Nothaphoebe coriacea (in Kalimantan, Indonesia, commonly known as gemor) is a native wetland-plants that can grow well in swampy areas. This tree is distributed throughout South and Central Kalimantan, Indonesia. The bark of Nothaphoebe coriacea is used for bioactive compound source for larvacide, mosquito repellent, hio and glue. The objective of this research was to investigate the ultrasound-assisted extraction methods for the extraction of antioxidants compounds of Nothaphoebe coriacea. The barks were subjected to ultrasound‐assisted ethanolic and hexane extraction followed by fractionation (using ethyl acetate, chloroform, and hexane) and the antioxidant activity test was performed. The activity of antioxidant was carried out through a DPPH method. The highest antioxidant activities are shown in ethanol extract with IC50 value of 72.36 μg/mL and chloroform fraction of ethanol extract with IC50 value of 54.18 μg/mL.

Pewarna alami lebih disukai karena tidak memiliki efek yang negatif terhadap tubuh manusia sehingga mempunyai tingkat keamanan pangan yang tinggi, selain itu juga pewarna alami mudah diuraikan. Bahan pewarna alami bisa diperoleh dari berbagai jenis sumber, salah satunya yaitu spesies alga yang merupakan tumbuhan tingkat rendah di perairan. Alga terdapat 2 jenis yaitu makroalga yang berukuran besar dan mikroalga yang berukuran kecil (renik). Spesies mikroalga yang dapat menghasilkan pewarna alami salah satunya adalah Spirulina. Jenis pewarna alami yang terkandung pada mikroalga tersebut yaitu fikosianin. Pada penelitian ini, dilakukan ekstraksi fikosianin dari Spirulina platensis dengan menggunakan tiga metode berbeda yaitu Maserasi, Ultrasound-Assisted-Extraction (UAE) dan Freezing untuk mengetahui metode mana untuk menghasilkan yield dan antioksidan tertinggi dari ekstrak yang dihasilkan. Hasil dari penelitian ini didapatkan bahwa berdasarkan nilai yield tertinggi dan kandungan fikosianin, freezing merupakan metode ekstraksi terbaik dengan kandungan fikosianin yang paling tinggi yaitu sebesar 26,53%. Sedangkan nilai aktivitas antioksidan terbesar dihasilkan oleh metode ekstraksi maserasi dengan nilai IC50 sebesar 49,59 ppm. Fikosianin yang dihasilkan dari ketiga metode ekstraksi termasuk antioksidan kuat.KATA KUNCI: ekstraksi, Spirulina plantesis, fikosianin, biopigmen, antioksidanEXTRACTION OF PHYCOCYANIN FROM SPIRULINA PLANTESIS FOR BIOPIGMENT AND ANTIOXIDANTABSTRACTRecently, biopigment known as a pigment which has no impact and easy to degradated in human body and better than artificial pigment. Biopigment can be resulted from the extraction of algae that are living in the water. Spirulina plantesis is one of algae resulting a pigment and known as phycocyanin. In this research, extraction of phycocyanin was done by three method of extractions; maceration, Ultrasound-Assisted-Extraction (UAE) and Freeze-thaw. Those extraction methods were compared to decide which method that resulted highest yield of extraction and has highest antioxidant activities. From the resulted data, the highest yield and highest phycocyanin concentration was obtained by freeze-thaw extraction method. Phycocyanin concentration from freeze-thaw ectract was 26,53% (w/w). However, the highest antioxidant activities was obtained by maceraion extraction mehod. The IC50 of extract from maceration was 49,59 ppm. Hence, all extracts from different extraction methods resulted high antioxidant activities.

Maserasi merupakan salah satu metode ekstraksi yang digunakan untuk menarik kandungan kimia dari bahan alam. Macerator Magnetic Stirrer (M-MS) merupakan alat yang dikembangkan untuk memaksimalkan proses maserasi melalui pengadukan secara berkesinambungan. Tujuan penelitian ini adalah untuk mengetahui efektivitas penggunaan M-MS dalam proses ekstraksi senyawa bahan alam. Parameter yang digunakan yaitu persentase zat terekstraksi (% rendemen) dan aktivitas antioksidan (IC50) ekstrak etanol Muntingia calabura (EEMC) menggunakan metode DPPH (2,2-Diphenyl-1-Picrylhydrazil). Pengujian dilakukan terhadap simplisia daun Muntingia calabura yang  dimaserasi selama 24 jam menggunakan etanol 95% dengan maserator konvensional, M-MS dengan laju pengadukan 200 rpm dan 300 rpm. Pengadukan dengan maserator konvensional dilakukan sebanyak 3 kali setiap 8 jam, sedangkan dengan M-MS dilakukan secara kontinyu selama 24 jam. Nilai % rendemen EEMC dari maserator konevnsional, M-MS 200 rpm dan 300 rpm secara berturut-turut adalah 7,6 %, 8,4 % dan 10,2 %. Hasil uji aktivitas antioksidan menghasilkan nilai IC50 EEMC dengan maserator konvensional sebesar 18,19 ppm, M-MS 200 rpm adalah 14,35 ppm dan 300 rpm adalah 7,85 ppm. Nilai IC50 untuk masing-masing uji menunjukan < 50 ppm yang termasuk dalam kategori sangat kuat. Ekstraksi EEMC paling efektif adalah menggunakan M-MS dengan laju pengadukan 300 rpm. The maceration is one of the extraction methods used to extract chemical content from natural compounds. Macerator Magnetic Stirrer (M-MS) is a tool developed to maximize the maceration process through continuous stirring. The purpose of this study was to determine the effectiveness of using M-MS in the process of extracting natural compounds. The parameters used were the percentage of extracted substance (% yield) and antioxidant activity (IC50) of ethanol extract of Muntingia calabura (EEMC) using the DPPH method (2,2-Diphenyl-1-Picrylhydrazil). Tests were carried out on the simplicia of Muntingia calabura leaves which were macerated for 24 hours using 95% ethanol with a conventional macerator, M-MS with a stirring rate of 200 rpm and 300 rpm. Stirring with a conventional macerator was carried out 3 times every 8 hours, while with M-MS it was done continuously for 24 hours. EEMC yield value respectively of conventional macerator, M-MS at 200 rpm and 300 rpm showed 7.6 %, 8.4% and 10.2 %. The results of the antioxidant activity test resulted in the IC50 EEMC value with a conventional macerator is 18.19 ppm, M-MS 200 rpm is 14.35 ppm and 300 rpm is 7.85 ppm. The IC50 value for each test shows <50 ppm which is included in the very strong category. The most effective EEMC extraction is using M-MS with a stirring rate of 300 rpm.Keywords: maceration, M-MS, EEMC, antioxidant

Object. Ultrasound-assisted extraction of total flavonoids from corn silk and their antioxidant activities were studied.Methods. Response surface methodology was adopted to optimize the extraction conditions and antioxidant activities of the extracted total flavonoids were detected through ferric reducing antioxidant power (FRAP) assay.Results. Through a three-level, three-variable Box-Behnken design of response surface methodology (RSM) adopting yield as response, the optimal conditions were determined as follows: ultrasonic power 500 W, extraction time 20 min, material solvent ratio 1 : 20, and ethanol concentration 30%. Under the optimum conditions, the extraction yield of total flavonoids was 1.13%. FRAP value of total flavonoids extracted from corn silk was 467.59 μmol/L.Conclusion. The total flavonoids of corn silk could be developed as food natural antioxidant reagents.

AbstractThe total content of polyphenols and flavonoids determined in the same plant and their corresponding antioxidant activities may vary widely, depending on the extraction conditions applied. This study was conducted to optimise the extraction conditions of phenolics and flavonoids from the mistletoe plant. Various extraction methods, i.e. ultrasound-assisted extraction technology, maceration, maceration with stirring, accelerated solvent extraction (ASE), and extraction under reflux were evaluated for their percentage extraction of polyphenols (TPC) and flavonoids (TFC) from Viscum album subsp. abietis. In addition, the anti-radical activity of extracts was analysed using the 2,2-diphenyl-1-picrylhydrazyl method. The effects of temperature, solvent type, and concentration on the phenolic extraction efficiency and antioxidant activity were studied using chemometric and statistical methods. The results showed that the extracts of V. album subsp. abietis contained large amounts of polyphenols and flavonoids (up to 57.673 mg g−1 and 9.955 mg g−1 of dry extract, respectively) and exhibited potent antioxidant activity, hence representing promising sources of powerful antioxidants. Due to its high extraction efficiency and considerable saving of time and solvent, ASE was more effective than the other extraction techniques. Extracts prepared with water-polar solvent mixtures displayed the highest TPC, TFC, and antioxidant activity, while organic polar solvents were the least efficient extractants.
