Subject. This article considers the issues of provision of supplementary education services for children and methodological tools for the formation of educational environment. Objectives. The article aims to assess the level of development of the system of supplementary education of children in Krasnoyarsk and offer a methodological approach to improving the management decision-making procedure in the formation of a portfolio of supplementary education services at the municipal level. Methods. For the study, we used the methods of theoretical, empirical, and logistic analyses, and sociological studies. Results. The article offers concrete solutions to the lack of a methodological approach to providing supplementary education services, considering one of the micro-districts of Krasnoyarsk as a case in point. It also offers tools that can be used by public authorities to organize educational space at the municipal level. Conclusions. A unified methodological approach should be used to provide a system of supplementary education, taking into account financial means, as well as differentiation in the distribution of educational facilities throughout the area. The relevance of the set of supplementary education services and consumer preferences should be taken into account, as well.

The priorities and problems of the implementation of the national project “Education” at the level of a comprehensive school in the region are investigated. The factors of the origin of conflict zones and types of conflicts based on the analysis of contradictions of the opinions of scientists are revealed. The role of unity and struggle of opposites as a phenomenon of overcoming conflicts is determined. Based on the author’s monitoring of the education management system in the Mari El Republic, the positive and negative dynamics of the number of educational organizations studying in them and in the system of additional education over the past five years, the enrollment of students in specialized education, as well as the dynamics of participants in the regional and final stage of the All-Russian Olympiad of schoolchildren were revealed. Particular attention is paid to the study of the state of modernization of the regional system of general education in the framework of the national project, highlighting such priorities as computerization, replenishment of the school library stock, improving the quality of education and developing the personnel potential of the industry. According to the authors, the resolution of such problems as the creation of effective mechanisms for financing educational services in organizations with a weak social infrastructure, the availability of high-quality education, the low attractiveness of working conditions and the level of salaries of teachers, and others remain incomplete. To eliminate them, it is necessary to apply such measures as the further development of innovative forms of the education system, the strengthening of interdepartmental bodies and organizations to increase the educational needs of the population, the creation of a specialized financing system for the modernization of educational institutions, as well as the increase in the level of professionalism of teachers. Their actualization at the present stage contributes to the reduction of conflict zones in the education system.

Acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) can serve as biochemical markers of various pathologies like liver disfunction and poisonings by nerve agents. Ellman’s assay is the standard spectrophotometric method to measure cholinesterase activity in clinical laboratories. The authors present a new colorimetric test to assess AChE and BChE activity in biological samples using chromogenic reagents, treated 3D-printed measuring pads and a smartphone camera as a signal detector. Multiwell pads treated with reagent substrates 2,6-dichlorophenolindophenyl acetate, indoxylacetate, ethoxyresorufin and methoxyresorufin were prepared and tested for AChE and BChE. In the experiments, 3D-printed pads containing indoxylacetate as a chromogenic substrate were optimal for analytical purposes. The best results were achieved using the red (R) channel, where the limit of detection was 4.05 µkat/mL for BChE and 4.38 µkat/mL for AChE using a 40 µL sample and a 60 min assay. The major advantage of this method is its overall simplicity, as samples are applied directly without any specific treatment or added reagents. The assay was also validated to the standard Ellman’s assay using human plasma samples. In conclusion, this smartphone camera-based colorimetric assay appears to have practical applicability and to be a suitable method for point-of-care testing because it does not require specific manipulation, additional education of staff or use of sophisticated analytical instruments.
