SALIVA AS HIGHLY INFORMATIVE SUBSTRATE FOR NON-INVASIVE ANALYSIS OF LIPOPEROXIDE PROCESSES AND MUSCLE DAMAGE IN HIQHLY SKILLED ATHLETES

The purpose of the investigation was to study the efficiency of measuring markers of oxidative stress and muscle damage in the oral fluid in highly skilled sportsmen under physical exercise for the assessment of their functional state. 70 highly qualified athletes at the age of 16-20 years specializing in the cyclic kinds of sports (track and field, swimming) took part in the investigation. Sportsmen performed the control test which consisted of the series of 3×100 m distances by a flat race with an active 45 s rest between them for the track and field athletes, and 4×50 m by the main swimming style with an active rest between the distances also for 45 s for the swimmers. Activity of creatine kinase, content of lipid peroxidation products in the blood and oral fluid were measured standard biochemical methods. The performance of the functional tests induces the excessive accumulation of toxic products of lipoperoxidation and increases activity of creatine kinase in the oral fluid of highly qualified athletes. Correlation analysis shows, that the intensity of free radical and peroxide processes in athletes can be evaluated by means of the method of measuring the content of lipid peroxidation products in the oral fluid.

Целью исследования было установить уровень диеновых конъюгатов в ротовой жидкости при интоксикации тетраметилтиурамдисульфидом. В результате было установлено, что на 28 сутки был выявлен наибольший токсический эффект ТМДТ, который превысил контрольные значения в 4 раза (400%). Концентрация ДК на 7, 14, 21 сутки возросла на 158,3%, 225,0%, 258,3% соответственно. Полученные результаты говорят о накоплении продуктов перекисного окисления липидов диеновых конъюгатов с увеличением сроков пестицидной интоксикации. The aim of the study was to determine the level of diene conjugates in the oral fluid during tetramethylthiuram disulfide intoxication. As a result, it was found that on day 28, the greatest toxic effect of TMDT was detected, which exceeded the control values by 4 times (400%).. The concentration of DC on days 7, 14, and 21 increased by 158.3%, 225.0%, and 258.3%, respectively. The results obtained indicate the accumulation of lipid peroxidation products of diene conjugates with an increase in the duration of pesticide intoxication.

Abstract The objectives of the current experiments were to evaluate the effect of feeding soybean oil (SO) with different levels of peroxidation on lipid, N, and GE digestibility, gut integrity, oxidative stress, and growth performance in nursery pigs. Treatments consisted diets containing 10% fresh SO (22.5 °C) or thermally processed SO (45 °C for 288 h, 90 °C for 72 h, or 180 °C for 6 h), each with an air infusion of 15 L/min, with postprocessing peroxide values of 7.6, 11.5, 19.1, and 13.4 mEq/kg and p-anisidine values of 1.92, 6.29, 149, and 159, for the 22.5 °C, 45 °C, 90 °C and 180 °C processed SO, respectively. In experiment 1, 64 barrows (7.1 ± 0.9 kg initial BW) were randomly allotted into 2 rooms of 32 pens and individually fed their experimental diets for 21 d, with a fresh fecal sample collected on day 20 for determination of GE and lipid digestibility. In experiment 2, 56 barrows (BW 9.16 ± 1.56 kg) were placed into individual metabolism crates for assessment of GE, lipid, and N digestibility and N retention. Urinary lactulose to mannitol ratio was assessed to evaluate in vivo small intestinal integrity, and urine and plasma were collected to analyze for markers of oxidative stress. Pigs were subsequently euthanized to obtain liver weights and analyze the liver for markers of oxidative stress. In experiment 1, pigs fed the SO thermally processed at 90 °C had reduced ADG (P = 0.01) and ADFI (P = 0.04) compared to pigs fed the other SO treatment groups, with no differences noted among pigs fed the 22.5 °C, 45 °C, and 180 °C SO treatments. No effects of feeding thermally processing SO on dietary GE or lipid digestibility (P > 0.10) were noted in either experiment. In experiment 2, there was no dietary effect of feeding peroxidized SO on the DE:ME ratio, N digestibility, or N retained as a percent of N digested, on the urinary ratio of lactulose to mannitol, on serum, urinary, or liver thiobarbituric acid reactive substances, on plasma protein carbonyls, or on urinary or liver 8-OH-2dG (P > 0.10). In experiment 2, pigs fed the SO thermally processed at 90 °C had the greatest isoprostane concentrations in the serum (P ≤ 0.01) and urine (P ≤ 0.05) compared to pigs fed the unprocessed SO. These results indicate that the change in fatty acid composition and/or the presence of lipid peroxidation products in peroxidized SO may reduce ADG and ADFI in nursery pigs, but appears to have no impact on GE, lipid, or N digestibility, or gut permeability. These data suggest that the presence of lipid peroxidation products may affect certain markers of oxidative stress.

One of the non-invasive methods for treating discoloration of hard tooth tissues is teeth whitening. The aim of this work is to assess the dynamics of changes in the acid resistance of enamel and hard tissues of teeth and the rate of its remineralization after the procedure of office teeth whitening. The study involved 123 patients aged 18 to 35 years with discoloration of various origins, with the color of hard tooth tissues on the Vita Classic A2 scale and darker. Before performing the office, teeth whitening procedure, all patients gave their written voluntary informed consent to participate in this study, as well as consent to the processing of personal data. Depending on the chosen method of office teeth whitening, patients were divided into 3 groups. The resistance of hard tooth tissues was judged based on the determination of TOER and CASRE tests. These indicators were determined at various times (5 days before the office teeth whitening procedure, 5 days after it, after 14, 30 days and 6 months). Regardless of the chosen whitening system, the office teeth whitening procedure is accompanied by a decrease in the enamel's resistance to acids and a decrease in the rate of its remineralization. The remineralizing function of oral fluid promotes the positive dynamics of the studied parameters after 14 days and after 30 days values increased due to the appointment of remineralizing therapy to all patients in 2 weeks after the teeth whitening procedure. After 6 months, all patients had high enamel resistance and the rate of its remineralization.

Abstract We previously determined that prenatal stress (PNS) differentially affected methylation of DNA from leukocytes of 28-d-old calves. Specifically, COX14 (cytochrome c oxidase (COX) assembly factor) and CKMT1B (mitochondrial creatine kinase U-type) were hypomethylated and COA5 (COX assembly factor 5), COX5A (COX subunit 5A), NRF1 (nuclear respiratory factor 1), and GSST1 (glutathione S-transferase theta-1) were hypermethylated in PNS compared to non-PNS calves (P ≤ 0.05). Our current objective was to test the hypothesis that PNS exhibit impaired mitochondrial function and greater oxidative stress than non-PNS calves. Blood and longissimus dorsi muscle samples were collected from yearling Brahman calves whose mothers were stressed by 2 h transportation at 60, 80, 100, 120, and 140 days of gestation (PNS; 8 bulls, 6 heifers) and non-PNS calves (4 bulls, 6 heifers). Serum was evaluated for the stress hormone, cortisol, and muscle damage marker, creatine kinase; muscle was analyzed for mitochondrial volume density and function by citrate synthase (CS) and COX activities, respectively, concentration of malondialdehyde, a lipid peroxidation marker, and activity of the antioxidant, superoxide dismutase (SOD). Data were analyzed using mixed linear models with treatment and sex as fixed effects. Serum cortisol was numerically higher in PNS than non-PNS calves but was not statistically different. Muscle CS and COX activities relative to protein were greater in PNS than non-PNS calves (P ≤ 0.03), but COX relative to CS activity was similar between groups. Activity of COX was greater in bulls than heifers (P = 0.03), but no other measure was affected by sex. All other measures were unaffected by PNS. Prenatal stress did not affect markers of muscle damage and oxidative stress in yearling Brahman calves at rest but mitochondrial volume density and function were greater in PNS calves. Acute stressors induce oxidative stress, so implications of differences in mitochondria in PNS calves following a stressor should be investigated.

Abstract The aim of the study was to determine the etiopathogenesis of reproductive dysfunction in highly productive cows. Studies were conducted on cows of black-motley breed, aged 3–5 lactations, with a live weight of 480–520 kg. Cows were divided into two groups. The first group included 20 cows effective inseminated after the first insemination. The second group included 17 cows that did not fertilize after the first insemination. Blood samples (n = 253) were taken from 4–7; 14–17; 24–34; 35–50; 88–95; 195–210 days of pregnancy; during the beginning of dry period (DP); 1–4 days before calving and on the first day after calving. The content of total serum lipids in pregnant animals was 15.8% higher compared to unfertilized animals. In non-pregnant cows, a decrease in free cholesterol (by 28.65%), especially in its ether-bound fraction (14.91%), indicates a lower synthetic function of the liver, which is confirmed by the low serum levels of vitamins A and E (a difference of 11.3 and 23.65%). The DP was characterized by a significant increase in the products of lipid peroxidation - malondialdehyde by 12–17%, diene and triene conjugates - by 10–15 and 8–15%, respectively, and 1–4 days before calving in cows, which subsequently had a pathological calving, in particular - retention of placenta (5 cows), were characterized by an increased content in the blood plasma of lipid peroxidation products. Thus, the difference in the number of dienes, trienes, and malondialdehyde products was 14.0, 12.0, and 23.5%, compared with animals without obstetric pathology. On the first day after calving, this difference increased even more. Cows with retention of placenta had a significantly higher level of lipid peroxidation products and were characterized by a predominance of oxidized products over the antiradical thiol disulfide and ascorbate systems restored in the antioxidant balance.

The modifications in the concentration of circulating myoglobin have been studied by means of a radioimmunoassay in 15 cancer patients undergoing polychemotherapy including adriamycin. In 8 patients significant increases in myoglobin levels were found after injection of low doses of the drug (25-50 mg/m2). Moreover, a disturbance of the normal biorhythm of the protein was evident in 12 patients. Creatine kinase-MB was evaluated by means of a radioimmunoassay, but there was no relation between an increase in the isoenzyme and an increase in myoglobin. No ECG modifications were detected. These data indicate that the measurement of myoglobin may offer an indication of myocardial or skeletal muscle damage caused by adriamycin.
