SDS-PAGE technique was used for the study of seed protein polymorphism among three genotypes of Cicer arietinum with different seed coat colour. A total of 24 polypeptide bands were recorded. Out of these 20 were common among all three genotypes and 4 (16.66%) were polymorphic. The data analysis using UPGMA clustering revealed that genotypes with C2 (dark brown) and C3 (black) were closer as compared to genotype with C1 (light brown) coat colour. Jaccard similarity coefficient value ranged from 0.87 to 0.92. The similarity matrix was subjected to UPGMA clustering to generate dendrogram. The most closely revealed genotypes were C2 (dark brown) and C3 (black) with the highest similarity index 0.92 whereas, C1 (light brown) showed minimum similarity index with C3 (black) genotype 0.87. Each of three genotypes of C.arietinum had some polypeptide bands which were peculiar to them only. This enabled distinguishing all three genotypes on the basis of specific polypeptide fragments using SDS-PAGE analysis.

Total seed proteins of 10 Acacia species were examined by SDS–PAGE. The protein patterns showed qualitative and quantitative differences among the taxa analysed. The main protein components of most species examined had MW's in the range of 38.5–49.0 × 103. Subgenus Aculeiferum differed from subg. Acacia in the presence of a high concentration of proteins in the range of 20–24.5 × 103. Hierarchical clustering of the 10 taxa was undertaken, based on Jaccard distances calculated from electrophoretic data. The species grouped in two main clusters, representing the two subgenera of Acacia that occur in America, namely subg. Acacia and subg. Aculeiferum. The taxonomic placement of Acacia emilioana, a species with uncertain sectional affinity within subg. Aculeiferum, is discussed.

The morphological diversity of North American specimens of Arctagrostis latifolia ssp. arundinacea (Trin.) Tzvelev was evaluated for possible varieties within the subspecies by considering (i) descriptions of three previously recognized taxa included in the subspecies, (ii) measurements of spikelet characters of the types of five taxa, (iii) Macoun's illustrations of spikelet diversity, (iv) a morphological study of 130 herbarium specimens, and (v) a transplant garden study. Characters thought to be uninfluenced by environment were examined. These included (i) the seed protein banding patterns revealed by SDS–PAGE, (ii) the guard cell sizes from plants of known chromosome number, and (iii) a study of anther lengths and pollen diameters. The data do not support the recognition of varieties. Key words: SDS–PAGE, transplant garden, pollen, chromosome number, guard cells, Arctagrostis, Poaceae, taxonomy.

This study aims to investigate the pollen and seed characters by light microscopy (LM) and scanning electron microscope (SEM) as related to biochemical data of seed protein and esterase isoenzymes by SDS-PAGE technique in the five Saudi Arabian Ocimum L. species. The detailed description for pollen and seed in addition electrophoretic patterns were conducted by means of numerical analyses based on thirty-nine parameters. The pollen grains were zonocolpate, hexacolpate, prolate to subprolate. Three types of exine ornamentation were recognized, perforate, reticulate and granulate. Also, The periclinal, the anticlinal and the boundaries between cells are described by the aid of SEM which exhibited four main distinct types of nutlets sculpture undulate, quirky, circular and straight. Molecular patterns of protein profiles and esterase (EC.3.1.1.1) showed that esterase could be considered as positive markers than protein, minimum and maximum gene / gene expression of esterase isoenzymes are demonstrated. The relationships between the studied taxa were demonstrated as a phenogram.

A field experiment was performed in order to evaluate the effects of drought stress and application of starter nitrogen fertilizer on seed storage proteins and protein pattern in chickpea cultivars. Experiment was performed in split-factorial using randomized complete block design with three replications. The experiment was laid out in a split-factorial design with drought stress in main plots and cultivar with nitrogen fertilizer in subplots with three replications. The experimental treatments consisted of three levels of drought stress [severe drought stress (S2), moderate drought stress (S1) and no drought stress(S0)] and two cultivars of chickpea consist of Azad and Bivanij and 2 N levels. Plants were either not given any N fertilizer (N0), or fertilized by N fertilizer at the rate of 25 kg ha-1 (N25). The results of this study showed that the effects of drought stress on seed storage proteins and protein yield, effect of cultivars on protein yield were significant. With increase of drought stress seed storage proteins was increased and protein yield decreased. Severe drought stress treatment has the highest seed protein and the control treatment has the lowest seed protein. Non stress treatment has the highest protein yield and the severe drought stress treatment has the lowest protein yield. The SDS- PAGE results revealed that no effects treatments on the protein banding patterns but the related severe drought stress bands were chromatic, because they have highest protein concentration in some protein bands.DOI: http://dx.doi.org/10.3126/ijls.v9i5.12704
