This study aimed to evaluate the Total Plate Count and total Staphylococcus aureus count of beef sold in wet markets in Mulyorejo sub-district below the National Standard Indonesia (SNI 7388:2009) about maximum limit of microbial contamination in food or not. Total of twenty four samples of beef purchased from traditional markets of Tempurejo, Krempyeng Yamuri, Pacar Keling, and Menur in Mulyorejo sub-district Surabaya were examined by Total Plate Count using pour plate method. The sample was also cultured in Mannitol Salt Agar. The colony suspected to be S. aureus were taken for identification. The identification of S. aureus consists of isolation in Mannitol Salt Agar, Gram staining, catalase test, and coagulase test. Total plate count result showed that four samples were exceeding the National Standard of Indonesia SNI 7388:2009 or 1x106 CFU/g and the rest were below the maximum Total Plate Count in SNI. The highest Total Plate Count result was 1,9x106 CFU/g and the lowest was 7,8x104 CFU/g. The result of identification showed that 100% samples examined were contaminated by S. aureus with the highest result was 2,9x104 CFU/g and the lowest result was 4,3x103 CFU/g or exceeding the SNI 7388:2009.

An investigation was carried out for detection and enumeration of coagulase-positive Staphylococcus aureus from fresh, fried and dried meat samples sold in the traditional markets in Benin City. Using Salt Milk Agar for presumptive detection and coagulase test for confirmation, it was found that all samples of meat from the traditional markets contained coagulase-positive staphylococci. Weekly sampling was done over a 6-month period to demonstrate the prevalence of the coagulase-positive staphylococci in both traditional market, supermarket and abattoir meat samples. The range of counts fell between 104 and 1011 cells per gram. Fresh pork from the traditional market had the highest average number, 6.4×1010/g, followed by dried beef 3.7×1010/g, fresh goat and fried chicken 6.8×109/g each and fresh beef 7.0×109/g. The supermarket meat showed that chilled beef had the highest, 4.5×105/g, followed by chilled pork 3.0×105/g and frozen chicken 2.0×104/g. The abattoir meat had 3.5×104/g. The effects of condiments on inhibition of S. aureus were determined in abattoir meat homogenates. Clove was the most active of the spices tested with 1% (w/v) preventing outgrowth of S. aureus for up to 18-h contact time. When used at 10% (w/v), clove was found to be bactericidal, reducing the S. aureus load from 2.9×103/ml of homogenate at 0 h to 1.2×101/ml after 18 h of contact time.

Abstract Waste management has consistently been challenged around the globe for decades, yet sophisticated technology has not managed the pile of trash, particularly in developing countries. One of the most often produced waste is the traditional market. The study aims to identify how the conventional market disposes of its regular waste and investigate the space quality as a crowded place during the pandemic of Covid-19. The research method is observing and analysing a case study located in Banda Aceh City, Indonesia. Pasar Al-Mahirah is one of the traditional markets which distributing its complete regular waste directly to landfills. The result indicates that waste management is not sustainable because it lacks sorting and processing before. In terms of C-19 spreading, the space inside the building is vulnerable even though it has excellent natural ventilation and sunlight. This study performed a visual design solution for distributing waste inside the building, sustainable management based on 3R principle methods, and composting through aerobic methods for organic waste. Moreover, the stalls arrangement and the circulation for users are purposed in a layout plan to prevent a higher risk of the rising of Coronavirus transmissions.
