The processes of genesis of coniferous forests are widely investigating using molecular markers. Many studies are based on the analysis of microsatellite loci, which are acknowledged as the most reproducible and polymorphic co-dominant molecular markers. This study is aimed to establish the possibility of using microsatellite markers to determine differentiation between Norway and Siberian spruce (Picea abies and Picea obovata). Another task of this research was to assess the degree of genetic isolation of regional spruce population in the territory of European part of the Russian Federation. This study shows that using a large number of microsatellite markers allows to divide the sample of spruce collected from European part of Russia and Siberia in two genetic clusters, corresponding to Picea abies and Picea obovata. The microsatellite loci analyzed in the study can be used to assess genetic diversity and geographic origin of spruce trees and determine the origin of wood and planting material of Norway and Siberian spruce.

AbstractCamellia oleifera is an important woody plant producing healthy edible oils. People need a large number of molecular markers, especially microsatellite, in breeding of C. oleifera. In this study, we sequenced the root transcriptomes of C. oleifera, and then designed a novel set of microsatellite markers based on the root-expressed genes. We assembled a total of 57,121 unigenes with a length of 42.63 Mb, which harboured 15,902 microsatellites. Among these microsatellites, di-nucleotide repeat motifs were the most abundant group (56.45%), then followed by tri- (25.20%), mono- (12.12%), hexa- (3.21%), penta- (2.18%) and quad-nucleotide ones (0.84%). In total, 6738 primer pairs were designed successfully to amplify the microsatellite loci. To test these microsatellite markers, 48 primer pairs were randomly selected and synthesized and validated in C. oleifera and its eight relatives. Up to 75% of the primer pairs amplified in C. oleifera and its relatives, and 62.5% displayed polymorphism. The transferability and diverse alleles across its eight relatives were detected for each polymorphic primer pair. The novel set of microsatellites derived from the root transcriptomes here provided a useful resource for future molecular genetics improvement of C. oleifera and its relatives.

Picea obovata Ledeb. is a slow-growing in the early stages of growth type of coniferous trees. The use of growth stimulants in the cultivation of spruce seedlings can accelerate the production of standard planting material. The paper presents the results of research of plant growth regulators influence on spruce seedlings within three years of cultivation in forest nursery conditions. Before sowing seeds of spruce soak in solutions of biological-active preparations Verva and Verva-spruce, obtaining from Abies and Picea wood greenery extracts. The plants growth regulator Verva from Abies needles (operating substance – triterpenic acids) is applied in plant growing at cultivation agricultural and commercial crops. Operating substance of a preparation Verva-spruce used for protection of agricultural plants from diseases, are phenolic compounds of Picea wood greenery possessing fungicidal and insecticidal activity. It is established that spruce seedlings processed before sowing by bio-preparation had higher parameters of growth (seedling height, diameter of seedling tree at root neck) in comparison with control plants within three years of cultivation in conditions of forest nursery. Three-year spruce seedlings which have been grown up with use of growth stimulators Verva and Verva-spruce exceeded control on 20–30% on height and on 19–20% on seedling tree thickness.  Bio-preparations Verva and Verva-spruce can be recommended for application in forest nurseries as effective growth stimulators at cultivation of Siberian spruce landing material.

We aimed to investigate inter-year variability in mating system and paternity in an experimental Norway spruce (Picea abies (L.) Karst.) clonal seed orchard that was established with five geographically distinct populations. Using five nuclear microsatellite markers, we analysed 1396 progeny of five mother trees representing each of the populations in three successive mast-years. Selfing varied among mother trees, ranging from 0% to 18.3% with a mean of 13.9% in 1994, 10.1% in 2004, and 7.6% in 2006. Considerable background pollination was observed in each of the three studied mast-years and reached ca. 58%. The paternity assignment conducted showed an uneven male contribution to the seed crop, what was partly related to the number of ramets per paternal clone. All provenances contributed to the seed crop, but with different input, and two provenances dominated. Effective pollen dispersal was significantly determined by distance, with 72.2% of the seed crop resulting from crosses at a distance of up to 15 m. Our study indicated that despite considerable differences in the geographic origin of populations, inter-mating between clones from each population was possible, which generally agrees with the expectations related to the establishment of this seed orchard.

On the basis of author's graphical analysis method, the typification of lake ecosystems transformation scenarios depending on the size of lakes was carried out. It was confirmed that the type of transformation depends not only on size of the lake, but also on the landscape and climatic region in which it is located. The distinctive features of lake ecosystems transformation types in the European part of Russia and in Western Siberia were analyzed and compared.

The results of a sintaxonomical study of plant communities of the Yamozero lake (the North-East of the European part of Russia) are presented. The diversity of the aquatic and helophytic vegetation of the Yamozero lake consists of 16 associations and 2 communities of 6 unions, 4 orders and 2 classes of the floristic classification: Potamogetonetea (7 associations, 2 communities), Phragmito-Magnocaricetea (9 associations). Many of described associations are widely distributed in the Central and the Eastern Europe. Some associations have the boundaries of their ranges. Some communities include 2 rare species of regional level: Isoetes setacea and Sagittaria natans.
