culture methods. Biodiversitas 21: 5845-5849. Several efforts have been made to preserve Citrus reticulata var. Tawangmangu and Citrus reticulata var. Garut as indigenous Indonesian mandarin cultivars, including in vitro tissue culture methods. This study aimed to determine growth differences of the stem explants of C. reticulata var. Tawangmangu and C. reticulata var. Garut,which planted on the same Murashige and Skoog (MS) media. The treatment groups were derived from different explants, grown in 4 separate culture bottles for 35 days at 25°C. The observed parameters included the percentage of explants indicating callus development browned-colored explants, and the contaminated explants. Among C. reticulata var. Tawangmangu explants, 23.53% indicated callus development, 29.42% were browned-colored explants, and 0% indicated contamination. In contrast, among the C. reticulata var. Garut explants, 0% indicated callus development, 7.14% brown-colored, and 7.14% indicated contamination. The stems explants from C. reticulata var. Tawangmangu showed a tendency to develop calluses, but the explants of C. reticulata var. Garut was able to support the growth of shoots. C. reticulata var. Tawangmangu and Garut mandarin stem explants showed differences of shoot growth because physiological conditions varied according to the variety.

The worldwide production of livestock embryos requires stable medium with long shelf life. In this experiment, we evaluated the impact of the freeze-dried in vitro culture (IVC) medium (Mdry) on in vitro embryo production. We compared the standard IVC and Mdry media for cleavage rate and embryo production. Media solutions (10 mL) were aliquoted into 50-mL conical tubes and lyophilized to form a powder concentrate using a Genesis freeze-dryer (VirTis, Gardener, NY, USA). Lyophilization consisted of a constant cooling from 20°C to –10°C at a constant rate of 1°C/min with a 2-h hold at –10°C before sublimation at 0°C. Mdry medium were held at –80°C for 4 months. When the IVC medium was rehydrated, the pH were adjusted to 7.4. Abattoir-derived Holstein oocytes (n = 618, in 7 replicates) were in vitro matured and fertilized with sexed semen, according to standard procedures (Rubessa et al. 2011 Theriogenology 76, 1347-1355). Twenty hours after IVF, presumptive zygotes were cultured in SOF medium with 5% BS at 39°C with 5% CO2, 7% O2, and 88% N2. On Day 7, embryo yields were assessed. All recorded parameters were subjected to a Chi-Square Test 2 × 2. The parameters compared were percent cleavage, blastocysts, and embryos/cleaved. The α level was set at 0.05. All data were expressed as quadratic means and mean standard errors. The results (Table 1) showed not a statistical difference between control and Mdry. The Mdry had a higher percentage of cleaved zygotes (65.4% v. 53.4%) but not enough for a statistical difference. However, when we compared embryo production, there was no difference between treatments. The ratio between blastocysts and cleaved embryos was higher in the control group but not significant according to our selected α level. These results indicate that it is possible lyophilize IVC medium without interfering with the potential quality of the medium. Further studies will be needed to better understand the positive effect of the lyophilization on the cleavage rate. Table 1.Mean (SD in parentheses) percentage cleavage and blastocysts
