Chemical Analysis, Phenolic Content, and Antioxidant Activities of Aqueous and Organic Moroccan Juglans regia L. Bark Extracts

Background: The present study was conducted on the Moroccan Juglans regia L. bark, in comparison with other Juglans regia collected in other countries, to study the effects of the use of different extraction solvents (water, ethanol, acetone, and diethyl ether), aqueous and organic extraction method, hot and cold extraction by maceration modalities on the presence and content of the different phytochemicals, and on the antioxidant activity. Methods: Various solvents of increasing polarity were used for the preparation of aqueous and organic extracts. Afterwards, a phytochemical screening, an estimation of phenolic compounds, flavonoids, and tannins contents was conducted. The in vitro antioxidant activity was carried out by five tests: H2O2, ABTS, FRAP, DPPH, and reducing power. Subsequently, Principal Component Analysis (PCA) was used to establish the correlation between phenol content and antioxidant activity. Results: Phytochemical analysis showed that the 9 extracts prepared are characterized by the presence of flavonoids, catechic tannins, saponins, anthraquinones, and quinones, and that the macerated acetone extract contains the highest content of phenolic compounds (327.972±0.06 μgEAG/mgE), flavonoids (1267.981±2.911 μgEQ/mgE), and tannins (38.056±1.886 μgEC/gE). The best antioxidant activity was obtained with the macerated acetone which was found to be the most active via the five different tests, with a percentage of scavenging of H2O2 equal at 24.13±1.81, a higher value of ABTS equal to 602.29±0.34 μgET/mgE. PCA allowed us to deduce that phenolic compounds, flavonoids, and tannins are strongly correlated with antioxidant activity. Conclusion: Moroccan Juglans regia may be one of the potential sources of antioxidant compounds.

Objective: The aim of this study was focussed on phytochemical analysis, in vitro antioxidant activities of different solvent extracts of Hypoestes phyllostachya leaf. Methods: The leaves were washed efficiently, shade dried and coarsely powdered. The powdered leaf material of Hypoestes phyllostachya was successively extracted with hexane, chloroform, methanol and water using Soxhlet apparatus. Preliminary phytochemical screenings for polyphenolics, proteins, phytosteroids, flavonoids, tannins, glycosides, alkaloids carbohydrates, polyphenolics, saponins were done by following standard procedure. In vitro antioxidant activities of various solvent extracts were assessed using DPPH, ABTS, Hydroxy radical, total antioxidant capacity and reducing power assay. Polyphenolics and flavonoids were also estimated. Results: The results showed that most of the phytochemical compounds such as carbohydrates, proteins, steroids, fixed oils and fatty acids, flavonoids, alkaloids, saponins, tannins, polyphenolics and glycosides present in the methanolic and aqueous extract of Hypoestes phyllostachya. The in vitro antioxidant activity of Hypoestes phyllostachya leaf clearly showed that it has better antioxidant activity, which was confirmed by different assays. Conclusion: From this work, it can be concluded that Hypoestes phyllostachya has the potential to be a good antioxidant but have only a lesser amount of flavonoids and polyphenolic contents. This was the first report on the pharmacological activity of the Hypoestes phyllostachya.

Objective: The current investigation was contemplated to evaluate the phytochemicals and in vitro antioxidant activities in peel and pulp of some commercially grown citrus fruits of South India, namely, lemon (Citrus aurantifolia), orange (Citrus reticulata), sour orange (Citrus aurantium), pomello (Citrus grandis), and citron (Citrus medica).Methods: The peel and pulp of the fruits were separated and subjected to cold extraction using 70% alcohol. The extracts obtained were screened for the presence of their phytoconstituents using various qualitative and further quantified for major constituents. Further, the in vitro antioxidant activity was assayed by different radical scavenging methods, namely, 2,2-diphenyl-1-picrylhydrazyl, superoxide anion, nitric oxide, lipid peroxidation inhibition, iron chelating activity, and reducing power assay at different concentrations.Results: All the citrus fruits have shown significant in vitro antioxidant activity for the parameters assessed, wherein peel extracts recorded superior antioxidant potential than their corresponding pulps. The broad range of activity of the extracts suggests that multiple mechanisms mediated by the phytoconstituents are responsible for the antioxidant activity.Conclusion: The study thus revealed that peel and pulp of citrus fruits are potential sources of bioactive compounds which are reflected in antioxidant activity and supports their health-promoting claims of plethora of investigations.

Amomum subulatum, commonly known as large or black cardamom, is a commonly used spice in Indian kitchens and is traditionally used to treat various ailments. To add more knowledge about the medicinal values of the fruit, this study was conducted to evaluate the in vitro antioxidant activities of aqueous, methanol, ethanol, hydro alcohol, ethyl acetate, acetone and chloroform extracts of the fruit. Preliminary assessment was done to detect the presence of phytoconstituents using identification tests. The antioxidant activity was measured by employing methods such as diphenylpicrylhydrazyl (DPPH) radical scavenging assay, total antioxidant activity equivalent to ascorbic acid, reducing power assay and superoxide anion scavenging assay. The antioxidant activities were compared with their respective phenol and flavonoid contents. Preliminary assessment revealed that large cardamom fruit is a good source of all the bioactive constituents as well as phenol and flavonoid essential for medicinal values. The extract obtained by polar solvents showed the highest antioxidant efficacy in relation to its phenol content. Also, all the solvent-soluble fractions showed a concentration-dependent antioxidant effect. Results from our study prove that large cardamom can alleviate oxidative stress, suggesting the potential of large cardamom as a functional food

Ten indeterminate tomato cultivars differing in fruit shape and typology, grown simultaneously in a greenhouse from Oltenia (Southwestern Romania) and harvested at red ripe stage, were investigated for the contents in ascorbic acid, lycopene, β-carotene, total phenolics, minerals (K, Na, Ca and Mg) and trace elements (Fe, Cu, Mn, Cr, Zn and B). In addition, their antioxidant activities were determined. Significant differences were found among tomato cultivars in all studied antioxidant compounds, as well as in the antioxidant activity. Ascorbic acid ranged from 91.9 to 329.7 mg kg-1 fw, lycopene ranged from 19.7 to 49.0 mg kg-1 fw, while total phenolic compounds varied between 300.2 and 557.8 mg kg-1 fw. Antioxidant activity ranged from 0.81 mmol Trolox kg-1 fw to 1.74 mmol Trolox kg-1 fw and it was significantly correlated to total phenolics content (r = 0.91; p< 0.05). The cocktail type cultivar ‘Tiger’ and the cherry type cultivar ‘Belle’ proved to be the most powerful in antioxidant activity and phenolic compounds while the rectangular plum shaped cultivar ‘Porto’ recorded the highest average lycopene content. A valuable cultivar proved to be the brownish red ‘Sacher’ which registered among the highest contents of phenolics, lycopene and ascorbic acid. Mineral and trace elements contents were also significantly affected by cultivars. Values recorded for K, Ca and Mg ranged from 2139.6 to 3056.9 mg kg-1, 137.7 to 325.8 mg kg-1 and 27.3 to 168.7 mg kg-1 respectively.

Dibenz[b,f]azepine and its five derivatives bearing different functional groups were synthesized by known methods. The compounds thus synthesized were evaluated for antioxidant potential through different in vitro models such as (DPPH) free radical scavenging activity,ß-carotene-linoleic acid model system, reducing power assay and phosphomolybdenum method. Under our experimental condition among the synthesized compounds dibenz[b,f]azepine (a) and 10-methoxy-5H-dibenz[b,f]azepine (d) exhibited potent antioxidant activity in concentration dependent manner in all the above four methods. Butylated hydroxyl anisole (BHA) and ascorbic acid (AA) were used as the reference antioxidant compounds. The most active compounds like dibenz[b,f]azepine and its methoxy group substituent have shown more promising antioxidant and radical scavengers compared to the standards like BHA and ascorbic acid. It is conceivable from the studies that the tricyclic amines,i.e. dibenz[b, f]azepine and some of its derivatives are effective in their antioxidant activity properties.

The aim of this work is to deepen the pharmacological effect of 4-phenyl-1,5-benzodiazepin-2-one derivatives which have a similar structure to nonionic surfactants: 4-phenyl-1,5-benzodiazepin-2-one is the hydrophilic head, and the carbon chain is hydrophobic tail. The antinociceptive activity of 4-phenyl-1,5-benzodiazepin-2-one derivatives was determined using acetic acid-induced writhing and tail immersion tests. In addition, the in vitro antioxidant activities of the tested derivatives were determined by using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging method and ferric reducing power assay. A single oral administration of these compounds at the doses of 50 and 100 mg/kg significantly reduced the number of abdominal writhes induced by acetic acid injection. Acute pretreatment with 4-phenyl-1,5-benzodiazepin-2-one derivatives at the dose of 100 mg/kg caused a significant increase in the tail withdrawal latency in the tail immersion test. Additionally, a significant scavenging activity in DPPH and reducing power was observed in testing antioxidant assays. Finally, we carried out a study of the antioxidant activity of these derivatives. The results of this study reveal that these compounds have a low antioxidant activity compared to the BHT. It decreases with the polarity of the molecule. The present study suggests that 4-phenyl-1,5-benzodiazepin-2-one derivatives possess potent antinociceptive and antioxidant effects, which suggest that the tested compounds may be useful in the treatment of pain and oxidation disorders.

Selected Australian native fruits such as Davidson’s plum, finger lime and native pepperberry have been reported to demonstrate potent antioxidant activity. However, comprehensive metabolite profiling of these fruits is limited, therefore the compounds responsible are unknown, and further, the compounds of nutritional value in these native fruits are yet to be described. In this study, untargeted and targeted metabolomics were conducted using the three fruits, together with assays to determine their antioxidant activities. The results demonstrate that targeted free and hydrolysed protein amino acids exhibited high amounts of essential amino acids. Similarly, important minerals like potassium were detected in the fruit samples. In antioxidant activity, Davidson’s plum reported the highest activity in ferric reducing power (FRAP), finger lime in antioxidant capacity (ABTS), and native pepperberry in free radical scavenging (DPPH) and phosphomolybdenum assay. The compounds responsible for the antioxidant activity were tentatively identified using untargeted GC×GC-TOFMS and UHPLC-QqQ-TOF-MS/MS metabolomics. A clear discrimination into three clusters of fruits was observed using principal component analysis (PCA) and partial least squares (PLS) analysis. The correlation study identified a number of compounds that provide the antioxidant activities. GC×GC-TOFMS detected potent aroma compounds of limonene, furfural, and 1-R-α-pinene. Based on the untargeted and targeted metabolomics, and antioxidant assays, the nutritional potential of these Australian bush fruits is considerable and supports these indigenous fruits in the nutraceutical industry as well as functional ingredients for the food industry, with such outcomes benefiting Indigenous Australian communities.

Antioxidant agents against reactive oxygen species can be used for several cosmetic and medicinal applications. This study’s objective was to evaluate the antioxidant activities of Polytrichastrum alpinum (Hedw.) G. L. Sm. (Polytrichaceae), an Antarctic moss species collected from King George Island (Antarctica). The identifi cation of the moss species was performed on the basis of morphological characteristics and molecular sequencing of the 18S rRNA gene. Two benzonaphthoxanthenones: ohioensins F and G, were isolated from the extract after several chromatographic procedures. The various in vitro antioxidant capacities of a methanolic extract of P. alpinum and the isolated compounds were evaluated by analyzing the scavenging capacities of free radicals of 2,2-azino-bis(3-ethylbenzthiazoline- 6-sulfonic acid) (ABTS) and 2,2-diphenyl-1-picrylhydrazyl (DPPH), the total phenol assay with Folin-Ciocalteu reagent, the ferric ion (Fe3+) reducing power and the nitric oxide (NO) scavenging activity and compared to those of commercial standards for each assay. The experimental data showed that even the crude extract of P. alpinum exhibited potent antioxidant activity. The antioxidant activity was increased two- to seven-fold for the purified compounds. The antioxidant activities of both purified compounds were found to be more or less the same in all experiments. However, the obtained data showed that the Fe3+ reducing power of the purified compounds and crude methanolic extract was almost the same suggesting the presence of other stronger reducing agents in the methanolic extract which could not be isolated in the present experiment. Therefore, further work on the isolation of these stronger antioxidant agents from this moss specimen of the extreme environment is warranted. Developments of laboratory mass culture techniques are anticipated to achieve bulk production of the active constituents for commercial application.

Abstract The objective of this work was to investigate the variability of physicochemical parameters, phenolic compounds and in vitro antioxidant activity of propolis collected from different apiaries in southeastern Mexico. A high variability was found in the moisture content (1.96–8.26%), ash (0.66–5.50%) and sensory characteristics of raw propolis from southeastern Mexico, but the raw propolis samples met the requirements of the quality regulations. In the same way, most of the ethanolic extracts also complied with the quality regulations. Of all the extracts, PE2 obtained from Santa Cruz showed the highest values for dry extract, content of total phenolic compounds (TPC), total flavonoids (TF) and antioxidant activity (DPPH and ABTS). The content of the individual phenolic compounds varied according to the geographical location of the apiary, but the PE2 extract resulted in the highest pinocembrin and chrysin content. A positive correlation was obtained between TPC and TF with antioxidant activity. Propolis extracts were classified into two groups through principal component analysis (PCA). These results indicate that the apiary location in southeastern Mexico influenced the characteristics of propolis.

Ayurveda contains a variety of medicines including the polyherbal fermented traditional medicines namely Arishta and Asava. It remains as a rich source of bioactive phytochemicals including antioxidants. There are scanty publications on the antioxidant activity of these medicines. There is a need to explore the natural antioxidants to replace synthetic counterparts which requires an assessment in terms of antioxidant activity by in vitro analysis. This study employs a range of antioxidant assay systems containing various free radicals (DPPH, hydroxyl, hydrogen peroxide, super oxide, nitric oxide and ABTS), Inhibition of free radical generation (metal chelating) and H+ – donating ability (reducing power) were performed. Ashokarishta, Aswagandharishta and Dasamoolarishta exhibited higher degree of antioxidant activities but less than the corresponding standards. Based on IC50 values, the hydrogen peroxide scavenging ability of Ashokarishta is comparable to the standard L-ascorbic acid. In ABTS assay, Aswagandharishta and Dasamoolarishta are comparable to the standard Butylated hydroxytoluene (BHT). These may be due to the phytochemical heterogeneity of the samples assayed. Thus study can lead to the sourcing of Ayuvedic polyherbal fermented medicines as a novel source of natural antioxidant molecules that might be acting as free radical quencher or scavenger and to treat human ailments.
