Abstract Background: Triple antibiotic pastes (TAP) has gained popularity as a root canal medicament in regenerative endodontic procedures and other endodontic treatment modalities. As this medicament changes the chemical structure of dentine, it may affect the bond strength of endodontic sealers to radicular dentine. This study aimed to evaluate the effect of TAP on the bond Strength of epoxy and methacrylate resin- based sealers to root canal dentine.Methods: In this in vitro study, eighty single-rooted human mandibular premolars were prepared using ProTaper rotary system. The specimens were randomly divided into a control group (without intracanal dressing) and an experimental group receiving TAP (n = 40). The intracanal dressing was removed after three weeks. Then samples of each group were randomly divided into four subgroups (n = 10) and obturated with gutta-percha and different resin-based sealers. G1: AH Plus, G2: Syntex, G3: EndoREZ, G4: MetaSEAL. After one week, 16 Slices of 1±0.1 mm thickness were obtained from the midroots of teeth in each subgroup and a push-out test was used to measure the bond strength. Slices were examined using a stereomicroscope at 30× to determine the mode of failure. The data were analyzed using two-way analysis of variance, one- way analysis of variance and Tukey post hoc tests (α = 0.05).Results: Compared to control group, TAP significantly increased the bond strength of MetaSeal and EndoREZ (p<0.05). In the control group, epoxy resin- based sealers showed higher bond strength compared to methacrylate ones (p=0.00). In TAP group, Syntex and EndoREZ showed significantly the greatest and the lowest bond strengths respectively (p<.05). The analysis of failure modes revealed a predominance of mixed failures in all groups except for Syntex group in which most failures were cohesive.Conclusions: TAP significantly increased the bond strength of methacrylate resin- based sealers.

ABSTRACT Background Smear layer is a negative factor which prevents adhesion of the filling material to the dentinal walls. Recent advances in dental research have incorporated lasers as a potential adjunct in root canal treatment by removing the smear layer before filling the root canal system, enhancing the adhesion of sealers to dentin and improving the sealing ability. Aim To evaluate the microtensile bond strength of AH-Plus resin-based sealer to dentin after treatment with 980 nm diode and 1,064 nm neodymium-doped:yttrium aluminum garnet (Nd:YAG) laser in vitro. Materials and methods Thirty specimens prepared for three groups namely group I (control), group II (980 nm diode–lased specimens) and group III (Nd:YAG–lased specimens). One tooth from each group was observed under scanning electron microscope for evaluation of intracanal root dentin morphology. Remaining specimens were used for making microsections by hard tissue microtome. Specimens for groups II and III were lased with 980 nm diode and 1,064 nm Nd:YAG laser. AH Plus sealer was applied onto specimens and mounted onto Instron universal testing machine for microtensile bond strength testing. Results were subjected to statistical analysis using one-way analysis of variance (ANOVA) and Tukey's test. Results Group III Nd:YAG had maximum mean microtensile bond strength values (11.558 ± 0.869), followed by group II diode (9.073 ± 0.468) and group I control (6.05 ± 0.036). Statistically significant differences were seen among all the groups. SEM analysis shows removal of smear layer in both groups II and III. Conclusion Both Nd:YAG and diode laser were more effective than control group in improving the microtensile bond strength of AH Plus sealer to dentin. Clinical significance Lasers have the potential to increase the adhesiveness of root canal sealer to dentin surface, thereby improving the quality of root canal obturation. The first aim of this paper was to evaluate the push-out bond strength of the gutta-percha coating of Thermafil and GuttaCore and compare it with that of gutta-percha used to coat an experimental hydroxyapatite/polyethylene (HA/PE) obturator. The second aim was to assess the thickness of gutta-percha around the carriers of GuttaCore and HA/PE obturators using microcomputed tomography (μCT). Ten (size 30) 1 mm thick samples of each group (Thermafil, GuttaCore, and HA/PE) were prepared. An orthodontic wire with a diameter of 0.5 mm was attached to the plunger of an Instron machine in order to allow the push-out testing of the gutta-percha coating. Five samples of (GuttaCore and HA/PE) were scanned usingμCT. The data obtained were analysed with one-way analysis of variance and Tukey post hoc test. HA/PE obturators exhibited significantly higher push-out bond strength (P<0.001) determined at 6.84 ± 0.96 than those of Guttacore around 3.75 ± 0.75 and Thermafil at 1.5 ± 0.63. GuttaCore demonstrated significantly higher bond strength than Thermafil (P<0.001).μCT imaging revealed that the thickness of gutta-percha around the experimental HA/PE carrier was homogeneously distributed. The bondability and thickness of gutta-percha coating around HA/PE carriers were superior to those of GuttaCore and Thermafil obturators.

Abstract Background: The aim of the current study was to evaluate and compare the influence of Dycal, Lime-lite, Theracal LC, Biodentine, Resin-modified glass ionomer cement (RMGIC), and Activa Bioactive as the pulp capping material on the shear bond strength of resin composite to dentin.Methods: A total of 70 extracted caries-free molars were randomly assigned to seven groups. Six test groups were covered with various protective liners: Dycal (GD), Theracal LC (GT), lime-lite (GL), Activa Bioactive (GA), Biodentine (GB), RMGIC (GR). The control group (GC)received no liner pretreatment. Each sample was bonded to resin composite using the total-etch tetric N bond adhesive. The samples were then tested for shear bond strength using the universal testing machine at a cross-head speed of 1 mm/min until bond failure occurred. The data were analyzed using the one-way ANOVA test followed by the Tamhane post-hoc test for pairwise comparisons of the groupsResults: Independent of the type of the applied liner, all groups exhibited inferior SBS to dentine compared to the control group. GT and GR showed significantly higher shear bond strength than GB and GD, which showed the lowest shear bond strength. GL and GA also had significantly lower SBS results than GT. The mode of fracture was predominantly cohesive in GD, GB, and GT and adhesive in GA.Conclusion: This present study concludes that the bond strength of resin-composite to dentine can be affected differently using various types of liners.

<sec> <title>Objective:</title> This laboratory study aimed to evaluate the effect of self-adhesive root canal sealers on the fracture strength of root canals filled with Resilon or EndoREZ. </sec> <sec> <title>Materials and Methods:</title> A total of eighty extracted mandibular premolar teeth were selected in this in-vitro study. All teeth were instrumented using a crown-down technique by FlexMaster rotary NiTi files. Specimens were divided into 4 test groups (n = 20) according to the sealer material: RS; RealSeal, RSS; RealSeal SE, MS; MetaSeal, and CG; (control group) zinc oxide eugenol-based sealer. Each main group was distributed into two subgroups (n = 10) according to the filling material either Resilon or EndoREZ and gutta-percha (n = 20) in the control group. Each root was mounted in acrylic resin blocks and subjected to fracture in a universal testing machine. The load values at root specimens fractured were registered in Newton’s and the data were analyzed using two-way analysis of variance and the Tukey HSD test (α = 0.05). </sec> <sec> <title>Results:</title> The two-way analysis of variance analysis indicated that the filling material had a significant effect on the fracture strength of endodontically treated teeth (p < 0.05) but not the sealer materials (p≥.05). Higher mean fracture strength was recorded in MetaSeal and Resilon group (1281.90±200.34 N) and lower mean fracture strength was shown in RealSeal and EndoREZ group (847.55±191.04 N). </sec> <sec> <title>Conclusion:</title> Self-adhesive (fourth-generation) resin sealers increased the fracture strength of root teeth more than self-etching (third-generation) root canal sealers when used with EndoREZ points. </sec>

Introduction: A pivotal issue to achieve success in the treatment of the root canal is root canal disinfection. One of the most important bacteria that infect the root canal is Enterococcus faecalis. This study seeks to examine the effectiveness of 3 methods for disinfecting the root canal: photodynamic therapy, modified triple antibiotic paste (MTAP), and calcium hydroxide. Methods: Sixty-two single-rooted extracted anterior teeth were collected. After cleaning and disinfecting the teeth, their crowns were cut at the CEJ point. The root canals were shaped to the working length up to file F3 ProTaper (F1, F2, F3). EDTA 17% and sodium hypochlorite 2.5% were used for 5 minutes to wash and remove the smear layer, and then the apical foramen was sealed using composite. After that, the teeth were sterilized in an autoclave at 121°C for 15 minutes. Then 10 samples were taken randomly as the negative control. The remaining samples were immersed and cultivated in a suspension containing E. faecalis for 21 days. Then the samples were divided into 5 groups: 2 positive control groups, 1 group treated with the antibiotic paste with the concentration of 1 mg/mL, 1 group treated with calcium hydroxide, and 1 group treated with photodynamic therapy. Then, to collect the biofilm, the ProTaper file F4 was used. After that, the microbial suspension was provided and counting the colonies was carried out to compare the groups. Results: The findings indicated that the amount of CFU/mg of MTAP samples, including clindamycin, metronidazole, ciprofloxacin in the concentration of 1 mg/mL and photodynamic therapy and calcium hydroxide was lower than that in the control group. Antibiotic paste cleansed the root canal up to 99.9%. Photodynamic therapy reduced the amount of CFU/mg to 98.8%, and calcium hydroxide reduced the amount of CFU/mg to 94.13%. Conclusion: Using photodynamic therapy causes a reduction in the biofilm and inhibits the growth of the E. faecalis bacterium. In addition, in this study, MTAP with a concentration of 1 mg/mL was used, which expunged the bacteria completely. Meanwhile, calcium hydroxide had the weakest effect of all on the E. faecalis bacterium.

This study evaluated comparatively the adhesion of Epiphany and AH Plus endodontic sealers to human root dentin treated with 1% NaOCl and 1% NaOCl+17% EDTA, using the push-out test. Sixty root cylinders obtained from maxillary canines had the canals prepared and were randomly assigned to 3 groups (n=20), according to root dentin treatment: GI - distilled water (control), GII - 1% NaOCl and GIII - 1% NaOCl+17% EDTA. Each group was divided into 2 subgroups (n=10) filled with either Epiphany or AH Plus. Bond strength push-out test data (kN) were obtained and analyzed statistically by ANOVA and Tukey's post-hoc test. There was statistically significant difference between sealers (AH Plus: 0.78 ± 0.13; Epiphany: 0.61 ± 0.19; p<0.01) and among root dentin treatments (distilled water: 0.58 ± 0.19; 1% NaOCl: 0.71 ± 0.12; 1% NaOCl+17% EDTA: 0.80 ± 0.17; p<0.05). In conclusion, AH Plus sealer presented greater adhesion to dentin than Epiphany, regardless of the treatment of root canal walls.

Abstract The aim of the present study was to evaluate the cytotoxicity of root canal sealers under conditions closely resembling a clinical reality. A primary human fibroblast cell line was seeded in 24-well acrylic plates with Dulbecco’s modified Eagle’s medium supplemented with 10% serum fetal bovine (SFB) and incubated for 24 h. Root canals from premolars were filled and individually attached to nylon devices to be stabilized in the wells with the already seeded cells. Specimens were divided into groups as follows: Control: gutta-percha cones (GPC); AH Plus+GPC; Sealapex+GPC; MTA Fillapex+GPC and Endofill+GPC. After 24 and 48 h, cell viability and morphology were evaluated by MTT assay and scanning electron microscopy (SEM), respectively. Statistical analysis was performed by Mann-Whitney test, complemented by Kruskal Wallis test (p<0.05). Only Endofill presented cytotoxicity after 24 h. MTA Fillapex and Endofill reduced the production of succinic desidrogenase after 48 h. AH Plus was non-toxic at any time point. SEM showed that the AH Plus and MTA Fillapex groups presented fibroblasts with morphology close to the control group, while the Endofill group presented few cells with thin extensions cells. The present study showed that good results were present in AH Plus and Sealapex, but not the Endofill group after 48 h. The method used enabled evaluation of the cytotoxicity of the studied sealers that diffused through the root apex.

ABSTRACT Objective: To evaluate the dentoskeletal effects of different anterior open bite treatment modalities in children. Materials and Methods: This cephalometric study assessed changes resulting from different treatment approaches on 77 growing children with anterior open bite. A control group (n = 30) was used for comparison. Lateral cephalograms were available before treatment and after 12 months. The sample was divided into four groups: removable palatal crib associated with a chincup (G1), bonded spurs associated with a chincup (G2), chincup (G3), and nontreated control (G4). Statistical comparisons among the four groups were performed on T1 and the treatment changes using analysis of variance with Tukey's post hoc tests. Results: No statistically significant changes in skeletal variables were found among the groups, except for lower anterior face height (LAFH) increase in G1. Overall, effects in all of the treated groups were exclusively dentoalveolar. A larger overbite (OB) increase was observed in G1 and G2 when compared with G3 and G4. The maxillary incisors in G1 showed increased palatal tipping, retrusion, and more vertical dentoalveolar development as well as increased lingual tipping among mandibular incisors. There was less vertical development of maxillary and mandibular molars in G3. Conclusions: A removable palatal crib provided an improvement in OB (97.5%), followed by the bonded spurs (84.5%). Conversely, the chincup-only group did not have positive OB effects.
