ABSTRACT Contaminated beef and beef products remain a frequent vehicle for the transmission of Escherichia coli O157:H7. The current U.S. Department of Agriculture (USDA) Food Safety and Inspection Service (FSIS) regulatory testing for E. coli O157:H7 uses the method described in the USDA-FSIS Microbiology Laboratory Guidebook (MLG), chapter 5. At times, described presumptive test results are nonconfirmable, suggesting that recent PCR technological advancements and presumed enhanced sensitivity and specificity may offer beneficial changes. Here, we have evaluated the precision and sensitivity of a fluorescence resonance energy transfer–based real-time PCR assay called ECO for the detection of E. coli O157:H7. ECO detects the gene target specific to both E. coli O157:H7 and E. coli O157:non-H7 but distinguishes the two by using a melt curve analysis. A total of 3,113 O157:H7 and O157:non-H7 isolates were used to define this melting temperature–based criteria. The simulated comparative performance evaluation in the spiked beef samples indicated detection of 3 of 3 samples by ECO at <3.3 log CFU/mL, whereas MLG only detected 1 of 3 (<3.3 log CFU/mL). Using modified tryptic soy broth–enriched natural beef and veal product samples (n = 452), the comparative sensitivity, specificity, false-positive rate, and false-negative rate against culture between MLG and ECO were 75 versus 92%, 91 versus 99%, 8.9 versus 0.77%, and 25 versus 8.3%, respectively. Positive predictive value, negative predictive value, and the overall accuracy were found to be 56 versus 94%, 96 versus 98%, and 88 versus 98%, for MLG and ECO, respectively. These data demonstrate that the ECO assay is comparable to MLG detection of E. coli O157:H7 and offers improved sensitivity.

Abstract Proper treatment of wastewater is key to the achievement of sustainable environmental management. The use of ultraviolet radiation and ultrasound have continued to be considered as some of the best sustainable practices in wastewater purification. However, despite the suitability of the two emerging techniques in sustainably increasing the purification efficiencies of wastewater, their application has not been fully understood, especially in eliminating faecal pathogenic microorganisms. Moreover, their combined potential in the elimination of Escherichia coli resistant genes from wastewater has not been adequately explored. This study was designed to evaluate the potential of individual and combined/integrated ultraviolet radiation and ultrasonic technologies in the removal of antibiotic-resistant E. coli from domestic effluents. There was a statistical difference in the mean log units of sulfonamide resistant E. coli between the different ultraviolet radiation and ultrasonic dosages (P < 0.05), showing that ultraviolet radiation technology was more effective in the removal of both sulfonamide and tetracycline resistant E. coli from the wastewater. However, the integrated ultraviolet radiation-ultrasonic technique was highly efficient and is recommended in the removal of antibiotic resistant E. coli from wastewater. Nonetheless, further studies also need to be performed to further evaluate the disinfection effectiveness on a different bacteria species under continuous operation.

No estado de Minas Gerais, as campanhas de coleta e análise da qualidade da água nos corpos hídricos contemplam até 51 variáveis, o que dificulta a análise e interpretação desse conjunto de dados e a identificação das variáveis determinantes para a qualidade da água. Diante disso, o objetivo deste trabalho foi identificar as principais fontes de poluição, bem como o comportamento da qualidade da água ao longo do tempo de monitoramento, por meio de diferentes análises estatísticas na circunscrição hidrográfica do rio Piranga. Pelos resultados obtidos, a análise fatorial/análise de componentes principais apontou a alta susceptibilidade que a bacia apresenta à erosão do solo, a contaminação pelo lançamento de efluentes domésticos e a variabilidade da qualidade das águas em virtude dos metais pesados. As variáveis Escherichia coli, ferro dissolvido, fósforo total e manganês total apresentaram os valores de violação da classe de enquadramento mais críticos. A análise de tendência mostrou padrões diferentes para o índice de qualidade da água e para as variáveis mais relevantes para a qualidade da água. Dentre as variáveis que compõe o índice, destacam-se as tendências de aumento de nitrato em todas as estações analisadas. De maneira geral, os resultados mostraram que a qualidade da água na área de estudo varia em função da erosão do solo, do alto grau de contaminação por efluentes domésticos, da poluição difusa advinda das áreas agrícolas e dos metais pesados, sendo as variáveis de qualidade da água vinculadas a estes fatores as mais importantes. Surface water quality assessment in the hydrographic region of the Piranga River using multivariate and non-parametric statistical analysis ABSTRACTIn the state of Minas Gerais, campaigns to collect and analyze water quality in water bodies include up to 51 variables, which makes it difficult to analyze and interpret this data set and to identify the determining variables for water quality. Therefore, the objective of this work was to identify the main sources of pollution, as well as the behavior of water quality over the monitoring time, through different statistical analyzes in the hydrographic region of the Piranga River. Based on the results obtained, the factor analysis/principal component analysis out the high susceptibility that the hydrographic region presents to soil erosion, contamination by the release of domestic effluents and the variability of water quality due to heavy metals. The variables Escherichia coli, dissolved iron, total phosphorus and total manganese presented the most critical values of violation of the framework class. The trend analysis showed different patterns for the water quality index and for the most relevant variables for water quality. Among the variables that make up the index, the trends of nitrate increase in all analyzed stations stand out. In general, the results showed that the water quality in the unit varies depending on soil erosion, the high degree of contamination by domestic effluents, the diffuse pollution from agricultural areas and heavy metals, with water quality variables being linked to these factors the most important.Keywords: environmental analysis, Minas Gerais, water pollution, water resources.

Smoked fish is traditionally processed by fish through an open process of hot smoked. Smoked fish process in Indonesia, especially in Maluku, is still carried out traditionally with capital and small business scale so that the use of tools is still simple, besides the sanitation and hygiene of handling and processing are still low. The raw material used in this study is smoke tuna loin produced by Dusun Air Manis,Laha village. This study aims to determine the microbiological and chemical quality of the smoked fish that was produced from Dusun Air Manis, Laha village. The test parameters carried out consisted of microbiological tests: 1) Total microbes on frozen tuna loin, washing water, immersion water, bamboo, smoked racks and smoked tuna, 2) Escherichia coli test on frozen tuna loin, washing water, soaking water, smoked tuna, 3) Salmonella test on smoked tuna. Chemical analysis of smoked tuna included moisture and pH. The results showed that the total plate count (TPC), Escherichia coli and Salmonella on tuna loin smoke still met microbiological standards according to SNI 2725: 2013. Moisture of smoked tuna was 59% and still meets the quality standard according to SNI 2725: 2013 which is a maximum of 60%. The pH of smoked tuna was 5.8.
