Maserasi merupakan salah satu metode ekstraksi yang digunakan untuk menarik kandungan kimia dari bahan alam. Macerator Magnetic Stirrer (M-MS) merupakan alat yang dikembangkan untuk memaksimalkan proses maserasi melalui pengadukan secara berkesinambungan. Tujuan penelitian ini adalah untuk mengetahui efektivitas penggunaan M-MS dalam proses ekstraksi senyawa bahan alam. Parameter yang digunakan yaitu persentase zat terekstraksi (% rendemen) dan aktivitas antioksidan (IC50) ekstrak etanol Muntingia calabura (EEMC) menggunakan metode DPPH (2,2-Diphenyl-1-Picrylhydrazil). Pengujian dilakukan terhadap simplisia daun Muntingia calabura yang  dimaserasi selama 24 jam menggunakan etanol 95% dengan maserator konvensional, M-MS dengan laju pengadukan 200 rpm dan 300 rpm. Pengadukan dengan maserator konvensional dilakukan sebanyak 3 kali setiap 8 jam, sedangkan dengan M-MS dilakukan secara kontinyu selama 24 jam. Nilai % rendemen EEMC dari maserator konevnsional, M-MS 200 rpm dan 300 rpm secara berturut-turut adalah 7,6 %, 8,4 % dan 10,2 %. Hasil uji aktivitas antioksidan menghasilkan nilai IC50 EEMC dengan maserator konvensional sebesar 18,19 ppm, M-MS 200 rpm adalah 14,35 ppm dan 300 rpm adalah 7,85 ppm. Nilai IC50 untuk masing-masing uji menunjukan < 50 ppm yang termasuk dalam kategori sangat kuat. Ekstraksi EEMC paling efektif adalah menggunakan M-MS dengan laju pengadukan 300 rpm. The maceration is one of the extraction methods used to extract chemical content from natural compounds. Macerator Magnetic Stirrer (M-MS) is a tool developed to maximize the maceration process through continuous stirring. The purpose of this study was to determine the effectiveness of using M-MS in the process of extracting natural compounds. The parameters used were the percentage of extracted substance (% yield) and antioxidant activity (IC50) of ethanol extract of Muntingia calabura (EEMC) using the DPPH method (2,2-Diphenyl-1-Picrylhydrazil). Tests were carried out on the simplicia of Muntingia calabura leaves which were macerated for 24 hours using 95% ethanol with a conventional macerator, M-MS with a stirring rate of 200 rpm and 300 rpm. Stirring with a conventional macerator was carried out 3 times every 8 hours, while with M-MS it was done continuously for 24 hours. EEMC yield value respectively of conventional macerator, M-MS at 200 rpm and 300 rpm showed 7.6 %, 8.4% and 10.2 %. The results of the antioxidant activity test resulted in the IC50 EEMC value with a conventional macerator is 18.19 ppm, M-MS 200 rpm is 14.35 ppm and 300 rpm is 7.85 ppm. The IC50 value for each test shows <50 ppm which is included in the very strong category. The most effective EEMC extraction is using M-MS with a stirring rate of 300 rpm.Keywords: maceration, M-MS, EEMC, antioxidant

Kepok banana peel (Musa x paradisiaca L.) contains flavonoids that act as antioxidants. The antioxidant activity of the ethanol extract of Kepok banana peel can be used as an active ingredient of nano gel. Nano-sized preparations can increase the efficiency of the delivery of active ingredients with lesser side effects and also decrease the stratum corneum to make it easier to penetrate. The objectives of this study were to determine whether the ethanol extract of Kepok banana peel (Musa x paradisiaca L.) can be formulated as nano gel and to measure its antioxidant activity. The method of nano gel preparation was stirring technique by using a magnetic stirrer and a sonicator. The antioxidant activity was measured using DPPH method. The results showed that the ethanol extract of kepok banana peel could be formulated as nano gel with size F0 = 161.9 nm; F1 = 171.3 nm; F2 = 165.6 nm; F3 = 163.9 nm which fulfilled the nano gel size requirement (20-200 nm) and the potential zeta value F0 = -43.4 mV; F1 = -43.7 mV; F2 = -46.9 mV; F3 = -47.0 mV fulfilled the requirements (smaller than -30 mV and greater than +30 mV). The antioxidant activity showed that the IC50 of ethanol extract of was stronger than IC50 of 1% nano gel (198.0279 µg/ml : 282.8933 µg/ml).

ABSTRACTKersen (Muntingia calabura L.) is a plant that has begun to be eliminated and was rarely used because it is often considered to have no economic value and lack of knowledge about its utilization, whereas kersen plants contain flavonoids, saponins, and tannins which were have high benefit for health. The content of metabolites is affected by soil nutrient elements and difference place of growth. This study aims to determine the potential of kersen leaves grown in North Minahasa based on phytochemical content, ability of antioxidant activity, and toxicity. Kersen leaves were extracted using sequential maceration method with n-hexane, ethyl acetate, and ethanol as solvents. Phytochemical Screening using several reagents which tailored to the type of phytochemical test. 1.1-diphenyl-2-picrylhydrazyl (DPPH) method is used to evaluate antioxidant activity, and Brine Shrimp Lethality Test (BSLT) method is used to evaluate toxicity. The result of this study indicate that the n-hexane extract contains phenols, flavonoids, and tannins, with IC50 value 12.54 μg/mL, and LC50 value 881 μg/mL. Ethyl acetate extract contains phenols, flavonoids, tannins, and saponins, with IC50 value 61.3 μg/mL, and LC50 value 1758 μg/mL. Ethanol extract has phenol, flavonoid, tannin, saponin, and terpenoid content, with IC50 value 9.01 μg/mL, and LC50 value 106 μg/mL. Keywords : Kersen leaves, Antioxidant, Toxicity, IC50, LC50 ABSTRAKKersen (Muntingia calabura L.) merupakan tanaman yang sudah mulai tersingkirkan dan jarang dimanfaatkan karena sering dianggap tidak punya nilai ekonomis dan kurangnya pengetahuan tentang pemanfaatannya, padahal tanaman kersen memiliki kandungan flavonoid, saponin, dan tanin yang bermanfaat tinggi untuk kesehatan. Kandungan senyawa metabolit dipengaruhi oleh unsur hara tanah dan perbedaan tempat tumbuh. Penelitian ini bertujuan untuk mengetahui potensi dari daun kersen yang tumbuh di Minahasa Utara berdasarkan kandungan fitokimia, kemampuan aktivitas antioksidan, dan toksisitasnya. Ekstrak daun kersen diekstraksi dengan metode maserasi sekuensial menggunakan pelarut n-heksan, etil asetat, dan etanol.  Skrining fitokimia menggunakan beberapa reagen yang disesuaikan dengan jenis uji fitokimia. Metode 1.1-diphenyl-2-picrylhydrazyl (DPPH) digunakan untuk mengevaluasi aktivitas antioksidan, dan metode Brine Shrimp Lethality Test (BSLT) digunakan untuk mengevaluasi toksisitas. Hasil penelitian ini menunjukkan bahwa ekstrak n-heksan memiliki kandungan fenol, flavonoid, dan tanin, nilai IC50 12,54 μg/mL, dan nilai LC50 881 μg/mL. Ekstrak etil asetat memiliki kandungan fenol, flavonoid, tanin, dan saponin, nilai IC50 61,3 μg/mL, dan nilai LC50 1758 μg/mL. Antioxidants are substances that can provide endogenous protection and exogenous oxidative stress by capturing free radicals. Many plants are efficacious as antioxidants, namely plants that contain polyphenols, especially flavonoids, so many are formulated as natural antioxidants. Plants such as Muntingia calabura, Syzygium cumini, Ocimum basilicum, and Eleutherine bulbosa contain polyphenol compounds, especially flavonoids which are efficacious as natural antioxidants. This research aimed to study antioxidant activity derived from some potential plants using the DPPH method by calculating the IC50 value of each plant extract. This research method starts from the determination process to prove the validity of the plants used, the extraction process using the maceration method with 70% ethanol solvent, then the antioxidant activity of extracts from each plant was carried out using the DPPH method. This research starts from the determination process to ensure the correctness of the plants used, then the extraction process is carried out using the maceration method with 70% ethanol solvent. After that the antioxidant activity was determined from the four plants using the DPPH method to see the strongest IC50 value among the four plants. IC50 is the concentration of the sample to inhibit 50% of free radicals. The results of IC50 values from ethanol extract of M. calabura leaves, Syzygium cumini leaves, Ocimum basilicum leaves and Eleutherine bulbosa bulbs, were 18.72; 63,84; 141.59 and 173.15 ppm. Ethanol extract of M. Diseases mediated by the immune system are difficult problems to treat such as human immunodeficiency virus (HIV) and other lethal viruses. Infections that occur in normal people are generally brief and rarely leave permanent damage. Treatment of this disease requires an aggressive and innovative approach to the development of new treatments so that it requires the role of immunomodulators to improve the immune system. A substance that acts as an enhancer or immune enhancer can be obtained by using herbs that are efficacious as immunostimulants. One of the herbs used is herbal binara (Artemisia vulgaris L) which has been studied as a potential immunomodulator with high antioxidant activity. Previous research also stated that red shoots (Syzygium oleana) were studied as potential immunomodulators with high antioxidant activity. Several other species such as Syzygium samarangense have 16 flavonoida compounds which show pharmacological immunological activity. The purpose of this study was to determine the content of secondary metabolites of ethanol extract of herbal binara (Artemisia vulgaris L.) with red shoots (Syzygium oleana) and to determine the best dose of extract from the ethanol extract of herbal binara (Artemisia vulgaris L.) with red shoots (Syzygium oleana) can reduce the volume of swelling of mouse feet. Examination of the chemical content of secondary metabolites from the ethanol extract of herbal binara (Artemisia vulgaris L.) with red shoots (Syzygium oleana) is carried out by chemical screening and characterization of simplicia and extract. The method used is the slow type hypersensitivity method. In this test the independent variable is the secondary metabolite of ethanol extract of herb binara (Artemisia vulgaris L.) with red shoots (Syzygium oleana) with four concentrations (50, 100, 200 and 400 mg / kgBB). The positive control used by Stimuno dose is 32.5 mg / kgBB

ABSTRACTHabitat of sponge Stylissa sp., were under the sea and these sponges contain active compounds, which are more active than the compounds produced by terrestrial plants. Antioxidants are inhibitors of oxidation reactions due to the free radicals, which can cause weak damage to unsaturated cells, cell wall membranes, blood vessels, DNA bases, and lipid tissue, that causing disease. The study was conducted to determine the antioxidant activity of ethanol extracts  of sponge Stylissa sp., which is located on the Bangka Island, Likupang District, North Minahasa Regency. Sponge Stylissa sp., was extracted by maceration with using ethanol. Testing of antioxidant activity was carried out by the DPPH method measured by a UV-Vis spectrophotometer. The results of the study showed that ethanol extracts of sponge Stylissa sp., has antioxidant activity in each concentration and the highest at a concentration of 100 mg / L. The conclusion is a ethanol extract of sponge Stylissa sp. have high antioxidant activity with a concentration of 25 mg/L (77,40%), concentration of 50 mg/L (85,63%), concentration of 75 mg/L (88,66%), and concentration 100 mg/L (88,96%). Key words: Stylissa sp. Antioksidan adalah zat penghambat reaksi oksidasi akibat radikal bebas yang dapat menyebabkan kerusakan lemah tak jenuh, membran dinding sel, pembuluh darah, basa DNA, dan jaringan lipid sehingga menimbulkan penyakit. Penelitian dilakukan untuk mengetahui aktivitas antioksidan ekstrak etanol biota laut spons Stylissa sp. yang terdapat di pulau Bangka, Kecamatan Likupang, Kabupaten Minahasa Utara. Spons Stylissa sp. ini diekstrak dengan metode maserasi menggunakan etanol. Pengujian aktivitas antioksidan ini dilakukan dengan metode DPPH yang diukur dengan alat spektrofotometer UV-Vis. Hasil dari penelitian menunjukan bahwa ekstrak etanol spons Stylissa sp. memiliki aktivitas antioksidan disetiap konsentrasi dan yang tertinggi pada konsentrasi 100 mg/L. Kesimpulan yang didapat bahwa ekstrak etanol spons Stylissa sp. memiliki aktivitas antioksidan yang tinggi dengan konsentrasi 25 mg/L (77,40%), konsentrasi 50 mg/L (85,63%), konsentrasi 75 mg/L (88,66%), dan konsentrasi 100 mg/L (88,96%). Kata Kunci : Spons Stylissa sp., Antioksidan, DPPH (1,1-difenil-2-pikrilhidrazil)

ABSTRACTDayak onion plant (Eleutherine americana Merr.) or commonly called forest onion is a typical plant of Central Kalimantan that has been used by local peoples for cure a various types of diseases. This study aims to determine the activity of antioxidant compounds from Dayak bulb ethanol extracts from Kotamobagu City. This research method is an experimental laboratory. Initially the Dayak bulb sample was extracted maceration with 96% ethanol solvent then evaporated until a thick extract was obtained. As a parameter, the DPPH method (1,1-diphenyl-2-picrylhydrazyl) was used to test the antioxidant activity of the samples at concentrations of 50, 60, 70, 80 and 90 mg / L and as a comparison standard used Vitamin C with concentrations of 2, 4, 6, 8 and 10 mg / L. The results obtained showed antioxidant activity in Dayak bulb ethanol extracts with IC50 values of 41.46 mg / L and on vitamin C IC50 values of 1.04 mg / L. Based on these results it can be concluded that the onion bulb ethanol extract has very strong antioxidant activity. Keywords: Dayak Onion bulbs, Activity, Antioxidant, DPPH  ABSTRAK Tanaman bawang Dayak (Eleutherine americana Merr.) atau biasa disebut bawang hutan merupakan tanaman khas Kalimantan Tengah yang sudah dipergunakan masyarakat lokal sebagai obat berbagai jenis penyakit. Penelitian ini bertujuan untuk mengetahui aktivitas senyawa antioksidan dari ekstrak etanol bulbus bawang Dayak yang berasal dari Kota Kotamobagu. Metode penelitian ini adalah eksperimental laboratorium. Awalnya sampel bulbus bawang Dayak diekstraksi maserasi dengan pelarut etanol 96% kemudian dievaporasi hingga didapatkan ekstrak kental. Sebagai parameter, digunakan metode DPPH (1,1-diphenyl-2-picrylhydrazyl) untuk pengujian aktivitas antioksidan sampel pada konsentrasi 50, 60, 70, 80 dan 90 mg/L dan sebagai standar pembanding digunakan Vitamin C dengan konsentrasi 2, 4, 6, 8 dan 10 mg/L. Hasil penelitian yang didapat menunjukkan aktivitas antioksidan pada ekstrak etanol bulbus bawang Dayak dengan nilai IC50 sebesar 41,46 mg/L dan pada vitamin C nilai IC50 sebesar 1,04 mg/L. Berdasarkan hasil tersebut dapat disimpulkan bahwa ekstrak etanol bulbus bawang Dayak memiliki aktivitas antioksidan yang sangat kuat. Kata kunci: Bulbus Bawang Dayak, Aktivitas, Antioksidan, DPPH

Green tea (Camellia sinensis L.) is known to have ability to protect skin against free radicals. This is supported by polyphenol compound catechin. This research aims to determine the optimum Hydrophilic-Lipophilic Balance (HLB) value of Tween 60 and Span 80 compositions on the optimum cream formula of ethanol extract of green tea leaves. Tea leaves are extracted by macerating using 70% ethanol. Catechin in extract is known from Thin Layer Chromatography (TLC) test with silica gel 60 F254 as stationary phase and ethyl acetate:aquadest:formic acid (18:1:1 v/v) as mobile phase. Antioxidant activity is determined by 2,2-Diphenyl-1-picryhydrazyl (DPPH) method and value of Inhibition Concentration 50% (IC50) is then calculated. Formula optimization using Design Expert® version 7.1.5 (DX 7) software, Simplex Lattice Design (SLD) method with two components Tween 60 and Span 80. Cream is characterized according to physical properties organoleptic, homogeneity, viscosity, pH, spreadability, adhesiveness, and cream type. The optimum formula obtained is then tested for physical stability for 4 weeks at room temperature (28±2°C) and data are statistically analyzed using one-way ANOVA. The extract contains catechin proved with Retention factor (Rf) value 0.8 and has antioxidant activity with IC50 value 56.35 ppm. 6.4% Tween 60 and 3.6% Span 80 result an optimum HLB value 11.1. It has viscosity 2897.50±35.94 mPa.s, spreadability 18.44±0.06 cm2, adhesiveness 0.85±0.05 seconds, and pH 4.530±0.002. Statistical test shows that the cream is significantly altered at pH, but does not significantly change in viscosity, spreadability, and adhesiveness after being stored for 4 weeks.

ABSTRACT Sponges are a component of coral reef biota that has bioactive compounds with a greater percentage of activity compared to compounds produced by terrestrial plants. This study aims to determine the presence of antioxidant activity in the ethanol extracts of sponge Ianthella basta from the waters of Tumbak Village. Ianthella basta sponge was extracted using maceration method with 96% ethanol solvent. Antioxidant activity testing was carried out using the DPPH method with a concentration of 100 µg / mL, 75 μg / mL, 50 μg / mL, and 25 μg / mL and Vitamin C p.a as positive control. Each sample was made three repetitions of the test. Testing using a UV-Vis spectrophotometer. The results showed that the ethanol extract of the Ianthella basta sponge had antioxidant activity with a percentage of 48.73% at a concentration of 100 µg / mL. Keywords: Ianthella basta sponge, Antioxidants, Extraction, DPPH  ABSTRAK Spons merupakan salah satu komponen biota penyusun terumbu karang yang mempunyai senyawa bioaktif dengan persentase keaktifan lebih besar dibandingkan dengan senyawa-senyawa yang dihasilkan oleh tumbuhan darat. Penelitian ini bertujuan untuk mengetahui adanya aktivitas antioksidan di dalam ekstrak etanol Spons Ianthella basta dari perairan Desa Tumbak. Spons Ianthella basta diekstraksi menggunakan metode maserasi dengan pelarut etanol 96%. Pengujian aktivitas antioksidan dilakukan dengan menggunakan metode DPPH dengan konsentrasi 100 µg/mL, 75 µg/mL, 50 µg/mL, dan 25 µg/mL dan Vitamin C p.a sebagai kontrol positif. Masing-masing sampel dibuat tiga kali pengulangan uji. Pengujian menggunakan alat spektrofotometer UV-Vis. The effect of Cu2+ and Mg2+ metal ions on the antioxidant activity of anthocyanins of ethanol extract from the red dragon skin fruit (Hylocereus polyrhizus) has been done. Anthocyanin pigments from ethanol extract the red dragon skin fruit, content and antioxidant activity of anthocyanins from the red dragon fruit skin with ethanol solvent, and the effect of Cu2+ and Mg2+ metal ions on the antioxidant activity of anthocyanins. Identification of anthocyanin was done by identification of color test, using spectrophotometer analysis of UV-Vis and FTIR. The anthocyanin content was determined using pH difference method, and determined effect of metal ions extract was done by adding 50 ppm, 100 ppm, and 150 ppm of metal ions Cu2+ and Mg2+ into the anthocyanin extract that was been acidified by HCl , using spectrophotometer analysis of FTIR. Antioxidant activity test using DPPH method. The results show that ethanol extracts the red dragon anthocyanin by content is 12.5241 mg / L and antioxidant activity (IC50) is 0.478 μg / mL and with addition of2+ and Mg2+ metal ions can be increased antioxidant activity (IC50) of anthocyanins to be 0.2259 μg / mL for Cu2+ at concentration 50 ppm and 0.3351 μg / mL for Mg2+ at concentration 50 ppm.

This is the first report on the extraction of cocoa bean shell (CBS) using deep eutectic solvents (DESs). Screening results with 16 different choline chloride-based DESs showed how choline chloride:oxalic acid DES was the most suitable solvent for the extraction of the bioactive compounds from CBS and that concentrations varied greatly depending on the used solvent. The DES extraction was compared to the DESs coupled with microwave extraction (MAE), and the yields of the extracted compounds were higher for DES/MAE. For theobromine, the obtained yields for DES extraction were 2.145–4.682 mg/g, and for caffeine, were 0.681–1.524 mg/g, whereas for DES/MAE, the same compounds were obtained in 2.502–5.004 mg/g and 0.778–1.599 mg/g. Antioxidant activity was also determined, using DPPH method, obtaining 24.027–74.805% activity for DES extraction and 11.751–55.444% for DES/MAE. Water content significantly influenced the extraction of targeted active compounds from CBS, whereas extraction time and temperature did not show statistically significant influence. The extraction temperature only influenced antioxidant activity. The study demonstrated how extraction using DES and microwaves could be of a great importance in the future trends of green chemistry for the production of CBS extracts rich in bioactive compounds.
