Targeted Nanostructured Lipid Carriers for Delivery of Paclitaxel to Cancer Cells: Preparation, Characterization, and Cell Toxicity

Aim: To sensitize cisplatin (Cis)-resistant ovarian cancer cells toward Cis using Cis-loaded nanostructured lipid carriers (CisNLCs). Materials & methods: CisNLCs were synthesized and characterized using dynamic light scattering, Fourier transform IR and x-ray diffraction (XRD). Sensitivity of PA-1 and CaOV3 cells to Cis and its biotoxicity were assessed. Further, expression of the Cis-resistance markers GSTPi and  ATP7B, and apoptotic markers Bax, Bcl2 and Cas9 were quantified by real-time PCR. Results: The size of synthesized CisNLCs was approximately 179.3 ± 2.32 nm and surface charge was -33.9 ± 1.47 mV. IC50 was 210 μg/ml in PA-1 and 500 μg/ml in CaOV3. CisNLCs modulated reactive oxygen species levels in CaOV3 cells. Reduced GSTPi and decreased Cis efflux via ATP7B sequestration caused Cis to accumulate in cytoplasm, thereby augmenting apoptosis in cells. Conclusion: CisNLCs sensitize CaOV3 by redox resetting, indicating their immense therapeutic potential.

To evaluate drug and metabolite efficacy on a target organ, it is essential to include metabolic function of hepatocytes, and to evaluate metabolite influence on both hepatocytes and the target of interest. Herein, we have developed a two-chamber microfabricated device separated by a membrane enabling communication between hepatocytes and cancer cells. The microscale environment created enables cell co-culture in a low media-to-cell ratio leading to higher metabolite formation and rapid accumulation, which is lost in traditional plate cultures or other interconnected models due to higher culture volumes. We demonstrate the efficacy of this system by metabolism of tegafur by hepatocytes resulting in cancer cell toxicity.

Drug delivery-based nanoparticle has been developing as a widespread innovation in cancer treatment protocols. Here, we investigated the role of Kaempferol (KAE) loaded in nanostructured lipid carriers (NLCs) to promote cytotoxicity, efficacy, and paclitaxel-dependent apoptosis in MDA-MB 468 breast cancer cells. Particle size distribution, scanning electron microscopy (SEM), zeta potential, and cellular uptake were harnessed to optimize and characterize of KAE -loaded NLCs. MTT assay was used to measure the cellular proliferation of cancer cells. The clarification of early and late apoptosis and their gene expression patterns was assessed by Annexin V/PI staining and real-time PCR, respectively. SEM images offered us a nasty particle size of 80 ± 3 nm to the Kaempferol formulated into NLCs. The IC50 values for KAE and paclitaxel determined 44 ± 0.52 μM and 1.75 ± 0.36 nM, respectively. The moderated cell proliferation from 56 ± 26.8% to 44 ± 3.9% (p < 0.05) was demonstrated by KAE loaded NLCs. Co-administration of KAE-loaded nanoparticles and paclitaxel into cancer cells significantly strengthens the percentage of apoptosis (p < 0.05). Our results recommend that KAE incorporated into NLCs as an anti-cancer adjuvant is a powerful technique that may be a useful delivery system to enhance chemotherapy agents' effect on breast cancer cells.
